Citrus yellow vein clearing virus(CYVCV)is a new citrus virus that has become an important factor restricting the development of China’s citrus industry,and the CYVCV coat protein(CP)is associated with viral pathogenicity.In this study,the Eureka lemon zinc finger protein(ZFP)ClDOF3.4 was shown to interact with CYVCV CP in vivo and in vitro.Transient expression of ClDOF3.4 in Eureka lemon induced the expression of salicylic acid(SA)-related and hypersensitive response marker genes,and triggered a reactive oxygen species burst,ion leakage necrosis,and the accumulation of free SA.Furthermore,the CYVCV titer in ClDOF3.4 transgenic Eureka lemon plants was approximately 69.4%that in control plants 6 mon after inoculation,with only mild leaf chlorotic spots observed in those transgenic plants.Taken together,the results indicate that ClDOF3.4 not only interacts with CP but also induces an immune response in Eureka lemon by inducing the SA pathways.This is the first report that ZFP is involved in the immune response of a citrus viral disease,which provides a basis for further study of the molecular mechanism of CYVCV infection. 展开更多

Yellow mealworm larvae(YML;Tenebrio molitor) are considered as a valuable insect species for animal feed due to their high nutritional values and ability to grow under different substrates and rearing conditions. Advances in the understanding of entomophagy and animal nutrition over the past decades have propelled research areas toward testing multiple aspects of YML to exploit them better as animal feed sources. This review aims to summarize various approaches that could be exploited to maximize the nutritional values of YML as an animal feed ingredient. In addition, YML has the potential to be used as an antimicrobial or bioactive agent to improve animal health and immune function in production animals. The dynamics of the nutritional profile of YML can be influenced by multiple factors and should be taken into account when attempting to optimize the nutrient contents of YML as an animal feed ingredient. Specifically, the use of novel land-based and aquatic feeding resources, probiotics, and the exploitation of larval gut microbiomes as novel strategies can assist to maximize the nutritional potential of YML. Selection of relevant feed supplies, optimization of ambient conditions, the introduction of novel genetic selection procedures, and implementation of effective post-harvest processing may be required in the future to commercialize mealworm production. Furthermore, the use of appropriate agricultural practices and technological improvements within the mealworm production sector should be aimed at achieving both economic and environmental sustainability. The issues highlighted in this review could pave the way for future approaches to improve the nutritional value of YML. 展开更多

Background:The experiment evaluated the effect of nutrition levels and sex on the growth performance,carcass characteristics and meat quality of F1 Angus × Chinese Xiangxi yellow cattle.Methods:During the background period of 184 d,23 steers and 24 heifers were fed the same ration,then put into a2×2×2 factorial arrangement under two levels of- dietary energy(TON:70/80%DM),protein(CP:11.9/14.3%DM)and sex(S:male/female) during the finishing phase of 146 d.The treatments were-(1) high energy/low protein(HELP),(2) high energy/high protein(HEHP),(3) low energy/low protein(LELP) and(4) low energy/high protein(LEHP).Each treatment used 6 steers and 6 heifers,except for HELP- 5 steers and 6 heifers.Results:Growth rate and final carcass weight were unaffected by dietary energy and protein levels or by sex.Compared with the LE diet group,the HE group had significantly lower dry matter intake(DMI,6.76 vs.7.48 kg DM/d),greater chest girth increments(46.1 vs.36.8 cm),higher carcass fat(19.9 vs.16.3%) and intramuscular fat content(29.9 vs.22.8%DM).The HE group also had improved yields of top and medium top grade commercial meat cuts(39.9 vs.36.5%).The dressing percentage was higher for the HP group than the LP group(53.4 vs.54.9%).Steers had a greater length increment(9.0 vs.8.3 cm),but lower carcass fat content(16.8 vs.19.4%) than heifers.The meat quality traits(shear force value,drip loss,cooking loss and water holding capacity) were not affected by treatments or sex,averaging 3.14 kg,2.5,31.5 and 52.9%,respectively.The nutritive profiles(both fatty and amino acid composition) were not influenced by the energy or protein levels or by sex.Conclusions:The dietary energy and protein levels and sex significantly influenced the carcass characteristics and chemical composition of meat but not thegrowth performance,meat quality traits and nutritive profiles. 展开更多

To investigate the effect of different dietary energy and protein levels on meat performance and meat quality of Jinghai yellow chickens, 480 43-day old Jinghai yellow chickens with similar weight were randomly divided into four experimental groups: experimental group 1 (protein 15%, metabolic energy 9.95 MJ/kg), experimental group 2 (protein 16%, metabolic energy 10.95 MJ/kg), experimental group 3 (protein 17%, metabolic energy 12.65 MJ/kg) and experimental group 4 ( protein 18%, metabolic energy 13.95 MJ/kg), respectively. All chickens were slaughtered at 112-day old. The breast and leg muscles of Jinghai yellow chickens were collected, to determine the slaughter performance, conventional meat quality and muscle chemical indicators. The results indicated that dressing-out percentage and eviscerated yield percentage in four experimental groups were above 87.27% and 67.00%, respectively; other slaughter performance indicators exhibited no significant differences among various groups (P 〉 0.05 ) ; breast muscle color of hens in experimental group 4 varied significantly from that in other three groups ( P 〈 0.05 ) ; leg muscle color of hens in experimental group 2 varied extremely significantly from that in other three groups ( P 〈 0.01 ) ; water-holding capacity of breast muscles of hens in experimental group 3 was significantly higher than that in experimental group 4 (P 〈 0.05 ) ; thiamine content of breast muscles of cocks in experimental group 3 was significandy higher than that in experimental group 2 ( P 〈 0.05 ) ; however, other properties exhibited no significant differenees among various groups (P 〉 0.05 ). 展开更多

We investigated the effects of fish protein hydrolysate (FPH) on growth performance and humoral immune response of the large yellow croaker (Pseudosciaena crocea R.). One thousand and two hundred large yellow croakers [initial average weight: (162.75±23.85) g] were divided into four groups and reared in floating sea cages (3 m×3 m×3 m). The animals were fed with 4 diets: basal diet only (control) or diets supplemented with 5%, 10% and 15% (w/w) FPH. The results show that dietary FPH levels significantly influenced the growth and immunity of the large yellow croaker. Compared with the control group, total weight gain (TWG) in all treatment groups, relative weight gain (RWG) and specific growth rate (SGR) in fish fed with diets supplemented with 10% and 15% FPH were significantly increased (P<0.05). Similar results were observed in immune parameters [lysozyme activity, serum complements, immunoglobulin M (IgM)]. Lysozyme activity, complement C4 and IgM were also significantly increased (P<0.05) in fish fed with diets supplemented with 10% and 15% FPH, while complement C3 level was significantly increased (P<0.05) in all treatment groups. In general, with the supplementation of FPH, particularly at dose of 10%, the growth performance and immunity of the large yellow croaker can be improved effectively. 展开更多

Distillers dried grains with solubles (DDGS) in combination with soy protein concentrate (SPC) with and without an essential amino acid (EAA) complex were assessed as protein alternatives in juvenile Yellow Perch Perca flavescens diets. Diets contained 5% FM, 40% SPC, and 20% or 40% DDGS each with or without EAA. No mortalities or health assessment differences were observed during the trial and all fish readily accepted the experimental diets. Diets supplemented with EAA produced greater weight gain, improved feed conversion, and apparent protein digestibility. Performance was consistently improved for fish fed diets containing amino acid supplements. Based on these results, Yellow Perch are able to utilize high levels of the plant proteins, accompanied with EAAs, as a FM replacer. 展开更多

[Objective]The paper was to investigate the effects of crude protein level of concentrate supplement on production performance and serum biochemical parameters of post-fattening Hainan Yellow Cattle.[Method]Twelve Hainan Yellow Cattles with the same genetic backgrounds and similar initial weight of(168.13±0.55)kg were randomly divided into three treatments.Cattle were fed with concentrate supplement containing different levels of crude protein.The trial lasted 60 d.[Result]The daily gain of 17.56%crude protein group was significantly higher than those of 15.55%and 13.56%crude protein groups(P<0.05),and the gross profit of 17.56%crude protein group was markedly higher than that of 13.56%crude protein group(P<0.05),but the feed gain ratio of 17.56%crude protein group was significantly lower than that of 13.56%crude protein group(P<0.05).Compared with 15.55%and 13.56%crude protein groups,the serum total protein content markedly increased(P<0.05),but the urea nitrogen level significantly decreased(P<0.05).[Conclusion]When crude protein level of concentrate supplement was 17.56%,post-fattening Hainan Yellow Cattle could obtain better production performance. 展开更多

The gene encoding for inf-y was amplified usingPCR technique. After the PCR product was retrieved and purified, it was digested withEcoR V and Not Ⅰ restriction endonuclease, and then cloned into the plasmidpIRES-EYFP. The recombinant plasmid plRES-EYFPIFN-γwas identified by restrictionendonuclease enzyme analysis and DNA sequence analysis.Results: The ifn-γ was successfully amplified and verified by partial DNA sequenceanalysis. 试验旨在研究低蛋白日粮对黄羽肉鸡生长性能、胴体品质及肉质的影响。试验选取1日龄慢速型母鸡360羽,设对照组、试验组,每组6个重复,每个重复30只鸡。试验组1~6周(w)、7~13 w、14~19 w阶段日粮粗蛋白浓度,分别设定为对照组的90%、95%、1... 试验旨在研究低蛋白日粮对黄羽肉鸡生长性能、胴体品质及肉质的影响。试验选取1日龄慢速型母鸡360羽,设对照组、试验组,每组6个重复,每个重复30只鸡。试验组1~6周(w)、7~13 w、14~19 w阶段日粮粗蛋白浓度,分别设定为对照组的90%、95%、100%,配方中增加玉米副产物比例、降低豆粕与大豆油用量。结果显示,相比于对照组,试验组鸡各阶段体重明显下降(P<0.05),1~6 w增重明显降低(P<0.01)、料重比增高(P<0.01),全程料重比无显著变化(P>0.05);鸡腹脂率显著下降(P<0.05),胸肌a值、肉色L值优于对照组(P<0.05);单位增重所需日粮粗蛋白下降1.7%,饲料成本下降7.1%。综上所述,在本研究条件下,下调生长前、中期日粮的粗蛋白浓度会明显降低黄羽肉鸡全期生长速度,但能节约增重饲料成本,对胴体品质、肉质有一定的改善作用。 展开更多

With artificially farmed and wild Henan Yellow River carps as the research objects, comparative study on muscle fiber diameter and density, routine nutritional composition, calcium and phosphorus contents and amino acids composition was conducted between the two groups of carps. The results showed that the moisture content was significantly higher (P 〈0.05), the crude fat and crude protein contents were significantly lower (P〈0.05), the essential amino acids and total amino acids contents were lower (P〉0.05), the phosphorus content was higher (P 〉0.05), and the calcium content was lower (P〉0.05) in the wild group compared with those in the farming group. The analysis of muscle fiber characteristics showed that there were significant differences in the average muscle fiber diameter (P〈0.05) and muscle fiber density (P〈0.01) between the two groups of Henan Yellow River carps. 展开更多

瓜类褪绿黄化病毒(cucurbit chlorotic yellows virus,CCYV)是近些年来出现的一种烟粉虱Bemisia tabaci传播的植物病毒,在瓜类生产中造成了严重损失。为了解该病毒外壳蛋白(coat protein,CP)在病毒侵染寄主过程中的亚细胞定位和致病作用... 瓜类褪绿黄化病毒(cucurbit chlorotic yellows virus,CCYV)是近些年来出现的一种烟粉虱Bemisia tabaci传播的植物病毒,在瓜类生产中造成了严重损失。为了解该病毒外壳蛋白(coat protein,CP)在病毒侵染寄主过程中的亚细胞定位和致病作用,本研究以CCYV山东黄瓜分离物为研究对象,构建了荧光表达载体CP-YFP和异源表达载体pGR-CP。浸润荧光表达载体48 h后的本氏烟Nicotiana benthamiana叶片在激光共聚焦显微镜下可以观察到荧光信号在细胞质和细胞核内均有分布;浸润异源表达载体的本氏烟植株7 d后上部叶片开始显现花叶症状,13 d后顶部新叶出现严重皱缩和坏死斑点。以上结果表明CCYV CP蛋白参与了病毒对寄主的侵染过程,可能是症状形成的致病相关因子。本研究为进一步解析该病毒的致病机理提供了初步的理论支持。 展开更多

红曲黄色素是常见的天然食用色素且具有多种生理功效,但其水溶性较差,且对光、热、酸碱敏感,限制了其在食品领域的广泛应用。以亚麻籽水溶蛋白(water-soluble flaxseed protein,FPW)为载体制备其与红曲黄色素的复合物,利用组合光谱分析... 红曲黄色素是常见的天然食用色素且具有多种生理功效,但其水溶性较差,且对光、热、酸碱敏感,限制了其在食品领域的广泛应用。以亚麻籽水溶蛋白(water-soluble flaxseed protein,FPW)为载体制备其与红曲黄色素的复合物,利用组合光谱分析技术研究两种红曲黄色素(monascin,MS和ankaflavin,AK)与FPW之间的相互作用行为,考察FPW同富含红曲黄色素的红曲色素(Monascus pigments,Mps)复合后,色素在水溶液中的分散性与稳定性变化情况。荧光光谱分析发现,MS和AK均以静态猝灭方式与FPW结合,且均只存在一个结合位点,FP W-AK体系的结合常数大于FP W-MS体系;热力学参数分析表明,维系FP W-MS体系的主要作用力为氢键和范德华力,维系FP W-AK体系的主要作用力为氢键和静电作用。同步荧光光谱、三维荧光光谱分析表明,MS或AK的加入使FPW的氨基酸残基微环境发生改变,从而引起FPW的构象改变。紫外-可见光谱和红外光谱研究结果进一步证明,MS和AK的添加使FPW的二级结构发生改变。此外,与FPW复合后,Mps在水中的分散性提高,且与不同浓度的FPW形成复合物后,在温度为25℃,光照强度为3900 Lux,光照时间为72 h时,与游离的Mps相比,FP W-Mps的保留率分别提高了9.53%、16.92%、31.37%;在温度为4、25、50℃的避光条件下,FP W-Mps较游离的Mps的保留率分别提高了25.65%、22.04%、25.98%;在温度为25℃,pH值为3、5、7、9、11,避光条件下,FP W-Mps较游离的Mps的保留率分别提高了10.27%、12.96%、12.06%、17.82%、7.27%。与FPW复合后,在不同光照时间、温度和pH值条件下,Mps的保留率均增加,表明FPW可以提高Mps的光稳定性、热稳定性和酸碱稳定性。研究旨在为红曲黄色素稳定性研究提供新的思路。 展开更多

红花黄色素对糖尿病小鼠足溃疡创面愈合的影响及机制

目的探讨红花黄色素(SY)对糖尿病小鼠足溃疡(DFU)创面愈合的影响及分子机制。方法取45只C57BL/6小鼠腹腔注射链脲佐菌素建立糖尿病模型。将糖尿病小鼠随机分为假手术组、模型组、低剂量SY干预组、高剂量SY干预组及高剂量SY联合胰岛素样... 目的探讨红花黄色素(SY)对糖尿病小鼠足溃疡(DFU)创面愈合的影响及分子机制。方法取45只C57BL/6小鼠腹腔注射链脲佐菌素建立糖尿病模型。将糖尿病小鼠随机分为假手术组、模型组、低剂量SY干预组、高剂量SY干预组及高剂量SY联合胰岛素样生长因子-1(IGF-1)组,每组9只。DFU模型制备前,模型组小鼠不给予干预处理,低剂量SY干预组和高剂量SY干预组分别腹腔注射5、20 mg·kg^(-1)SY,高剂量SY联合IGF-1组腹腔注射20 mg·kg^(-1)SY和0.03 mg·kg^(-1)IGF-1。除假手术组外,其余4组小鼠通过切开足背部皮肤制备DFU模型;假手术组不制作足背部皮肤伤口,余手术步骤与模型组一致。于造模后第14天,用电子秤测量各组小鼠的体质量,采集尾静脉血测定空腹血糖,并使用小型游标卡尺测量创面宽度,然后处死小鼠收集创面组织,采用苏木精-伊红染色检测各组小鼠创面组织病理学变化,反转录-实时荧光定量聚合酶链反应检测各组小鼠创面组织中血小板衍生生长因子(PDGF)、血管内皮生长因子(VEGF)、α-平滑肌肌动蛋白(α-SMA)和I型胶原蛋白(collagen I)、蛋白酪氨酸磷酸酶1B(PTP1B)、晚期糖基化终产物(AGEs)mRNA的相对表达量,蛋白质印迹法检测各组小鼠创面组织中增殖标志物Ki-67、增殖细胞核抗原(PCNA)、凋亡相关蛋白(caspase-3、caspase-6、caspase-7)、p85磷脂酰肌醇-3-激酶(PI3K)和磷酸化蛋白激酶B(p-AKT)蛋白的相对表达量,酶联免疫吸附试验检测各组小鼠创面组织中肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β及IL-6水平。结果假手术组、模型组、低剂量SY干预组、高剂量SY干预组及高剂量SY联合IGF-1组小鼠血糖和体质量比较差异均无统计学意义(P>0.05)。高剂量SY干预组小鼠创面组织愈合率显著大于模型组、低剂量SY干预组及高剂量SY联合IGF-1组(P<0.01)。模型组、低剂量SY干预组及高剂量SY联合IGF-1组创面组织愈合率比较差异无统计学意义(P>0.05)。高剂量SY干预组小鼠创面组织中出现大量胶原纤维密集有序排列,并伴有大量新生血管;模型组、低剂量SY干预组及高剂量SY联合IGF-1组小鼠创面组织中胶原纤维稀疏,并伴有少量新生血管。高剂量SY干预组小鼠创面组织中PDGF、VEGF、α-SMA及collagen I mRNA相对表达量显著高于模型组、低剂量SY干预组、高剂量SY联合IGF-1组(P<0.01);高剂量SY干预组小鼠创面组织中PTP1B和AGEs mRNA相对表达量显著低于模型组、低剂量SY干预组、高剂量SY联合IGF-1组(P<0.01)。模型组、低剂量SY干预组、高剂量SY联合IGF-1组小鼠创面组织中PDGF、VEGF、α-SMA、collagen I、PTP1B及AGEs mRNA相对表达量比较差异无统计学意义(P>0.05)。高剂量SY干预组小鼠创面组织中Ki-67和PCNA蛋白相对表达量显著高于模型组、低剂量SY干预组、高剂量SY联合IGF-1组(P<0.01);caspase-3、caspase-6、caspase-7、p85 PI3K及p-AKT蛋白相对表达量显著低于模型组、低剂量SY干预组、高剂量SY联合IGF-1组(P<0.01)。模型组、低剂量SY干预组、高剂量SY联合IGF-1组中Ki-67、PCNA、caspase-3、caspase-6、caspase-7、p85 PI3K及p-AKT蛋白相对表达量比较差异无统计学意义(P>0.05)。高剂量SY干预组小鼠创面组织中TNF-α、IL-1β及IL-6水平显著低于模型组、低剂量SY干预组、高剂量SY联合IGF-1组(P<0.01)。模型组、低剂量SY干预组、高剂量SY联合IGF-1组小鼠创面组织中TNF-α、IL-1β及IL-6水平比较差异无统计学意义(P>0.05)。结论高剂量SY干预可通过提高血管生成、胶原形成及细胞增殖,降低胰岛素抵抗、炎症反应和细胞凋亡来促进小鼠DFU创面愈合,其机制可能与抑制PI3K/AKT通路有关。 展开更多

低压静电场协同微冻对小黄鱼贮藏过程中品质的影响

目的探讨添加低压静电场协同微冻技术对小黄鱼贮藏保鲜过程中的鱼肉品质及理化特性的影响。方法以纯低温微冻的小黄鱼组为对照,讨论添加低压静电场的低温环境对小黄鱼的持水率、挥发性盐基氮(total volatile basic nitrogen,TVB-N)值、... 目的探讨添加低压静电场协同微冻技术对小黄鱼贮藏保鲜过程中的鱼肉品质及理化特性的影响。方法以纯低温微冻的小黄鱼组为对照,讨论添加低压静电场的低温环境对小黄鱼的持水率、挥发性盐基氮(total volatile basic nitrogen,TVB-N)值、盐溶性蛋白、硫代巴比妥酸(thiobarbituric acid,TBA)值、钙离子-ATP酶(Ca2+-ATPase)活性、十二烷基硫酸钠-聚丙烯酰胺(sodium dodecyl sulfate-polyacrylamide gel,SDS-PAGE)凝胶电泳、总巯基、菌落总数(total viable count,TVC)、质构特性等重要指标的影响。结果添加低压静电场能有效减缓持水率的下降和TVB-N值的上升(延缓6 d上升至25 mg/100 g);降低了盐溶性蛋白的变性程度(贮藏结束时含量比对照组高79.02%),第34 d盐溶性蛋白含量为32.01 mg/g,远高于对照组(17.88 mg/g);保持鱼肉更高的Ca2+-ATPase活性(对照组的活性下降了57.6%,在电场协同微冻技术作用下只降了26.2%);有效减缓TBA的上升,第34 d时对照组和电场组的TBA值分别为1.24 mg/kg和0.70 mg/kg;有效抑制细菌的繁殖;减少了肌肉组织中裂隙的产生;减少了质构的破坏(贮藏期间硬度与咀嚼性下降更少)和色泽的变化(贮藏期间白度变化更小)。结论低压静电场叠加微冻的贮藏效果明显优于普通微冻贮藏,通过低压静电场叠加微冻的贮藏方式有利于维持水产品品质。 展开更多

红花黄色素对大鼠心梗后心肌重构的影响研究

目的观察红花黄色素(SY)对急性心肌梗死(AMI)后心肌重构的影响,并探究其与腺苷酸活化蛋白激酶(AMPK)/哺乳动物西罗莫司靶蛋白(mTOR)信号通路的关系。方法利用冠状动脉左前支结扎法构建AMI大鼠模型,随机分为Model组、氯沙坦组(10 mg/kg)... 目的观察红花黄色素(SY)对急性心肌梗死(AMI)后心肌重构的影响,并探究其与腺苷酸活化蛋白激酶(AMPK)/哺乳动物西罗莫司靶蛋白(mTOR)信号通路的关系。方法利用冠状动脉左前支结扎法构建AMI大鼠模型,随机分为Model组、氯沙坦组(10 mg/kg)、SY低剂量组(8 mg/kg)、SY高剂量组(32 mg/kg)、SY+Compound C组(32 mg/kg SY+10 mg/kg Compound C),并设假手术(Sham)组,每组12只。连续治疗4周后,使用超声心动图仪测定大鼠心功能指标;采用HE、Masson染色观察大鼠心肌组织变化,计算胶原容积积分(CVF)、测定大鼠心肌细胞直径;酶联免疫吸附法(ELISA)测定心肌组织乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)水平;Werstern blot法检测大鼠心肌组织Ⅰ、Ⅲ型胶原(COL-Ⅰ、COL-Ⅲ)、B型钠尿肽(BNP)、心房钠尿肽(ANP)、微管相关蛋白1轻链3Ⅱ/LC3-Ⅰ(LC3-Ⅱ/LC3-Ⅰ)、AMPK、自噬相关蛋白P62、Beclin-1、磷酸化AMPK(p-AMPK)、mTOR、磷酸化mTOR(p-mTOR)蛋白表达水平。结果与Sham组相比,Model组大鼠心肌细胞肥大,心肌纤维排列紊乱,左室短轴缩短率(LVFS)、左室射血分数(LVEF)及LC3-Ⅱ/LC3-I、Beclin-1、p-AMPK、p-mTOR蛋白表达水平及SOD水平降低(P<0.05),左室舒张末期内径(LVEDD)、左室收缩末期内径(LVESD)、心肌细胞直径、CVF、COL-Ⅰ、COL-Ⅲ、BNP、ANP及P62蛋白表达水平、LDH水平升高(P<0.05);与Model组相比,氯沙坦组、SY低、高剂量组大鼠心肌细胞有所缩小,心肌纤维排列趋于正常,LVFS、LVEF及LC3-Ⅱ/LC3-I、Beclin-1、p-AMPK、p-mTOR蛋白表达水平及SOD水平升高(P<0.05),LVEDD、LVESD、心肌细胞直径、CVF、COL-Ⅰ、COL-Ⅲ、BNP、ANP及P62蛋白表达水平、LDH水平降低(P<0.05);与SY高剂量组相比,SY+Compound C组大鼠心肌细胞肥大,心肌纤维排列紊乱,LVFS、LVEF及LC3-Ⅱ/LC3-I、Beclin-1、p-AMPK、p-mTOR蛋白表达水平及SOD水平降低(P<0.05),LVEDD、LVESD、心肌细胞直径、CVF、COL-Ⅰ、COL-Ⅲ、BNP、ANP及P62蛋白表达水平、LDH水平升高(P<0.05)。结论SY可提高AMI心肌组织抗氧化能力,降低心肌肥厚程度,缓解心肌纤维化进程,逆转心肌重构,可能与激活AMPK/mTOR信号通路,上调心肌自噬有关。 展开更多

目的:探讨花黄色素(SY)调控腺苷酸活化蛋白激酶(AMPK)/沉默信息调节因子1(SIRT1)/核转录因子-κB(NF-κB)对冠心病(CHD)大鼠心肌损伤的影响。方法:将60只SD大鼠随机分为正常对照组(NC组,生理盐水)、模型组(Model组,生理盐水)、SY组(100 ... 目的:探讨花黄色素(SY)调控腺苷酸活化蛋白激酶(AMPK)/沉默信息调节因子1(SIRT1)/核转录因子-κB(NF-κB)对冠心病(CHD)大鼠心肌损伤的影响。方法:将60只SD大鼠随机分为正常对照组(NC组,生理盐水)、模型组(Model组,生理盐水)、SY组(100 mg/kg SY)、EX-527组(47 mg/kg EX-527)、SY+EX-527组(100 mg/kg SY+47 mg/kg EX-527),每组12只,持续4周给予相应药物。试剂盒检测血清三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)水平和心肌白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)水平;苏木素-伊红(HE)染色法进行心肌组织病理学观察;末端脱氧核苷酸转移酶介导的dUTP缺口末端标记(TUNEL)法检测心肌细胞凋亡情况;蛋白免疫印迹(Western Blot)法检测心肌组织AMPK/SIRT1/NF-κB通路相关蛋白表达。结果:与NC组相比,Model组大鼠心肌细胞数量少且排列杂乱,细胞核皱缩,TC、TG、LDL-C、氧化修饰低密度脂蛋白(ox-LDL)、IL-6、TNF-α水平、心肌细胞凋亡率及NF-κB蛋白水平明显上升(P<0.05),SOD、GSH-Px水平及磷酸化-AMPK(p-AMPK)/AMPK、SIRT1蛋白水平明显下降(P<0.05)。与Model组比较,SY组大鼠心肌细胞数量增多且排列整齐有序,TC、TG、LDL-C、ox-LDL、IL-6、TNF-α水平、心肌细胞凋亡率及NF-κB蛋白水平明显下降(P<0.05),SOD、GSH-Px水平及p-AMPK/AMPK、SIRT1蛋白水平明显上升(P<0.05);而EX-527组以上指标呈现相反趋势。与SY组相比,SY+EX-527组大鼠心肌损伤加重,TC、TG、LDL-C、ox-LDL、IL-6、TNF-α水平、心肌细胞凋亡率及NF-κB蛋白水平明显上升(P<0.05),SOD、GSH-Px水平及p-AMPK/AMPK、SIRT1蛋白水平明显下降(P<0.05)。结论:SY可能通过调控AMPK/SIRT1/NF-κB通路降低氧化应激,减轻炎症反应,对冠心病大鼠心肌损伤起到一定改善作用。 展开更多
