Methanol oxidation reaction (MOR) at Pt and Pt electrode surface deposited with various amounts of Ru (denoted as PtxRuy, nominal coverage y is 0.17, 0.27, and 0.44 ML) in 0.1 mol/L HClO4+0.5 mol/L MeOH has been studied under potentiostatic conditions by in situ FTIR spectroscopy in attenuated-total-reflection con guration and di erential electro-chemical mass spectrometry under controlled flow conditions. Results reveal that (i) CO is the only methanol-related adsorbate observed by IR spectroscopy at all the Pt and PtRu electrodes examined at potentials from 0.3 V to 0.6 V (vs. RHE); (ii) at Pt0.56Ru0.44, two IR bands, one from CO adsorbed at Ru islands and the other from COL at Pt substrate are detected, while at other electrodes, only a single band for COL adsorbed at Pt is observed; (iii) MOR activity decreases in the order of Pt0.73Ru0.27〉Pt0.56Ru0.44〉Pt0.83Ru0.17〉Pt; (iv) at 0.5 V, MOR at Pt0.73Ru0.27 reaches a current e ciency of 50% for CO2 production, the turn-over frequency from CH3OH to CO2 is ca. 0.1 molecule/(site sec). Suggestions for further improving of PtRu catalysts for MOR are provided. 展开更多

Methanol Perturbing Modeling Cell Membranes Investigated using Linear and Nonlinear Vibrational Spectroscopy 被引量:1

Cell membranes play a crucial role in many biological functions of cells. A small change in the composition of cell membranes can strongly influence the functions of membrane-associated proteins, such as ion and water channels, and thus mediate the chemical and physical balance in cells. Such composition change could originate from the introduction of short-chain alcohols, or other anesthetics into membranes. In this work, we have applied sum frequency generation vibrational spectroscopy (SFG-VS), supplemented by attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR), to investigate interaction between methanol and 1,2-dimyristoyl-d54-sn-glycero-3-phosphocholine (d54-DMPC) lipid bilayers. Lipid's hydrocarbon interior is deuterated while its head group is hydrogenated. At the same time, CH3 symmetric stretch from methanol and lipid head amine group has different frequency, thus we can distinguish the behaviors of methanol, lipid head amine group, and lipid hydrocarbon interior. Based on the spectral feature of the bending mode of the water molecules replaced by methanol, we determined that the methanol molecules are intercalated into the region between amine and phosphate groups at the lipid hydrophilic head. The dipole of CH3 groups of methanol and lipid head, and the water O-H M1 adopt the same orientation directions. The introduction of methanol into the lipid hydrophilic head group can strongly perturb the entire length of the alkyl chains, resulting that the signals of CD2 and CD3 groups from both leaflets can not cancel each other. 展开更多

Besides carbon solubility, the carbide formation possibility is another important factor to differentiate various substrate materials in graphene growth. A recent experiment indicates that the formation of transition metal carbides (TMCs) can suppress carbon precipitation. In this study, Mo2C, a representative of TMCs, is used to study the effects of carbide formation in graphene growth from first principles. Carbon diffusion in Mo2C bulk turns out to be very difficult and it becomes much easier on the Mo2C(001) surface. Therefore, carbon precipitation suppression and graphene growth can be realized simultaneously. A direction depended diffusion behavior is observed on the Mo2C(101) surface, which makes it less favorable for graphene growth compared to the (001) surface. 展开更多

结合表面等离波子共振(SPR)技术与免疫检测技术,研究和建立了一种响应速度快、免标记、低成本的地表水微囊藻毒素(MC-LR)检测方法。基于SpreetaTM传感器构建了小型SPR免疫检测系统,采用共价偶联方法在传感器金膜表面修饰MC-LR-BSA抗原... 结合表面等离波子共振(SPR)技术与免疫检测技术,研究和建立了一种响应速度快、免标记、低成本的地表水微囊藻毒素(MC-LR)检测方法。基于SpreetaTM传感器构建了小型SPR免疫检测系统,采用共价偶联方法在传感器金膜表面修饰MC-LR-BSA抗原为生物敏感膜;开展了MC-LR的SPR免疫检测方法实验研究,结果表明该方法的相对标准偏差为1.0%(n=6),定量范围为2~32ng/mL,检测限为0.63ng/mL,半抑制浓度CI50为10.7ng/mL,空白加标回收率和样品加标回收率在90%~113%之间。实样检测实验中,管道末梢饮用自来水水样未能检测出MC-LR,而某湖水水样中MC-LR的质量浓度为2.46ng/mL。实验研究与结果表明,MC-LR的SPR免疫检测方法可以满足世界卫生组织(WHO)对于饮用水中MC-LR最低含量检测的需求。 展开更多

蹭行运动在生物被膜形成过程中对细菌适应表面环境以及后续生物被膜三维结构的形成起重要作用。因此,对蹭行运动的原位表征、量化是生物被膜研究中的重要科学问题之一。我们通过高通量数据采集、自动化图像处理、数据库建立以及图形化... 蹭行运动在生物被膜形成过程中对细菌适应表面环境以及后续生物被膜三维结构的形成起重要作用。因此,对蹭行运动的原位表征、量化是生物被膜研究中的重要科学问题之一。我们通过高通量数据采集、自动化图像处理、数据库建立以及图形化输出等技术手段,建立了一整套基于单细菌的统计分析方法。利用这一方法对蹭行运动中的行走、弹射过程进行了详细分析,发现弹射运动过程中存在以0.9 s为周期的周期性弛豫。并定量比较了群体感知信号分子对蹭行运动的影响,发现加入信号分子后蹭行运动在高速区明显增强。该方法的建立为后续蹭行运动分子机制以及调节方式的研究奠定了基础。 展开更多

[目的]表达纯化布氏锥虫自噬蛋白TbATG12,并对其二级结构进行预测。[方法]克隆TbATG12编码序列,采用酶切连接方法构建表达载体TbATG12(pET-28a),转化进入BL21(DE3)表达菌株诱导蛋白表达,利用Ni^(2+)-NAT亲和层析及分子筛层析方法纯化蛋白。采用核磁手段,采集一维和二维谱图,利用JPred 4预测其二级结构。[结果]16℃、0.5 mmol/L IPTG诱导20 h,TbATG12以可溶蛋白形式存在。纯化后蛋白纯度可达95%。TbATG12谱峰分布均匀,强度较强,二级结构预测其含有3个α-螺旋和5个β-折叠,为后续解析TbATG12溶液结构提供指导。[结论]克隆、表达纯化得到纯度>95%可溶TbATG12蛋白,二级结构预测和比对发现其结构在进化上很保守。 展开更多
