通过基因工程技术 ,使白曲酶菌 (AspergillusKawachii)的耐酸性α-淀粉酶和糖化酶基因多拷贝化 ,获得重组体生产菌株TR12。用此菌株进行制曲 ,进行了实验室规模的米烧酒发酵试验考察。发现发酵糟经减压蒸馏制取米烧酒后 ,在蒸馏废液中... 通过基因工程技术 ,使白曲酶菌 (AspergillusKawachii)的耐酸性α-淀粉酶和糖化酶基因多拷贝化 ,获得重组体生产菌株TR12。用此菌株进行制曲 ,进行了实验室规模的米烧酒发酵试验考察。发现发酵糟经减压蒸馏制取米烧酒后 ,在蒸馏废液中单是投入蒸米就能再循环发酵和可制取优质米烧酒。其后的蒸馏废液经过再次的再循环发酵 ,即半固体状态的发酵和蒸馏后 ,与常规曲组的对照实验相比 ,米烧酒的发酵收率及香味成分浓度进一步提高。发酵收率达到 470mL/kg以上 ,乙酸异戊酯、乙酸苯乙酯等特异性芳香成分的浓度分别达到 2 1 4mg/ 10 0mL和 8 6mg/ 10 0mL的极高水平。并且 ,蒸馏废液的排除量仅仅为传统方法的 33%或者更低。 展开更多

With the transformant TR12 containing multi copies of acid stable α-amylase and gflucoamylase genes in fermentation,it was possible to produce quality rice liquor from the post-distillation slurry used repeatedly not drain However,it did’t do the same with the white qu mold Aspergillus Kawachii Furthermore,saccarifying enzymes as much as that contained in the post-distillation derived from fermented mash of using TR12 strain were added to the post-distillation slurry derived from that of using conventional white qu mold,the value of alcohol yield and the quality of rice liquor yet were lower in fermentation Thus,after assaying some composition of rice qu and commercialized enzymes,a semi-solid atate fermentation test that the post-distillation slurry was used in second time was carried out by adding saccarifying enzymes with a low activity of acid protease As a result,comparing with the contrast,the fermentation yield increased to 460 mL/kg,the concentration of isoamy1 acetate and β-phenylethy1 acetate increased to 20 0 mg/L and6.2mg/L,approached the level of using TR12 strain And the discharge of distillation wastewater was only 33% that of conventional method or even lower。 展开更多
