目的:观察阿魏菇多糖(polysaccharides from Pleurotus ferulae,PFP)、顺铂(DDP)及两者联合对小鼠宫颈癌细胞株U14移植瘤的抑制作用及凋亡诱导作用,并初步探讨阿魏菇多糖的抗肿瘤作用机制。方法:建立昆明小鼠U14宫颈癌模型,并随机分为四...目的:观察阿魏菇多糖(polysaccharides from Pleurotus ferulae,PFP)、顺铂(DDP)及两者联合对小鼠宫颈癌细胞株U14移植瘤的抑制作用及凋亡诱导作用,并初步探讨阿魏菇多糖的抗肿瘤作用机制。方法:建立昆明小鼠U14宫颈癌模型,并随机分为四组:模型对照组、PFP组、DDP组、PFP+DDP组,治疗10天后检测各组瘤重及抑瘤率,采用TUNEL法检测各组瘤组织凋亡指数(apoptosis index,AI)。结果:模型组小鼠体内瘤体积、瘤重明显高于各干预组(P<0.01),PFP+DDP组小鼠体内瘤重和瘤体积分别为[(0.40±0.61)g,(197.84±11.99)mm3]低于DDP组[(0.61±0.26)g,(211.46±14.58)mm3];PFP组、DDP组、PFP+DDP组瘤组织凋亡指数均高于模型对照组,其中以PFP+DDP组最明显。结论:PFP联合DDP比两药单独应用抑制肿瘤生长及诱导肿瘤细胞凋亡的作用更强,二者具有协同抗癌作用。PFP可以增强DDP的药物敏感性,其作用机制可能是通过诱导宫颈癌细胞凋亡而实现的。展开更多
