Expression of PKC Iota Affects Neuronal Differentiation of PC12 Cells at Least Partly Independent of Kinase Function 被引量:3

Atypical PKC (aPKC) plays a role in establishing cell polarity and has been indicated in neuronal differentiation and polarization, including neurite formation in rat pheochromocytoma PC12 cells, albeit by unclear mechanisms. Here, the role of the aPKC isoform, PKC iota (PKCι), in the early neuronal differentiation of PC12 cells, was investigated. NGF-treated PC12 cells with stably expressed exogenous wild-type PKCι showed decreased expression of a neuroendocrine marker, increased expression of a neuronal marker and increased neurite formation. Stable expression of a kinase-inactive PKCι, but not constitutively active PKCι lacking a regulatory domain, had similar though less potent effects. Pharmacological inhibition of endogenous aPKC kinase activity in parental PC12 cells did not inhibit neurite formation, suggesting that some of the observed effects of PKCι expression on neuronal differentiation are kinase-independent. Interestingly, exogenous expression of wild-type and kinase-inactive PKCι had little effect on overall PKCι activity, but caused a decrease in PKC zeta (PKCζ) kinase activity, suggesting an interplay between the two isoforms that may underlie the observed results. Overall, these findings suggest that in PC12 and perhaps other neuroendocrine precursor cells, PKCι influences an early differentiation decision between the neuroendocrine (chromaffin) and sympathetic neuron cell lineages, potentially by affecting PKCζ function. 展开更多

Could Sequential Residual Centering Resolve Low Sensitivity in Moderated Regression? Simulations and Cancer Symptom Clusters

Multicollinearity constitutes shared variation among predictors that inflates standard errors of regression coefficients. Several years ago, it was proven that the common practice of mean centering in moderated regression cannot alleviate multicollinearity among variables comprising an interaction, but merely masks it. Residual centering (orthogonalizing) is unacceptable because it biases parameters for predictors from which the interaction derives, thus precluding interpretation of moderator effects. I propose and validate residual centering in sequential re-estimations of a moderated regression—sequential residual centering (SRC)—by revealing unbiased multicollinearity conditioning across the interaction and its related terms. Across simulations, SRC reduces variance inflation factors (VIF) regardless of distribution shape or pattern of regression coefficients across predictors. For any predictor, the reduced VIF is used to derive a lower standard error of its regression coefficient. A cancer sample illustrates SRC, which allows unbiased interpretations of symptom clusters. SRC can be applied efficiently to alleviate multicollinearity after data collection and shows promise for advancing synergistic frontiers of research. 展开更多

The Value of Positive Pressure Ventilations for Clients in Acute Respiratory Distress as a Result of Cardiac and Pulmonary Issues

Objective: Research was conducted to examine benefits to using non-invasive ventilation (NIV) or continuous positive airway pressure (CPAP) early in the treatment of respiratory distress caused by pulmonary edema, chronic obstructive pulmonary disease (COPD) and asthma. Limitations to successful NIV and CPAP therapy were evaluated to determine how prolonged initiation of treatment may lead to hypoxemia (decreased oxygen in the blood) and hypercapnia (increased carbon dioxide in the blood) resulting in poor outcomes. Method: Reviews of literature from nursing and allied health data bases (CINAHL and ProQuest) with terms pulmonary edema, positive pressure device and non-invasive ventilation from 2010 to 2014 were used. Studies were conducted in the hospital and prehospital settings. Results: The literature search located 7 articles from CINAHL and 25 articles from ProQuest. A total of 6 of these articles were analyzed. Additional sources of data were obtained from Ignatavicius and Workman (2013) Medical-Surgical Nursing Patient-Centered Collaborative Care 7th edition and American Journal of Nursing (02/2013) Volume 113: 2. Conclusion: All of the articles concluded that early initiation of continuous positive airway pressure ventilations in the short-term was beneficial;however, late initiation of therapy required additional interventions. The studies indicated that early use of positive airway pressure in acute respiratory distress improved breath rate, heart rate and blood pressure. The use of positive airway pressure for respiratory distress may decrease the need for endotracheal intubation. 展开更多

目的:调查分析维持性血液透析患者动静脉内瘘穿刺疼痛的现状及相关影响因素。方法使用疼痛视觉模拟法评分、疼痛自我效能量表及患者一般情况调查表对180例使用动静脉内瘘进行维持性血液透析治疗的患者进行调查。结果回收有效问卷169份... 目的:调查分析维持性血液透析患者动静脉内瘘穿刺疼痛的现状及相关影响因素。方法使用疼痛视觉模拟法评分、疼痛自我效能量表及患者一般情况调查表对180例使用动静脉内瘘进行维持性血液透析治疗的患者进行调查。结果回收有效问卷169份,有效回收率93.89%(169/180)。动静脉内瘘穿刺患者穿刺疼痛强度中位数评分为5分,疼痛自我效能得分为(31.42±14.59)分;逐步多元回归分析显示,穿刺疼痛主要与患者性别、内瘘使用时间及疼痛自我效能有关(β=0.152、-0.017、-0.409,P<0.05),OR值(95%CI)分别为2.069(1.206~3.148)、0.803(0.710~0.984)、1.004(0.886~1.431);穿刺疼痛强度与疼痛自我效能呈负相关,r=-0.647,P<0.01。结论动静脉内瘘穿刺维持性血液透析患者穿刺疼痛普遍存在,患者的性别、内瘘使用时间及疼痛自我效能可影响患者对疼痛的感知。 展开更多

The mechanism by which the plant hormone auxin regulates gene expression has been shown to involve regulated degradation, through the ubiquitin-proteasome pathway, of transcriptional repressor proteins. However, the key first component in this pathway, the receptor that binds auxin and initiates auxin signaling, has remained a mystery. Two recent papers identify the F-box protein TIR1, part of the complex that attaches ubiquitin to its targets, as an auxin receptor. This breakthrough reveals a new mode of signal transduction and lays the groundwork for a more complete understanding of auxin physiology. 展开更多

This paper considers rearrangeable multihop lightwave networks whereby each network node is equipped with a number p of transmitters and receivers, and a spectrum of wavelengths is accessible by, and shared among, all nodes by using the Wavelength Division Multiplexing (WDM). Depending on input traffic flow, nodal transmitters and receivers can be re-tuned to create virtual connectivity best suited with respect to a given optimization criterion. We present an efficient heuristic algorithm that combines two criteria for optimization: throughput maximization, as well as total flow minimization. Throughput maximization criterion is equivalent to congestion minimization, while minimizing total flow under the assumption of having links with equal lengths implies minimization of the average number of hops. Taking into account lengths of the links (i.e. link costs proportional with distances), the total flow minimization becomes equivalent to the total delay minimization.
