Ecological Planning of Resource-based City——A Case Study of Huaibei City,Anhui Province

Ecological planning plays an important role in the construction of cities. Huaibei is a typical coal resource-based city, the abundant coal resources lay a solid foundation for its economic development. Coal mining has induced a series of social problems, and affected the regional social and economic development. Ecological planning of Huaibei City concentrates on ecological restoration of coal mining subsidence, and applies circular economy to develop eco-agriculture, eco-tourism and eco-industry, which will contribute considerably to the economic transformation of the city. Analytic Hierarchy Process and mathematical statistical model were applied to calculate sustainable development indexes of Huaibei from 2002 to 2010. By evaluating the indexes, it was found that Huaibei City has a low sustainable development level, and 3 imbalanced subsystems as the major obstacle. The authors proposed countermeasures according to specific conditions of the city. 展开更多

Effects of Sucrose on Total RNA Concentration during Formation of Microtubers of Pinellia ternata in vitro

[ Objective] This study aimed to investigate the variation rule of total RNA concentration during formation of microtubers of PineUia ternata induced by sucrose. [ Method ] Plantlets of P. ternata were used as experimental materials. Total RNA concentration was determined with spectrophotometer, and the variation of total RNA was analyzed before and after formation of P. ternata microtubers treated with different concentrations of sucrose using DDRT-PCR technology. [ Result] Total RNA concentration of P. ternata treated with different concentrations of sucrose varied greatly, which showed an upward trend in the first 15 d, de- clined from 15~ d to 25th d, and slightly raised from 25'h d to 30'8 d. Microtubers formed under various sucrose treatments showed an increase in RNA concentration after induction, and it was assumed that the treatment of high concentrations of sucrose might have activated the expression of development-related' genes in P. ter- nata microtubers. [ Conclusion] This study provides the basis for revealing the related gene expression information and molecular mechanism during formation of P. ternata microtubers induced by sucrose. 展开更多

A magnetosome-deleted mutant NM21 of Magnetospirillum gryphiswaldense MSR-1 was generated by mini-Tn5 lacZ2 transposon mutagenesis, and a 3073-bp fragment flanking mini-Tn5 lacZ2 in NM21 was cloned by Anchored PCR. Sequencing analysis showed that this fragment involved three putative ORFs; the mini-Tn5 lacZ2 was inserted into ORF1. Functional complementary test indicated that the 3073-bp fragment was required for biosynthesis of magnetosomes in M. gryphiswaldense MSR-1. The majority of proteins, which had homology with the protein encoded by ORF1, were the cation transporter. Transmembrane domain analysis showed that the protein encoded by ORF1 contained four trans-membrane domains. It may be a transmembrane protein. The protein encoded by ORF1 contained two putative conserved domains: COG0053 and PRK09509. The MMT1 and FieF, containing conserved do-mains COG0053 and PRK09509 too, were Fe2+ transporter (cation diffusion facilitator superfamily). It was suggested that the protein encoded by ORF1 might take part in the magnetosomes biosynthesis as Fe2+ transporter. 展开更多
