[ Method] The model of piglets with spleen deficiency was produced by the methods of injecting reserpine into piglets, and alternating between no feed and good feed. The determinations of serum amylase, lipase and xanthine oxidase of piglets with spleen deficiency were done before injecting reserpine, on the day when reserpine was injected last time, the next day of finishing giving powder, the eighth day after finishing giving powder. [ Result] After the experiment was finished, all 3 enzymes activities of Guchangcuzhangsan group had no significant differ- ence from that of normal contrast group, enzyme activities of spleen deficiency group and smecta group had significant difference compared with that of normal contrast group. [ Conclusion] Guchangcuzhangsan can promote the recovery of serum amylase, lipase and xanthine oxidase of piglets with spleen deficiency which was caused by reserpine.展开更多

The mechanism and rapid detection method of this disease has not been reported. We applied bulked sergeant analysis (BSA) in combination with RAPD method to analyze a molecular genetic marker linked with self-biting trait in mink group. The molecular marker was converted into sequence-characterized amplified regions (SCAR) marker for rapid detection of this disease. A single RAPD marker A8 amplified a specific band of 263bp in self-biting minks, which was designated as SRA8-250, and non-specific band of 315bp in both self-biting and healthy minks. The sequences of the bands exhibited 75% and 88% similarity to Canis familiarizes major histocompatibility complex (MHC) class II region and Macaca mulatta MHC class I region, respectively. A SCAR marker SCAR-A8 was designed for the specific fragment SRA8-250 and validated in 30 self-biting minks and 30 healthy minks. Positive amplification of SCAR-A8 was detected in 24 self-biting minks and 12 healthy minks. χ2 test showed significant difference (p〈0.01) in the detection rate between the two groups. This indicated that SRA8-250 can be used as a positive marker to detect self-biting disease in minks. Furthermore, the finding that self-biting disease links with MHC genes has significant implications for the mechanism of the disease.展开更多

Overexpression of Hoxa10 in mouse endometrium can increase litter size. Moreover, Hoxa10 plays a key role in regulating the embryo implantation of sows. This study aimed to generate transgenic pigs using Hoxa10 via somatic cell nuclear transfer(SCNT). We established seven Hoxa10-transgenic cell lines, and two of the cell lines were selected as nuclear donors for the transfer. A total of 1 270 cloned embryos were generated and transferred to five surrogate mothers(Landrace×Yorkshire). Eight cloned male piglets were produced including one with cryptorchidism. Six transgenic piglets grew up healthy and produced 56 offspring. Finally, we obtained six transgenic male pigs and 26 transgenic positive offspring that can be used to further study the regulatory mechanism of Hoxa10 on the reproductive performance of pigs.展开更多

Hepatitis A virus is a pathogen associated with water pollution.Contaminated drinking water can cause hepatitis A outbreaks,lead to economic losses,and even threaten human lives.It is difficult to detect low levels of...Hepatitis A virus is a pathogen associated with water pollution.Contaminated drinking water can cause hepatitis A outbreaks,lead to economic losses,and even threaten human lives.It is difficult to detect low levels of hepatitis A virus in water,so the virus must be concentrated in order to quantify it accurately.Here,we present a simple,rapid,efficient technique for the concentration and detection of hepatitis A virus in water.Our data showed that adding phosphate-buffered saline to the water,pre-filtering the water,and adding Trizol reagent directly to the filtration membrane can significantly improve concentration efficiency.Of three types of filtration membranes studied(mixed cellulose ester membrane,polyvinylidene fluoride membrane,and nylon membrane),the concentration efficiency using mixed cellulose ester membrane with a 0.1-μm pore size was the highest,reaching 92.62 ± 5.17%.This method was used to concentrate hepatitis A virus in water samples from Donghu Lake.Using SYBR Green real-time reverse transcription polymerase chain reaction analysis,the detection sensitivity of this method reached 10~1 copies/μL and its concentration efficiency reached 79.45 ± 9.88%.展开更多
