【目的】木薯(Manihot esculenta Crantz)中含有潜在毒性的生氰糖苷,其食用安全性受到影响且导致加工成本增加。因此,利用生物技术开展低生氰糖苷木薯的培育具有重要意义。【方法】利用CRISPR/Cas9技术对木薯醇氰酶基因MeHNL进行了编辑... 【目的】木薯(Manihot esculenta Crantz)中含有潜在毒性的生氰糖苷,其食用安全性受到影响且导致加工成本增加。因此,利用生物技术开展低生氰糖苷木薯的培育具有重要意义。【方法】利用CRISPR/Cas9技术对木薯醇氰酶基因MeHNL进行了编辑。该基因编码催化生氰糖苷分解的α-羟基腈裂解酶,编辑靶点位于第1个外显子上,通过农杆菌介导的稳定转化获得了27株阳性植株。【结果】测序分析显示,其中26个株系被编辑,编辑效率高达96.3%。编辑类型主要包括碱基的插入和缺失,少数为碱基替换和大片段缺失。氰化物检测试剂盒染色和HPLC测定分析表明,编辑株系中的氢氰酸和生氰糖苷含量均显著降低。与非编辑植株相比,编辑植株的叶片细长,暗示了MeHNL可能对木薯的生长发育产生影响。【结论】利用CRISPR/Cas9技术获得了低氰化物的木薯种质,为开展生氰糖苷代谢影响木薯生长发育的研究提供了材料。 展开更多

通过浸渍法制备出一系列氧化钙负载铂金属催化材料,用于生物质基糠醇选择性加氢制备戊二醇研究。研究采用X射线衍射仪、扫描电子显微镜、CO_(2)-程序升温脱附、N2等温吸附-脱附、X射线光电子能谱、热重-差热分析法等一系列表征手段,对... 通过浸渍法制备出一系列氧化钙负载铂金属催化材料,用于生物质基糠醇选择性加氢制备戊二醇研究。研究采用X射线衍射仪、扫描电子显微镜、CO_(2)-程序升温脱附、N2等温吸附-脱附、X射线光电子能谱、热重-差热分析法等一系列表征手段,对合成的催化剂样品物理化学性质进行了系统表征。实验结果表明,Pt/CaO-600催化剂表现出优异的催化活性,在4MPa氢压、210℃反应温度条件下,糠醇转化率为99.8%,1,2-戊二醇和1,5-戊二醇收率分别可达48.6%和21.5%。催化剂的优异性能可归因于作为载体的CaO提供了适宜的碱性位点,有利于糠醇开环反应的发生,进而提高了糠醇转化率与戊二醇选择性。同时,催化剂还表现出了优异的循环使用性能,经重复使用4次后,其催化糠醇转化率可达96.5%,1,2-戊二醇和1,5-戊二醇的收率分别为40.3%和17.0%。 展开更多

当前锂辉石提锂工艺主要采用高温转型—硫酸化焙烧法,通常需要1100℃左右转晶焙烧及硫酸化焙烧工序,存在能耗高、工艺流程长等问题。提出了一种以CaO和Na_(2)SO_(4)为混合助剂的一步低温焙烧法从α-锂辉石提取锂的工艺,进一步优化了助... 当前锂辉石提锂工艺主要采用高温转型—硫酸化焙烧法,通常需要1100℃左右转晶焙烧及硫酸化焙烧工序,存在能耗高、工艺流程长等问题。提出了一种以CaO和Na_(2)SO_(4)为混合助剂的一步低温焙烧法从α-锂辉石提取锂的工艺,进一步优化了助剂用量及焙烧温度。结果表明:在焙烧温度900℃、焙烧时间2 h,50%Na_(2)SO_(4)和6%CaO混合助剂作用下,锂浸出率达94.97%。借助XRD、FT-IR、SEM-EDS等分析方法明确了混合助剂对α-锂辉石一步焙烧提锂过程的矿相调控机理为:CaO在低温可与α-锂辉石反应生成钙长石及石英,破坏锂辉石的稳定结构,降低了α-锂辉石转型温度,Na_(2)SO_(4)与石英、锂辉石进一步反应生成钠长石和可溶性NaLiSO 4。该工艺显著降低了锂辉石的焙烧温度,实现了锂的短程高效提取,具有重要的应用价值。 展开更多

鹦嘴鱼科(Scaridae)鱼类参与珊瑚礁生态系统诸多关键生态过程,在维持珊瑚礁生态系统稳定与平衡中发挥着重要作用。由于以往研究手段的限制,对鹦嘴鱼的食物来源及生态功能价值认识不足,在其功能定位方面存在较多争议。本研究选择珊瑚分... 鹦嘴鱼科(Scaridae)鱼类参与珊瑚礁生态系统诸多关键生态过程,在维持珊瑚礁生态系统稳定与平衡中发挥着重要作用。由于以往研究手段的限制,对鹦嘴鱼的食物来源及生态功能价值认识不足,在其功能定位方面存在较多争议。本研究选择珊瑚分布的典型区域—南沙群岛中的东门礁和南薰礁为研究海域,对该海域鹦嘴鱼优势种类黑斑鹦嘴鱼(Scarus globiceps)摄食的藻类多样性进行全面分析。通过18S rDNA和16S rDNA多基因条形码技术,分别对黑斑鹦嘴鱼肠含物的真核藻类和原核藻类DNA进行高通量测序分析。18S rDNA的测序结果发现,黑斑鹦嘴鱼类的肠含物中真核藻类以甲藻(Dinoflagellata)、红藻(Rhodophyta)、绿藻(Chlorophyta)、褐藻(Ochrophyta)为主,共计77个OTU(operational taxonomic units)。甲藻相对序列丰度和多样性较高,序列比例占真核藻类序列总数的51.42%,其中网甲藻科(Suessiaceae)OTU_5在东门礁和南薰礁的样品中均超过20%。16S rDNA测序鉴定发现肠含物含有原核藻类(蓝藻)的序列,共计21个OTU,其中以念珠藻目(Nostocales)的相对序列丰度最高,达到39.33%。本研究表明,黑斑鹦嘴鱼在摄食过程中会摄入一定量大型藻类,但微藻(甲藻)序列占据优势地位,蓝藻在肠含物中也有较高的检出率,说明需要重新考虑微藻(甲藻和蓝藻)在鹦嘴鱼食源中的重要贡献以及对珊瑚礁生态系统结构与功能的影响。 展开更多

The use of‘Electrostatic tweezers'is a promising tool for droplet manipulation,but it faces many limitations in manipulating droplets on superhydrophobic surfaces.Here,we achieve noncontact and multifunctional droplet manipulation on Nepenthes-inspired lubricated slippery surfaces via triboelectric electrostatic tweezers(TETs).The TET manipulation of droplets on a slippery surface has many advantages over electrostatic droplet manipulation on a superhydrophobic surface.The electrostatic field induces the redistribution of the charges inside the neutral droplet,which causes the triboelectric charged rod to drive the droplet to move forward under the electrostatic force.Positively or negatively charged droplets can also be driven by TET based on electrostatic attraction and repulsion.TET enables us to manipulate droplets under diverse conditions,including anti-gravity climb,suspended droplets,corrosive liquids,low-surface-tension liquids(e.g.ethanol with a surface tension of 22.3 mN·m^(-1)),different droplet volumes(from 100 nl to 0.5 ml),passing through narrow slits,sliding over damaged areas,on various solid substrates,and even droplets in an enclosed system.Various droplet-related applications,such as motion guidance,motion switching,droplet-based microreactions,surface cleaning,surface defogging,liquid sorting,and cell labeling,can be easily achieved with TETs. 展开更多

基于FlowCam影像和YOLOv3深度学习模型的多纹膝沟藻检测

海洋中的有害藻华,包括微藻形成的赤潮和褐潮以及大型藻类形成的绿潮和金潮,已成为一类突出的海洋生态灾害问题。2021年11—12月,山东半岛东北部海域发生多纹膝沟藻(Gonyaulax polygramma)形成的大规模赤潮,导致海带养殖严重受损,亟待... 海洋中的有害藻华,包括微藻形成的赤潮和褐潮以及大型藻类形成的绿潮和金潮,已成为一类突出的海洋生态灾害问题。2021年11—12月,山东半岛东北部海域发生多纹膝沟藻(Gonyaulax polygramma)形成的大规模赤潮,导致海带养殖严重受损,亟待构建有针对性的赤潮监测预警体系。本文以多纹膝沟藻为对象,尝试应用流式影像仪(FlowCam)结合卷积神经网络模型YOLOv3,分别对含有多纹膝沟藻的浓缩海水样品、含有多纹膝沟藻和链状亚历山大藻(Alexandrium catenella)的浓缩海水样品,以及含有多纹膝沟藻活细胞及空壳的浓缩海水样品进行了检测实验,探究了基于藻种影像的深度学习模型在赤潮原因种检测方面的应用潜力。结果表明,在以FlowCam获取的影像数据集训练3万次后,YOLOv3对多纹膝沟藻表现出较好的识别能力,对浓缩海水样品中目标藻种识别的平均精度为88.2%;当样品中存在与多纹膝沟藻形态相似的链状亚历山大藻时,模型对多纹膝沟藻识别的平均精度下降为76.6%,对两种微藻同时进行训练和检测,可将对多纹膝沟藻识别的平均精度提高至84.6%。对于低温下出现脱壳现象的多纹膝沟藻,模型对海水中活细胞识别的平均精度为86.7%,对空壳识别的平均精度87.8%,将多纹膝沟藻活细胞与空壳分别识别的平均精度均值(87.3%)高于将两者统一作为多纹膝沟藻进行检测的识别精度(84.2%),整体识别精度提升3.1%。综合相关结果可以看出,FlowCam与深度学习模型相结合在赤潮监测和研究中具有重要应用潜力。 展开更多

Transition metal phosphides(TMPs)have been regarded as alternative hydrogen evolution reaction(HER)and oxygen evolution reaction(OER)catalysts owing to their comparable activity to those of noble metal-based catalysts.TMPs have been produced in various morphologies,including hollow and porous nanostructures,which are features deemed desirable for electrocatalytic materials.Templated synthesis routes are often responsible for such morphologies.This paper reviews the latest advances and existing challenges in the synthesis of TMP-based OER and HER catalysts through templated methods.A comprehensive review of the structure-property-performance of TMP-based HER and OER catalysts prepared using different templates is presented.The discussion proceeds according to application,first by HER and further divided among the types of templates used-from hard templates,sacrificial templates,and soft templates to the emerging dynamic hydrogen bubble template.OER catalysts are then reviewed and grouped according to their morphology.Finally,prospective research directions for the synthesis of hollow and porous TMP-based catalysts,such as improvements on both activity and stability of TMPs,design of environmentally benign templates and processes,and analysis of the reaction mechanism through advanced material characterization techniques and theoretical calculations,are suggested. 展开更多

Radiotherapy for hyoid bone metastasis from lung adenocarcinoma:A case report 被引量:1

BACKGROUND Metastasis to the hyoid bone is an exceptionally rare occurrence,with documented cases limited to breast,liver,colon,skin,lung,and prostate cancers.This report highlights an unusual case involving the metastasis of lung adenocarcinoma to the hyoid bone,accompanied by a distinctive headache.Previous documentation involved surgical resection of the hyoid mass.We present a case displaying the benefits of palliative radiotherapy.CASE SUMMARY A 72-year-old non-smoking,non-alcoholic woman,initially under investigation for a year-long elevation in absolute lymphocyte count,presented with a monthlong history of intermittent throat pain.Despite negative findings in gastroenterological and otolaryngologic examinations,a contrast-enhanced chest computed tomography scan revealed a mediastinal mass and questionable soft tissue thickening in her left anterolateral neck.Subsequent imaging and biopsies confirmed the presence of lung adenocarcinoma metastasis to the hyoid bone.The patient was treated with platinum-based chemo-immunotherapy along with pembrolizumab.Ultimately,the lung cancer was unresponsive.Our patient opted for palliative radiation therapy instead of surgical resection to address her throat pain.As a result,her throat pain was alleviated,and it also incidentally resolved her chronic headaches.This is the second documented case of lung adenocarcinoma metastasizing to the hyoid bone.CONCLUSION Palliative radiotherapy may add to the quality of life in symptomatic patients with cancer metastatic to the hyoid bone. 展开更多

Jetting-based bioprinting facilitates contactless drop-on-demand deposition of subnanoliter droplets at well-defined positions to control the spatial arrangement of cells,growth factors,drugs,and biomaterials in a highly automated layer-by-layer fabrication approach.Due to its immense versatility,jetting-based bioprinting has been used for various applications,including tissue engineering and regenerative medicine,wound healing,and drug development.A lack of in-depth understanding exists in the processes that occur during jetting-based bioprinting.This review paper will comprehensively discuss the physical considerations for bioinks and printing conditions used in jetting-based bioprinting.We first present an overview of different jetting-based bioprinting techniques such as inkjet bioprinting,laser-induced forward transfer bioprinting,electrohydrodynamic jet bioprinting,acoustic bioprinting and microvalve bioprinting.Next,we provide an in-depth discussion of various considerations for bioink formulation relating to cell deposition,print chamber design,droplet formation and droplet impact.Finally,we highlight recent accomplishments in jetting-based bioprinting.We present the advantages and challenges of each method,discuss considerations relating to cell viability and protein stability,and conclude by providing insights into future directions of jetting-based bioprinting. 展开更多

This study employs advanced electrochemical and surface characterization techniques to investigate the impact of electrochemical hydrogen charging on the corrosion behavior and surface film of the Ti-6Al-4V alloy.The findings revealed the formation ofγ-TiH andδ-TiH_(2) hydrides in the alloy after hydrogen charging.Prolonging hydrogen charging resulted in more significant degradation of the alloy microstructure,leading to deteriorated protectiveness of the surface film.This trend was further confirmed by the electrochemical measurements,which showed that the corrosion resistance of the alloy progressively worsened as the hydrogen charging time was increased.Consequently,this work provides valuable insights into the mechanisms underlying the corrosion of Ti-6Al-4V alloy under hydrogen charging conditions. 展开更多

In order to better understand the specific substituent effects on the electrochemical oxidation process of β-O-4 bond, a series of methoxyphenyl type β-O-4 dimer model compounds with different localized methoxyl groups, including 2-(2-methoxyphenoxy)-1-phenylethanone, 2-(2-methoxyphenoxy)-1-phenylethanol, 2-(2-methoxyphenoxy)-1-(4-methoxyphenyl)ethanone, 2-(2-methoxyphenoxy)-1-(4-methoxyphenyl)ethanol, 2-(2,6-dimethoxyphenoxy)-1-(4-methoxyphenyl)ethanone, 2-(2,6-dimethoxyphenoxy)-1-(4-methoxyphenyl)ethanol have been selected and their electrochemical properties have been studied experimentally by cyclic voltammetry, and FT-IR spectroelectrochemistry. Combining with electrolysis products distribution analysis and density functional theory calculations, oxidation mechanisms of all six model dimers have been explored. In particular, a total effect from substituents of both para-methoxy(on the aryl ring closing to Cα) and Cα-OH on the oxidation mechanisms has been clearly observed, showing a significant selectivity on the Cα-Cβbond cleavage induced by electrochemical oxidations. 展开更多

For microelectronic devices,the on-chip microsupercapacitors with facile construction and high performance,are attracting researchers'prior consideration due to their high compatibility with modern microsystems.Herein,we proposed interchanging interdigital Au-/MnO_(2)/polyethylene dioxythiophene stacked microsupercapacitor based on a microfabrication process followed by successive electrochemical deposition.The stacked configuration of two pseudocapacitive active microelectrodes meritoriously leads to an enhanced contact area between MnO_(2)and the conductive and electroactive layer of polyethylene dioxythiophene,hence providing excellent electron transport and diffusion pathways of electrolyte ions,resulting in increased pseudocapacitance of MnO_(2)and polyethylene dioxythiophene.The stacked quasi-solid-state microsupercapacitors delivered the maximum specific capacitance of 43 mF cm^(-2)(211.9 F cm^(-3)),an energy density of 3.8μWh cm^(-2)(at a voltage window of 0.8 V)and 5.1μWh cm^(-2)(at a voltage window of 1.0 V)with excellent rate capability(96.6%at 2 mA cm^(-2))and cycling performance of 85.3%retention of initial capacitance after 10000 consecutive cycles at a current density of 5 mA cm^(-2),higher than those of ever reported polyethylene dioxythiophene and MnO_(2)-based planar microsupercapacitors.Benefiting from the favorable morphology,bilayer microsupercapacitor is utilized as a flexible humidity sensor with a response/relaxation time superior to those of some commercially available integrated microsensors.This strategy will be of significance in developing high-performance on-chip integrated microsupercapacitors/microsensors at low cost and environment-friendly routes. 展开更多

In the present study,we propose the synonymization of Kurixalus silvaenaias with Kurixalus qionglaiensis.This conclusion is based on morphological examination and phylogenetic analysis of a series of newly collected specimens,as well as the type series of both nomenclatures.The publication dates of the two species were determined according to the related articles of the International Code of Zoological Nomenclature.Of note,the ZooBank registrations for both species are invalid.Consequently,the publication of K.silvaenaias does not qualify as a published work and the correct publication date of K.qionglaiensis should be the date on which it was physically printed.Furthermore,based on the results presented in this study and its original description,the proposal of K.inexpectatus is deemed untenable and should,therefore,be considered a junior synonym of K.idiootocus.We further provide suggestions for the authors,editors,and publishers who are working on taxonomic publications,as well as for ZooBank to improve the registering service. 展开更多

A compound varifocal lens based on electromagnetic drive technology is designed and fabricated, where the polydimethylsiloxane(PDMS) film acts as a driving component, while the PDMS biconvex lens and the plane-concave lens form a coaxial compound lens system. The plane-concave lens equipped with driving coils is installed directly above the PDMS lens surrounded by the annular magnet. When different currents are applied, the annular magnet moves up and down, driving the PDMS film to undergo elastic deformation, and then resulting in longitudinal movement of the PDMS lens. The position change of the PDMS lens changes the focal length of the compound lens system. To verify the feasibility and practicability of this design, a prototype of our compound lens system is fabricated in experiment. Our proposed compound lens shows that its zoom ability reaches 9.28 mm when the current ranges from -0.20 A to 0.21 A. 展开更多
