Phytochemical composition of Caesalpinia crista extract as potential source for inhibiting cholinesterase and β-amyloid aggregation: Significance to Alzheimer's disease

Objective: To screen plant extract fractions and elucidate the components present in Caesalpinia crista(C. crista) leaves for cholinergic and anti-amyloidogenic activities for the treatment of Alzheimer's diseases. Methods: This work has been carried out to study the action of C. crista extracts from nonpolar to polar solvents toward inhibition of oxidative stress, cholinergic and amyloidosis. The antioxidant activity was studied using DPPH total antioxidant assay; cholinergic assay by Ellman's method and anti-amyloidogenic assay by thioflavin-T fluorescence and transmission electron microscopy. Results: The quantification of polyphenols was carried out following C. crista methanolic extract(CCMeOH) HPLC fingerprinting, along with LC-MS and elucidated by MS LAMPS database. GC-MS of CCMeOH was screened for potential moieties. In vitro experimental results showed that the CCMe OH was potential extract that exhibited active inhibition of antioxidant property, cholinergic enzymes acetylcholinesterase and butyrylcholinesterase. For anti-amyloidogenic evaluations, among all the extracts, the CCMe OH was found to have the potential toward inhibiting the oligomers, fibrillation of Aβ42 with good defibrillation of amyloid cascading properties. Conclusions: These results are also supported by the presence of polyphenols as the active ingredients. Multi-potent target drug therapy is a promising option in treating the Alzheimer's diseases. Methanolic extract of C. crista shows potential activity against cholinergic enzymes, Aβ42 aggregation with antioxidant activity. 展开更多

Heat energy generated during machining has been found to have a greater influence on determining the machinability of the materials.In this work,magnesium-based silicon carbide composite,which has been identified as a suitable lightweight application material,is prepared with the weight ratio of 90:10 by a stir casting process.Conventionally available HSS drill tools with different diameters of 4,6 and 8 mm are used to perform the drilling operations with governing parameters of spindle speed,feed rate and constant depth of cut.Thermal image camera of the FLIR E60 series is used to measure the temperature variation in the cutting zone at different operating conditions.The influences of machining temperature on chip morphology,tool wear and surface profile of the machined samples are investigated.Spindle speed has been found to have a significant effect on machining temperature.When spindle speed increases,the diameter of drill tool increases the tool wear and surface profile,respectively.Both abrasion and adhesive type of wears are observed in the drill tool.Further,change from abrasion to adhesive wear is noticed with the increase of the diameter of the drill tool.Surface plots are drawn with respect to the interaction of governing parameters along with the working temperatures obtained under different machining conditions. 展开更多

Nitrogen-doped titanium dioxide(N/TiO2)nanophotocatalysts were successfully synthesized in the presence of environmentally benign nitrogen dopant source,guanidinium chloride,by the sol-gel method.The effect of calcination temperature(300–600℃)on their physicochemical properties was investigated by means XRD,XPS,FESEM,HRTEM,Raman spectroscopy,UV-vis DRS,PL and BET.Moreover,their photocatalytic activities were evaluated against rhodamine B(RhB)degradation under direct sun light.Results showed that the crystal phase of spheroidal N/TiO2 nanoparticles was changed from anatase(300℃)to rutile(600℃)via an intermediate anatase/rutile(A/R)mixed phase(400–500℃),and the RhB photodegradation performance was increased with the decrease of the calcination temperature.Notably,N/TiO2 prepared at 400℃demonstrated the best degradation performance(99%)after 5 h irradiation.The enhanced performance with high photostability was mainly attributed to its higher surface area and pore volume,stronger light absorption,and lower recombination rate.Such nanomaterials have practical applications for environmental remediation. 展开更多
