One of the least selected contents by teachers of physical education (PE) in high school is the combat sport. Several authors have reported the neglect suffered by these activities in the Spanish school environment, stating a number of social prejudices and beliefs that affect their selection in school, e.g., they are aggressive and dangerous. Fencing is usually not included for several reasons; lack of knowledge about sport, its complexity, the cost of practice materials, lack of teaching materials, and the limited researches. This paper presents the results of a survey of 106 PE teachers of Valencia (Spain) in order to know what are the main causes that affect the non-selection of fencing as a sport in school, with emphasis on the lack of training and materials, both specific in fencing's learning. 展开更多

Purpose: The present study aimed to investigate the relationship between different types of sport expertise (externally-paced vs. self-paced sports) and vigilance performance in children by evaluating the cardiovascular fitness level of the participants. Methods: Three groups of children (11.0 ± 0.2 years) differentiated in terms of their regular sport participation (football players, n = 20; track and field athletes, n = 20; non-athletic controls, n = 20) took part in the study. In one session, participants performed the Leger Multi-stage fitness test to estimate their aerobic fitness level. In another session, participants completed the Psychomotor Vigilance Task (PVT) to evaluate their vigilance performance under 2 conditions of velocity demands (normal vs. speed). Results: The results revealed that both groups of sport practitioners had higher cardiovascular fitness than non-athlete controls. In contrast, no significant differences in the performance PVT were found between track and field athletes and controls. Crucially, football players showed better performance in the PVT than track and field athletes and controls. These between-group differences were not modulated by the speed demands of the task. Conclusion: The major novel finding of this research points to a positive relationship between sport participation and vigilance performance during childhood. We discuss our results in terms of the different hypotheses put forward in the literature to explain the relationship between regular exercise and cognitive fimctioning: the "cardiovascular fitness" and the "cognitive component skills" hypotheses.2018 Published by Elsevier B.V. on behalf of Shanghai University of Sport. Estimation of Partition Coefficients of Fe-S Protein Models: Se Substitutions in Fe4S4Cysn

The organic solvent-water partition coefficients P are calculated. For FeaS4Cysn, the organic solvent-water partition coefficients for l-octanol Po, cyclohexane Pch and chloroform Pcf decay 4.46, 6.25 and 4.60 log units per Cys, respectively. The Po is of the same order of magnitude as CDHI (conformationally dependent hydrophobic index) calculations and Pch-cf as computations performed with a method by Leo and Hansch. The logPo-ch-cf mean relative errors are 17%, 25% and-17%, which represent mean and unsigned relative errors of -3% and 20%. On varying the number of Cys, the structures show hydrophobic moments indicative of amphipathic compounds. For Se substitutions in Fe4SeaCys,, Po-ch-cf drop 4.52, 6.30 and 4.66 exponents per Cys. With the reference calculations, Po-ch-cf decay 4.03, 4.80 and 5.76 exponents per Cys. For FeaS4 mSemCys4, the calculated similar partition coefficients and hydrophobic moments, and lower distance in dendrogram suggest a role of Fe-Se clusters in physiological processes. The analysis is extended to lysozyme and its secondary structures. 展开更多

The purpose is to extend the studies to anionic lipid bilayers. Electrostatics is studied in cationic protein adsorption to zwitterionic PC and anionic mixed PC/PG SUVs. Protein adsorption is investigated in SUVs along with changes of fluorescence emission spectra. Partition coefficients and cooperativity parameters are calculated. At pl binding obtains maximum while at lower or higher pHs binding decreases. In Gouy-Chapman formalism activity coefficient goes with square charge, which deviations indicate asymmetric location of anionic phospholipid in the inner leaflet, in mixed SUVs for lysozyme- and myoglobin-PC/PG systems, in agreement with experiments and molecular dynamics simulations. Vesicles bind myoglobin anti-cooperatively while lysozyme-BSA cooperativitivey. A model is proposed for both, which composes two protein sub-layers with different structures and properties. Hill coefficient reflects subunit cooperativity of bi and tridomain proteins. 展开更多

Carbamoyl phosphate synthetase i (CPS1) deficiency (CPS1D) is an inborn error of the urea cycle having autosomal (2q34) recessive inheritance that can cause hyperammonemia and neonatal death or mental retardation. We analyzed the effects on CPS1 activity, kinetic parameters and enzyme stability of missense mutations reported in patients with CPS1 deficiency that map in the 20-kDa C-terminal domain of the enzyme. This domain turns on or off the enzyme depending on whether the essential allosteric activator of CPS 1, N-acetyl- L-glutamate (NAG), is bound or is not bound to it. To carry out the present studies, we exploited a novel system that allows the expression in vitro and the purification of human CPS1, thus permitting site-directed mutagenesis. These studies have clarified disease causation by individual mutations, identifying functionally important residues, and revealing that a number of mutations decrease the affinity of the enzyme for NAG. Patients with NAG affinity-decreasing mutations might benefit from NAG site saturation therapy with N-carbamyl-L- glutamate (a registered drug, the analog of NAG). Our results, together with additional present and prior site-directed mutagenesis data for other residues mapping in this domain, suggest an NAG-triggered conformational change in the 134-~4 loop of the C-terminal domain of this enzyme. This change might be an early event in the NAG activation process. Molecular dynamics simulations that were restrained according to the observed effects of the mutations are consistent with this hypothesis, providing further backing for this structurally plausible signaling mechanism by which NAG could trigger urea cycle activation via CPS1. 展开更多

The study of sperm used: transmission electron microscopy (both patients); apoptotic markers, DNA fragmentation test and fluorescence in-situ hybridization (patient 1) and immunoeytochemistry study of sperm flagellum using anti-β tubulin antibodies and ciliary activity test (patient 2).Results Increased DNA fragmentation (52.6%) and apoptosis biomarkers were detected in patient 1, and loss of the central pair of mierotubules in patient 2 (‘9+0' axoneme); the nasal ciliary activity was normal. Conclusion Results suggest an apoptotic origin of the abnormalities in the sperm from patient 1 and dysplasia of the fibrous sheath in patient 2. 展开更多
