瞄准:调查氮的氧化物的角色(没有) 在像使用费的受体 2 (TLR2 ) 在尖锐出血性的引起坏死胰腺炎(AHNP ) 的肝的 /4mRNA 表示老鼠。方法:110 只 SD 雄的老鼠随机被划分成假冒操作组(n=10 ) , AHNP 组(n=30 ) ,氯奎(CQ ) 对待组(n=30 )... 瞄准:调查氮的氧化物的角色(没有) 在像使用费的受体 2 (TLR2 ) 在尖锐出血性的引起坏死胰腺炎(AHNP ) 的肝的 /4mRNA 表示老鼠。方法:110 只 SD 雄的老鼠随机被划分成假冒操作组(n=10 ) , AHNP 组(n=30 ) ,氯奎(CQ ) 对待组(n=30 ) 和 L-Arg-treated 组(n=40 ) 。在 AHNP 老鼠的肝的 TLR2/4mRNA 表示被 RT-PCR 测量。结果:TLR2/4mRNA 的表示能在假冒操作组(0.155E-5+/-0.230E-6 和 0.115E-2+/-0.545E-4 ) 在 AHNP 老鼠的肝被检测,但是显著地在在 12 h 达到顶点的 AHNP 组(0.197E-2+/-0.114E-3 和 0.175+/-0.349E-2 ) 在 3 h 被增加(0.294E-2+/-0.998E-4 和 2.673+/-2.795E-2, P【0.01 ) 。肝的损害被加重,在肝的 TNF-alpha 集中被增加,没有集中被减少(P【0.05 或 P【0.01 ) 。当 TLR2/4mRNA 表示被 CQ 禁止时(3 h:1.037E-4+/-3.299E-6 和 0.026+/-3.462E-3;6 h:1.884E-4+/-4.679E-6 和 0.108+/-6.115E-3;12 h:2.443E-4+/-7.714E-6 和 0.348+/-6.807E-3;P【0.01 ) ,肝的损害被减轻,在肝的集中都没被增加, TNF-alpha 集中被减少(P【0.05 或 P【0.01 ) 。当有 AHNP 的老鼠与 L-Arg 被对待时,在肝的 TLR2/4mRNA 表示能有效地被禁止(50 mg-T:0.232E-2+/-0.532E-4 和 0.230+/-6.883E-3;100 mg-T:0.210E-2+/-1.691E-4 和 0.187+/-0.849E-2;200 mg-T:0.163E-2+/-0.404E-4 和 0.107+/-0.195E-2;400 mg-T:0.100E-2+/-0.317E-4 和 0.084+/-0.552E-2;P【0.01 ) 并且肝的损害被减轻。同时,在肝的集中都显著地没被增加, TNF-alpha 集中被减少(P【0.05 或 P【0.01 ) 。结论:TLR2/4mRNA 的表示被增加,肝的损害在 AHNP 老鼠的肝被加重。在 AHNP 老鼠的肝的 TLR2/4mRNA 基因表示能显著地被禁止由没有,导致肝的损害的消除。 展开更多

Chloroquine Relieves Acute Lung Injury in Rats with Acute Hemorrhagic Necrotizing Pancreatitis 被引量:5

Summary: This study preliminarily investigated the mechanism by which chloroquine (CQ) relieves acute lung injury (ALI) complicated in acute hemorrhagic necrotizing pancreatitis (AHNP). Sixty male Wistar rats were randomized into sham-operated group (group A, n=10), AHNP group (group B, n=10), L-arginine-treated group (group C, n=10), L-N-nitro-L-arginine methyl ester (NAME)-treated group (group D, n=10), CQ-treated group (group E, n=10) and CQ+L-NAME-treated group (group F, n=10). TLR4 expression was measured by using real time-PCR and Western blotting respectively. The results showed that, in the group B, the expression of TLR4 and the levels of TNF-α and IL-6 in the lungs were significantly increased, and the nitric oxide (NO) concentration was reduced, as compared with those in the group A (P〈0.05 or P〈0.01). Lung injury was aggravated with the increased expression of TLR4. When the inhibitor and stimulator of TLR4, namely L-Arg and L-NAME, were added respectively, lung injury was correspondingly relieved or aggravated (P〈0.05 or P〈0.01). In the group E, TLR4 expres- sion was substantially lower and NO concentration higher than those in the group B (P〈0.05 or P〈0.01). However, in the group F, NO concentration was markedly decreased, and the inhibitory effect of CQ on TLR4 expression and the relief of lung injury were weakened when compared with those in the group E (P〈0.05 or P〈0.01). It was concluded that TLR4 may play an important role in the pathogenesis and development of ALl complicated in AHNP. CQ could relieve ALl by decreasing the TLR4 expression and increasing the NO release. 展开更多

Relationship between the Apoptosis and Notch-1 Expression in Acute Pancreatitis 被引量:3

In order to investigate the expression of Notch-1 in rats with acute pancreatitis (AP) and its relation with apoptosis of pancreatic cells, mild AP (MAP) and severe AP (SAP) models were established by retrograde injection of different concentrations of sodium taurocholae into pancreatic duct. The apoptosis index and the expression of Notch-1 protein and mRNA were detected by using TUNEL, Western blot and real-time PCR in MAP and SAP at different time intervals. The results showed that in MAP group the apoptosis index was significantly elevated during 4 to 24 h after induction of pancreatitis, but there was no significant difference among different time intervals. In SAP group, the apoptosis index reached the peak at 4th h after induction of pancreatitis, then gradually declined. There was significant difference in apoptosis index between MAP and SAP groups. Starting from 4th after induction of pancreatitis, the expression of Notch-1 in both MAP and SAP group was increased at different time intervals, but that in SAP group was significantly higher than in MAP group (P〈0.05). The expression of Notch in MAP and SAP groups reached the peak at 12th and 8th h respectively after induction of pancreatitis. It was concluded that there was significant difference in apoptosis index and the Notch-1 expression in different types of AE The overexpression of Notch-1 could aggravate AP by inhibiting the apoptosis of pancreatic cells. 展开更多
