In order to understand the yield performance and nitrogen (N) response of hybrid rice under different ecological conditions in southern China, field experiments were conducted in Huaiji County of Guangdong Province, Binyang of Guangxi Zhuang Autonomous Region and Changsha City of Hunan Province, southern China in 2011 and 2012. Two hybrid (Liangyoupeijiu and Y-liangyou 1) and two inbred rice cultivars (Yuxiangyouzhan and Huanghuazhan) were grown under three N treatments (N1,225 kg ha-l; N2, 112.5-176 kg ha-l; N3, 0 kg ha-1) in each location. Results showed that grain yield was higher in Changsha than in Huaiji and Binyang for both hybrid and inbred cultivars. The higher grain yield in Changsha was attribut- ed to larger panicle size (spikelets per panicle) and higher biomass production. Consistently higher grain yield in hybrid than in inbred cultivars was observed in Changsha but not in Huaiji and Binyang. Higher grain weight and higher biomass production were responsible for the higher grain yield in hybrid than in inbred cultivars in Changsha. The better crop perfor- mance of rice (especially hybrid cultivars) in Changsha was associated with its temperature conditions and indigenous soil N. N2 had higher internal N use efficiency, recovery efficiency of applied N, agronomic N use efficiency, and partial factor productivity of applied N than N1 for both hybrid and inbred cultivars, while the difference in grain yield between N1 and N2 was relatively small. Our study suggests that whether hybrid rice can outyield inbred rice to some extent depends on the ecological conditions, and N use efficiency can be increased by using improved nitrogen management such as site-specific N management in both hybrid and inbred rice production. 展开更多

Polyethylene glycol (PEG)-mediated DNA transformation for transient gene expression in protoplasts and Agrobacterium tumefaciens-mediated transformation in lower epidermis of leaves are readily available in several plant species. In the study, these two versatile tools were used in rapeseed. A simple and efficient method was established for isolating protoplasts from rapeseed cotyledons and leaves, and found that cotyledons might be better than true leaves. Transient expression analysis showed that yellow fluorescent protein (YFP) and luciferase (LUC) could be expressed in rapeseed protoplasts. Moreover,GUS histochemical assays indicated that Agrobacterium-mediated DNA transient expression was achievable only in lower epidermis of rapeseed cotyledons and expression signal was the highest on the 5th day after injection with the bacterial suspension (OD600=0.8).These methods might provide valuable tools for rapid functional gene analysis in rapeseed. 展开更多

Traits such as grain shape, panicle length and seed shattering, play important roles in grain yield and harvest. In this study, the cloning and functional analysis of PANICLE TRAITS 2 (PT2), a novel gene from the Indica rice Chuandali (CDL), is reported. PT2 is synonymous with Growth-Regulating Factor 4 (OsGRF4), which encodes a growth-regulating factor that positively regulates grain shape and panicle length and negatively regulates seed shattering. Higher expression of OsGRF4 is correlated with larger grain, longer panicle and lower seed shattering. A unique OsGRF4 mutation, which occurs at the OsmiRNA396 target site of OsGRF4, seems to be associated with high levels of OsGRF4 expression, and results in phenotypic difference. Further research showed that OsGRF4 regulated two cytokinin dehydrogenase precursor genes (CKX5 and CKX1) resulting in increased cytokinin levels, which might affect the panicle traits. High storage capacity and moderate seed shattering of OsGRF4 may be useful in high-yield breeding and mechanized harvesting of rice. Our findings provide additional insight into the molecular basis of panicle growth. 展开更多

Glucosinolates are amino acid-derived secondary metabolites that act as chemical defense agents against pests.However,the presence of high levels of glucosinolates severely diminishes the nutritional value of seed meals made from rapeseed(Brassica napus L.).To identify the loci affecting seed glucosinolate cont ent(SGC),we con ducted genome-wide resequencing in a population of 307 diverse B.napus accessions from the three B.napus ecotype groups,namely,spring,winter,and semi-winter.These resequencing data were used for a genome-wide association study(GWAS)to identify the loci affecting SGC.In the three ecotype groups,four comm on and four ecotype-specific haplotype blocks(HBs)were significantly associated with SGC.To identify candidate genes controlling SGC,transcriptome analysis was carried out in 36 accessions showing extreme SGC values.Analyses of haplotypes,genomic variation,and candidate gene expression pointed to five and three candidate genes in the common and spring group-specific HBs,respectively.Our expression analyses dem on strated that additive effects of the three candidate genes in the spring group-specific HB play important roles in the SGC of B.napus. 展开更多
