The first step of fermentation was done using Saccharomyces cerevisae KVL013 for 7 days. The second step, an acetic fermentation was realized using two acetic acid bacteria: A. tropicalis CRSBAN-BVA1 and A. tropicalis CRSBAN-BVK2 for 21 days. Several parameters of the vinegar produced such as physico-chemical and sensory properties were determined. Microbial density during each step was monitored. Results showed that pH, alcoholic and acetic acid contents of vinegar were respectively 2.97%, 7% and 4.54% g/ml respectively by using A. tropicalis CRSBAN-BVA1 and 3.02%, 7% and 4.32% g/ml with A. tropicalis CRSBAN-BVK2. Sensory evaluation revealed that the vinegar was acceptable to the panellists. Results of microbial density showed that the maximum concentration of cell biomass produced was 4.32 × 10<sup>8</sup> and 4.25 × 10<sup>8</sup> CFU/ml respectively for CRSBAN-BVA1 and CRSBAN-BVK2. 展开更多

Background: The assessment of iron status using a single biomarker of iron metabolism is not enough sensitive and specific to reliably diagnose iron deficiency associated with multiple comorbidities. The objective of this study was to describe the iron status of people living with HIV in sub-Saharan Africa using a multi-criteria approach based on the determination of blood ferritin, sTfR, CRP and the calculation of sTfR-F index. Methods: This study was conducted using a retrospective panel of 933 sera/plasmas. We determined serum ferritin concentration, serum sTfR concentration, and C-reactive protein (CRP) by immunoturbidimetry for each subject. The sTfR-F index was determined by calculating the sTfR/log ferritin ratio. The statistical test used was Chi<sup>2</sup>. Results: Regardless of the inflammatory syndrome, we determined 3.80%, 30.29%, and 42.70% iron deficiency based on the separate interpretation of ferritin concentration, sTfR, and sTfR-F calculation, respectively. We used those biomarkers in addition to CRP in an algorithm for the diagnosis of iron deficiency. Subjects without inflammatory syndrome, had iron deficiency of 2.89% (n = 26). Taking into account the presence of an inflammatory syndrome, the frequency obtained was n = 88 (9.78%). Overall, iron deficiency was diagnosed in 114 (12.67%) patients when we used the diagnostic algorithm. Conclusion: The use of diagnostic algorithms combining several biomarkers of iron metabolism and taking into account the presence or absence of an inflammatory syndrome is a good approach to detect a large number of iron deficiencies in a population. Therefore, an assessment of the effectiveness of different diagnostic algorithms is necessary. 展开更多

Over the past three decades, genomic and epigenetic sciences have identified more than 70 genes involved in the molecular pathophysiology of Alzheimer’s disease (AD). DNA methylation, abnormal histone and chromatin regulation and the action of various miRNAs induce AD. The identification of mutated genes has paved the way for the development of diagnostic kits and the initiation of gene therapy trials. However, despite major advances in neuroscience research, there is yet no suitable treatment for AD. Therefore, the early diagnosis of this neurodegenerative disease raises several ethical questions, including the balance between the principle of non-maleficence and the principle of beneficence. The aims of this research were to present the genomic and ethical aspects of AD, and to highlight the ethical principles involved in its presymptomatic diagnosis and therapy. A systematic review of the literature in PubMed, Google Scholar and Science Direct was carried out to outline the genomic aspects and ethical principles relating not only to the presymptomatic diagnosis of AD, but also to its gene therapy. A total of 16 publications were selected. AD is a multifactorial disease that can be genetically classified into Sporadic Alzheimer’s Disease and Familial Alzheimer’s Disease based on family history. Gene therapy targeting specific disease-causing genes is a promising therapeutic strategy. Advancements in artificial intelligence applications may enable the prediction of AD onset several years in advance. While early diagnosis of AD may empower patients with full decision competence for early decision-making, it also carries implications for the patient’s family members, who are at risk of developing the disease, potentially becoming a source of confusion or anxiety. AD has a significant impact on the life of individuals at risk and their families. Given the absence of disease modifying therapy, genetic screening and early diagnosis for this condition raise ethical issues that must be carefully considered in the context of fundamental bioethical principles, including autonomy, beneficence, non-maleficence, and justice. 展开更多

Excessive use of mineral fertilizers in maize farming negatively affects farmers’ income and impacts long-term soil health. This study aims to appreciate the effectiveness of biostimulant based on native Glomeraceae arbuscular mycorrhizal fungi on the production and uptake of phosphorus, nitrogen and potassium of maize (Zea mays L.) plants in central Benin. The trials were set up in a farming environment with thirty-four producers. The experimental design was composed of three treatments installed at 34 producers. Three growth parameters were evaluated on 60 ème days after sowing. Grain yield, nutritional status of maize plants and mycorrhization parameters were determined at harvest. The results showed that the Glomeraceae + 50% NPK (NPK: azote-phosphore-potassium)_Urea treatment improved the height, the crown diameter and the leaf area by 17.85%, 21.79% and 28.32% compared to the absolute control and by 0.41%, 1.11% and 1.46% compared to the 100% NPK_Urea treatment, respectively. Similarly, grain yield improved by 45.87% with the use of Glomeraceae + 50% NPK_Urea compared to the absolute control and by 3.96% compared to the 100% NPK_Urea treatment. The Glomeraceae + 50% NPK_Urea significantly improved the phosphorus and potassium uptake of maize plants. With respect to nitrogen uptake, no statistical difference was observed between treatments. The mycorrhizae strains used improved root infection in the maize plants. We recorded 66% frequency and 40.5% intensity of mycorrhization. The biostimulant based on indigenous Glomeraceae combined with 50% NPK_Urea can be used as a strategy to restore soil health and improve maize productivity in Benin. 展开更多

Most cases remain asymptomatic. The majority of patients have mild symptoms while about 15% develop a severe form. The clinical spectrum of SARS-CoV-2 infection appears broad, encompassing asymptomatic infection, upper respiratory tract symptoms, and severe viral pneumonia with respiratory failure that can lead to death. Laboratory tests play an important role in the management of COVID-19 patients. In addition to being essential for the diagnosis, several biological analyses make it possible to identify the inflammatory processes and the potential complications of this disease. This study attempted to identify biochemical assays that could help in the prognosis of the disease to ensure early management. Methods: This was a descriptive study. It focused on patients hospitalized for COVID-19 from March 19, 2020, to January 26, 2021, at the Infectious Disease Management Centre in Lomé (Togo). Medians were compared using the (Mann-Whitney and Wilcoxon) test and frequencies were compared using the Chi-square test or Fisher’s exact test. Results: We included 782 patients. The median age was 41 years IQR from 32 to 55. We observed several biochemical abnormalities in varying proportions for all biochemical parameters studied. Compared to non-serious patients, critically ill patients at admission had more frequently elevated urea, creatinine, transaminases, TG, GGT, CRP and blood glucose. Also, they had more frequent decreases in total cholesterol, HDL-c, blood chloride, and blood calcium. As for patients who died during hospitalization, compared with healed patients, they had more frequent elevations of urea, creatinine, AST, ALT, GGT. CRP and blood glucose. They also had a more frequent decrease in total cholesterol, HDL-c, blood chloride, blood calcium, and blood glucose (p = 0.025). Conclusion: This study shows that COVID-19 is a multi-organ systemic inflammatory viral disease that should be systematically investigated once the diagnosis is confirmed. 展开更多

The wild type maize genotype, B73, is not amenable for callus production and an efficient protocol for B73 maize callus induction has never been reported up-to-date. Scientific efforts in producing B73 maize callus using all known callus inducible media have been unsatisfactory. Here we developed and described an efficient protocol for callus induction from B73 maize seedlings. The protocol is based on well known callus inducible media CM4C where we have sequentially subtracted some chemical compounds and added some new compounds mediating cell proliferations. This newly described protocol was able to induce callus production in a wide range of crop species including rice and soybean. We found that cell proliferation factors, NAA (auxin analog) and 2,4 D (auxin influx carrier) were not only very crucial but required for positive B73 maize callus induction. The absence of one or the other will lead to the failure of B73 maize callus production. The well known CM4C callus induction composition lacks NAA. Our findings will advance genetic studies of maize mutants generated from B73 genotype background. 展开更多

Our study aimed at assessing the effects of 3 Plants Growth Promoting Rhizobacteria (PGPR) either singly or in combination on maize growth under laboratory and greenhouse conditions. Seeds were inoculated with single and combined solution of 108 CFU/ml of Rhizobacteria. Seeds were not inoculated for the control variant. The highest germination percentage was obtained with the combination of Pseudomonas fluorescens and Pseudomonas putida. This combination also recorded the best vigor index, plants circumferences number of leaves and the leaf area. The maximal heights of plants were observed with seeds treated with Azospirillum lipoferum with an increase of 37.32%. The highest rates of underground dry matter were recorded with A. lipoferum, with an increase of more than 56% comparative to control, while the combination P. fluorescens and P. putida increased the aerial dry matter of 59.11%. Finally, the highest value of the aerial biomass was obtained with the plants treated with the combination of P. fluorescens and P. putida and the highest underground biomass was obtained with plants treated only with A. lipoferum. These results suggest that specific combinations of PGPR can be considered as efficient alternative biofertilizers to promote maize seed germination, biomass and crop yield. 展开更多

The ichnovirus TrIV, transmitted by the endoparasitic wasp Tranosema rostrale to its lepidopteran host during oviposition, replicates asymptomatically in wasp ovaries and causes physiological dysfunctions in parasitized caterpillars. The need to identify ichnoviral genes responsible for disturbances induced in lepidopteran hosts has provided the impetus for the sequencing and annotation of ichnovirus genomes, including that of TrIV. In the latter, 86 putative genes were identified, including 35 that could be assigned to recognized ichnoviral gene families. With the aim of assessing the relative importance of each TrIV gene, as inferred from its level of expression, and evaluating the accuracy of the gene predictions made during genome annotation, the present study builds on an earlier qPCR quantification of transcript abundance of TrIV rep ORFs, in both lepidopteran and wasp hosts, extending it to other gene families as well as to a sample of unassigned ORFs. We show that the majority (91%) of putative ORFs assigned to known gene families are expressed in infected larvae, while this proportion is lower (67%) for a sample taken among the remaining ORFs. Selected members of the TrV and rep gene families are shown to be transcribed in infected larvae at much higher levels than genes from any other TrIV gene family, pointing to their likely involvement in host subjugation. In wasp ovaries, the transcriptional profile is dominated by a rep gene and a member of a newly described gene family encoding secreted proteins displaying a novel cysteine motif, which we identified among previously unassigned ORFs. 展开更多

The effect of the ethanolic extract Khaya senegalensis on some biochemical parameters was tested in rabbits with normal glucose and glucose overload. Phytochemical screening after three batches of 6 rabbits race (TBD) were treated with the extract in oral doses of respectively 10, 25 and 50 mg/kg body weight for 30 days;three other groups receiving respectively these doses of the extract were placed under a priori temporary overload of glucose;a control group received instead distilled water treatment. Blood glucose, triglycerides and total cholesterol were measured by spectrophotometry and weight gain of the animals was performed at the beginning of the experiment and every 2 weeks during the study period. The phytochemical screening revealed the presence of polyphenolic compounds (majority) of saponins of anthracene derivatives and steroids in the extract. The test results indicate that under the dose of 50 mg/kg body weight (BW) there was a significant decrease (p < 0.05) in blood glucose after 24 hours. The extract had no effect on the lipid profile or body weight of rabbits. 展开更多

Many enzymes which catalyze the conversion of large substrates are made of several structural domains belonging to the same polypeptide chain. Transfer RNA (tRNA), one of the substrates of the multidomain aminoacyl-tRNA synthetases (aaRS), is an L-shaped molecule whose size in one dimension is similar to that of its cognate aaRS. Crystallographic structures of aaRS/tRNA complexes show that these enzymes use several of their structural domains to interact with their cognate tRNA. This mini review discusses first some aspects of the evolution and of the flexibility of the pentadomain bacterial glutamyl-tRNA synthetase (GluRS) revealed by kinetic and interaction studies of complementary truncated forms, and then illustrates how stable analogues of aminoacyl-AMP intermediates have been used to probe conformational changes in the active sites of Escherichia coli GluRS and of the nondiscriminating aspartyl-tRNA synthetase (ND-AspRS) of Pseudomonas aeruginosa. 展开更多

The better knowledge of a plant genetic biodiversity is based on a prior study of its agro-morphological characteristics. The objective of this study was to investigate the morphological diversity of maize accessions and its structure on the basis of 14 variables. For the experimentation, 43 and 98 maize accessions were planted in three replicates respectively in central and northern Benin following an incomplete randomized block. The mixed model analysis of two factors variance revealed a significant difference for all accessions considering each agro-morphological characteristic evaluated except the germination day of two areas seed accessions. The numerical classification grouped the accessions into four groups in each zone. The stepwise discriminant analysis showed that early characters related to plant height and ear insertion were the variables that discriminated accessions in both zones. Maturity and recovery ears, sensitivity to streak and the germination days are the variables that discriminate accessions of the two areas. These results provide a database for the creation of improved maize varieties that meet the needs of 展开更多

Does Haptoglobin Phenotype Impact Infection Mortality?

Introduction: The physiological status of a subject and the pathophysiology in some diseases might be under the influence of haptoglobin phenotype. The objective of this work was to determine the relationship between mortality from HIV/AIDS infection and haptoglobin phenotype in a black population in C&#244;te d’Ivoire. Methods: The study was conducted from a retrospective panel of 933 sera/plasma from the previous workup of the ANRS 12136 TEMPRANO trial at month 0 of patients in deferred-ART arms. For each subject, we determined the serum haptoglobin concentration, haptoglobin phenotype, and other variables from patient files from the TEMPRANO trial database. Statistical tests used were Chi-2, Fischer, and Kruskal-Wallis tests for non-gaussian distribution. We used the Kaplan-Meier method for survival analysis. Results: The distributions of the haptoglobin phenotypes were 32.3% for Hp 1-1, 39.5% for Hp 2-1 and 27.2% for Hp 2-2. The blood haptoglobin concentration seemed to be associated with haptoglobin phenotypes (p-value > 5%). The survival rate at M30 and for an extended follow-up up to 6 years was independent of haptoglobin phenotype (p-value > 5%). Besides, the haptoglobin phenotypes do not appear to be associated with CD4+ T-cell count and with hemoglobin concentration. Conclusion: Haptoglobin phenotype seems to not impact the mortality of HIV/AIDS infection. However, given the antioxidant and immunomodulatory properties of some haptoglobin phenotypes, it would be relevant to seek out possible confounding factors indirectly associated with haptoglobin phenotypes and clinical or biological infection variables. 展开更多

Malnutrition is still a widespread public health problem worldwide. The objective of this review is to identify the most effective nutrition programs used in the prevention and management of child malnutrition in children under five years. Also, it is to suggest courses of action to improve interventions in the fight against malnutrition. The promotion of exclusive breastfeeding and food diversification improves the mothers’ knowledge and contributes to prevention of malnutrition. Food supplementation increases the weight/height ratio of malnourished children. The two programs combined bring better results on mothers’ feeding practices and children’s anthropometric data. This review has highlighted the importance of food supplementation and nutritional education in the management and prevention of child malnutrition. In addition, the combination of several approaches to good dietary practices brings better results. Maize (Zea mays L.) is the major cereal cultivated in Benin and it is important to know its genetic diversity to improve the yield. The genetic markers of important traits are evaluated in order to improve the maize inbred lines. The aim of this study was to evaluate the genetic diversity of Benin’s maize accessions by SSR marker. Thus, one hundred eighty seven maize accessions from three areas (South, Center and North) were analyzed using three SSR markers. A total of 227 polymorphic bands were produced and showed high genetic diversity (Shannon index = 0.51). The polymorphic information content (PIC) values for the SSR loci ranged from 0.58 to 0.81, with an average of 0.71. Genetic distance-based UPGMA dendrogram showed a genetic differentiation between accessions and they were grouped into four clusters in each area. This work provides necessary information that can be used not only to improve the maize production and conservation but also to better manage genetic species resources in Benin. 展开更多

The aim of the study was to investigate the effects of five plant growth-promoting rhizobacteria (PGPR) (Bacillus panthothenicus;Pseudomonas Cichorii;Pseudomonas Putida;Pseudomonas syringae and Serratia marcescens) on the growth and yield of maize on a ferruginous soil under field condition. Maize seeds were inoculated with 10 ml of bacterial suspension. Study was conducted in a completely randomized design with fifteen treatments and three replicates. A half-dose of recommended (13, 17, 17 kg?ha?1) NPK was applied 15 days after emergence. The results show that the Serratia marcescens + 50% NPK treatment yielded the best results for height, fresh underground biomass, dry aboveground biomass, dry underground biomass, and grain yield with respective increases of 41.09%, 217.5%, 213.34%, 93.82%, and 39.05% compared to the control. Maximum stem diameter (increases of 49.65%) was recorded in the plants treated with 100% NPK (full dose NPK) while the highest leaf area (466.36 ± 9.57 cm2), obtained on plant treated with Pseudomonas putida + 50% NPK was 32.08% greater than in the non-inoculated control. Our results suggest the use of these rhizobacteria as biological fertilizers for enhancing the growth and maize seed yield in ferruginous soil in the North of Benin. 展开更多

Objective: The study aims to investigate some physical and technological parameters of varieties of maize collected from the most representative areas of maize production in the southern part of Benin, and mainly used by the populations of those areas. Method: The chemical composition, physical and technological characteristics of the grains were determined using standard methods. Results: The results showed that the weight of 1000 grains ranges from 158.01 to 305.12 g, the length from 0.79 to 1.81 cm, the average width from 0.74 to 1.51 cm, thickness from 0.36 to 0.85 cm and density from 1.07 to 1.25. Regarding the technological parameters, the hardness of the grains before and after cooking was between 16.78 and 47.20 kgf, and 0.23 and 0.71 kgf respectively, while the cooking time varies from 71.00 and 158.33 mn. There was a positive and significant correlation (r = 0.405;p < 0.026) between the hardness after cooking and the weight of 1000 grains, between the hardness after cooking and grain width (r = 0.460;p < 0.011). A positive and highly significant correlation (r = 0.527, p < 0.003) was also observed between the hardness after cooking and the grain length, between the hardness after cooking and the thickness of grain (r = 0.476, p < 0.008), and between the grain density and the weight of 1000 grains (r = 0.481, p < 0.007). A positive and highly significant correlation (r = 0.923, p < 0.000) was also observed between maize grain length and width, and between grain length and grain thickness (r = 0.764, p < 0.000) and between the width and thickness (r = 0.764, p < 0.000). There is growing concern in Sub-Saharan Africa about the spread of the Ebola virus disease(EVD),formerly known as Ebola haemorrhagic fever,and the public health burden that it ensues.Since 1976,there have been 885,343 suspected and laboratory confirmed cases of EVD and the disease has claimed 2,512 cases and 932 fatality in West Africa.There are certain requirements that must be met when responding to EVD outbreaks and this process could incur certain challenges.For the purposes of this paper,five have been identified:(i)the deficiency in the development and implementation of surveillance response systems against Ebola and others infectious disease outbreaks in Africa;(ii)the lack of education and knowledge resulting in an EVD outbreak triggering panic,anxiety,psychosocial trauma,isolation and dignity impounding,stigmatisation,community ostracism and resistance to associated socio-ecological and public health consequences;(iii)limited financial resources,human technical capacity and weak community and national health system operational plans for prevention and control responses,practices and management;(iv)inadequate leadership and coordination;and(v)the lack of development of new strategies,tools and approaches,such as improved diagnostics and novel therapies including vaccines which can assist in preventing,controlling and containing Ebola outbreaks as well as the spread of the disease.Hence,there is an urgent need to develop and implement an active early warning alert and surveillance response system for outbreak response and control of emerging infectious diseases.Understanding the unending risks of transmission dynamics and resurgence is essential in implementing rapid effective response interventions tailored to specific local settings and contexts.Therefore,the following actions are recommended:(i)national and regional inter-sectorial and trans-disciplinary surveillance response systems that include early warnings,as well as critical human resources development,must be quickly adopted by allied ministries and organisations in African countries in epidemic and pandemic responses;(ii)harnessing all stakeholders commitment and advocacy in sustained funding,collaboration,communication and networking including community participation to enhance a coordinated responses,as well as tracking and prompt case management to combat challenges;(iii)more research and development in new drug discovery and vaccines;and(iv)understanding the involvement of global health to promote the establishment of public health surveillance response systems with functions of early warning,as well as monitoring and evaluation in upholding research-action programmes and innovative interventions. 展开更多

The number of surveillance networks for infectious disease diagnosis and response has been growing.In 2000,the World Health Organization(WHO)established the Global Outbreak Alert and Response Network,which has been endorsed by each of the 46 WHO African members since then.Yet,taming the dynamics and plague of the vicious Ebola virus disease(EVD)in African countries has been patchy and erratic due to inadequate surveillance and contact tracing,community defiance and resistance,a lack of detection and response systems,meager/weak knowledge and information on the disease,inadequacies in protective materials protocols,contact tracing nightmare and differing priorities at various levels of the public health system.Despite the widespread acceptance of syndromic surveillance(SS)systems,their ability to provide early warning alerts and notifications of outbreaks is still unverified.Information is often too limited for any outbreak,or emerging or otherwise unexpected disease,to be recognized at either the community or the national level.Indeed,little is known about the role and the interactions between the Ebola infection and exposure to other syndemics and the development of acquired immunity,asymptomatic reservoir,and Ebola seroconversion.Can lessons be learnt from smallpox,polio,and influenza immunity,and can immunization against these serve as a guide?In most endemic countries,community health centers and disease control and prevention at airports solely relies on passive routine immunization control and reactive syndromic response.The frontline and airport Ebola SS systems in West Africa have shown deficiencies in terms of responding with an alarming number of case fatalities,and suggest that more detailed insights into Ebola,and proactive actions,are needed.The quest for effective early indicators(EEE)in shifting the public and global health paradigm requires the development and implementation of a comprehensive and effective community or regional integrated pandemic preparedness and surveillance response systems tailored to local contexts.These systems must have mechanisms for early identification,rapid contact tracing and tracking,confirmation,and communication with the local population and the global community,and must endeavor to respond in a timely manner. 展开更多

Climate change is generating range shifts in many organisms,notably along the elevational gradient in mountainous environments.However,moving up in elevation exposes organisms to lower oxygen availability,which may reduce the successful reproduction and development of oviparous organisms.To test this possibility in an upwardcolonizing species,we artificially incubated developing embryos of the viperine snake(Natrix maura)using a split-clutch design,in conditions of extreme high elevation(hypoxia at 2877 m above sea level;72%sea-level equivalent O2 availability)or low elevation(control group;i.e.normoxia at 436 m above sea level).Hatching success did not differ between the two treatments.Embryos developing at extreme high elevation had higher heart rates and hatched earlier,resulting in hatchlings that were smaller in body size and slower swimmers compared to their siblings incubated at lower elevation.Furthermore,post-hatching reciprocal transplant of juveniles showed that snakes which developed at extreme high elevation,when transferred back to low elevation,did not recover full performance compared to their siblings from the low elevation incubation treatment.These results suggest that incubation at extreme high elevation,including the effects of hypoxia,will not prevent oviparous ectotherms from producing viable young,but may pose significant physiological challenges on developing offspring in ovo.These early-life performance limitations imposed by extreme high elevation could have negative consequences on adult phenotypes,including on fitness-related traits. 展开更多
