To investigate the distribution of hepatitis B virus (HBV) genotypes and subgenotypes among the Bai nationality in Dali, a total of 100 serum samples from patients with chronic HBV-infection were collected for the det...To investigate the distribution of hepatitis B virus (HBV) genotypes and subgenotypes among the Bai nationality in Dali, a total of 100 serum samples from patients with chronic HBV-infection were collected for the detection of HBV genotypes and subgenotypes by genotype-specific primers and restriction fragment length polymorphism (RLFP), respectively. Among the 100 samples, the proportions of genotype B, C and mixed genotype (B+C) were 41%, 25% and 34%, respectively. All the genotype B strains belonged to subgenotype Ba. In genotype C, 84% were Subgenotype Cs and 12% were subgenotype Ce. The distribution of genotypes B, C and B+C showed no significant difference between male and female patients (P=0.182) and among the age groups of patients (P=0.812). The rates of HBeAg/HBeAg positivity were no significantly different among genotypes B, genotype C and mixed genotype (B+C) (P=0.077/P=0.663). In Dali, genotypes B, B+C and C existed among Bai nationality with chronic HBV-infection, and genotype B was the major genotype. Subgenotypes Ba and Cs were the predominant strains in patients with HBV genotype B/C infection. The most prominent characteristic was the higher prevalent rate of mixed genotype (B+C) in patients.展开更多

The start date of rainy season occurred on May 8th, 2001 is the earliest;The start date of rainy season occurred on July 1st, 2015 is the latest;The average value of the start date of rainy season from 1962 to 2015 is May 31st. During the past 54 years, the annual, seasonal and monthly rainfall showed the different degrees of reduction. Linear trend analysis showed that the trend of annual rainfall was decreasing with linear trend rate of -14.02 mm/10a, the annual rainfall decreases with the number of years is not significant. Seasonal rainfall decrease with the year number is not significant. The rainfall in summer and autumn decreased with the year number. The decreasing trend of rainfall in summer is greater than that in autumn. The rainfall in Winter and Spring increased with the year number. The increasing trend of rainfall in summer is greater than that in autumn. By using the method of cumulative anomaly and Morlet wavelet, the abrupt change of annual rainfall was analyzed, and the change of annual rainfall were found in 1974, 2002 and 2008. The annual rainfall was mainly in the period of 6 and 30. Objective To analyze the hepatitis B vaccine(HepB)vaccination rate and immunelevel among children aged eight months to 14 years in Dali Bai Autonomous Prefecture,and to evaluate the effect of immune prevention and con...Objective To analyze the hepatitis B vaccine(HepB)vaccination rate and immunelevel among children aged eight months to 14 years in Dali Bai Autonomous Prefecture,and to evaluate the effect of immune prevention and control.Methods A stratified and cluster sampling was designed to survey and collect serum samples of the 17236 cases from 12 counties(cities)in Dali Bai Autonomous Prefecture.The cases were divided into four age groups(eight to 17 months,18 to 35 months,three to six years,seven to 14 years).Serum levels of hepatitis B surface antigen(HBsAg)and hepatitis B surface antibody(anti-HBs)were detected by enzyme linked immunosorbent assay(ELISA).Statistical analysis was conducted by Spearman rank correlation analysis and chi-square test.Results The overall HepB vaccination rate,timely vaccination rate,and full course vaccination rate among 17236 children were 98.78%(17025/17236).展开更多
背景:溶质载体家族1成员5(solute carrier family 1 member 5,SLC1A5)在多种疾病中发挥了潜在作用,但确切作用机制尚不清楚。构建稳定的SLC1A5过表达和敲低细胞模型可为深入研究SLC1A5在疾病中的确切作用机制以及发现潜在治疗靶点提供...背景:溶质载体家族1成员5(solute carrier family 1 member 5,SLC1A5)在多种疾病中发挥了潜在作用,但确切作用机制尚不清楚。构建稳定的SLC1A5过表达和敲低细胞模型可为深入研究SLC1A5在疾病中的确切作用机制以及发现潜在治疗靶点提供有力的实验工具。目的:构建小鼠SLC1A5过表达和敲低的慢病毒载体,以建立稳定转染的RAW264.7细胞株,为深入探讨SLC1A5在炎症中的作用提供实验基础。方法:根据SLC1A5基因序列设计合成引物并使用聚合酶链反应扩增该基因片段。将目的基因定向接入经Age I/Nhe I酶切的载体质粒GV492中构建重组慢病毒质粒,对阳性克隆进一步筛选后测序比对结果;pHelper1.0质粒载体、pHelper2.0质粒载体、目的质粒载体与293T细胞共同培养并转染,获得慢病毒原液进行包装和滴度测定;在此基础上,通过体外培养RAW264.7细胞,确定嘌呤霉素工作质量浓度;不同滴度的慢病毒分别与RAW264.7细胞共同培养,根据荧光强度确定转染效率;用嘌呤霉素挑选出稳定转染细胞,实时荧光定量聚合酶链反应和蛋白免疫印迹方法检测稳定转染细胞株的SLC1A5基因和蛋白表达水平。结果与结论:(1)测序序列与目的序列一致提示重组慢病毒载体构建成功;(2)过表达SLC1A5慢病毒的滴度为1×10~9 TU/mL,敲低SLC1A5慢病毒的滴度为3×10~9 TU/mL;(3)确定RAW264.7细胞嘌呤霉素工作质量浓度为3μg/mL;(4)过表达/敲低SLC1A5慢病毒转染RAW264.7细胞的最佳条件皆为HiTransG P转染增强液且感染复数值等于50;(5)过表达SLC1A5稳转细胞株中SLC1A5基因和蛋白的表达量明显上调,而敲低SLC1A5稳转细胞株中SLC1A5基因和蛋白的表达量显著下调。结果表明,成功构建了小鼠SLC1A5过表达和敲低的慢病毒载体并获得稳定转染的RAW264.7细胞株。展开更多
