Comprehensive bioinformatics analysis of CYB561 expression in breast cancer:Link between prognosis and immune infiltration 被引量:2

Background:Cytochrome b561(CYB561)plays a critical role in neuroendocrine function,cardiovascular regulation,and tumor growth;however,the prognostic value of CYB561 in patients with breast cancer and the relationship between CYB561 expression and immune infiltration in breast cancer remain unclear.Methods:The mRNA expression and clinical data of patients with breast cancer were obtained from The Cancer Genome Atlas database.Functional enrichment analysis was used to explore underlying biological functions associated with CYB561.The methylation status of CYB561 was analyzed using the MethSurv database.The enrichment score of immune cell infiltration for CYB561 in breast cancer was calculated using single-sample gene set enrichment analysis.The prognostic value of CYB561 was evaluated using Kaplan-Meier method and Cox regression analysis.Based on the results of the multivariate Cox analysis,a nomogram was constructed to predict the effect of CYB561 expression on overall survival(OS).Results:The results showed that CYB561 was highly expressed in breast cancer tissues.Hypomethylation of CYB561 is associated with an unfavorable prognosis.In multivariate Cox regression analysis,CYB561 was an independent prognostic factor for OS.Functional enrichment analysis indicated that estrogen signaling pathway,inflammatory response,KRAS signaling pathway,epithelial-mesenchymal transition,leukocyte migration,and regulation of lymphocyte activation were strongly enriched in the low CYB561 expression group.Additionally,CYB561 expression was negatively correlated with immune infiltration of B cells,plasmacytoid dendritic cells,dendritic cells,and neutrophils.Conclusion:CYB561 may serve as a potential biomarker for breast cancer diagnosis and prognosis. 展开更多

BACKGROUND Benign lymphoepithelial cyst(BLEC)of the parotid gland is a rare benign embryonic-dysplastic cystic tumor in the anterolateral neck that occurs most commonly in human immunodeficiency virus(HIV)-positive adults and rarely in non-acquired immune deficiency syndrome patients.The main presentation is a slow-growing,painless mass,and secondary infection may cause acute inflammatory symptoms.CASE SUMMARY A 44-year-old Chinese male patient presented with a 1-year history of a mass in the left side of the neck.On physical examination,a mass similar in size and shape to a quail egg was found in the left parotid gland.The mass was tough,without tenderness,and easily moveable.The results of HIV tests,including antibody and nucleic acid tests and CD4+T cell examination,were negative.Imaging examination revealed a left parotid gland mass.The patient underwent surgical treatment,and BLEC was diagnosed based on postoperative pathology.After 2years of follow-up,the patient survived well without related discomfort.CONCLUSION The detailed characteristics of a BLEC in a patient without HIV infection contribute to an improved understanding of this rare disease. 展开更多

Objective:To explore the relationship between calcification of human vascular system and local inflammation using PET/CT.Methods:Patients who were treated with concurrent PET/CT in our hospital were collected continuously,and their arterial image data were analyzed in parallel with standardized uptake values.(SUV)and corresponding slice calcification score(CS)measurement.SUV values were collected and measured for slices with CS greater than 100.Patients were grouped according to CS,and the correlation analysis between SUV values and CS in each group was performed.Results:Overall CS and SUVmax(r=0.62,P<0.05)and SUVmean(r=0.59,P<0.05)were positively correlated,among which CS and SUV values were in the SUVmax(r=0.72,P<0.05),SUVmean(r=0.59,P<0.05),SUVmax of CS 200-299 group(r=0.71,P<0.05),SUVmean(r=0.80,P<0.05),SUVmax of CS 300-399 group(r=0.47,P<0.05),SUVmean(r=0.53,P<0.05)were positively correlated,SUVmax(r=-0.17,P>0.05),SUVmean(r=-0.07,P>0.05)were negatively correlated in CS 400-499 group,CS There was no significant correlation between SUVmax(r=0.22,P>0.05)and SUVmean(r=0.18,P>0.05)in the 500 group.Conclusion:There is a certain correlation between the local vascular calcification area and the corresponding local inflammatory response in the calcification progress PET/CT has the potential to monitor early calcification. 展开更多

A total of 80 patients with refractory epilepsy were recruited from the Inner Mongolia Medical College Affiliated Hospital. The loci of 60% of the patients could be positioned using a combined positron emission tomography/CT imaging modality. Hyper- and hypometabolism foci were examined as part of this study. Patients who had abnormal metabolism in positron emission tomography/CT imaging were divided into intermittent-phase group and the seizure-phase group. The intermittent-phase group was further divided into a single-focus group and a multiple-foci group according to the number of seizure foci detected by imaging. Following gamma knife treatment, seizure frequency was significantly lower in the intermittent-phase group and the seizure-phase group. Wieser's classification reached Grade I or II in nearly 40% of patients. Seizure frequency was significantly lower following treatment, but Wieser's classification score was significantly higher in the seizure-phase group compared with the intermittent-phase group. Seizure frequency was significantly lower following treatment in the single-focus group, but Wieser's classification score was significantly higher in the single-focus group as compared with the multiple-foci group. 展开更多

Background We previously reported that iodine-131(131^I)-Iabeled anti-pro-gastrin-releasing peptide (ProGRP(31-98)) monoclonal antibody D-D3 could selectively accumulate in the tumor sites of nude mice bearing small cell lung cancer (SCLC) xenografts. However, 1311-D-D3 was cleared slowly from the body, and the best radioimmunoimaging time for SCLC was 72-96 hours after injection. The aims of this study were to radiolabel anti-ProGRP(31-98) D-D3 monoclonal antibody with technetium-99m(99m^Tc) and to investigate the biodistribution of this antibody in healthy ICR mice. Methods D-D3was labeled with 99m^Tc via the 2-mercaptoethanol reduction method. 99m^Tc-D-g3 was purified by the gel column separation method. The labeling efficiency and radiochemical purity were measured by thin-layer chromatography. The immunological activity of 99m^Tc-D-O3 was determined with cell conjugation assays. 99m^Tc-D-D3 was injected into healthy ICR mice via a tail vein, and all the healthy ICR mice were sacrificed by cervical dislocation at a designated time. Then, the blood and major organs were removed and weighed, and counted in a gamma scintillation counter to determine the percentage of the injected dose per gram (%ID/g). Results The labeling rate and the radiochemical purity of 99m^TC-D-D3 were (73.87±2.89)% and (94.13±4.49)%, respectively. The immunobinding rates of 99m^Tc-O-D3 to the human small cell lung cancer NCI-H446 cell line and lung adenocarcinoma A549 cell line were (81.2±2.37)% and (24.3±1.46)%, respectively. The distribution data of normal ICR mice demonstrated that 99m^TC-D-D3was mainly distributed in the liver, kidney and lung, and less in the brain tissue and muscle. Conclusions 99m^Tc-D-D3 antibody not only had high radiochemical purity, but also had good stability both in vitro and in vivo, and maintained good immunological activity. 99m^Tc-D-D3 was metabolized mainly in the kidney and liver, and the blood radioactivity decreased rapidly. Thus, 99m^Tc-O-D3 is conducive to the radioimmunoimaging of SCLC. 展开更多

That tetraacetonitrile copper perchlorate catalyzes intramolecular amidation of arenes was found to be a new strategy for construction of nitrogen-containing heterocycles and resulted in the formation of a series of novel benzo[1,4]oxazin-3-one derivatives from N-(1,3-diphenyl-1H-1,2,4-triazol-5-yl)-2-phenoxyacetamides.This approach of heterocyclic construction proceeds in a chemoselective manner in which only benzo[1,4]oxazin-3-one derivatives were obtained by C—N bonds formation with cheap and simple copper salt catalyst under mild conditions in moderate to good yields.The biological assay of some of benzo[1,4]oxazin-3-one derivatives showed that they had moderate antiproliferative activity toward MDA-MB231 and HeLa cancer cell lines. 展开更多

To determine the relationship between DNP level after human severe brain injury and hyponatremia as well as isorrhea. Methods : The peripheral venous plasma as control was collected from 8 volunteers. The peripheral venous plasma from 14 severe brain injury patients were collected in the 1, 3, 7 days after injury. Radloimmunoassay was used to detect the DNP concentration. Meanwhile, daily plasma and urine electrolytes, osmotic pressure as well as 24 h Hquid intake and output volume were detected. Results: The normal adult human plasma DNP level was 62. 46 pg/ml ±27. 56 pg/ml. In the experimental group, the plasma DNP levels were higher from day 1 to day 3 in 8 of the 14 patients than those in the control group (P1 =0.05, P3 =0.03). Negative fluid balance occurred in 8 patients and hyponatremia in 7 patients. The increase of plasma DNP level was significantly correlated with the development of a negative fluid balance (r=-0.69, P〈0.01) and hyponatremia (χ^2 =4.38, P〈0.05). Conclusions: The increase of plasma DNP level is accompanied by the enhancement of natriuretic and diuretic responses in severe brain-injured patients, which is associated with the development of a negative fluid balance and hyponatremia after brain injury. 展开更多
