This study was designed to measure the pressure distribution of the intervertebral disc under different degrees of distraction of the interspinous process, because of a suspicion that the degree of distraction of the ...This study was designed to measure the pressure distribution of the intervertebral disc under different degrees of distraction of the interspinous process, because of a suspicion that the degree of distraction of the spinous process may have a close relationship with the disc load share. Six human cadaver lumbar spine L2-L5 segments were loaded in flexion, neutral position, and extension. The L3-L4 disc load was measured at each position using pressure measuring films. Shape-memory interspinous process implants (SMID) with different spacer heights, ranging in size from 10 to 20 mm at 2 mm increments, were used. It was found that a SMID with a spacer height equal to the distance of the interspinous process in the neutral position can share the biomechanical disc load without a significant change of load in the anterior annulus. An interspinous process stabilizing device (IPD) would not be appropriate to use in those cases with serious spinal stenosis because the over-distraction of the interspinous process by the SMID would lead to overloading the anterior annulus which is a recognized cause of disc degeneration.展开更多
Objective:To investigate the expression of miR—218 and its clinical significance in osteosarcoma tissues and explore its effect on proliferation and apoptosis in osteosarcoma cells.Methods:miR-218 expression was dete...Objective:To investigate the expression of miR—218 and its clinical significance in osteosarcoma tissues and explore its effect on proliferation and apoptosis in osteosarcoma cells.Methods:miR-218 expression was detected in 76 samples of surgically resected osteosarcoma and matched normal tumor-adjacent tissues using quantitative reverse transcription polymerase chain reaction(qRT-PCR).MiR-218 was over-expressed by exogenous miR-218 plasmids in Saos-2 cells,and then BrdU cell proliferation assay and flow cytometry were used to determine cell proliferation and apoptosis.Results:The expression of miR-218 in osteosarcoma tissues was significantly lower than those in normal tumor-adjacent tissues(t=8.735.P<0.001).MiR-218 expression in tumor tissues was significantly correlated with tumor size(x^2=5.380,P=0.020).clinical stage(x^2=6.692,P=0.010) and distant metastasis(x^2=4.l80.P=0.041).MiR-218 was obviously overexpressed by exogenous miR-218 plasmids(t= 19.42.P<0.001),and miR-218 overexpression significantly reduced cell proliferation(t=9.045.P<0.001) and induced apoptosis(t=12.38,P<0.001) in Saos-2 cells.Conclusions:The low-expression of miR-218 is correlated with the poor clinicopathological features in osteosarcoma.Moreover.miR-218 overexpression reduces cancer cell proliferation and induces apoplosis in Saos-2 cells,suggesting that miR-218 may play a key role in the progression of human osteosarcoma.展开更多

Methods:A rat model of deep venous thrombosis was established by directly clamping femoral vein.Based on the different biological situations of femoral vein thrombosis and observation phases,150 SD rats were divided into 7 groups.Inflammatory cells in vein wall of each group were counted.The fold change and cluster analysis were applied to study the change of gene expression during the development of venous thrombosis.Especially,the genes related to inflammation,fibrinolysis,coagulation of endotbelium were analyzed in detail. Results:The inflammation cells in femoral vein wall were mostly neutrophilic granuiocytes in Groups B,C and D,while they were lymphocytes in Groups E,F and G. Compared with Groups A,B,E and G,the inflammation cell counts in Groups C,D and F were much higher(P<0. 05).The results of fold-change analysis showed that 2 504 genes(Log 2 ratio≥1 or≤-1)presented different expressions in the process of TDVT,Most of these genes' functions were not clarified so far and the genes with known functions were involved in inflammation,DNA-dependent transcription regulation,blood coagulation,fibrinolysis, etc.Among them,23 genes related to inflammation had different expressions during TDVT.The cluster analysis showed that the expression changes of several genes,such as IL-1α,IL-1β,IL-6,Cinc2,corresponded with the development of femoral vein thrombosis.
