The loss of these cells causes permanent functional disorders in both the cochlear and vestibular systems. Transplantation of retinal precursor cells, R28 cells, into inner ear tissue may help replace missing cells. The aim of the current project was to induce R28 cell transdifferentiation into cochlear and vestibular cell types under culture conditions. The first part was related to R28 cell labeling with DiI fluorescence that would help identify and track R28 cells. The second part involved co-culturing R28 cells in cochlear and vestibular organotropic cultures or isolated spiral ganglion neurons. The results suggest that R28 cells have the potential to differentiate into supporting cell types and spiral ganglion neurons in serum free medium, probably under the influence of diffusible signals from inner ear tissues. This information is useful for future efforts in inducing stem cell differentiation in the inner ear to replace lost sensory and neural cells.展开更多

An effective method for CEC cultivation was proposed. The isolation of human choroidal CECs using micro dissection followed by the use of superparamagnetic beads (Dynabeads) coated with the CD 31, which selectively binds to the endothelial cell surface. Cells bound to beads were isolated using a magnetic particle concentrator. The CECs were planted into type Ⅳ collagen coated 24 well plates. The results show that the primary cultured CEC is induced to tube formation in collagen Ⅳ coated environment, which can be presented as an in vitro model of choroidal neovascularization.展开更多

METHODS:We retrospectively observ...PURPOSE: To report the incidence of back pain after photodynamic therapy,which suggests methods for prevention that are related to its pathogenesis. DESIGN: Retrospective case series. METHODS:We retrospectively observed 548 patients who had undergone photodynamic therapy with verteporfin. RESULTS: Of 548 patients at the first treatment,14 patients (2.6% ) experienced pain during the infusion. Eleven patients were being treated for age-related macular degeneration; their mean age was 81 years,which significantly differed from the mean age of the overall age-related macular degeneration group (P=.003). The pain was mild in eight patients,moderate in four patients,and severe in two patients,with dyspnea and precordial pain. Five of the 14 patients had further courses of photodynamic therapy. After being treated prophylactically 60 minutes before photodynamic therapy,only one patient reported further mild pain. CONCLUSIONS: The biologicmechanisms of back pain may involve a high level of circulating thromboxanes that are induced by the liposomal composition of verteporfin. Prevention may include hydration,nonsteroidal anti-inflammatory drugs,and halving the infusion rate.展开更多

To examine the safety and efficacy of a modified scleral fixated intraocular lens (SFIOL) technique combining a scleral tunnel for intraocular lens (IOL) insertion, horizontal passage of sutures, and performing anterior vitrectomy through self-sealing pars plana sclerostomies. Methods. Retrospective reviewof all patients who underwent SFIOL for aphakic correction at the Department of Ophthalmology and Visual Sciences, The Chinese University of Hong Kong, Prince of Wales Hospital, Hong Kong (a tertiary referral centre) between October 2000 and December 2001 with either the modified or conventional techniques. Results. A total of 20 eyes were identified. Eight modified (Group 1) and 12 conventional (Group 2) with a mean follow-up of 18.3 months. Postoperatively, all the eyes improved on unaided preoperative acuity and they all had a stable and well-positioned IOL. In all, 85% (7/8) of eyes in Group 1 maintained or improved best-corrected visual acuity. Conclusions. The modified SFIOL technique was found to be as safe and effective as conventional techniques. The main advantages of this technique include easy intraoperative suture management and superior globe maintenance.展开更多

Design: Crosssectional study. Methods: Concentrations of α - tocop- herol were measured by high- performance liquid chromatography in blood and in human lenses with and without cataract. Lenses were obtained during cataract surgery and from a regional eye bank. Peripheral α - tocopherol status in patients was assessed as plasma α - tocopherol (total and corrected for low- density lipoproteins) and as red blood cell bound α - tocopherol. Data (mean± standard error of the mean) are expressed as μ M α - tocopherol/g lens protein, μ Mα - tocopherol/l plasma, μ M α - tocopherol/g low- den- sity lipoproteins, and μ M α - tocopherol/T red blood cells. Results: Concentrations of α - tocopherolwere measured in 27 lenses of cataract patients, 8 cadaver lenses with cataract and in 14 clear cadaver lenses. The concentration of α - tocopherol was significantly higher in cataract than in control cadaver lenses (0.49± 0.04 vs 0.35± 0.03, P< .05). The difference between α - tocopherol in lenses of cataract patients and control cadaver lenses was even higher (0.7± 0.1 vs 0.35 ± 0.03, P< .01). No significant correlation was observed between plasma α - tocopherol or red blood cell bound α - tocopherol and lens α - tocopherol in patients.
