Background:Patients with pancreatic ductal adenocarcinoma(PDAC)who undergo surgical resection and receive effective chemotherapy have the best chance for longterm survival.Unfortunately,because of the heterogeneity of...Background:Patients with pancreatic ductal adenocarcinoma(PDAC)who undergo surgical resection and receive effective chemotherapy have the best chance for longterm survival.Unfortunately,because of the heterogeneity of pancreatic cancer,it is difficult to find a personalized treatment strategy for patients.Organoids are ideal preclinical models for personalized medicine.Therefore,we explore the cultivation conditions and construction methods of PDAC organoid models to screen the individualized therapy strategy.Methods:Fresh PDAC tissues from surgical resection were collected and digested with digestive enzymes;then the tumor cells were embedded in Matrigel with a suitable medium to establish the PDAC organoid models.The genetic stability of the organoids was analyzed using whole exon sequencing;hematoxylin and eosin staining and immunohistochemistry of organoids were performed to analyze their consistency with the pathological morphology of the patient's tumor tissue;After 2 days of organoid culture,we selected four commonly used clinical chemotherapy drugs for single or combined treatment to analyze drug sensitivity.Results:Two cases of PDAC organoid models were successfully established,and the results of their pathological characteristics and exome sequencing were consistent with those of the patient's tumor tissue.Both PDAC organoids showed more sensitivity to gemcitabine and cisplatin,and the combined treatment was more effective than monotherapy.Conclusion:Both organoids better retained the pathological characteristics,genomic stability,and heterogeneity with the original tumor.Individual PDAC organoids exhibited different sensitivities to the same drugs.Thus,this study provided ideal experimental models for screening individualized therapy strategy for patients with PDAC.展开更多
The cumulative damage caused by repetitive mild traumatic brain injury can cause long-term neurodegeneration leading to cognitive impairment.This cognitive impairment is thought to result specifically from damage to t...The cumulative damage caused by repetitive mild traumatic brain injury can cause long-term neurodegeneration leading to cognitive impairment.This cognitive impairment is thought to result specifically from damage to the hippocampus.In this study,we detected cognitive impairment in mice 6 weeks after repetitive mild traumatic brain injury using the novel object recognition test and the Morris water maze test.Immunofluorescence staining showed that p-tau expression was increased in the hippocampus after repetitive mild traumatic brain injury.Golgi staining showed a significant decrease in the total density of neuronal dendritic spines in the hippocampus,as well as in the density of mature dendritic spines.To investigate the specific molecular mechanisms underlying cognitive impairment due to hippocampal damage,we performed proteomic and phosphoproteomic analyses of the hippocampus with and without repetitive mild traumatic brain injury.The differentially expressed proteins were mainly enriched in inflammation,immunity,and coagulation,suggesting that non-neuronal cells are involved in the pathological changes that occur in the hippocampus in the chronic stage after repetitive mild traumatic brain injury.In contrast,differentially expressed phosphorylated proteins were mainly enriched in pathways related to neuronal function and structure,which is more consistent with neurodegeneration.We identified N-methyl-D-aspartate receptor 1 as a hub molecule involved in the response to repetitive mild traumatic brain injury,and western blotting showed that,while N-methyl-D-aspartate receptor 1 expression was not altered in the hippocampus after repetitive mild traumatic brain injury,its phosphorylation level was significantly increased,which is consistent with the omics results.Administration of GRP78608,an N-methyl-D-aspartate receptor 1 antagonist,to the hippocampus markedly improved repetitive mild traumatic brain injury-induced cognitive impairment.In conclusion,our findings suggest that N-methyl-D-aspartate receptor 1 signaling in the hippocampus is involved in cognitive impairment in the chronic stage after repetitive mild traumatic brain injury and may be a potential target for intervention and treatment.展开更多

The byte stream is widely used in malware detection due to its independence of reverse engineering.However,existing methods based on the byte stream implement an indiscriminate feature extraction strategy,which ignore...The byte stream is widely used in malware detection due to its independence of reverse engineering.However,existing methods based on the byte stream implement an indiscriminate feature extraction strategy,which ignores the byte function difference in different segments and fails to achieve targeted feature extraction for various byte semantic representation modes,resulting in byte semantic confusion.To address this issue,an enhanced adversarial byte function associated method for malware backdoor attack is proposed in this paper by categorizing various function bytes into three functions involving structure,code,and data.The Minhash algorithm,grayscale mapping,and state transition probability statistics are then used to capture byte semantics from the perspectives of text signature,spatial structure,and statistical aspects,respectively,to increase the accuracy of byte semantic representation.Finally,the three-channel malware feature image is constructed based on different function byte semantics,and a convolutional neural network is applied for detection.Experiments on multiple data sets from 2018 to 2021 show that the method can effectively combine byte functions to achieve targeted feature extraction,avoid byte semantic confusion,and improve the accuracy of malware detection.展开更多
For black shales,laminae and bedding are hard to identify,grain size is difficult to measure,and trace fossils do not exist.Taking the Ordovician Wufeng–Silurian Longmaxi shale in southern Sichuan Basin,China,as an e...For black shales,laminae and bedding are hard to identify,grain size is difficult to measure,and trace fossils do not exist.Taking the Ordovician Wufeng–Silurian Longmaxi shale in southern Sichuan Basin,China,as an example,the types,characteristics and models of microfacies in epicontinental shale are analyzed by means of full-scale observation of large thin sections,argon-ion polishing field emission-scanning electron microscopy(FE-SEM),and kerogen microscopy.The epicontinental sea develops delta,tidal flat and shelf facies,with black shale found in microfacies such as the underwater distributary channel and interdistributary bay under delta front facies,the calcareous and clayey flats under intertidal flat facies,the calcareous and clayey shelfs under shallow shelf facies,the deep slope,deep plain and deep depression under deep shelf facies,and the overflow under gravity flow facies.Basinward,silty lamina decreases and clayey lamina increases,the grain size changes from coarse silt to fine mud,the silica content increases from about 20%to above 55%,the carbonate and clay minerals content decreases from above 40%to around 10%,and the kerogen type changes from type II2 to type II1 and type I.Provenance and topography dominate the types and distribution of shale microfacies.The underwater distributary channel,interdistributary bay,clayey flat,clayey shelf,and overflow microfacies are developed in areas with sufficient sediment supply.The calcareous flat and calcareous shelf are developed in areas with insufficient sediment supply.The deep shelf shale area is divided into deep slope,deep plain,and deep depression microfacies as a result of three breaks.The formation of epicontinental shale with different microfacies is closely related to the tectonic setting,paleoclimate,and sea level rise.The relatively active tectonic setting increases the supply of terrigenous clasts,forming muddy water fine-grained sediment.The warm and humid paleoclimate is conducive to the enrichment of organic matter.The rapid sea level rise is helpful to the widespread black shale.展开更多
Objective:To investigate the mechanism of action and material basis of AiTongXiao granule in the treatment of hepatocellular carcinoma(HCC)based on network pharmacology and transplanted liver cancer rat model.Methods:...Objective:To investigate the mechanism of action and material basis of AiTongXiao granule in the treatment of hepatocellular carcinoma(HCC)based on network pharmacology and transplanted liver cancer rat model.Methods:TCMSP database was used to screen out effective components and its corresponding potential pharmaceutical targets,and databases including Gene Cards,OMIM,Drugbank and TTD were further used to collect HCC-related drug targets.The intersecting targets were obtained by mapping the drug and disease targets.The component-targets network was constructed and visualized by Cytoscape 3.8.2 software.Protein-protein interaction(PPI)network was built by STRING online platform,and the topological relationship and core targets was analyzed and screened by using CytoNCA software.In addition,Metascape database was used to perform gene ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis of the core targets.At last,rat liver transplanted liver cancer model was established by using Walker-256 cell line and treated by AiTongXiao granule for 15 days.Western blot was used to further compare the expression levels of AKT,pAKT,p53,p-p53,ERK1/2 and ERK1/2 in the tumor between treatment group and the control group.Results:257 active components were obtained from AiTongXiao granule,corresponding to 294 drug targets.Meanwhile,233 of the 7993 HCC disease targets were screened out between AiTongXiao granule drug and HCC disease targets.11 core targets including AKT1,IL6,TP53,MAPK3,TNF,JUN,CASP3,MAPK1,MYC,PTGS2,MMP9 were further obtained by median screening.GO and KEGG analysis results showed that these core targets enriched to HBV,TNF and cancer related pathways.The rat transplanted liver cancer model results indicated significant down regulation for AKT,p-AKT,pERK1/2,and significant up regulation of p-p53 after AiTongXiao granule treatment(P<0.05).Conclusion:AiTongXiao granule could act to multiple cancer related pathways,and AKT,p53 and ERK1/2 were validated to be regulated by ATXF in rat model.The mechanism may be through the regulation of the above signaling pathways to exert anti-liver cancer effect.展开更多
In this study,the effect of sintering temperature and the addition of kaolin,a sintering agent,on the microscopic,phase,and mechanical properties of ceramics were investigated using secondary aluminum dross(SAD)as the...In this study,the effect of sintering temperature and the addition of kaolin,a sintering agent,on the microscopic,phase,and mechanical properties of ceramics were investigated using secondary aluminum dross(SAD)as the main component in the manufacturing of ceramics.The basic phases of the ceramics were Al_(2)O_(3),MgAl_(2)O_(4),NaAl_(11)O_(17),and SiO_(2)without the addition of kaolin.The diffraction peaks of MgAl_(2)O_(4),NaAl_(11)O_(17),and SiO_(2)kept decreasing while those of Al_(2)O_(3)kept increasing with an increase in temperature.In addition,the increase in temperature promoted the growth of the grains.The grains were uniform in size and regular in distribution,with a shrinkage of 2.2%,porosity of 72.5%,bulk density of 1.076 g/cm^(3),and compressive strength of 1.12 MPa.When the sintering temperature was 1450°C,the basic phases of the ceramic after the addition of kaolin were Al_(2)O_(3),MgAl_(2)O_(4),NaAl_(11)O_(17),and SiO_(2).With the increase of kaolin,the diffraction peaks of NaAl_(11)O_(17)and SiO_(2)decreased until they disappeared,while the diffraction peaks of Al_(2)O_(3)increased significantly.When kaolin was added at 30 wt.%,the ceramics obtained had shrinkage of 18%,a porosity of 47.26%,a bulk density of 1.965 g/cm^(3),and compressive strength of 31.9 MPa.Cracks existed inside the ceramics without the addition of kaolin,while the addition of kaolin significantly changed this defect.It is shown that SAD can obtain porous ceramics with good properties at a sintering temperature of 1450°C and a kaolin addition of 30 wt.%.展开更多
The objective of this study was to investigate the main active ingredients,potential targets,and possible mechanisms of action of the combination of Radix Astragali and Caulis Spatholobi for the treatment of atheroscl...The objective of this study was to investigate the main active ingredients,potential targets,and possible mechanisms of action of the combination of Radix Astragali and Caulis Spatholobi for the treatment of atherosclerosis using network pharmacology.The study aimed to provide a reference basis for the development of new formulations and clinical use of Chinese medicine.The main components of Radix Astragali and Caulis Spatholobi were obtained from the TCMSP,BATMAN-TCM database,and literature reports.The targets corresponding to the main components were imported into the Uniprot database to standardize the names,and target information was supplemented with the Swiss Target Prediction database.Disease-related targets were obtained from DrugBank,OMIM,CTD,GeneCards,and DisGeNET online databases.Venn tools were used to obtain the potential targets of Radix Astragali and Caulis Spatholobi for the treatment of AS.The intersecting genes were imported into the STRING 11.5 database to construct protein-protein interaction network maps and analyze their interactions.Cytoscape 3.7.1 software was used to mine their core targets.GO function and KEGG signaling pathway enrichment analysis were performed using the DAVID v2023q1 database.The results were imported into the“Bioinformatics Cloud Platform”to generate enrichment bubble maps.Finally,the“component-target-pathway”diagram was constructed using Cytoscape 3.7.1 software.The study found that 78 major active ingredients and 527 potential targets were obtained from Radix Astragali and Caulis Spatholobi.The main active components of the two in combination for the treatment of AS are quercetin,stigmasterol,kaempferol,luteolin,formononetin,etc.The key targets involve CDKN1A,E2F1,CDK4,CDK2,CDK1,RB1,TP53,CDKN1B,IL6,JUN,etc.The main pathways involved the AGE-RAGE signaling pathway in diabetic complications,cancer pathway,etc.The biological processes involved include positive regulation of gene expression,negative regulation of apoptotic process,etc.The study initially verified the feasibility of the combination of Radix Astragali-Caulis Spatholobi by Qi-invigorating(promoting human metabolic activity)and blood-activating for the treatment of AS.It demonstrated that the combination of Chinese medicine has multi-level,multi-target,and multi-pathway mechanisms of action to treat the disease,providing a reference basis for the development and utilization of new drugs.展开更多
Background:Traditional Chinese medicine(TCM)syndrome is the basic unit of TCM treatment,which help clinicians assess the disease progression and treatment preoperative of tumor patients.However,the prognostic signific...Background:Traditional Chinese medicine(TCM)syndrome is the basic unit of TCM treatment,which help clinicians assess the disease progression and treatment preoperative of tumor patients.However,the prognostic significance of TCM syndrome is still unclear.This study aims to detect the differences in overall survival between different TCM syndrome and further develop a new nomogram with TCM syndrome for predicting overall survival in colorectal cancer.Methods:A total of 324 patients with colorectal cancer were enrolled and categorized into three groups based on TCM syndrome:deficiency,excess,and deficiency-excess.The prognosis of colorectal cancer patients with different TCM syndromes was evaluated using Kaplan-Meier analysis and Cox regression analysis.Results:The proportion of advanced stage and lymph metastasis in the patients with deficiency syndrome was higher,and the overall survival was shorter than other syndromes.Meanwhile,the TCM syndrome(P<0.001),tumor invasion depth(P<0.001),lymph metastasis(P=0.018),organic metastasis(P=0.005)and tumor node metastasis(TNM)stage(P=0.029)were the independent prognostic factor.Then,a new nomogram with TCM syndrome was established and assessed.324 colorectal cancer patients were randomly divided into training(n=215)and validation cohorts(n=109).A nomogram incorporating preoperative TCM syndrome,gender,age,T,N,and M status was developed,which had good discrimination and calibration.Conclusion:Taken together,our results indicated that TCM syndrome could assess the prognosis of colorectal cancer.The nomogram incorporating TCM syndromes and tumor information is helpful for risk stratification and prognostic predictions in colorectal cancer preoperatively.展开更多
Na_(3)V_(2)(PO_(4))_(3)(NVP)cathode material of the sodium ion battery(1 C=117 mAh g-1)has a NASICON-type structure,which not only facilitates the rapid migration of sodium ions,but also has a small volume deformation...Na_(3)V_(2)(PO_(4))_(3)(NVP)cathode material of the sodium ion battery(1 C=117 mAh g-1)has a NASICON-type structure,which not only facilitates the rapid migration of sodium ions,but also has a small volume deformation during sodium ion de-intercalation and the main frame mechanism remains unchanged,and thus is seen as an energy storage material for a wide range of applications,but has a limited electronic conductivity due to its structure.In this paper,NVP cathode materials with finer primary particles are successfully prepared using a simple hydrothermal treatment-assisted sol-gel method.The increased pore size of the NVP materials prepared under the hydrothermal process allows for more active sites and more effective resistance to the volume deformation of sodium ions during insertion/extraction processes,effectively facilitating the diffusion of ions and electrons.The Na_(3)V_(2)(PO_(4))_(3) material obtained by the optimized process exhibited good crystallinity in XRD characterization,as well as superior electrochemical properties in a series of electrochemical tests.A specific capacitance of 106.3 mAh g^(-1) at 0.2 C is demonstrated,compared to 96.5 mAh g^(-1) for Na_(3)V_(2)(PO_(4))_(3) without hydrothermal treatment,and cycling performance is also improved with 93%capacity retention.The calculated sodium ion diffusion coefficient(DNa=5.68×10^(-14))obtained after EIS curve fitting of the improved sample illustrates that the pore structure is beneficial to the performance of the Na_(3)V_(2)(PO_(4))_(3)cathode material.展开更多
Gas diffusion in the shale matrix has a dominant effect on late-stage production from shale gas reservoirs.However,adequate research on the mechanisms and contributions of gas diffusion for varied pore size population...Gas diffusion in the shale matrix has a dominant effect on late-stage production from shale gas reservoirs.However,adequate research on the mechanisms and contributions of gas diffusion for varied pore size populations in shale matrix under recreated in situ stress is lacking.We report gas-diffusion measurements under constant in situ stress but variable gas pressures for contrasting non-adsorbent(helium(He))and adsorbed(methane(CH_(4)))gases to investigate the impact of effective stress on the evolution of dominant mechanisms of diffusion.An intact sample replicates true pore-network topology and diffusion paths.An integrated diffusion model is proposed that combines the effects of slip flow,Knudsen flow,and surface diffusion to constrain the evolution of these flow regimes and their respective contributions to the observational data.Finally,a probability density function(PDF)is employed to separate the gas content distributions of macropores and micropores from the total gas content and to investigate gas contributions in various pores.The results reveal that the diffusion coefficients of both He and CH_(4) in macropores and micropores increase with gas pressure but decrease with increasing effective stress.The diffusion coefficients of He and CH_(4) are different in macropores but remain nearly the same in micropores.The diffusion coefficients of slip flow and surface diffusion increase with decreasing effective stress except for CH_(4) diffusion in the micropores,while the evolution of Knudsen diffusion shows the opposite trend.Slip flow plays a dominant role in He and CH_(4) diffusion within macropores(pore size 45 nm).Knudsen diffusion gradually becomes significant for He diffusion in the micropores(pore size 4 nm),conversely,for CH_(4) diffusion in the micropores,surface diffusion becomes significant.Related to gas production from reservoirs,the contributions of the micropores will increase gradually with the duration of gas recovery,indicating the significant role of gas diffusion in micropores to steady supply during latestage production.展开更多

Test materials with different...The influence of rare earth (RE) elements on the solidification process and eutectic transformation and mechanical properties of the high-V type cast, high-speed steel roll was studied. Test materials with different RE additions were prepared on a horizontal centrifugal casting machine. The solidification process, eutectic structure transformation, carbide morphology, and the elements present, were all investigated by means of differential scanning calorimetry (DSC) and scanning electron microscopy energy dispersive spectrometry (SEM-EDS). The energy produced by crack initiation and crack extension was analyzed using a digital impact test machine. It was found that rare earth elements increased the tensile strength of the steel by inducing crystallization of earlier eutectic γ-Fe during the solidification process, which in turn increased the solidification temperature and thinned the dendritic grains. Rare earth elements with large atomic radius changed the lattice parameters of the MC carbide by forming rare earth carbides. This had the effect of dispersing longpole M C carbides to provide carbide grains, thereby, reducing the formation of the gross carbide and making more V available, to increase the secondary hardening process and improve the hardness level. AIM To explore the role of macrophages in chronic pancreatitis(CP) and the effect of Dachaihu decoction(DCHD) on pancreatic fibrosis in mice.METHODS Kun Ming mice were randomly divided into a control group, CP group, ...AIM To explore the role of macrophages in chronic pancreatitis(CP) and the effect of Dachaihu decoction(DCHD) on pancreatic fibrosis in mice.METHODS Kun Ming mice were randomly divided into a control group, CP group, and DCHD group. In the CP and DCHD groups, mice were intraperitoneally injected with 20% L-arginine(3 g/kg twice 1 d/wk for 6 wk). Mice in the DCHD group were administered DCHD intragastrically at a dose of 14 g/kg/d 1 wk after CP induction. At 2 wk, 4 wk and 6 wk post-modeling, the morphology of the pancreas was observed using hematoxylin and eosin, and Masson staining. Interleukin-6(IL-6) serum levels were assayed using an enzyme-linked immunosorbent assay. Double immunofluorescence staining was performed to observe the co-expression of F4/80 and IL-6 in the pancreas. Inflammatory factors including monocyte chemoattractant protein-1(MCP-1), macrophage inflammatory protein-1α(MIP-1α) and IL-6 were determined using real time-polymerase chain reaction. Western blot analysis was used to detect fibronectin levels in the pancreas. RESULTS Compared with the control group, mice with 20% L-arginine-induced CP had obvious macrophage infiltration and a higher level of fibrosis. IL-6 serum concentrations were significantly increased. Double immunofluorescence staining showed that IL-6 and F4/80 were co-expressed in the pancreas. With the administration of DCHD, the infiltration of macrophages and degree of fibrosis in the pancreas were significantly attenuated; IL-6, MCP-1 and MIP-1α m RNA, and fibronectin levels were reduced. CONCLUSION The dominant role of macrophages in the development of CP was mainly related to IL-6 production. DCHD was effective in ameliorating pancreatic fibrosis by inhibiting macrophage infiltration and inflammatory factor secretion in the pancreas.展开更多

The electrochemistry properties of CS-CMC BM were also studied. Hematoxylin-eosin staining...This study aimed to explore the pathological change to hippocampal neurons and the expression of growth associated protein 43 in 21-day-old young rats following chronic intermittent hypoxia. Hematoxylin-eosin staining results showed varying degrees of degeneration and necrosis in hippocampal neurons depending on the modeling time. Immunohistochemistry revealed that growth associated protein 43 expression in young rats following chronic intermittent hypoxia decreased, but that levels were still higher than those of normal rats at each time point, especially 4 weeks after modeling. During 1 5 weeks after modeling, a slow growth in rat weight was observed. Experimental findings indicate that chronic intermittent hypoxia may induce growth dysfunction and necrosis of hippocampal neurons, as well as increase the expression of growth associated protein 43 in young rats.展开更多
文摘With the development of cutting-edge multi-object spectrographs,fiber positioners located in the focal plane are being scaled down in size,and miniature hollow-cup Permanent Magnet motors are now being considered as a suitable replacement for Faulhaber Precistep stepper motors.However,the small electrical time constant of such coreless motors poses a challenge,as the problem of severe commutation torque ripple in a fiber positioner running a position loop has been tricky.To overcome this challenge,it is advised to increase the Pulse Width Modulation(PWM)frequency as much as possible to mitigate the effects of the current fluctuation.This must be done while ensuring adequate resolution of the PWM generator.By employing a voltage open-loop field-oriented control based on a modulation frequency of 1 MHz,the drive current only costs 25 m A under a 3.3 V power supply.The sine degree of phase current is immaculate,and the repeat positioning accuracy can reach 2μm.Moreover,it is possible to further shrink the bill of devices and the layout area of the Printed Circuit Board,especially in sizesensitive applications.This device has been developed under the new generation of The Large Sky Area MultiObject Fiber Spectroscopic Telescope.

文摘High-content screening(HCS)technology combines automated high-speed imaging hardware and single-cell quantitative analysis.It can greatly accelerate data acquisition in cellular fluorescence imaging and is a powerful research technique in traditional Chinese medicine(TCM).An increasing number of laboratories and platforms,including TCM laboratories,have begun utilizing HCS systems.However,this technology is still in its infancy in TCM research and there is a lack of sufficient experience with the associated concepts,instrument configurations,and analysis methods.To improve the understanding of HCS among researchers in the field of TCM,this paper summarizes the concept of HCS,software and hardware configuration,the overall research process,as well as common problems and related solutions of HCS in TCM research based on our team’s previous research experience,providing several research examples and an outlook on future perspectives,aiming to provide a technical guide for HCS in TCM research.

文摘The acceleration of LMXB 4U 1820-30 derived from its orbital-period derivative P_(b)was supposed to be the evidence for an Intermediate-mass black hole(IMBH)in the Galactic globular cluster(GC)NGC 6624.However,we find that the anomalous P_(b)is mainly due to the gravitational wave emission,rather than the acceleration in cluster potential.Using the standard structure models of GCs,we simulate acceleration distributions for pulsars in the central region of the cluster.By fitting the acceleration of J1823-3021 A with the simulated distribution profiles(maximum values),it is suggested that an IMBH with mass M■950_(-350)^(+550)M_(⊙) may reside in the cluster center.We further show that the second period derivative P of J1823-3021 A is probably due to the gravitational perturbation of a nearby star.

文摘Background:Patients with pancreatic ductal adenocarcinoma(PDAC)who undergo surgical resection and receive effective chemotherapy have the best chance for longterm survival.Unfortunately,because of the heterogeneity of pancreatic cancer,it is difficult to find a personalized treatment strategy for patients.Organoids are ideal preclinical models for personalized medicine.Therefore,we explore the cultivation conditions and construction methods of PDAC organoid models to screen the individualized therapy strategy.Methods:Fresh PDAC tissues from surgical resection were collected and digested with digestive enzymes;then the tumor cells were embedded in Matrigel with a suitable medium to establish the PDAC organoid models.The genetic stability of the organoids was analyzed using whole exon sequencing;hematoxylin and eosin staining and immunohistochemistry of organoids were performed to analyze their consistency with the pathological morphology of the patient's tumor tissue;After 2 days of organoid culture,we selected four commonly used clinical chemotherapy drugs for single or combined treatment to analyze drug sensitivity.Results:Two cases of PDAC organoid models were successfully established,and the results of their pathological characteristics and exome sequencing were consistent with those of the patient's tumor tissue.Both PDAC organoids showed more sensitivity to gemcitabine and cisplatin,and the combined treatment was more effective than monotherapy.Conclusion:Both organoids better retained the pathological characteristics,genomic stability,and heterogeneity with the original tumor.Individual PDAC organoids exhibited different sensitivities to the same drugs.Thus,this study provided ideal experimental models for screening individualized therapy strategy for patients with PDAC.

文摘The cumulative damage caused by repetitive mild traumatic brain injury can cause long-term neurodegeneration leading to cognitive impairment.This cognitive impairment is thought to result specifically from damage to the hippocampus.In this study,we detected cognitive impairment in mice 6 weeks after repetitive mild traumatic brain injury using the novel object recognition test and the Morris water maze test.Immunofluorescence staining showed that p-tau expression was increased in the hippocampus after repetitive mild traumatic brain injury.Golgi staining showed a significant decrease in the total density of neuronal dendritic spines in the hippocampus,as well as in the density of mature dendritic spines.To investigate the specific molecular mechanisms underlying cognitive impairment due to hippocampal damage,we performed proteomic and phosphoproteomic analyses of the hippocampus with and without repetitive mild traumatic brain injury.The differentially expressed proteins were mainly enriched in inflammation,immunity,and coagulation,suggesting that non-neuronal cells are involved in the pathological changes that occur in the hippocampus in the chronic stage after repetitive mild traumatic brain injury.In contrast,differentially expressed phosphorylated proteins were mainly enriched in pathways related to neuronal function and structure,which is more consistent with neurodegeneration.We identified N-methyl-D-aspartate receptor 1 as a hub molecule involved in the response to repetitive mild traumatic brain injury,and western blotting showed that,while N-methyl-D-aspartate receptor 1 expression was not altered in the hippocampus after repetitive mild traumatic brain injury,its phosphorylation level was significantly increased,which is consistent with the omics results.Administration of GRP78608,an N-methyl-D-aspartate receptor 1 antagonist,to the hippocampus markedly improved repetitive mild traumatic brain injury-induced cognitive impairment.In conclusion,our findings suggest that N-methyl-D-aspartate receptor 1 signaling in the hippocampus is involved in cognitive impairment in the chronic stage after repetitive mild traumatic brain injury and may be a potential target for intervention and treatment.

文摘Background:Alzheimer’sdisease(AD)is a prevalent neurodegenerative disorder causing progressive dementia.Research suggests that microRNAs(miRNAs)could serve as biomarkers and therapeutic targets for AD.Reduced levels of miR-137 have been observed in the brains of AD patients,but its specific role and down stream mechanisms remain unclear.This study sought to examine the therapeutic potential of miR-137-5p agomir in alleviating cognitive dysfunction induced in AD models and explore its potential mechanisms.Methods:This study utilized bioinformatic analysis and a dual-l uciferase reporter assay to investigate the relationship between miR-137-5p and ubiquitin-specific peptidase 30(USP30).In vitro experiments were conducted using SH-SY5Y cells to assess the impact of miR-137-5p on Aβ1-42 neurotoxicity.In vivo experiments on AD mice evaluated the effects of miR-137-5p on cognition,Aβ1-42 deposition,Tau hyperphosphorylation,and neuronal apoptosis,as well as its influence on USP30 levels.Results:It was discovered that miR-137-5p mimics efficiently counteract Aβ1-42 neurotoxicity in SH-SY5Y cells,a protective effect that is negated by USP30 overexpression.In vivo experiments demonstrated that miR-137-5p enhances the cognition and mobility of AD mice,significantly reducing Aβ1-42 deposition,Tau hyperphosphorylation,and neuronal apoptosis within the hippocampus and cortex regions.Mechanistically,miR-137-5p significantly suppresses USP30 levels in mice,though USP30 overexpression partially buffers against miR-137-5p-i nduced AD symptom improvement.Conclusion:Our study proposes that miR-137-5p,by instigating the downregulation of USP30,has the potential to act as a novel and promising therapeutic target for AD.

文摘The wave functions,energy levels and matrix elements of Yb+ions are calculated using the relativistic configuration interaction plus core polarization(RCICP)method.The static and dynamic electric dipole polarizabilities of the ground state and low-lying excited states are determined.Then,the magic wavelengths of the magnetic sublevel 6s_(1/2,m=1/2)→5d_(3/2,m=±3/2,±1/2)and 6s_(1/2,m=1/2)→5_(d5/2,m=±5/2,±3/2,±1/2)transitions in the linearly,right-handed,and left-handed polarized light are further determined.The dependence of the magic wavelengths upon the angle between the direction of magnetic field and the direction of laser polarization is analyzed.

文摘The byte stream is widely used in malware detection due to its independence of reverse engineering.However,existing methods based on the byte stream implement an indiscriminate feature extraction strategy,which ignores the byte function difference in different segments and fails to achieve targeted feature extraction for various byte semantic representation modes,resulting in byte semantic confusion.To address this issue,an enhanced adversarial byte function associated method for malware backdoor attack is proposed in this paper by categorizing various function bytes into three functions involving structure,code,and data.The Minhash algorithm,grayscale mapping,and state transition probability statistics are then used to capture byte semantics from the perspectives of text signature,spatial structure,and statistical aspects,respectively,to increase the accuracy of byte semantic representation.Finally,the three-channel malware feature image is constructed based on different function byte semantics,and a convolutional neural network is applied for detection.Experiments on multiple data sets from 2018 to 2021 show that the method can effectively combine byte functions to achieve targeted feature extraction,avoid byte semantic confusion,and improve the accuracy of malware detection.

文摘For black shales,laminae and bedding are hard to identify,grain size is difficult to measure,and trace fossils do not exist.Taking the Ordovician Wufeng–Silurian Longmaxi shale in southern Sichuan Basin,China,as an example,the types,characteristics and models of microfacies in epicontinental shale are analyzed by means of full-scale observation of large thin sections,argon-ion polishing field emission-scanning electron microscopy(FE-SEM),and kerogen microscopy.The epicontinental sea develops delta,tidal flat and shelf facies,with black shale found in microfacies such as the underwater distributary channel and interdistributary bay under delta front facies,the calcareous and clayey flats under intertidal flat facies,the calcareous and clayey shelfs under shallow shelf facies,the deep slope,deep plain and deep depression under deep shelf facies,and the overflow under gravity flow facies.Basinward,silty lamina decreases and clayey lamina increases,the grain size changes from coarse silt to fine mud,the silica content increases from about 20%to above 55%,the carbonate and clay minerals content decreases from above 40%to around 10%,and the kerogen type changes from type II2 to type II1 and type I.Provenance and topography dominate the types and distribution of shale microfacies.The underwater distributary channel,interdistributary bay,clayey flat,clayey shelf,and overflow microfacies are developed in areas with sufficient sediment supply.The calcareous flat and calcareous shelf are developed in areas with insufficient sediment supply.The deep shelf shale area is divided into deep slope,deep plain,and deep depression microfacies as a result of three breaks.The formation of epicontinental shale with different microfacies is closely related to the tectonic setting,paleoclimate,and sea level rise.The relatively active tectonic setting increases the supply of terrigenous clasts,forming muddy water fine-grained sediment.The warm and humid paleoclimate is conducive to the enrichment of organic matter.The rapid sea level rise is helpful to the widespread black shale.

文摘Objective:To investigate the mechanism of action and material basis of AiTongXiao granule in the treatment of hepatocellular carcinoma(HCC)based on network pharmacology and transplanted liver cancer rat model.Methods:TCMSP database was used to screen out effective components and its corresponding potential pharmaceutical targets,and databases including Gene Cards,OMIM,Drugbank and TTD were further used to collect HCC-related drug targets.The intersecting targets were obtained by mapping the drug and disease targets.The component-targets network was constructed and visualized by Cytoscape 3.8.2 software.Protein-protein interaction(PPI)network was built by STRING online platform,and the topological relationship and core targets was analyzed and screened by using CytoNCA software.In addition,Metascape database was used to perform gene ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis of the core targets.At last,rat liver transplanted liver cancer model was established by using Walker-256 cell line and treated by AiTongXiao granule for 15 days.Western blot was used to further compare the expression levels of AKT,pAKT,p53,p-p53,ERK1/2 and ERK1/2 in the tumor between treatment group and the control group.Results:257 active components were obtained from AiTongXiao granule,corresponding to 294 drug targets.Meanwhile,233 of the 7993 HCC disease targets were screened out between AiTongXiao granule drug and HCC disease targets.11 core targets including AKT1,IL6,TP53,MAPK3,TNF,JUN,CASP3,MAPK1,MYC,PTGS2,MMP9 were further obtained by median screening.GO and KEGG analysis results showed that these core targets enriched to HBV,TNF and cancer related pathways.The rat transplanted liver cancer model results indicated significant down regulation for AKT,p-AKT,pERK1/2,and significant up regulation of p-p53 after AiTongXiao granule treatment(P<0.05).Conclusion:AiTongXiao granule could act to multiple cancer related pathways,and AKT,p53 and ERK1/2 were validated to be regulated by ATXF in rat model.The mechanism may be through the regulation of the above signaling pathways to exert anti-liver cancer effect.

文摘In this study,the effect of sintering temperature and the addition of kaolin,a sintering agent,on the microscopic,phase,and mechanical properties of ceramics were investigated using secondary aluminum dross(SAD)as the main component in the manufacturing of ceramics.The basic phases of the ceramics were Al_(2)O_(3),MgAl_(2)O_(4),NaAl_(11)O_(17),and SiO_(2)without the addition of kaolin.The diffraction peaks of MgAl_(2)O_(4),NaAl_(11)O_(17),and SiO_(2)kept decreasing while those of Al_(2)O_(3)kept increasing with an increase in temperature.In addition,the increase in temperature promoted the growth of the grains.The grains were uniform in size and regular in distribution,with a shrinkage of 2.2%,porosity of 72.5%,bulk density of 1.076 g/cm^(3),and compressive strength of 1.12 MPa.When the sintering temperature was 1450°C,the basic phases of the ceramic after the addition of kaolin were Al_(2)O_(3),MgAl_(2)O_(4),NaAl_(11)O_(17),and SiO_(2).With the increase of kaolin,the diffraction peaks of NaAl_(11)O_(17)and SiO_(2)decreased until they disappeared,while the diffraction peaks of Al_(2)O_(3)increased significantly.When kaolin was added at 30 wt.%,the ceramics obtained had shrinkage of 18%,a porosity of 47.26%,a bulk density of 1.965 g/cm^(3),and compressive strength of 31.9 MPa.Cracks existed inside the ceramics without the addition of kaolin,while the addition of kaolin significantly changed this defect.It is shown that SAD can obtain porous ceramics with good properties at a sintering temperature of 1450°C and a kaolin addition of 30 wt.%.

文摘The objective of this study was to investigate the main active ingredients,potential targets,and possible mechanisms of action of the combination of Radix Astragali and Caulis Spatholobi for the treatment of atherosclerosis using network pharmacology.The study aimed to provide a reference basis for the development of new formulations and clinical use of Chinese medicine.The main components of Radix Astragali and Caulis Spatholobi were obtained from the TCMSP,BATMAN-TCM database,and literature reports.The targets corresponding to the main components were imported into the Uniprot database to standardize the names,and target information was supplemented with the Swiss Target Prediction database.Disease-related targets were obtained from DrugBank,OMIM,CTD,GeneCards,and DisGeNET online databases.Venn tools were used to obtain the potential targets of Radix Astragali and Caulis Spatholobi for the treatment of AS.The intersecting genes were imported into the STRING 11.5 database to construct protein-protein interaction network maps and analyze their interactions.Cytoscape 3.7.1 software was used to mine their core targets.GO function and KEGG signaling pathway enrichment analysis were performed using the DAVID v2023q1 database.The results were imported into the“Bioinformatics Cloud Platform”to generate enrichment bubble maps.Finally,the“component-target-pathway”diagram was constructed using Cytoscape 3.7.1 software.The study found that 78 major active ingredients and 527 potential targets were obtained from Radix Astragali and Caulis Spatholobi.The main active components of the two in combination for the treatment of AS are quercetin,stigmasterol,kaempferol,luteolin,formononetin,etc.The key targets involve CDKN1A,E2F1,CDK4,CDK2,CDK1,RB1,TP53,CDKN1B,IL6,JUN,etc.The main pathways involved the AGE-RAGE signaling pathway in diabetic complications,cancer pathway,etc.The biological processes involved include positive regulation of gene expression,negative regulation of apoptotic process,etc.The study initially verified the feasibility of the combination of Radix Astragali-Caulis Spatholobi by Qi-invigorating(promoting human metabolic activity)and blood-activating for the treatment of AS.It demonstrated that the combination of Chinese medicine has multi-level,multi-target,and multi-pathway mechanisms of action to treat the disease,providing a reference basis for the development and utilization of new drugs.

文摘Background:Traditional Chinese medicine(TCM)syndrome is the basic unit of TCM treatment,which help clinicians assess the disease progression and treatment preoperative of tumor patients.However,the prognostic significance of TCM syndrome is still unclear.This study aims to detect the differences in overall survival between different TCM syndrome and further develop a new nomogram with TCM syndrome for predicting overall survival in colorectal cancer.Methods:A total of 324 patients with colorectal cancer were enrolled and categorized into three groups based on TCM syndrome:deficiency,excess,and deficiency-excess.The prognosis of colorectal cancer patients with different TCM syndromes was evaluated using Kaplan-Meier analysis and Cox regression analysis.Results:The proportion of advanced stage and lymph metastasis in the patients with deficiency syndrome was higher,and the overall survival was shorter than other syndromes.Meanwhile,the TCM syndrome(P<0.001),tumor invasion depth(P<0.001),lymph metastasis(P=0.018),organic metastasis(P=0.005)and tumor node metastasis(TNM)stage(P=0.029)were the independent prognostic factor.Then,a new nomogram with TCM syndrome was established and assessed.324 colorectal cancer patients were randomly divided into training(n=215)and validation cohorts(n=109).A nomogram incorporating preoperative TCM syndrome,gender,age,T,N,and M status was developed,which had good discrimination and calibration.Conclusion:Taken together,our results indicated that TCM syndrome could assess the prognosis of colorectal cancer.The nomogram incorporating TCM syndromes and tumor information is helpful for risk stratification and prognostic predictions in colorectal cancer preoperatively.
文摘Na_(3)V_(2)(PO_(4))_(3)(NVP)cathode material of the sodium ion battery(1 C=117 mAh g-1)has a NASICON-type structure,which not only facilitates the rapid migration of sodium ions,but also has a small volume deformation during sodium ion de-intercalation and the main frame mechanism remains unchanged,and thus is seen as an energy storage material for a wide range of applications,but has a limited electronic conductivity due to its structure.In this paper,NVP cathode materials with finer primary particles are successfully prepared using a simple hydrothermal treatment-assisted sol-gel method.The increased pore size of the NVP materials prepared under the hydrothermal process allows for more active sites and more effective resistance to the volume deformation of sodium ions during insertion/extraction processes,effectively facilitating the diffusion of ions and electrons.The Na_(3)V_(2)(PO_(4))_(3) material obtained by the optimized process exhibited good crystallinity in XRD characterization,as well as superior electrochemical properties in a series of electrochemical tests.A specific capacitance of 106.3 mAh g^(-1) at 0.2 C is demonstrated,compared to 96.5 mAh g^(-1) for Na_(3)V_(2)(PO_(4))_(3) without hydrothermal treatment,and cycling performance is also improved with 93%capacity retention.The calculated sodium ion diffusion coefficient(DNa=5.68×10^(-14))obtained after EIS curve fitting of the improved sample illustrates that the pore structure is beneficial to the performance of the Na_(3)V_(2)(PO_(4))_(3)cathode material.

文摘Gas diffusion in the shale matrix has a dominant effect on late-stage production from shale gas reservoirs.However,adequate research on the mechanisms and contributions of gas diffusion for varied pore size populations in shale matrix under recreated in situ stress is lacking.We report gas-diffusion measurements under constant in situ stress but variable gas pressures for contrasting non-adsorbent(helium(He))and adsorbed(methane(CH_(4)))gases to investigate the impact of effective stress on the evolution of dominant mechanisms of diffusion.An intact sample replicates true pore-network topology and diffusion paths.An integrated diffusion model is proposed that combines the effects of slip flow,Knudsen flow,and surface diffusion to constrain the evolution of these flow regimes and their respective contributions to the observational data.Finally,a probability density function(PDF)is employed to separate the gas content distributions of macropores and micropores from the total gas content and to investigate gas contributions in various pores.The results reveal that the diffusion coefficients of both He and CH_(4) in macropores and micropores increase with gas pressure but decrease with increasing effective stress.The diffusion coefficients of He and CH_(4) are different in macropores but remain nearly the same in micropores.The diffusion coefficients of slip flow and surface diffusion increase with decreasing effective stress except for CH_(4) diffusion in the micropores,while the evolution of Knudsen diffusion shows the opposite trend.Slip flow plays a dominant role in He and CH_(4) diffusion within macropores(pore size 45 nm).Knudsen diffusion gradually becomes significant for He diffusion in the micropores(pore size 4 nm),conversely,for CH_(4) diffusion in the micropores,surface diffusion becomes significant.Related to gas production from reservoirs,the contributions of the micropores will increase gradually with the duration of gas recovery,indicating the significant role of gas diffusion in micropores to steady supply during latestage production.

文摘The influence of rare earth (RE) elements on the solidification process and eutectic transformation and mechanical properties of the high-V type cast, high-speed steel roll was studied. Test materials with different RE additions were prepared on a horizontal centrifugal casting machine. The solidification process, eutectic structure transformation, carbide morphology, and the elements present, were all investigated by means of differential scanning calorimetry (DSC) and scanning electron microscopy energy dispersive spectrometry (SEM-EDS). The energy produced by crack initiation and crack extension was analyzed using a digital impact test machine. It was found that rare earth elements increased the tensile strength of the steel by inducing crystallization of earlier eutectic γ-Fe during the solidification process, which in turn increased the solidification temperature and thinned the dendritic grains. Rare earth elements with large atomic radius changed the lattice parameters of the MC carbide by forming rare earth carbides. This had the effect of dispersing longpole M C carbides to provide carbide grains, thereby, reducing the formation of the gross carbide and making more V available, to increase the secondary hardening process and improve the hardness level. The presence of rare earth elements in the steel raised the impact toughness by changing the mechanism of MC carbide formation, thereby increasing the crack initiation energy.

文摘AIM To explore the role of macrophages in chronic pancreatitis(CP) and the effect of Dachaihu decoction(DCHD) on pancreatic fibrosis in mice.METHODS Kun Ming mice were randomly divided into a control group, CP group, and DCHD group. In the CP and DCHD groups, mice were intraperitoneally injected with 20% L-arginine(3 g/kg twice 1 d/wk for 6 wk). Mice in the DCHD group were administered DCHD intragastrically at a dose of 14 g/kg/d 1 wk after CP induction. At 2 wk, 4 wk and 6 wk post-modeling, the morphology of the pancreas was observed using hematoxylin and eosin, and Masson staining. Interleukin-6(IL-6) serum levels were assayed using an enzyme-linked immunosorbent assay. Double immunofluorescence staining was performed to observe the co-expression of F4/80 and IL-6 in the pancreas. Inflammatory factors including monocyte chemoattractant protein-1(MCP-1), macrophage inflammatory protein-1α(MIP-1α) and IL-6 were determined using real time-polymerase chain reaction. Western blot analysis was used to detect fibronectin levels in the pancreas. RESULTS Compared with the control group, mice with 20% L-arginine-induced CP had obvious macrophage infiltration and a higher level of fibrosis. IL-6 serum concentrations were significantly increased. Double immunofluorescence staining showed that IL-6 and F4/80 were co-expressed in the pancreas. With the administration of DCHD, the infiltration of macrophages and degree of fibrosis in the pancreas were significantly attenuated; IL-6, MCP-1 and MIP-1α m RNA, and fibronectin levels were reduced. CONCLUSION The dominant role of macrophages in the development of CP was mainly related to IL-6 production. DCHD was effective in ameliorating pancreatic fibrosis by inhibiting macrophage infiltration and inflammatory factor secretion in the pancreas.

文摘CS-CMC bipolar membrane was prepared and the cross-section photograph of CS-CMC BM was observed by SEM. FT-IR spectrum indicated that CS-CMC BM contained -N=CRH2^+ and -COO^- functional groups. The charge density of -N--CRH2^+ in CS membrane was about 14.13 mmol/g and the charge density of -COO in CMC membrane was about 9.01 mmol/g. The electrochemistry properties of CS-CMC BM were also studied. CS-CMC BM not only can effectively stop FeO4^2- from diffusing into the cathode chamber, but also plays an important role in the supply of OH" consumed during the electro-generated FeO4^2- process.

文摘This study aimed to explore the pathological change to hippocampal neurons and the expression of growth associated protein 43 in 21-day-old young rats following chronic intermittent hypoxia. Hematoxylin-eosin staining results showed varying degrees of degeneration and necrosis in hippocampal neurons depending on the modeling time. Immunohistochemistry revealed that growth associated protein 43 expression in young rats following chronic intermittent hypoxia decreased, but that levels were still higher than those of normal rats at each time point, especially 4 weeks after modeling. During 1 5 weeks after modeling, a slow growth in rat weight was observed. Experimental findings indicate that chronic intermittent hypoxia may induce growth dysfunction and necrosis of hippocampal neurons, as well as increase the expression of growth associated protein 43 in young rats.
