Chlorhexidine Digluconate Formulations Used for Skin Antisepsis

Aims: The representativeness of European standards phase 2, step 1 regarding bactericidal and yeasticidal activities was used for the comparison of two marketed antiseptic solutions, one containing chlorhexidine digluconate (0.5%) and the other combining chlorhexidine digluconate (0.25%), benzalkonium chloride (0.025%) and benzylic alcohol (4%). Methods: The bactericidal activity of the antiseptic solutions used pure or diluted was assessed according to the European standards NF EN 13727 and NF EN 13624 for the bactericidal and yeasticidal activity respectively. The contact time was 1 min at 20°C. Interfering substances used correspond to soiling conditions i.e. bovine serum albumin and sheep erythrocytes. A reduction of colony-forming units by ≥5 log10 was deemed to meet the requirements to conclude bactericidal activity and ≥4 log10 for yeasticidal activity. Results: Regarding all the mandatory strains, both solutions are bactericidal and yeasticidal even after a 40% dilution and even under “dirty” conditions. Conclusions: The present study demonstrated the efficient bactericidal and yeasticidal activity of aqueous solutions containing chlorhexidine digluconate either alone at a concentration of 0.5% (w/v) or at a concentration of 0.25% (w/v) when combined with benzalkonium chloride at 0.025% (w/v) and benzylic alcohol 4%. Nowadays, the circulation of poor quality medicines is becoming an alarming worldwide phenomenon with serious public health and socio-economic concerns. The situation is particularly critical in developing countries where drug quality assurance and regulatory systems for drug manufacturing, importation, distribution and sales are weak. A sustained vigilance on poor quality medicines that regroup counterfeit/falsified, substandard and degraded medicines is therefore required to ensure patient safety and genuine medicines integrity. A case situation is illustrated including a strategic approach and analytical tools that were found useful to detect poor quality medicines, identify unknown components, and timely alerts for appropriate measures against the spread of those harmful products. Several suspected medicines randomly sampled in several strategic Rwandan areas were firstly check-controlled by means of visual inspection and then applying several analytical techniques from simple to more complex ones. The following medicines were studied: quinine sulfate tablets, artemisinin-based combination tablets, and artesunate powders for injection. Taking into account the pharmaceutical forms and the chemical characteristics, the following tests were applied: uniformity of mass, friability, disintegration, fluorescence, identification and assay. They were followed by more complex analytical techniques that allowed more comprehension of abnormal findings among which the presence of a wrong active pharmaceutical ingredient in quinine sulfate tablets which is mainly discussed in this paper to illustrate a strategic approach and various analytical tools that can be used in detecting and identifying unknown component in poor quality medicines. 展开更多

Anatomical and TLC study of three Chinese peonies were conducted to make a comparative analysis. Peonies (Paeonia suffruticosa (tree peony), Paeonia lactiflora (Chinese peony) and Paeonia veitchii (Chinese peony)) are traditionally used on the Qinghai-Tibet Plateau in China. Recent studies have shown that the peonies have different pharmacological activities and clinical applications. To distinguish these three species of peonies and ensure the safety and effectiveness in their use, the microscopic characteristics and chromatographic profile of their roots and the corresponding powder were studied. Plant materials sectioned and stained and the raw powder were studied with an optical microscope using standard techniques in microscopy. The results of microscopic features and TLC were described and illustrated. The three species have different microscopic characteristics and TLC profiles, which allow us to distinguish them. In fact, with the help of features semi-quantitative and qualitative, an identification key was developed in our work and illustrated with photos and a table. The aim of our work was to show that the optical microscopy and related techniques provide a achievable practicality, which can be applied without ambiguity to the authentication of species peonies. 展开更多
