Glycosides of Cistanche(GC)is a preparation used extensively for its neuroprotective effect against neurological diseases,but its mechanisms of action remains incompletely understood.Here,we established a bilateral common carotid artery occlusion model of vascular dementia in rats and injected the model rats with a suspension of GC(10 mg/kg/day,intraperitoneally)for 14 consecutive days.Immunohistochemistry showed that GC significantly reduced p-tau and amyloid beta(Aβ)immunoreactivity in the hippocampus of the model rats.Proteomic analysis demonstrated upregulation of mitochondrial precursor protein and downregulation of keratin type II cytoskeletal6A after GC treatment compared with model rats that had received saline.Western blot assay confirmed these findings.Our results suggest that the neuroprotective effect of GC in vascular dementia occurs via the promotion of neuronal cytoskeleton regeneration. 展开更多

MiRNAs are integral for maintaining immune homeostasis and self-tolerance.In this study,qPCR analyses were performed to determine which miRNAs play an important role in wound healing.Next,an experiment in a model of wound healing was performed,and histology,mRNA expression and T-cell subpopulations in wound tissue were analyzed.The accelerated experiments were performed by local injection of either rIL-17A and/or rlL-9 after wound healing.In vitro,the differentiation of Thl7/Th9 in miR-155^+/+ or miR-155^-/- mice was investigated,and the target genes of miR155 were analyzed.From our findings,miR-155^-/- in mice promoted wound healing and weakened T cell-mediated inflammation,especially in IL-17/IL-9,and less severe skin fibrosis developed in the mice.rIL-17A and/or rIL-9 could exacerbate inflammatory injury and delay wound healing.We also demonstrated that miR-155^-/- led to a defect in the differentiation of Th17/Th9 in vitro,and this effect of IL-17/IL-9 might be related to the expression of C-maf,which is a target gene of miR155.MiR-155 regulated IL-17/IL-9-related inflammation in wound healing and might be a potential therapeutic target to attenuate the inflammatory response in wound tissue and promote the closure of wound injuries. 展开更多

Objective:To probe into the relationships between PTEN gene expression,the promoter methylation and gastric cancer and its clinical pathological specific features.Methods:We analyzed the PTEN gene promoter methylation and mRNA expression status in gastric cancer tissues and its adjacent normal tissues by methylation specific PCR(MSP) and reverse transcription-polymerase chain reaction(RT-PCR) techniques.Results:PTEN promoters in 48.2%(27/56) gastric cancer tissues and 3.6(2/56) adjacent normal tissues were methylated and the PTEN promoter methylation rate in carcinoma tissues was obviously higher(P < 0.05).Of the 2 cases where the adjacent gastric tissues were methylated,the gastric cancer tissues were both methylated.Of the 29 gastric cancers with lymph node metastasis,19 had their PTEN gene promoters methylated and the PTEN gene promoter methylation in cases with lymph node metastasis was obviously higher than that without lymph node metastasis(P < 0.05).RT-PCR result showed that no expression of PTEN mRNA existed in any of the methylated gastric cancer tissues.Conclusion:The expression loss of PTEN gene mRNA in gastric cancers is related to their promoter methylation and might be one of the reasons for the generation,development and metastasis of gastric cancers. 展开更多

BACKGROUND The tunnel endoscopic technique is the treatment of choice for submucosal tumors. However, the use of tunnel endoscopy to diagnose adenocarcinoma of the esophagus originating from the submucosa has not been well studied.CASE SUMMARY A 74-year-old man who presented with dysphagia for half a year underwent a series of checks, such as gastroendoscopy, X-ray contrast examination of the upper digestive tract, endoscopic ultrasonography, high-resolution esophageal manometry, and positron emission computed tomography. It should be noted that the stenosis of the esophagus was too narrow for endoscopic ultrasoundguided fine needle aspiration. The cause remained undiagnosed. Eventually, the tunnel endoscopic technique was perform for the pathological examination in the submucosa and the final diagnosis was adenocarcinoma of the esophagus. The patient and family members chose expectant treatment due to the patient's age and the high costs of surgical treatment.CONCLUSION Tunnel endoscopy could be used to diagnose tumors. Moreover, we review the literature to provide guidance regarding the causes of esophagostenosis. 展开更多

Summary: Glycoprotein (GP) Ibα ectodomain shedding has important implications for thrombosis and hemostasis. A disintegrin and metalloproteinase 17 (ADAM17) was identified to play an essen- tial role in agonist induced GPIbαshedding. The relationship of GPIbu shedding and ADAM 17 in the acute stage of atherosclerotic ischemic stroke (AIS) patients has not been thoroughly studied. A total of 306 patients and 230 controls matched for age, sex, race, history of hypertension and diabetes mel- litus were enrolled in the study. GPIbα, ADAM17, glycocalicin were detected by flow cytometry, Western blotting, and enzyme-linked immunosorbent assay (ELISA) respectively. Compared with the control group, the expression of GPIbα in patients with acute ischemic stroke was significantly lower (P=0.000, P〈0.01). Plasma glycocalicin and ADAM17 in AIS group were higher than those in con- trol group (P=0.699, P=0.000). Pearson's analysis showed glycocalicin bore no correlation with GPIbu in AIS patients (r=0.095, P〉0.05). GPIbα and National Institute of Health Stroke Scale (NIHSS) had negative correlation (r=-0.514, P〈0.01). Our findings indicate that ADAM 17 may be a risk factor for ischemic stroke in Chinese and the expression of GPIbα can serve as a measure for stroke severity. 展开更多

Methods: Experimental rats were divided into normal control group(NC group), diabetic control group (DC group )and diabetic group treated with 3-aminobenzamide (DT group ).Nerve conduction velocity (NCV), serum superoxide dismutase (SOD) activity and serum malondialdehyde (MDA) concentration,phosphocreatine (Pcr) ,creatine (Cr) concentration in sciatic nerves were evaluated after 4 weeks. Results: SOD,Pcr activity,and NCV were higher (P 〈 0.05) and MDA concentration were significantly lower in DT group, compared with DC group (P 〈 0.01 ). Meanwhile, ATP and Cr in sciatic nerves were similar in DT group, compared with DC group (P 〉 0.05). Conclusion: 3-aminobenzamide could alleviate the established functional and metabolic abnormalities of early DPN in the streptozotocin-induced diabetic rat models,which pro- vided a novel approach for prevention and treatment of diabetic neuropathy. 展开更多

To investigate whether LL-37 and human beta defensin-2 (hBD-2) is related to the patients with psoriasis seldom having skin infections and explore the role of the two peptides and CCR6 (the receptor of hBD-2) in the pathogenesis of psoriasis, the expression levels of mRNA of LL-37, hBD-2, and CCR6 in skin lesions of patients with psoriasis vulgaris were detected by using RT-PCR. The results showed that the mRNA expression levels of the two peptides and CCR6 in psoriatic lesions all increased compared with the normal skin (P<0.001). It was suggested that up-regulated expression of LL-37 and hBD-2 might be the main reason that result in the the skin of patients with psoriasis being seldom infected, and the two peptides and CCR6 might play crucial roles in the pathogenesis of psoriasis. 展开更多

Objective: To explore the effect of Daine-35 on serum hormone and antral follicle count of different subtypes of polycystic ovarian syndrome (PCOS). Methods: 138 cases of PCOS were recruited and typed according to Rotterdam diagnostic criteria;78 cases of tubal infertility without hyperandrogenism and menstrual disorders during the same period were collected as control. Serum reproductive hormones including follicle-stimulating hormone (FSH), luteotropic hormone (LH), prolactin (PRL), estradiol (E2), dehydroepiandrosterone (DHEAS), total testosterone (TT) and fast insulin (INS) were determined by chemiluminescent immunoassay. Ovarian antral follicle count was determined by ultrasonography. The effects on above indexes were evaluated for different subtypes of PCOS after 3 cycles of treatment with Daine-35. Results: The cases of Type A and Type B were respectively 43 (31.2%) and 73 (52.9%). They were the most common clinical phenotypes of PCOS in the central region ofChina. Total testosterone (TT) and DHEAS levels of Types A, C and D were higher than those of the control group (p < 0.05). The LH, LH/FSH, INS and HOMA-IR levels in all types of PCOS were higher than those of the control group (p < 0.05). Antral follicle counts of Types A, B and D were more than those of Type C and the control group (p < 0.05). After treatment with Daine-35 for 3 cycles, the serum TT levels of Types A, C and D decreased (p < 0.05). DHEAS values also decreased, but not significantly (p > 0.05). The serum LH and LH/FSH as well as the antral follicle count of all the PCOS types significantly decreased (p < 0.01). Conclusion: Type A and Type B are the most common clinical phenotypes of PCOS in the central region of China. Taking Daine-35 for 3 cycles can significantly reduce the serum androgen, serum LH, LH/FSH ratio, and antral follicle count of different PCOS types. 展开更多

Effect of epidermal growth factor on the migration of neural stem cells

BACKGROUND: Recently, researches on neural stem cells (NSCs) are focus on differentiation and migration ot stem cells. How to regulate and control differentiation and migration of NSCs based on human wills is still a hot topic.OBJECTIVE : To investigate the effect of epidermal growth factor (EGF) on the migration and proliferation of NSCs and analyze duration of the effect. DESIGN : Contrast study based on cells SETTING : Department of Neurological Surgery, the First Hospital of Wuhan MATERIALS: Healthy SD rats aged 13-14 embryonic days. EGF (Sigma Company) METHODS: The experiment was carried out in the Animal Laboratory of Experimental Center Affiliated to the First Hospital of Wuhan from October 2004 to July 2006. NSCs selected from embryonic striatum of rats with 13-14 embryonic days were cultured; 7 days later, suspended neural sphere was used to make simple cell suspension and cultured once more. Then, DMEM-F12+20 μg/L EGF was added into culture medium; 14 days later, the rats were divided into experimental group and control group. Rats in the experimental group were cultured with the same medium mentioned above; however, rats in the control group were cultured with only DMEM-F12. Migration of cells was observed under microscope every day. MAIN OUTCOME MEASURES: NSCs migration in both experimental group and control group RESULTS: Cell spheres in primary culture were NSCs. In addition, 14 days later, proliferation of stem cells were observed remarkably in EGF culture, and size of cell sphere was about that of 100 cells. In experimental group, proliferation of cell sphere was slow down on the 14^th culture day, and apophysis was erupted to neighbor cell sphere. Moreover, NSCs migrated from big cell sphere to small cell sphere during 14-17 culture days, and then, cell migration was disappeared at 17 days after culture. In control group, cell migration was not observed. CONCLUSION: EGF can induce proliferation and migration of NSCs during a special time (14-17 days). However, NSCs do not immigrate over the duration even they are caused by EGF. Migration has a transparent direction, i.e., from big cell sphere to small one. 展开更多

AIM: To characterize patterns of gastric cancer recurrence and patient survival and to identify predictors of early recurrence after surgery.METHODS: Clinicopathological data for 417 consecutive patients who underwent curative resection for gastric cancer were retrospectively analyzed. Tumor and node status was reclassified according to the 7th edition of the American Joint Committee on Cancer tumor-node-metastasis classification for carcinoma of the stomach. Survival data came from both the patients' follow-up records and telephone followups.Recurrent gastric cancer was diagnosed based on clinical imaging, gastroscopy with biopsy, and/or cytological examination of ascites, or intraoperative findings in patients who underwent reoperation.Predictors of early recurrence were compared in patients with pT1 and pT2-4a stage tumors. Pearson's χ 2 test and Fisher's exact test were used to compare differences between categorical variables. Survival curves were constructed using the Kaplan-Meier method and compared via the log-rank test. Variables identified as potentially important for early recurrence using univariate analysis were determined by multivariate logistic regression analysis.RESULTS: Of 417 gastric cancer patients, 80(19.2%)were diagnosed with early gastric cancer and the remaining 337(80.8%) were diagnosed with locally advanced gastric cancer. After a median follow-up period of 56 mo, 194 patients(46.5%) experiencedrecurrence. The mean time from curative surgery to recurrence in these 194 patients was 24 ± 18 mo(range, 1-84 mo). Additionally, of these 194 patients,129(66.5%) experienced recurrence within 2 years after surgery. There was no significant difference in recurrence patterns between early and late recurrence(P < 0.05 each). For pT1 stage gastric cancer, tumor size(P = 0.011) and pN stage(P = 0.048) were associated with early recurrence of gastric tumors.Patient age, pT stage, pN stage, Lauren histotype,lymphovascular invasion, intraoperative chemotherapy,and postoperative chemotherapy were independent predictors of early recurrence in patients with pT2-4a stage gastric cancer(P < 0.05 each).CONCLUSION: Age, pT stage, pN stage, Lauren histotype, lymphovascular invasion, intraoperative chemotherapy, and postoperative chemotherapy are independent factors influencing early recurrence of pT2-4a stage gastric cancer. 展开更多

The efficacy and applied value of endoscopic hematoma evacuation vs. external ventricular drainage (EVD) in the treatment of severe ventficular hemorrhage (IVH) were explored and compared.From Jan.2015 to Dec.2016,the clinical data of 42 cases of IVH were retrospectively analyzed,including 18 patients undergoing endoscopic hematoma evacuation (group A),and 24patients receiving EVD (group B).The hematoma clearance rate was calculated by 3D Slicer software,and complications and outcomes were compared between the two groups.There were no significant differences in age,sex and Graeb score between groups A and B (P>0.05).The hematoma clearance rate was 70.81%±27.64% in group A and 48.72%±36.58% in group B with a statistically significant difference (P<0.05).The operative time in groups A and B was 72.45±25.26 min and 28.54±15.27min,respectively (P<0.05).The Glasgow Coma Scale (GCS) score increased from 9.28±2.72 at baseline to 11.83±2.91 at 1 week postoperatively in group A,and from 8.25±2.62 at baseline to 10.79±4.12 at 1 week postoperatively in group B (P<0.05).The length of hospital stay was 12.67±5.97 days in group A and 17.33±8.91 days in group B with a statistically significant difference (P<0.05).The GOS scores at 6 months after surgery were 3.83±1.12 in group A,and 2.75±1.23 in group B (P<0.05). These results suggested that endoscopic hematoma evacuation has an advantage of a higher hematoma clearance rate,fewer complications and better outcomes in the treatment of severe IVH,indicating it is a safe,effective and promising approach for severe IVH. 展开更多

Plasmids GV144-MEG3 (MEG3 overexpression plasmid) and GV144 (control plasmid) were stably transfected into 786-0 cells by using lipofectamine 2000. Cell viabilities were determined by MTT, cell apoptosis rates by flow cytometry following PE Annexin V and 7AAD staining, apoptosis-related protein expressions by Western blotting, and Bcl-2 mRNA by RT-qPCR in the transfected cells. The results showed that MEG3 was evidently downregulated in RCC tissues (P〈0.05) and RCC cell lines (P〈0.05). The viabilities of 786-0 cells were decreased significantly after transfection with GV144-MEG3 for over 24 h (P〈0.05). Consistently, the apoptosis rate was significantly increased in 786-0 cells transfected with GV144-MEG3 for 48 h (P〈0.05). Furthermore, overexpression of MEG3 could reduce the expression of Bcl-2 and procaspase-9 proteins, enhance the expression of cleaved caspase-9 protein, and promote the release of cytochrome c protein to cytoplasm (P〈0.05). Additionally, Bcl-2 mRNA level was declined by MEG3 overexpression (P〈0.05). It was concluded that MEG3 induces the apoptosis of RCC cells possibly by activating the mitochondrial pathway. 展开更多

The present study examined von Willebrand factor (vWF) levels and ADAMTS13 activity in pregnant and severe preeclamptic women in order to shed light on the prothrombotic state in severe preeclampsia.Thirty healthy women of childbearing age,22 second trimester pregnant women,30 third trimester pregnant women and 10 severe preeclamptic patients were recruited in this study.ADAMTS13 activity was determined by the FRETS-vWF73 assay and vWF antigen (vWF:Ag) levels by an enzyme-linked immunosorbent assay.The results showed that there were statistically significant differences in plasma vWF antigen levels between the severe preeclamptic and third trimester pregnant women,between third and second trimester pregnant women (P【0.05).The third trimester pregnant women had significantly lower plasma ADAMTS13 activity than second trimester pregnant women (P【0.05).Nevertheless,no significant differences in plasma ADAMTS13 activity were found between severe preeclamptic patients and the third trimester pregnant women (P】0.05).In conclusion,plasma ADAMTS13 activity is normal in severe preeclampsia despite the increased vWF:Ag levels.Prothrombotic state is involved in the pathogenesis of severe preeclampsia,as a result of endothelial injury. 展开更多

Eukaryotic initiation factor 5A2(eIF5A2),as one of the two isoforms in the family,is reported to be a novel oncogenic protein that is involved in multiple aspects of many types of human cancer.Overexpression or gene amplification of EIF5A2 has been demonstrated in many cancers.Accumulated evidence shows that eIF5A2 initiates tumor formation,enhances cancer cell growth,increases cancer cell metastasis,and promotes treatment resistance through multiple means,including inducing epithelial–mesenchymal transition,cytoskeletal rearrangement,angiogenesis,and metabolic reprogramming.Expression of eIF5A2 in cancer correlates with poor survival,advanced disease stage,as well as metastasis,suggesting that eIF5A2 function is crucial for tumor development and maintenance but not for normal tissue homeostasis.All these studies suggest that eIF5A2 is a useful biomarker in the prediction of cancer prognosis and serves as an anticancer molecular target.This review focuses on the expression,subcellular localization,post-translational modifications,and regulatory networks of eIF5A2,as well as its biochemical functions and evolving clinical applications in cancer,especially in human digestive system neoplasms. 展开更多

BACKGROUND Paradoxical psoriasis induced by tumor necrosis factor alpha antagonists is a rare side effect of those drugs and has similarities with and differences from classical psoriasis in clinical and pathological characteristics.Treating severe paradoxical psoriasis is challenging because the reported cases are rare,with treatment experience being only anecdotal.CASE SUMMARY We report 2 cases of paradoxical psoriasis caused by infliximab.Both cases manifested with a significant number of pustular lesions and had protracted and complicated clinical courses.In case 1,secukinumab alone could not control the eruptions,but colchicine supplementation markedly decreased disease activity.In case 2 miscellaneous medications were administered,including the systemic drug acitretin,the immunosuppressive drug cyclosporine,and the biologic agent ustekinumab.However,multiple applications of those medications failed to prevent new lesions from occurring.Both cases showed moderate-to-high antinuclear antibody titers.CONCLUSION Based on these cases,moderate-to-high anti-nuclear antibody titer seems to be a risk factor for paradoxical psoriasis.In addition,extensive pustular presentation may be a negative prognostic indicator and may portend a protracted clinical course refractory to therapy. 展开更多

In this case-control study,the relationship between M196R(676 T→G) variant in exon 6 of tumor necrosis factor receptor type 2(TNFR2) gene and genetic susceptibility of acne vulgaris in Han Chinese was investigated.A total of 93 acne vulgaris patients and 90 healthy subjects from Han Chinese ethnic group were enrolled in this study.Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) technique was adopted to analyze the single nucleotide polymorphisms(SNPs) of TNFR2 M196R gene,and to examine the association between acne vulgaris and the polymorphisms in TNFR2 M196R gene.The relationship between different genotypes and the susceptibility of acne vulgaris was analyzed.The results showed that there was significant difference in the frequency of the genotype M/R+R/R in the TNFR2 M196R genetic polymorphisms between acne vulgaris patients and healthy controls(χ2=4.343;P=0.037;OR=1.899;95% CI:1.036-3.445);and there was significant difference in the allele(R) frequency between acne vulgaris patients and healthy controls(χ2=5.588;P=0.018;OR=1.838;95% CI:1.105-3.057).It was concluded that the high frequency of 196R allele in the functional M196R polymorphism of TNFR2 is a risk factor for acne vulgaris in Han Chinese. 展开更多

MicroRNAs(miRNAs)are a novel class of small,non-coding RNAs that play a significant role in both inflammatory and cardiovascular diseases.Immune cells,especially T helper(Th)cells,are critical in the development of atherosclerosis and the onset of acute coronary syndrome(ACS).To assess whether inflammation-related miRNAs(such as miR-155,146a,21,125a-5p,125b,31)are involved in the imbalance of Th cell subsets in patients with ACS,we measured the expression of related miRNAs in patients with acute myocardial infarction(AMI),unstable angina(UA),stable angina(SA)and chest pain syndrome(CPS);analyzed the relationship between miRNA expression and the frequency of Th cell subsets;and observed the co-expression of miR-155 and IL-17A in peripheral blood mononuclear cells(PBMCs)of patients with ACS.The results showed that the expression of miR-155 in the PBMCs of patients with ACS was decreased by approximately 60%,while the expression of both miR-21 and miR-146a was increased by approximately twofold.The expression patterns of miRNAs in plasma correlated with those in PBMCs,except for miR-21,which was increased by approximately sixfold in the AMI group and showed no significant difference between the UA group and the CPS group.We also found that the expression of miR-155 inversely correlated with the frequency of Th17 cells(r520.896,P,0.01)and that miR-155 was co-expressed with IL-17A in patients with ACS.In conclusion,our study revealed the expression patterns of inflammation-related miRNAs in patients with ACS and found that miR-155 may be associated with Th17 cell differentiation. 展开更多
