Investigating the molecular mechanisms of oocyte maturation and ovulation in Nile tilapia:A focus on the steroidogenic enzyme Cyp17a2

Previous research has highlighted the significant role of progestins and glucocorticoids in fish oocyte maturation and ovulation.To clarify the molecular mechanisms underlying these processes,comprehensive investigations were conducted using a cyp17a2 mutant Nile tilapia(Oreochromis niloticus) model.Analysis revealed pronounced Cyp17a2 expression in ovarian somatic cells of the tilapia.Female cyp17a2-deficient mutants exhibited markedly reduced levels of 17,20β-dihydroxy-4-pregnen-3-one(DHP) and cortisol/cortisone,leading to delayed meiotic initiation and impaired oocyte maturation and spawning.Notably,supplementation with human chorionic gonadotrophin(hCG),DHP,and cortisol effectively induced germinal vesicle breakdown(GVBD) and facilitated oocyte release with follicular cell layers in cyp17a2^(-/-)females.Additionally,cyp17a2-/-and rescued cyp17a2-/-females showed elevated transcription of steroidogenic enzymes involved in 17β-estradiol(E2)production compared to spawning wild-type females.Moreover,the reduction in Akt phosphorylation observed in cyp17a2-deficient females and upon inhibitor treatment impaired hCG-induced oocyte maturation.Conversely,activation of the phosphoinositide 3-kinase/protein kinase B(PI3K-Akt) signaling pathway partially rescued the oocyte maturation impairment caused by cyp17a2mutation.Overall,these findings provide functional evidence supporting the critical role of Cyp17a2 in DHP and cortisol biosynthesis,which,in turn,facilitates oocyte maturation and ovulation through activation of the PI3K-Akt signaling pathway in fish. 展开更多

Fishing boats are usually anchored side by side in the harbor because of the small structural size and poor resistance to wind and waves.A series of physical model experiments are conducted to investigate the motion characteristics of multiple fishing boats that are moored together.A decay test in calm water is conducted to study the natural period and damping coefficients.Regular wave experiments are performed to analyze the roll motion response of each boat for four modes(different numbers of boats side-by-side).The results indicate that the“natural period”of each boat for the mode of multi-boats especially three or four boats,is slightly smaller than that of a single boat,whereas the damping coefficient is visibly larger than that of a single boat.The maximum roll angle of each boat does not appear at the same time under a 90°incident wave.Small roll motion energy is generated at low frequencies and high frequencies when multiple boats are moored together.The energy decreases with the increasing wave period.The roll motion responses of each boat in four modes exhibit different trends with the increasing wave frequency.The number of boats and boat position have significant effects on roll motion. 展开更多

Glass catfish(Kryptopterus vitreolus)are notable in the aquarium trade for their highly transparent body pattern.This transparency is due to the loss of most reflective iridophores and light-absorbing melanophores in the main body,although certain black and silver pigments remain in the face and head.To date,however,the molecular mechanisms underlying this transparent phenotype remain largely unknown.To explore the genetic basis of this transparency,we constructed a chromosome-level haplotypic genome assembly for the glass catfish,encompassing 32 chromosomes and 23344 protein-coding genes,using PacBio and Hi-C sequencing technologies and standard assembly and annotation pipelines.Analysis revealed a premature stop codon in the putative albinism-related tyrp1b gene,encoding tyrosinase-related protein 1,rendering it a nonfunctional pseudogene.Notably,a synteny comparison with over 30 other fish species identified the loss of the endothelin-3(edn3b)gene in the glass catfish genome.To investigate the role of edn3b,we generated edn3b^(−/−)mutant zebrafish,which exhibited a remarkable reduction in black pigments in body surface stripes compared to wild-type zebrafish.These findings indicate that edn3b loss contributes to the transparent phenotype of the glass catfish.Our high-quality chromosome-scale genome assembly and identification of key genes provide important molecular insights into the transparent phenotype of glass catfish.These findings not only enhance our understanding of the molecular mechanisms underlying transparency in glass catfish,but also offer a valuable genetic resource for further research on pigmentation in various animal species. 展开更多

Two predominant currents, the warm Kuroshio Current and the cold Oyashio Current, meet in the North- west Pacific Ocean. The dynamics of physical oceanographic structures in this region, including frontal zones and meandering eddies, result in a highly productive habitat that serves as a favorable feeding ground for various commercially important species. Neon flying squid, Ommastrephes bartramii, is an im- portant oceanic squid, which is widely distributed in the North Pacific Ocean. Based on the catch data col- lected by Chinese squid jigging fleets and relevant environmental data, including sea surface temperature (SST) and fronts (represented by gradients of SST and thermocline) during 1998-2009, the variations of oceanic fronts and their influence on the fishing grounds of O. bartramii were evaluated, and the differ- ences in distribution of fishing grounds of O. bartramii in 2000 and 2002 were compared by describing the differences in vertical temperature between 0-300 m. It was found that the preferred horizontal tem- perature gradient of SST for O. bartramii tended to be centered at 0.01-0.02~C/nm, which attracted nearly 80% of the total fishing effort, and the preferred horizontal temperature gradients at the 50 m and 105 m layers were mainly located at 0.01-0.03~C/nm, which accounted for more than 70% of the total fishing effort during August-October. The preferred vertical temperature gradient within the 0-50 m layer for O. bartramii tended to be centered at 0.15-0.25~C/m during August and September and at 0.10-0.15~C/m in October, implying that the mixed surface layer was distributed at depths of 0-50 m. It was concluded that the vertical temperature gradient was more important than the horizontal temperature gradient in playing a role in forming the fishing ground. The results improved our understanding of the spatial dynamics of the 0. bartramii fishery. 展开更多

Objective:To evaluate the antibacterial effect of aqueous and ethanolic moringa leaf extracts (Moringa oleifera) on the growth of gram-positive and negative bacteria.Methods:Paper disks were soaked with 100,200,300 and 400μL of extract at 20 g/180mL and 10 g/190 mL.All extracts were tested against Escherichia coli(ATCC25922),Staphylococcus aureus(ATCC25923),Vibrio parahaemolyticus,Enterococcus faecalis(ATCC29212),Pseudomonas aeruginosa(ATCC27853), Salmonella enteritidis(IH) and Aeromonas caviae.The susceptibility tests were performed using the modified disk diffusion method.Results:The strains E.coli,P.aeruginosa and S.enteritidis (IH) were resistant to all treatments.In general,disks with 400μL extract were the most efficient against S.aureus,V.parahaemolyticus,E.faecalis and A.caviae.Conclusions:The study indicates a promising potential for aqueous and ethanolic Moringa leaf extracts as alternative treatment of infections caused by the tested strains. 展开更多

During 1995-2011, annual production of winter-spring cohort of Ommastrephes bartramii for Chinese squidjigging fishery has greatly fluctuated, which is closely related to the environmental conditions on the spawning and fishing grounds. To better understand how squid recruitment and abundance were infuenced by ocean environmental conditions, biological and physical environmental variables including sea surface temperature (SST), SST anomaly (SSTA), chlorophyll a (Chl a) concentration and the Kuroshio Current were examined during years with the highest (1999), intermediate (2005), and lowest (2009) catches. Catch per unit effort (CPUE) of the squid-jigging vessels was used as an indicator of squid abundance. The results indicated that high SST and Chl a concentration on the spawning ground in 1999 resulted in favorable incubation and feeding conditions for squid recruitment. Whereas the suitable spawning zone (SSZ) in 2009 shifted southward and coincided with low SST and Chl a concentration, resulting in a reduction in the squid recruitment. The small difference of SSZ area in the three years suggested the SSZ provided limited influences on the variability in squid recruitment. Furthermore, high squid abundance in 1999 and 2005 was associated with warm SSTA on the fishing ground. While the cool SSTA on the fishing ground in 2009 contributed to adverse habitat for the squid, leading to extremely low abundance. It was inferred that strengthened intensity of the Kuroshio force generally yielded favorable environmental conditions for O. bartramii. Future research are suggested to focus on the fundamental research oil the early life stage of O. bartramii and mechanism of how the ocean-climate variability affects the squid abundance and spatial distribution by coupling physical model with squid biological process to explore transport path and abundance distribution. 展开更多

The generalized linear model (GLM) and generalized additive model (GAM) were applied to the standardization of catch per unit effort (CPUE) for Chilean jack mackerel from Chinese factory trawl fishing fleets in the Southeast Pacific Ocean from 2001 to 2010 by removing the operational, environmental, spatial and temporal impacts. A total of 9 factors were selected to build the GLM and GAM, i.e., Year, Month, Vessel, La Nifia and E1 Nifio events (ELE), Latitude, Longitude, Sea surface temperature (SST), SST anomaly (SSTA), Nino3.4 index and an interaction term between Longitude and Latitude. The first 5 factors were significant components in the GLM, which in combination explained 27.34% of the total variance in nominal CPUE. In the stepwise GAM, all factors explained 30.78% of the total variance, with Month, Year and Vessel as the main factors influencing CPUE. The higher CPUE occurred during the period April to July at a SST range of 12-15℃ and a SSTA range of 0.2-1.0℃. The CPUE was significantly higher in normal years compared with that in La Nifia and E1 Nifio years. The abundance of Chilean jack mackerel declined during 2001 and 2010, with an increase in 2007. This work provided the relative abundance index of Chilean jack mackerel for stock as- sessment by standardizing catch and effort data of Chinese trawl fisheries and examined the influence of temporal, spatial, environ- mental and fisheries operational factors on Chilean jack mackerel CPUE. 展开更多

The spatial scale(?shing grid) of ?sheries research af fects the observed spatial patterns of?sheries resources such as catch-per-unit-ef fort(CPUE) and ?shing ef fort. We examined the scale impact of high value(HH) clusters of the annual ?shing ef fort for Dosidicus gigas of fshore Peru from 2009 to 2012.For a multi-scale analysis, the original commercial ?shery data were tessellated to twelve spatial scales from 6′ to 72′ with an interval of 6′. Under these spatial scales, D. gigas clusters were identi?ed using the Anselin Local Moran's I. Statistics including the number of points, mean CPUE, standard deviation(SD),skewness, kurtosis, area and centroid were calculated for these HH clusters. We found that the z-score of global Moran's I and the number of points for HH clusters follow a power law scaling relationship from2009 to 2012. The mean ef fort and its SD also follow a power law scaling relationship from 2009 to 2012.The skewness follows a linear scaling relationship in 2010 and 2011 but ?uctuates with spatial scale in2009 and 2012; kurtosis follows a logarithmic scale relationship in 2009, 2011 and 2012 but a linear scale relationship in 2010. Cluster area follows a power law scaling relationship in 2010 and 2012, a linear scaling relationship in 2009, and a quadratic scaling relationship in 2011. Based on the peaks of Moran's I indices and the multi-scale analysis, we conclude that the optimum scales are 12′ in 2009 ? 2011 and 6′ in 2012, while the coarsest allowable scales are 48′ in 2009, 2010 and 2012, and 60′ in 2011. Our research provides the best spatial scales for conducting spatial analysis of this pelagic species, and provides a better understanding of scaling behavior for the ?shing ef fort of D. gigas in the of fshore Peruvian waters. 展开更多

Modeling Monthly Spatial Distribution of Ommastrephes bartramii CPUE in the Northwest Pacific and Its Spatially Nonstationary Relationships with the Marine Environment 被引量:5

There are substantial spatial variations in the relationships between catch-per-unit-effort(CPUE) and oceanographic conditions with respect to pelagic species. This study examines the monthly spatiotemporal distribution of CPUE of the neon flying squid, Ommastrephes bartramii, in the Northwest Pacific from July to November during 2004–2013, and analyzes the relationships with oceanographic conditions using a generalized additive model(GAM) and geographically weighted regression(GWR) model. The results show that most of the squids were harvested in waters with sea surface temperature(SST) between 7.6 and 24.6℃, chlorophyll-a(Chl-a) concentration below 1.0 mgm^(-3), sea surface salinity(SSS) between 32.7 and 34.6, and sea surface height(SSH) between-12.8 and 28.4 cm. The monthly spatial distribution patterns of O. bartramii predicted using GAM and GWR models are similar to observed patterns for all months. There are notable variations in the local coefficients of GWR, indicating the presence of spatial non-stationarity in the relationship between O. bartramii CPUE and oceanographic conditions. The statistical results show that there were nearly equal positive and negative coefficients for Chl-a, more positive than negative coefficients for SST, and more negative than positive coefficients for SSS and SSH. The overall accuracies of the hot spots predicted by GWR exceed 60%(except for October), indicating a good performance of this model and its improvement over GAM. Our study provides a better understanding of the ecological dynamics of O. bartramii CPUE and makes it possible to use GWR to study the spatially nonstationary characteristics of other pelagic species. 展开更多

The cephalopod beak is a vital hard structure with a stable configuration and has been widely used for the identification of cephalopod species. This study was conducted to determine the best standardization method for identifying different species by measuring 12 morphological variables of the beaks of Illex argentinus, Ommastrephes bartramii, and Dosidicus gigas that were collected by Chinese jigging vessels. To remove the effects of size, these morphometric variables were standardized using three methods. The average ratios of the upper beak morphological variables and upper crest length of O. bartramii and D. gigas were found to be greater than those of I. argentinus. However, for lower beaks, only the average of LRL(lower rostrum length)/LCL(lower crest length), LRW(lower rostrum width)/LCL, and LLWL(lower lateral wall length)/LCL of O. bartramii and D. gigas were greater than those of I. argentinus. The ratios of beak morphological variables and crest length were found to be all significantly different among the three species(P < 0.001). Among the three standardization methods, the correct classification rate of stepwise discriminant analysis(SDA) was the highest using the ratios of beak morphological variables and crest length. Compared with hood length, the correct classification rate was slightly higher when using beak variables standardized by crest length using an allometric model. The correct classification rate of the lower beak was also found to be greater than that of the upper beak. This study indicates that the ratios of beak morphological variables to crest length could be used for interspecies and intraspecies identification. Meanwhile, the lower beak variables were found to be more effective than upper beak variables in classifying beaks found in the stomachs of predators. 展开更多

The mitre squid(U roteuthis chinensis) and the swordtip squid( U. edulis) are Indo-Pacific cephalopod species that are abundant in the western Pacific Ocean. They are currently exploited in the East and South China Seas and make up a significant portion of the Chinese neritic squid catch. Beaks, the feeding organs of squid, are important for individual size and biomass estimation because of their high resistance to degradation in predator stomachs and consistent dimensions. In this study, 104 U. chinensis and 143 U. edulis individuals were sampled from northern South China Sea with mantle length from 70 to 260 mm and 96 to 284 mm, respectively. The results indicated that morphological beak values were greater for U. edulis, compared to U. chinensis, for upper hood length(UHL), upper crest length(UCL), upper lateral wall length(ULWL), lower crest length(LCL), and lower lateral wall length(LLWL). According to principal component analysis, UHL/ML, UCL/ML, ULWL/ML, LCL/ML, LLWL/ML and LWL/ML could represent the characteristics of beaks for U. chinensis, while UHL/ML, UCL/ML, ULWL/ML, LHL/ML, LCL/ML and LLWL/ML could represent it for U. edulis. According to Akaike's information criterion(AIC) values, a power function was the most suitable model for U. chinensis, while a linear function was the most suitable model for U. edulis. The beak variable-mantle length ratio(beak variable/mantle length) declined with the increasing of mantle length and declined sharply at the early stage of growth in both beaks and species. The ratio changed quickly after achieving the mantle length of 140 mm for U. chinensis, while the ratio changed quickly after 170 mm for U. edulis. Beaks in both species experienced sharper changes through maturity stage I to II than other maturity stages. This study gives us basic beak morphology information for U. chinensis and U. edulis in the East and South China Seas. Geometric morphological methods combined with dietary analysis should be used in the future. 展开更多

The daily catch of D. gigas in the two survey cruises ranged from 0 to 5.5 t and was mostly obtained from the areas bounded by 6°–9°N and 91°–94°W and by 6°30′–7°30′N and 96°–97°W. The sea surface temperature in the areas yielding the most catch ranged from 27.5 to 29℃. The sex ratio of the total catch was 3.75:1(female: male). The mantle length of the squid ranged from 211 to 355 mm(male) and from 204 to 429 mm(female) with an average of 297.9 and 306.7 mm, respectively. In the relationship of the mantle length(mm) and body weight(g) of the squid, there was no significant difference between sexes. The female and male were at a similar maturity, and most individuals are maturing or have matured with a few females being spent. The size(mantle length) and age at the first sexual maturity were 297 mm and 195 d in females, and less than 211 mm and 130 d in males, respectively. Most of the sampled stomachs(70.6%) had no food remains. The major preys of the squids were fish, cephalopods and crustaceans, with the most abundant Myctophum orientale and D. gigas. The preys in more than 65% of the non-empty sampled stomachs evidenced the cannibalism of D. gigas. The results improved current understanding of the fishery biology of D. gigas off the Costa Rica Dome, which may facilitate the assessment and management of relative fishery resources. 展开更多

We examined the scale impacts on spatial hot and cold spots of CPUE for Ommastrephes bartramii in the northwest Pacific Ocean. The original fishery data were tessellated to 18 spatial scales from 5′×5′ to 90′×90′ with a scale interval of 5′ to identify the local clusters. The changes in location, boundaries, and statistics regarding the Getis-Ord Gi* hot and cold spots in response to the spatial scales were analyzed in detail. Several statistics including Min, mean, Max, SD, CV, skewness, kurtosis, first quartile(Q1), median, third quartile(Q3), area and centroid were calculated for spatial hot and cold spots. Scaling impacts were examined for the selected statistics using linear, logarithmic, exponential, power law and polynomial functions. Clear scaling relations were identified for Max, SD and kurtosis for both hot and cold spots. For the remaining statistics, either a difference of scale impacts was found between the two clusters, or no clear scaling relation was identified. Spatial scales coarser than 30′ are not recommended to identify the local spatial patterns of fisheries because the boundary and locations of hot and cold spots at a coarser scale are significantly different from those at the original scale. 展开更多

The Humboldt squid Dosidicus gigas has a short life span, and environmental variability plays a significant role in regulating its population dynamics and distribution. An analysis of 1 096 samples of D. gigas collected by the Chinese commercial fishing vessels during 2013, 2014, and 2016 off the Peruvian Exclusive Economic Zone, was conducted to evaluate the impacts of El Nino events on the somatic condition of D. gigas. This study indicates that the slopes of all beak variables in relation to mantle length (ML) for females were greater than those of males during 2013, 2014, and 2016, and slopes of the upper crest length and the lower rostrum length significantly differed between females and males in 2013 (P<0.05). Variation in the slopes for beak variables among years was studied;no significant difference was observed (ANCOVA, P>0.05). The Fulton's condition coefficients (K) of females and males in 2013 and 2014 were significantly greater than those in 2016 (P<0.01). The K values of females were greater than those of males in 2013, 2014, and 2016, and K values significantly differed between females and males in 2013. In normal years, the chlorophyll a (Chl a) concentration showed an N-shaped variability from January to December. However, in the El Nino period, it tended to weaken the upwelling coupled with warm and low Chl a concentration waters. We suggest that the poor somatic condition of D. gigas during the El Nino year was resulted from the low Chl a concentration in the waters, and the abundance of D. gigas would decrease due to the unfavourable environment and the lack of prey items in the El Nino year. 展开更多

The modern fishery stock assessment could be conducted by various models,such as Stock Synthesis model with high data requirement and complicated model structure,and the basic surplus production model,which fails to incorporate individual growth,maturity,and fishery selectivity,etc.In this study,the Just Another Bayesian Biomass Assessment(JABBA)Select which is relatively balanced between complex and simple models,was used to conduct stock assessment for yellowfin tuna(Thunnus albacares)in the Atlantic Ocean.Its population dynamics was evaluated,considering the influence of selectivity patterns and different catch per unit effort(CPUE)indices on the stock assessment results.The model with three joint longline standardized CPUE indices and logistic selectivity pattern performed well,without significant retrospective pattern.The results indicated that the stock is not overfished and not subject to overfishing in 2018.Sensitivity analyses indicated that stock assessment results are robust to natural mortality but sensitive to steepness of the stock-recruitment relationship and fishing selectivity.High steepness was revealed to be more appropriate for this stock,while the fishing selectivity has greater influence to the assessment results than life history parameters.Overall,JABBA-Select is suitable for the stock assessment of Atlantic yellowfin tuna with different selectivity patterns,and the assumptions of natural mortality and selectivity pattern should be improved to reduce uncertainties. 展开更多

The goal of this paper is to provide a comparative analysis of two commonly used approaches to discretize offshore fish cages: the lumped-mass approach and the finite element technique. Two case studies are chosen to compare predictions of the LMA(lumped-mass approach) and FEA(finite element analysis) based numerical modeling techniques. In both case studies, we consider several loading conditions consisting of different uniform currents and monochromatic waves. We investigate motion of the cage, its deformation, and the resultant tension in the mooring lines. Both model predictions are sufficient close to the experimental data, but for the first experiment, the DUT-Flex Sim predictions are slightly more accurate than the ones provided by Aqua-FETM. According to the comparisons, both models can be successfully utilized to the design and analysis of the offshore fish cages provided that an appropriate safety factor is chosen. 展开更多

Spatial-temporal distribution of marine fishes is strongly influenced by environmental factors.To obtain a more continuous distribution of these variables usually measured by stationary sampling designs,spatial interpolation methods(SIMs)is usually used.However,different SIMs may obtain varied estimation values with significant differences,thus affecting the prediction of fish spatial distribution.In this study,different SIMs were used to obtain continuous environmental variables(water depth,water temperature,salinity,dissolved oxygen(DO),p H,chlorophyll a and chemical oxygen demand(COD))in the Changjiang River Estuary(CRE),including inverse distance weighted(IDW)interpolation,ordinary Kriging(OK)(semivariogram model:exponential(OKE),Gaussian(OKG)and spherical(OKS))and radial basis function(RBF)(regularized spline function(RS)and tension spline function(TS)).The accuracy and effect of SIMs were cross-validated,and two-stage generalized additive model(GAM)was used to predict the distribution of Coilia nasus from 2012 to 2014 in CRE.DO and COD were removed before model prediction due to their autocorrelation coefficient based on variance inflation factors analysis.Results showed that the estimated values of environmental variables obtained by the different SIMs differed(i.e.,mean values,range etc.).Cross-validation revealed that the most suitable SIMs of water depth and chlorophyll a was IDW,water temperature and salinity was RS,and p H was OKG.Further,different interpolation results affected the predicted spatial distribution of Coilia nasus in the CRE.The mean values of the predicted abundance were similar,but the differences between and among the maximum value were large.Studies showed that different SIMs can affect estimated values of the environmental variables in the CRE(especially salinity).These variations further suggest that the most applicable SIMs to each variable will also differ.Thus,it is necessary to take these potential impacts into consideration when studying the relationship between the spatial distribution of fishes and environmental changes in the CRE. 展开更多

The southern Patagonian stock(SPS) of Argentinian shortfin squid, Illex argentinus, is an economically important squid fishery in the Southwest Atlantic. Environmental conditions in the region play an important role in regulating the population dynamics of the I. argentinus population. This study develops an environmentally dependent surplus production(EDSP) model to evaluate the stock abundance of I. argentines during the period of 2000 to 2010. The environmental factors(favorable spawning habitat areas with sea surface temperature of 16–18°C) were assumed to be closely associated with carrying capacity(K) in the EDSP model. Deviance Information Criterion(DIC) values suggest that the estimated EDSP model with environmental factors fits the data better than a Schaefer surplus model without environmental factors under uniform and normal scenarios.The EDSP model estimated a maximum sustainable yield(MSY) from 351 600 t to 685 100 t and a biomass from 1 322 400 t to1 803 000 t. The fishing mortality coefficient of I. argentinus from 2000 to 2010 was smaller than the values of F(0.1) and F(MSY). Furthermore, the time series biomass plot of I. argentinus from 2000 to 2010 shows that the biomass of I.argentinus and this fishery were in a good state and not presently experiencing overfishing. This study suggests that the environmental conditions of the habitat should be considered within squid stock assessment and management. 展开更多
