The hydrogen production from acetate is lower than that from lactate, probably because of its low ability to metabolize acetate. In this study, gene of acetaldehyde dehydrogenase (ACDH, EC 1.2.1.10) that catalyzes the reversible conversion of acetaldehyde and CoA to acetyl-CoA with the concurrent reduction of NAD to NADH, is overexpressed in the RV strain. The produced acetyl-CoA can be oxidized to carbon dioxide in the tricarboxylic acid (TCA) cycle, wherein electrons are generated and used for hydrogen production. The byproduct NADH can be used as reducing agent for acetate to produce acetaldehyde by acetate dehydrogenase. The recombinant RV strain (RVAC) expressing the ACDH gene showed ACDH activity with a specific activity of 3.2 mU/ mg, and the RV and the recombinant RV strain that harbored the intact (empty) plasmid pLP-1.2 (RVI) showed no detectable ACDH activity. The hydrogen yields of the RVAC strain from 21-mM acetate were 1.5-fold higher than that of the wild type RV strain and also that of the RVI strain. In contrast, hydrogen yield from 21-mM lactate was 30% lower than that in the control strains.展开更多

The absorption maximum of subphthalocyanines, called the Q band, appears around 560 - 630 nm, which is shifted by approximately 100 nm to shorter wavelengths compared to phthalocyanines. Subphthalocyanines are used as precursors to prepare unsymmetric phthalocyanines for ring enlargement reaction. In this decade, phthalocyanines are used for dye-sensitized solar cells (DSSCs), which require strong absorption of far-red light between 700 and 850 nm because of their highly efficiency. Non-peripheral thioaryl-substituted phthalocyanines have been synthesized. They show near-infrared absorption around 780 - 870 nm and have excellent electron transfer properties. However, their lack of affinity to basal plats inhibits their use as DSSC photosensitizer. Therefore, to synthesize unsymmetrical non-peripheral thioaryl-substituted phthalocyanines possessing good affinity to basal plates, the authors prepared subphthalocyanines having thioaryl substituents as precursors. Spectroscopic properties and electron transfer abilities to synthesize non-peripheral thioaryl-substituted subphthalocyanines were estimated using cyclic voltammetry. The Q band of non-peripheral thioaryl-substituted subphthalocyanines shows around 650 nm shifted to longer wavelength by 86 nm in comparison to subphthalocyanine. The compounds show many reduction potentials. They are acceptable electrons in the subphthalocyanine ring, meaning that the compounds have good electron transfer properties.展开更多

Aβ1-42 self-assembles and is highly toxic to neurons. Thus, recognizing aggregated Aβ1-42 is very important for elucidation of Aβ1-42 structure and for the diagnosis of AD. In this study, the specificity of the 79-3 monoclonal antibody against soluble aggre- gate Aβ1-42 was measured by sandwich Enzyme-Linked Immuno Sorbent Assay (ELISA). Eight monoclonal antibodies against both soluble aggregates and amorphous aggregates were used as primary antibodies. Soluble aggregates and amorphous aggregates were used as antigen. As secondary antibody, HRP was labeled with the 79-3 monoclonal antibody. The reactivity of the 79-3 monoclonal antibody against soluble aggregates was confirmed in all combinations, but little reactivity against amorphous aggregates was found. Furthermore, we performed the above sandwich ELISA using the 37-11 antibody, which is reactive against large oval aggregates (LOA) that occur in micro aggregates, instead of the 79-3 antibody. The 77-3 antibody is 1 of the 8 monoclonal antibodies against soluble aggregates;amorphous aggregates also reacted with the 37-11 antibody. These results indicated that soluble aggregates are specifically recognized by a combination of different antibodies. The combined use of these antibodies can be applied to the diagnosis of AD and to defining the structure of the Aβ1-42.展开更多

Our previous reports showed that the monoclonal antibodies 37-11 and 77-3 react with conformational epitopes on the surface of the soluble aggregates of Aβ42 and that sandwich ELISA using these two monoclonal antibodies yields high reactivity to detect soluble aggregates of Aβ42. Here, the reactivity of the sandwich ELISA was shown to increase in the presence of 50 μM Cu2+. However, the addition of Cu2+ had only a small effect on the reactivity of a direct ELISA using antibody 37-11 or 77-3, suggesting that Cu2+ has a small effect on the number of epitopes on the surface of the aggregates. Atomic force microscopy images showed that larger aggregates were formed in the presence of Cu2+, as shown in the other reports. Cu2+ may gather the aggregates with distinct epitopes recognized by antibodies 37-11 and 77-3, leading to increased signal intensity of the sandwich ELISA.展开更多
