Sequential neoadjuvant chemotherapy using pegylated liposomal doxorubicin and cyclophosphamide followed by taxanes with complete trastuzumab and pertuzumab treatment for HER2-positive breast cancer: A phase Ⅱ single-arm study

Objective: Despite cardiotoxicity overlap, the trastuzumab/pertuzumab and anthracycline combination remains crucial due to significant benefits. Pegylated liposomal doxorubicin(PLD), a less cardiotoxic anthracycline, was evaluated for efficacy and cardiac safety when combined with cyclophosphamide and followed by taxanes with trastuzumab/pertuzumab in human epidermal growth factor receptor-2(HER2)-positive early breast cancer(BC).Methods: In this multicenter, phase II study, patients with confirmed HER2-positive early BC received four cycles of PLD(30-35 mg/m^(2)) and cyclophosphamide(600 mg/m^(2)), followed by four cycles of taxanes(docetaxel,90-100 mg/m^(2) or nab-paclitaxel, 260 mg/m^(2)), concomitant with eight cycles of trastuzumab(8 mg/kg loading dose,then 6 mg/kg) and pertuzumab(840 mg loading dose, then 420 mg) every 3 weeks. The primary endpoint was total pathological complete response(tp CR, yp T0/is yp N0). Secondary endpoints included breast p CR(bp CR),objective response rate(ORR), disease control rate, rate of breast-conserving surgery(BCS), and safety(with a focus on cardiotoxicity).Results: Between May 27, 2020 and May 11, 2022, 78 patients were treated with surgery, 42(53.8%) of whom had BCS. After neoadjuvant therapy, 47 [60.3%, 95% confidence interval(95% CI), 48.5%-71.2%] patients achieved tp CR, and 49(62.8%) achieved bp CR. ORRs were 76.9%(95% CI, 66.0%-85.7%) and 93.6%(95% CI,85.7%-97.9%) after 4-cycle and 8-cycle neoadjuvant therapy, respectively. Nine(11.5%) patients experienced asymptomatic left ventricular ejection fraction(LVEF) reductions of ≥10% from baseline, all with a minimum value of >55%. No treatment-related abnormal cardiac function changes were observed in mean N-terminal pro-BNP(NT-pro BNP), troponin I, or high-sensitivity troponin.Conclusions: This dual HER2-blockade with sequential polychemotherapy showed promising activity with rapid tumor regression in HER2-positive BC. Importantly, this regimen showed an acceptable safety profile,especially a low risk of cardiac events, suggesting it as an attractive treatment approach with a favorable risk-benefit balance. 展开更多

In order to explore the criminal psychological mechanism of burglars,this study adopts the qualitative research method of grounded theory to conduct in-depth interviews with 41 burglars in two prisons in Jiangxi Province,China.Nvivo 11.0 was used to code-construct and qualitatively analyze the interview content in order to refine the influencing factors and psychological evolution process of burglary behavior.Thefindings revealed that(1)burglary risk factors include burglary cognition,burglary motivation,burglary decision-making,delinquent peers,burglary opportunity,and incomplete reformation.(2)There are three stages in the psychological evolution pro-cess of burglars:cognitive formation,motivational dominance,and behavioral decision.(3)The interpretation of the criminal psychological mechanism of burglars is a comprehensive and dynamic outcome of the interaction of internal and external factors that shape the individual.Participants’inspection and non-participants’inspection were adapted to verify the research results’validity,which showed that the results were reliable. 展开更多

Objective:To explore and analyze the effect and safety of personalized nursing in improving thrombocytopenia after lymphoma chemotherapy.Methods:80 lymphoma patients with thrombocytopenia after lymphoma chemotherapy in the Department of Hematology of our hospital from May 2021 to May 2023 were selected as the research objects,and they were divided into an experimental group and reference group by random drawing,with 40 cases in each group.The experimental group received individualized nursing,and the reference group received routine nursing.The platelet count,platelet-related indicators,incidence of adverse reactions,and life scores were compared between the two groups.Results:Before the intervention,there was no significant difference in platelet count between the groups(P>0.05);after the intervention,the platelet count in the experimental group was significantly higher compared to the reference group(P<0.05).In the experimental group,the duration for platelet decline and the time for platelets to normalize were notably shorter compared to the reference group(P<0.05).Moreover,the personality group displayed a significantly lower incidence of adverse reactions than the reference group(P<0.05).Prior to the intervention,there were no statistically significant differences(P>0.05)in the life scores between the two groups,such as functional condition,symptom manifestation,and health scores among the groups.However,post-intervention,the personality group exhibited a significant improvement in those scores compared to the reference group(P<0.05).Conclusion:Individualized nursing can optimize the platelet level in thrombocytopenia after lymphoma chemotherapy,improve symptoms,and reduce the occurrence of adverse reactions. 展开更多

LncRNAH19 transcripts also serve as a precursor for miR-675. However, it is unknown whether LncRNAH19 and miR-675 are involved in the migration and invasion of hepatocellular carcinoma (HCC) cells. The purpose of this study was to investigate the effect and mechanism of LncRNAH19 and miR-675 on migration and invasion of HCC cells. The migration and invasion of HCC cells were measured by Transwell migration and invasion assays after transfection of HCC cells with miR-675 inhibitors and LncRNAH19siRNA. The levels of LncRNAH19 and miR-675 were detected by quantitative reverse transcriptase real-time polymerase chain reaction (qRT-PCR), and the protein expression of AKT, GSK-3[3 and Cdc25A by Western blotting analysis. The expression levels of LncRNAHI9 and miR-675 were higher in MHCC-97H cells than in L02, Huh-7 and HepG2 cells. Transwell migration assay revealed that the miR-675 inhibitor and LncRNAH19siRNA could significantly increase the migration of HCC cells (P〈0.01) as compared with the control group. Transwell invasion assay demonstrated that the miR-675 inhibitor and LncRNAH19siRNA could significantly increase the invasion of HCC cells (P〈0.01) as compared with the control group. Western blotting analy- sis showed that the expression levels of AKT and Cdc25A were significantly increased (P〈0.05), and the expression level of GSK-313 was significantly decreased (P〈0.05) after treatment with miR-675 inhibitors and LncRNAH19siRNA as compared with the control group. These findings suggested that inhibition of LncRNAH 19 and miR-675 expression can promote migration and invasion of HCC cells via AKT/GSK-3[3/Cdc25A signaling pathway. 展开更多

Objective:To investigate the expression and the regulation effect of cell growth of microRNA-577 in hepatocellular carcinoma(HCC).Methods:qRT-PCR was applied to detect the relative expression of miR-577 in 70 paired HCC and matched tumor adjacent tissues collecting from resection between March 2011 and March 2014.Pearson chi-square test was used to analyze the relationship between the miR-577 expression and clinical features.The miR-577 mimics were transfected into HepG2 cells:cell cycles were detected by flow cytometry,cell proliferation was measured by MTT assay and BrdU incorporation assay,and cell apoptosis was determined by flow cytometry and Caspase 3/7 activity analysis.The expressions ofβ-catenin were measured by immunohistochemistry.Spearman correlation analysis was used to analyze the relationship between miR-577 and p-catenin.qRT-PCR and westernblot were used to detect the expression of p-catenin in transfected HepG2 cells.Results:The relative expressions of miR-577 was significantly lower in HCC tissues compared to the matched normal tumor-adjacent tissues(P<0.05).Low expression of miR-577 was significantly associated with large tumor size(≥5 cm,P<0.05) and advanced tumor node metastasis stage(Ⅲ+Ⅳ,P<0.05).Transfection of miR-577 mimics could inhibit repress cell proliferation,enhance cell apoptosis and block the cell cycles in G_1/G_1 phase(P<0.05).miR-577 in HCC group had a significant negative correlation relationship with the expression of downstream target of β-catenin(P<0.05).Both the mRNA and protein expression in HepG2 cells were down-regulated after transfection(P<0.05).Conclusions:Low expression of miR-577 is related to the malignant clinicopathological features in HCC tissues,and miR-577 may suppress HCC growth through down-regulating p-catenin. 展开更多

Objective:To investigate whether there is an association between diameter of bacille CalmetteGuerin(BCG)sears and effect of purified protein derivative(PPD)reaction anil to determine whether vitamin A(VA)combined vitamin I)(VD)supplementation influences the immune response to BCG revueeinated in Chinese infants.Methods:A cross-section and 3-month community-randomised trial was conducted.A total of 5 629 infants at 3,6 and 12 months of age in Junan County of China were examined for BCG scar fonnation.Then,597 revuccinated infants were randomly assigned to supplementation(n=307)and control(n=290)groups.The supplementation group were daily assigned to 1 500 IU VA and 500 IU VD for 3 months.Then all infants were subjected to skin test with PPD.Results:The diameter of BCG sears was positively con-elated with diameter of skin indurations of PPD(r=0.17,P<0.05)in the 5 629 infants.The rate of positive response to PP1)was higher in the supplementation group than in the control group(96.1%versus 89.7%,P<0.05,prevalence ratio 1.07.95%CI 1.02-1.12).The prevalence ralio of PPD response for the supplementation group compared with that for the control group was 1.07(95%CI 1.01-1.13)for the males and 1.08(95%CI 1.00-1.17)for the females.For the supplementation group,the males got larger tuberculin induration than the females[(0.73±0.2l)cm versus(0.67±0.20)cm.P<0.05)after intervention.Conclusions:The diameter of BCG scars was effectively correlated with PPD response,which indicates BCG scar formation may be an useful tool Io evaluate the effect of tuberculosis prevention.VA combined VD supplementation may play an immunoregulatory rale in BCG revuecination.This may contribute to the prevention of childhood tuberculosis. 展开更多

To study the effect of titanium alloy cage on the treatment of the ischemic necrosis of femoral head in dog, the model of the ischemic necrosis of femoral head was made with the liquid nitrogen in 15 hybrid adult dogs. The titanium alloy cage made of a hollow cylinder was driven into the subchondral bone of necrotic femoral head via central channel. The dogs were divided into 3 groups, each group was sacrificed 3, 6, 12 weeks after the operation respectively. No collapse of femoral head was observed after the operation. The position of the cages was good on radiograph. Microscopically, the cancellous bone of necrotic femoral head rebuilt gradually and grew into cage. After 12 weeks of creeping substitution, the cancellous bone filled up the hollow cavity and holes of the cages. It is concluded that the titanium alloy cage can provide structural support for the subchondral bone and prevent collapse and can be used for the treatment of the ischemic necrosis of femoral head. 展开更多

Objective: To investigate patients' perception of service quality at hospitals in nine Chinese cities and propose some measures for improvement. Methods: The ServQ ual scale method was used in a survey involving patients at out-patient and in-patient facilities in Shanghai, Chongqing, Chengdu, Nanning, Guilin and Laibin of Guangxi, Honghezhou of Yunnan, Wulumuqi of Xinjiang and Zhongshan of Guangdong. The data collected were entered and analyzed using SPSS 20.0. Statistical analyses included descriptive statistics, factor analyses, reliability analyses, product-moment correlations, independent-sample t-tests, One-way ANOVA and regression analyses. Results: The Kaiser-Meyer-Olkin value for the factor analysis of the scale was 0.979. The Cronbach's α for the reliability analysis was 0.978. All the Pearson correlation coei cients were positive and statistically signii cant. Visitors to out-patient facilities reported more positive perception tacilities on tangibles(t = 4.168, P(t = 1.979, P <han visitors to in-patient f 0.05). Patients of 60 years of age and above reported mor< 0.001) and reliability e positive perception th<an those between 40 and 49 on reliability(F = 3.311, P = 0.010), assurances(F = 2.751, P 0.05) and empathy(F = 4.009, P = 0.003). For the i ve dimensions of the scale, patients in Laibin, Guangxi reported the most positive perceived service quality, followed by patients in Shanghai. On the other hand, patients in Chongqing and Nanning and Guilin of Guangxi reported relatively poor perceptions of service quality. Standardized regression coei cients showed statistically significant(P < 0.001) positive values for all Serv Qual dimensions. Empathy(β = 0.267) and reliability(β uality. = 0.239) most strongly predicted perception of service qConclusions: Chinese patients perceived service quality as satisfactory. The roles of NF-kappaB (NF-κB) expression, Bax activity and cytochrome C (Cyt C) release, apoptosis of islet cells induced by high concentration glucose were explored in vitro. Pancreatic islet cells, which were isolated from Kunming mice, were cultured with different concentrations of glucose in DMEM, and divided into the following groups: G1, G2, G3, G4, G5, and G6 groups, corresponding to the glucose concentrations of 5.6, 7.8, 11.1, 16.7, 22.5, and 27.6 mmol/L, respectively. After culture for 120 h, insulin secretion was evaluated by radioimmunoassay, and the NF-rd3 expression was detected by immunocytochemistry. Bax activity and Cyt C release were measured by immunofluorescence, and apoptosis was examined by Hoechst33342 assay. The results showed that in GI, G2 and G3 groups, insulin secretion was enhanced with the increase of glucose concentration, and the NF-κB expression was also increased (P〈0.05), but Bax activity, Cyt C release and apoptosis rate showed no significant difference among them. However, in G4, G5, and G6 groups, apoptosis rate of islet cells, NF-rd3 expression, Bax activity, and Cyt C release were all significantly increased, and insulin secretion was impaired as compared with G1, G2, and G3 groups (P〈0.05). It was concluded that the exposure of islet cells to high glucose could induce islet cells apoptosis as well as impaired insulin secretion. The NF-κB signaling pathway and mitochondria pathway in islet cells might play some roles in the progressive loss of islet cells in diabetes. The inhibition of the NF-κB expression could be an effective strategy for protecting pancreatic islet cells. 展开更多

Objective: To investigate the effects and underlying mechanisms of different doses of nerve growth factor(NGF) pretreatment on neuron apoptosis and the expressions of the apoptosis-related protein, Bcl-2 and Bax, in the gerbil cerebral prefrontal cortex following global cerebral ischemia-reperfusion(I/R) injury. Methods: Fifty-four gerbils were randomly divided into five groups, group C: sham operation(n = 6); group I/R(n = 12), group L(n = 12): low-dose NGF+I/R, group M(n = 12): medium-dose NGF+I/ R and group H (n = 12): high-dose NGF+I/R. Groups I/R, L, M and H were further divided into 2 subgroups according to the duration of reperfusion(24 h and 72 h). The global cerebral I/R injury model was induced by bilateral carotid artery occlusion for 20 min, followed by removal of the clamps to permit reperfusion. In groups L, M and H, NGF was injected into the lateral ventricle 24 h prior to ischemia. Terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling(TUNEL) and immunohistochemical staining were performed to detect neuron apoptosis and the expressions of Bcl-2 and Bax protein in the cerebral cortex, respectively. Results: In the I/R group and NGF pretreatment groups(L, M and H groups), reperfusion for 72 h caused higher percentages of both neurons exhibiting apoptosis and Bax positive cells(P 〈 0.01), but lower percentages of Bcl-2 positive cells compared with the corresponding 24 h reperfusion groups(P 〈 0.05). All NGF pretreatment groups exhibited lower percentages of neurons exhibiting apoptosis and Bax positive cells but a higher percentage of Bcl-2 positive-cells relative to the I/R group(P 〈 0.01). Moreover, the high-dose NGF pretreatment group had a greater decreased neuronal apoptosis and Bax protein expression and increased Bcl-2 protein expression than either the low-or medium-dose groups. Conclusion: Neuron apoptosis participates in the progression of cerebral ischemia-reperfusion injury. The protective effect of NGF pretreatment against ischemia-reperfusion-induced neuron apoptosis seems to be both time- and dose-dependent. This anti-apoptosis mechanism may be associated with upregulation of Bcl- 2 protein expression and downregulation of Bax protein expression. 展开更多

Methods: Polymerase Chain Reaction (PCR) andenzyme-linked immunosorbent assay (ELISA) were used witha standard curve of DNA copies of HSV as quantitativecontrast. Results: Ninety-three cases were confirmed HSV positiveand 7 cases were found to be negative. There were 58 cases ofHSV-2 (62.4%) and 35 cases of HSV-1(37.6%) among the 93positive cases. The number of DNA plasmids ranged from 115to 1.1×10~5 per 250μL among the 93 positive samples (mean=7.1×10~4/250μL). The number of HSV DNA plasmids rangedfrom 136 to 1.1×10~5 copies per 250μL(mean=7.6×10~4) amongthose with HSV-2, and 115 to 9.4×10~4 per 250μL(mean=6.3×10~4) among those with HSV-1. Meanwhile 10μL ofextracted and dissolved DNA randomly taken from 8 each ofHSV-2 and HSV-1 samples were tested. The number of HSV-2DNA plasmids ranged from 35 copies to 2.7×10~4 (Mean=1.8×10~4) and the number of HSV-1 DNA ranged from 29 to2.5×10~4 (Mean=1.6×10~4). In the 7 negative cases, the quantityof HSV plasmids was zero. Conclusion: The sensitivity of ELISA quantitation (93%) isequal to that of Southern blot. The sensitivity of PCR fordiagnosis is 91%, and 88% for PCR typing. 展开更多

Objective:The inter-α-trypsin inhibitor heavy chain 4(ITIH4)protein is involved in the development of tumors.However,the relationship between ITIH4 and ovarian cancer(OC)has not been extensively examined.This study aimed to explore the effect of ITIH4 on OC and to identify its underlying mechanism.Methods:Expressions of ITIH4 in OC tissues and cells were determined using quantitative reverse transcription polymerase chain reaction(RT-qPCR)and western blots.The function of ITIH4 in the OC cell line HO8910 pm was tested via ITIH4 knockdown.The cell growth rate was measured using MTT and colony formation assays.Flow cytometry was performed to evaluate cell cycle progression.Cell migration and invasion abilities were observed using the transwell migration assay.Results:ITIH4 was downregulated in OC tissues and cells.ITIH4 knockdown promoted cell growth and cell cycle progression.Consistent with these results,inhibition of ITIH4 in OC cells significantly increased cell migration and invasion abilities.Cox regression analysis suggests that ITIH4 expression alone is not a good predictor of the prognosis of malignant ovarian tumors in patients.Conclusions:ITIH4 inhibits the progression of OC,suggesting that ITIH4 may be a useful biomarker for OC.This study may provide a potential novel target for the treatment of OC. 展开更多

According to the structural analysis, the Co( Ⅱ ) ion adopts a slightly distorted six-coordinated octahedral geometry. One N atom of the Schiff base of each molecule was hydrogenated to form hydrogen bond with O atom. B vitamins are enzyme cofactors that play an important role in energy metabolism.The aim of this study was to elucidate whether B vitamin administration can reduce body weight(BW)gain by improving energy metabolism-related enzyme activities in rats fed on a highfat diet.Fifty rats were randomly assigned to one of the following five groups:control group(C),including rats fed on standard rat chow;four treatment groups(H0,H1,H2,and H3),in which rats were fed on a high-fat diet.Rats in the H1 group were treated daily with 100 mg/kg BW thiamine(VB1),100 mg/kg BW riboflavin(VB2),and 250 mg/kg BW niacin(VPP);rats in the H2 group were treated daily with 100 mg/kg BW pyridoxine(VB6),100 mg/kg BW cobalamin(VB12),and 5 mg/kg BW folate(FA);and rats in the H3 group were treated daily with all of the B vitamins administered to the H1 and H2 groups.After 12 weeks,the BW gains from the initial value were 154.5±58.4 g and 159.1±53.0 g in the H1 and C groups,respectively,which were significantly less than the changes in the H0 group(285.2±14.8 g,P〈0.05).In the H0 group,the plasma total cholesterol(CHO)and triglyceride(TG)levels were 1.59±0.30 mmol/L and 1.55±0.40 mmol/L,respectively,which were significantly greater than those in the H1 group(1.19±0.18 mmol/L and 0.76±0.34 mmol/L,respectively,P〈0.05).The activities of transketolase(TK),glutathione reductase,and Na^+/K^+adenosine triphosphatase were significantly increased in the B vitamin-treated groups and were significantly greater than those in the H0 group(P〈0.05).Furthermore,the glucose-6-phosphate dehydrogenase,pyruvic acid kinase,and succinate dehydrogenase activities also were increased after treatment with B vitamins.Supplementation with B vitamins could effectively reduce BW gain and plasma levels of lipids by improving energy metabolism-related enzyme activities in rats,thus possibly providing potential benefits to humans. 展开更多

OBJECTIVE It has been recognized that HBV infection and alcohol consumption are two important risk factors for primary hepatocellular carcinoma (HCC). Recently, the role of clonorchiasis as a risk factor for HCC is controversial. We aimed to investigate whether these factors increase the risk of HCC in Guangxi, China. METHODS A hospital-based, case-control study of HCC was conducted from July 2005 to July 2007. We enrolled 500 consecutive patients with HCC as an experimental group and 500 patients without tumor in liver as a control group. The risk factors that the patients were exposed to were assessed. RESULTS Comparing the risks of developing the HCC, we found out the following results. The risk of developing HCC for the patients with clonorchiasis was 5 folds of that for the patients without clonorchiasis (OR = 5.0; 95% CI: 3.1-8.1), and the risk for the patients with alcohol consumption was 3 folds of that for the patients without drinking alcohol (OR = 3.4; 95% CI: 2.3-4.9), and similarly, the risk for the patients with HBV infection was 21 times of that for the patients without HBV infection (OR = 20.6; 95% CI: 14.3-29.7). According to crossover analysis, there was significant interaction among clonorchiasis, HBV infection and alcohol consumption, with synergistic indices greater than 1. The etiologic fractions attributed to these interactions [EF (A × B)] are 0.7465, 0.5789 and 0.5506, respectively. CONCLUSION Clonorchiasis, HBV infection and heavy alcohol consumption are independent risk factors for developing HCC in our population in Guangxi, and as they can interact synergistically, the risk of developing HCC is increased. Data from this study may indicate new prevention strategies of developing HCC in high-risk individuals. 展开更多

Summary: To explore the expression of heme oxygenase-1 (HO-1) in the peripheral blood mononuclear cells (PBMCs) and its relationship with pulmonary ventilation function in asthmatic patients, 18 asthmatic patients and 18 healthy subjects were selected. HO-1 protein and mRNA levels in PBMCs were measured by immunohistochemical staining and reverse transcription-polymerase chain reaction (RT-PCR), respectively. Blood carbon monoxide Hb (COHb), serum total IgE and pulmonary ventilatory function were observed. Our results showed that the percentage of cells positive for immunohistochemical staining of HO-1 were significantly higher in asthmatic patients (41.72±7.44) % than that in with healthy subjects (10.45±4.36)% (P〈0.001) and the optical density of PBMC HO-1 mRNA was higher in asthmatic patients (26.05±4. 14) than that in healthy subjects (10.82±4.26) (P〈0.001). The relation analysis showed that PBMC HO-1 protein and mRNA levels had significantly negative relation with FEV, %, PEFR, MEFR50 , respectively (r=-0.51-0.89, P〈0.05-0. 001, respectively) and a positive relation with COHb and serum total IgE (r=0.48-0. 85, 0.05-0. 001, respectively). It is concluded that the expression of PBMC HO-1 protein and mRNA increased significantly in asthmatic patients, and HO-1 may play a significant role in the pathogenesis of asthma. [ Objective ] This study aimed to obtain Phytolacca acinosa Roxb. polyploids and provide high-efficiency raw materials for the preparation of biological pesticides. [Method] Terminal bud growing points of Phytolacca acinosa Roxb. seedlings were treated with 0.1%, 0.2%, 0.3% colchicine and 1% agar solution for 24, 48 and 72 h, respectively, while Phytolacca acinosa Roxb. without treatment was adopted as the control. The mutagenic effect of various concentrations of colchicine was calculated. Phytolacca acinosa Roxb. mutants were identified using morphological, anatomical and cytological methods. [ Result] Induction with 0. 2% colchicine for 48 h led to the highest mntagenic effect. Compared with the control, leaf width and leaf thickness of mutants were enhanced by 40. 15% and 33. 25%, respectively; vertical and horizontal diameter of stomata of mutants were enhanced by 30.25% and 60.82%, respectively ; the stomata density of mutants was improved by 150%. Identification of meiotic chromosome of pollen mother cells using compression method showed that the chromosome number of microspore cells of Phytolacca acinosa Roxb. mutants was n = 2 x = 36, while that of the control was n = x = 18, indicating that the mutants were tetraploids. [ Conclusion ] Phytolacca acinosa Roxb. polyploids were induced using the traditional liquid droplet method, which was easy and direct with simple operation and good effect to acquire a large number of polyploid materials within a short time. 展开更多

Objective: We have continued previous work in which we demonstrated that #117 and #372 amino acids contributed to the high activities of human CYP2A13 in catalyzing 4-methylnitrosamino-1-(3-pyridyl)-1-butanone(NNK) and aflatoxin BI(AFB1) carcinogenic activation. The present study was designed to identify other potential amino acid residues that contribute to the different catalytic characteristics of two CYP2A enzymes, CYP2A6 and CYP2A13, in nicotine metabolism and provide insights of the substrate and related amino acid residues interactions. Methods: A series of reciprocally substituted mutants of CYP2A6lle^300→ Phe, CYP2A6Gly^301aAla, CYP2A6Ser^369 → Gly, CYP2A13Phe^300→ Ile, CYP2A13Ala^301 → Gly and CYP2A13Gly^369 → Set were generated by site-directed mutagenesis/baculovirus-Sf9 insect cells expression. Comparative kinetic analysis of nicotine 5'hydroxylatin by wild type and mutant CYP2A proteins was performed. Results:All amino acid residue substitutions at 300, 301 and 369 caused significant kinetic property changes in nicotine metabolism. While CYP2A6Ile^300→ Phe and CYP2A6Gly^301→Ala mutations had notable catalytic efficiency increases compared to that for the wild type CYP2A6, CYP2A13Phe^300→Ile and CYP2A13Ala^301→Gly replacement introduced remarkable catalytic efficiency decreases. In addition, all these catalytic efficiency alterations were caused by Vmax variations rather than Km changes. Substitution of #369 residue significantly affected both Km and Vmax values. CYP2A6Ser^369 → Gly increase the catalytic efficiency via a significant Km decrease versus Vmax enhancement, while the opposite effects were seen with CYP2A13Gly^369 → Ser. Conclusion:#300, #301 and #369 residues in human CYP2A6/13 play important roles in nicotine 5' -oxidation. Switching #300 or #301 residues did not affect the CYP2A protein affinities toward nicotine, although these amino acids are located in the active center. Set369 to Gly substitution indirectly affected nicotine binding by creating more space and conformational flexibility for the nearby residues, such as Leu^370 which is crucial for many hydroxylations. 展开更多

EMMPRIN enhances the metastatic ability of CT26 murine colon adenocarcinoma

Many recent results about metastasis research indicated that EMMPRIN played an important role in cancer metastasis. So, we designed this experiment to investigate whether EMMPRIN can enhance the metastatic ability of murine colon adenocarcinoma cell, CT26. Methods: EMMPRIN was over expressed in CT26 cells through transfecting pCMV-HA2-EMMPRIN into the CT26 cells. Invasion assay, wound migration assay and adhesion assay were utilized to analyze the metastasis of CT26 cells in vitro after EMMPRIN over expression. Results: After EMMPRIN over expression, invasion assay showed that invasive cells were 103.33 + 8.49 in EMMPRIN group and 48.67 + 5.3 in control group (P < 0.001). Migration assay showed that migrating cells were 40.67 + 2.49 in EMMPRIN group and 18.33 + 2.05 in control group (P < 0.001). CCK-8 absorbance value in adhesion assay were 3.33 + 0.17 in EMMPRIN group and 2.10 + 0.22 in control group (P < 0.001). Conclusion: Over expression of EMMPRIN could enhance the CT26 cell capacity of invasion and migration, and inhibit CT26 cell capacity of adhesion remarkably. The results suggest that EMMPRIN may be involved in cancer metastasis and play an important role in promotion of cancer metastasis. 展开更多

Objective:To investigate the predictive value of of serum NSE,umbilical cord blood albumin levels in hyperbilirubinemia full-term neonates with brain injury.Methods:300 cases of hyperbilirubinemia full-term neonates in our hospital during January 2018 to November 2019 were chosen as hyperbilirubinemia group,200 cases of healthy full-term neonates who were delivered in our hospital during the same period and whose general information matched were chosen as normal control group.According to whether the hyperbilirubinemia group had brain injury,they were further divided into bilirubin encephalopathy group(n=34)and non-bilirubin encephalopathy group(n=266).NSE,umbilical cord blood albumin levels in hyperbilirubinemia group and normal control group were compared.Clinical data,NSE and umbilical cord blood albumin levels between bilirubin encephalopathy group,non-bilirubin encephalopathy group were compared.Risk factors associated with brain injury in full-term newborn with hyperbilirubinemia were analyzed by logistics regression,predictive value of NSE and umbilical cord blood albumin on brain injury in full-term newborn with hyperbilirubinemia were evaluated by ROC curve.Results:NSE level in hyperbilirubinemia group was higher than that in normal control group(P<0.05).There was no significant difference in umbilical cord blood albumin between two groups(P>0.05).Incidence of jaundice,anemia,indirect bilirubin peak,total bilirubin peak and NSE level in bilirubin encephalopathy group were higher than those in non-bilirubin encephalopathy group,level of umbilical cord blood albumin was lower than that in non-bilirubin encephalopathy group(P<0.05).Logistic regression analysis showed that jaundice,anemia,indirect bilirubin peak,total bilirubin peak,NSE and umbilical cord blood albumin within 24h after birth were risk factors for hyperbilirubinemia with brain injury(P<0.05).When NSE was 67.09 ng/mL,the yoden index was the highest,with sensitivity and specificity of 79.70%,55.88%respectively.When the umbilical cord blood albumin was 4.20mg/mL,the jorden index was the highest,sensitivity and specificity were 76.32%,82.35%respectively.Conclusion:Abnormal changes in serum NSE,umbilical cord blood albumin levels are observed in full-term neonates with hyperbilirubinemia,moreover they have value of early prediction for subsequent brain injury. 展开更多
