Effect of Huanglian Jiedu Decoction on the Behavior of Zebrafish with Alzheimer's Disease

[Objectives]To study the effect of Huanglian Jiedu Decoction on the behavior of zebrafish with Alzheimer's disease caused by AlCl 3.[Methods]Each portion of Huanglian Jiedu Decoction was prepared according to the proportion of Coptis chinensis∶Phellodendron chinense Rupr.∶Scutellaria baicalensis Georgi∶Gardenia jasminoides Ellis=63 g∶42 g∶42 g∶63 g.After that,each portion of Huanglian Jiedu Decoction was soaked in 6.3 L water for 30 min and boiled twice at 100℃.The extracts were combined twice and filtered,then concentrated to 308 g/L and put into refrigerator for later use.Before training,zebrafishes were put into T-maze to adapt for 2 d,and then behavioral training was carried out for 7 d.After video recording,the behavior of zebrafish was analyzed by Smart 3.0,and qualified zebrafishes were selected for follow-up experiments.Then 60 successfully trained zebrafishes were randomly divided into control group,model group,positive group,low-dose group,medium-dose group and high-dose group of Huanglian Jiedu Decoction.Except for the control group,all the other groups were exposed to 100μg/L AlCl 3 for 7 d.After that,video was recorded,and behavioral analysis was carried out with behavioral record and analysis software Smart 3.0.And then the zebrafishes in the other four groups except the model group were treated with Huperzine A(4μg/L)and Huanglian Jiedu Decoction(154,308,616 mg/L)for 6 d,respectively.After that,it was recorded and the behavior of each group was analyzed.[Results]There was a significant difference in the time spent in the left area and the percentage of time in the left area between the control group and the model group(P<0.001).The time spent in the left area and the percentage of time in the left area in the model group and positive group,low,medium and high dose groups of Huanglian Jiedu Decoction decreased significantly(P<0.05,P<0.01,P<0.001).The swimming distance in the left area and the percentage of swimming distance in the left area in the model group were significantly higher than those in the control group(P<0.001).There was a significant difference in swimming distance between model group and positive group,low,medium and high dose groups of Huanglian Jiedu Decoction(P<0.01,P<0.001).In the percentage of swimming distance in the left area,there was a significant difference between the model group and the low and high dose groups of Huanglian Jiedu Decoction(P<0.01,P<0.001).[Conclusions]Huanglian Jiedu Decoction can improve the behavior of zebrafish with Alzheimer's disease.

The Mechanism by Which Pereskia aculeata Mill.Regulates Ferroptosis and Interferes with Colon Cancer Based on Network Pharmacology

[Objectives]The paper was to investigate the mechanism by which Pereskia aculeata Mill.regulates ferroptosis and interferes with colon cancer using network pharmacology.[Methods]The effect of P.aculeata on HCT116 cells was observed by examining cell morphology.The intersection of P.aculeata,colon cancer,and ferroptosis targets was determined using the Venny 2.1.0 online platform.DAVID database was used to perform GO and KEGG enrichment analyses.Cytoscape_v3.10.0 was used for network mapping and PPI analysis.[Results]The observation of cell morphology indicated that P.aculeata suppressed the growth of HCT116 cells.GO and KEGG analysis identified 67 genes involved in potential pathways,including the MAPK signaling pathway and Apoptosis,18 of which were located on the MAPK pathway.After conducting PPI visualization analysis and ranking based on Degree values,we identified TP53,TNF,EGFR,MAPK14,and AKT1 as the top five targets with the highest Degree values.[Conclusions]P.aculeata may activate the p53 gene through the MAPK signaling pathway,inducing ferroptosis and ultimately resulting in tumor cell death.

TLC Identification of Laggerae Alatae Herba and Extraction Process of Chlorogenic Acid

[Objectives]Taking chlorogenic acid as index component,TLC and content determination method of Laggerae Alatae Herba were established.[Methods]TLC identification used silica gel G thin-layer plate,and butyl acetate∶formic acid∶water(7∶2.5∶2.5)was taken as developing agent,and it was inspected under ultraviolet lamp(365 nm).The content was determined by chromatographic column Inertsil ODS-3 C_(18)(4.60 mm×250 mm,5μm).Mobile phase:methanol-0.1%phosphoric acid(28∶72);detection wavelength:329 nm;flow speed:1.0 mL/min;column temperature:25℃;injection volume:10μL.[Results]Chlorogenic acid can be detected by TLC,with clear spots and good specificity.When injection volume of chlorogenic acid was between 0.099 and 0.990μg(R^(2)=0.9999),there was good linear relationship.In low,medium and high sample adding groups of Laggerae Alatae Herba,average recovery rates of chlorogenic acids were 98.80%(RSD=2.09%),98.24%(RSD=1.96%)and 99.65%(RSD=2.15%).[Conclusions]The method could effectively identify medicinal material Laggerae Alatae Herba,and accurately measure the content of chlorogenic acid in Laggerae Alatae Herba,thereby providing a scientific basis for developing and using medicinal resources of Laggerae Alatae Herba.

Fever glove hand-shake method safe blood collection from children's fingertips in COVID-19 fever clinic

BACKGROUND During the coronavirus disease 2019(COVID-19)epidemic,the fever clinic is an important link for screening and diagnosing whether a patient is infected with the novel coronavirus.Blood collection from children’s fingertips is a commonly used detection method;however,in children,the blood collection process may cause discomfort and resistance.To address this problem,the use of heating gloves combined with hand swinging can be considered for fingertip blood collection in children.AIM To explore the application of fever gloves with the handshaking method for fingertip blood collection from children in fever clinics during the COVID-19 epidemic.METHODS A total of 100 children were selected for fingertip blood collection at the fever clinic of our hospital from June 2022 to June 2023 and were divided into two groups using a randomized numerical table method,with 50 cases in each group,including the control and observation groups.The patients in the control group followed the doctor's instructions to cooperate with the routine fingertip blood collection method,and the patients in the observation group followed the doctor's instructions to cooperate with the static fever gloves with the shaking hands method of children's fingertip blood collection.The level of the six blood routine and collection indexes,and the satisfaction of the examination of the patients in the peripheral blood group and the fever gloves with the shaking hands method of the children's fingertip blood collection group were compared.RESULTS The red and white blood cell count,hemoglobin,and red blood cell pressure volume in the observation group were higher than those in the control group(P<0.05);the platelet count in the control group was lower than that in the observation group(P<0.05);the number of times of squeezing the fingertip,the average time of blood collection,and the score of puncture pain in the observation group were significantly better than those in the control group(P<0.05);and satisfaction with the routine blood examination in the observation group was greater than that in the control group.CONCLUSION The application value of the fever gloves with shaking hands method for children's fingertip blood collection was better,the accuracy of examination indexes was higher,and patient satisfaction with the examination was greater.
