Cymbidium goeringii is an economically important ornamental plant, and flower color is one of the main features of C.goeringii that contributes to its high economic value. To clarify the molecular mechanisms underlying the role of anthocyanins in mediating differences in color among varieties, liquid chromatography–tandem mass spectrometry was used to perform anthocyanin-targeted metabolomics of seven C.goeringii varieties, including‘Jin Qian Yuan’(JQY),‘Jin Xiu Qian Yuan’(JXQY),‘Miao Jiang Su Die’(MJSD),‘Qian Ming Su’(QMS),‘Shi Chan’(SC),and‘Yang Ming Su’(YMS),as well as the C.goeringii.We detected 64 anthocyanins, including cyanidins, delphinidins, malvidins, pelargonidins, peonidins, petunidins, procyanidins, and flavonoids. We identified six shared differentially accumulated metabolites(DAMs),including cyanidin-3-O-rutinoside,delphinidin-3-Osophoroside,pelargonidin-3-O-rutinoside,peonidin-3-O-(6-O-malonyl-beta-D-glucoside),peonidin-3-Osophoroside,and chalcone. Most DAMs were enriched in the anthocyanin biosynthesis pathway.Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed that the differentially expressed metabolites were significantly enriched in the anthocyanin biosynthesis pathway.Analysis of the content of differentially expressed metabolites indicated that peonidin-3-O-(6-O-malonyl-beta-D-glucoside)was the key metabolite underlying color differences among C.goeringii varieties.Procyanidin B2,pelargonidin-3-O-galactoside,and naringenin might also affect the color formation of JQY and QMS,SC,and MJSD,respectively. The results of this study shed light on the metabolic mechanism underlying flower color differences in C.goeringii at the molecular level. Our findings will aid future studies of the mechanism of flower color regulation in C.goeringii and have implications for the breeding of new varieties. 展开更多

At present, colorectal cancer depends on surgery, radiotherapy and chemotherapy. Due to the characteristics of the elderly, postoperative recovery is slow, prone to postoperative complications and difficult to tolerate radiotherapy and chemotherapy have become the main problem hindering the disease. Based on the theory of prevention, the use of drugs from the perspective of constitution and etiology can prevent the occurrence of colorectal cancer in the elderly. Traditional Chinese medicine (TCM) combined with radiotherapy and chemotherapy can alleviate adverse reactions and improve treatment efficacy. TCM with surgical treatment can reduce the occurrence of postoperative complications and promote postoperative recovery. For patients with advanced colorectal cancer, TCM can play a role in fighting cancer and improving the quality of life of patients. For patients in stable phase, TCM can prevent tumor recurrence and metastasis. 展开更多

Ulcerative colitis(UC)is a chronic recurrent inflammatory bowel disease.Despite ongoing advances in our understanding of UC,its pathogenesis is yet unelu-cidated,underscoring the urgent need for novel treatment strategies for patients with UC.Exosomes are nanoscale membrane particles that mediate intercellular communication by carrying various bioactive molecules, such as proteins,RNAs,DNA,and metabolites. The NOD-like receptor family pyrin domain containing 3(NLRP3)inflammasome is a cytosolic tripartite protein complex whose activation induces the maturation and secretion of proinflammatory cytokines interleukin-1β(IL-1β)and IL-18,triggering the inflammatory response to a pathogenic agent or injury.Growing evidence suggests that exosomes are new modulators of the NLRP3 inflammasome, with vital roles in the pathological process of UC.Here, recent evidence is reviewed on the role of exosomes and NLRP3 inflammasome in UC.First, the dual role of exosomes on NLRP3 inflammasome and the effect of NLRP3 inflammasome on exosome secretion are summarized. Finally,an outlook on the directions of exosome-NLRP3 inflammasome crosstalk research in the context of UC is proposed and areas of further research on this topic are high-lighted. 展开更多

Seed germination with selenium(Se)is promising for producing Se-biofortified foods.Mungbean(Vigna radiata(L.)Wilczek)sprout is freshly eaten as a salad dressed with sauce, making it superior for Se biofortification.Since the Se safety range for the human body is extremely narrow,it is imperative to evaluate the genotypic responses of mungbean sprouts to Se.This study evaluated the Se enrichment capacity and interaction withflavonoids and antioxidant systems in sprouts of 20 mungbean germplasms.Selenium treatment was done by immersing mung-bean seeds in 20μM sodium selenite solution for 8 h.Afterward, the biomass,Se amounts, flavonoid(particularly vitexin and isovitexin)contents, antioxidant capacity, and key biosynthetic gene expressions were measured.Sprout Se content was 2.0-7.0μg g^(-1) DW among the 20 mungbean germplasms.Selenium treatment differentially affected the biomass, totalflavonoid, vitexin, isovitexin, antioxidant enzyme activities, and antioxidant capacities of the mungbean germplasms.Eight germplasms showed increased biomass(p<0.05),the highest increasing by 127%,but 13 did not phenotypically respond to Se treatment.Seven and six germplasms showed varied levels of vitexin and isovitexin increment after Se treatment, the highest measuring 2.67-and 2.87-folds for vitexin and isovitexin, respectively. Two mungbeanflavonoid biosynthesis genes, chalcone synthase(VrCHS)and chalcone isomerase(VrCHI)were significantly up-regulated in the germplasms with increased vitexin and isovitexin levels(p<0.05).Moreover,Se enrichment capacity was significantly correlated with the vitexin, isovitexin, and antiox-idant capacities. In conclusion, mungbean sprouts could be a useful Se-biofortified food, but the Se enrichment capacity and nutritional response must be determined for each germplasm before commercialization. 展开更多

BACKGROUND Malancao(MLC)is a traditional Chinese medicine with a long history of utilization in treating ulcerative colitis(UC).Nevertheless, the precise molecular mechanisms underlying its efficacy remain elusive. This study leveraged ultrahigh-performance liquid chromatography coupled with exactive mass spectrometry(UHPLC-QE-MS),network pharmacology, molecular docking(MD),and gene microarray analysis to discern the bioactive constituents and the potential mechanism of action of MLC in UC management.AIM To determine the ingredients related to MLC for treatment of UC using multiple databases to obtain potential targets for fishing.METHODS This research employs UHPLC-QE-MS for the identification of bioactive compounds present in MLC plant samples.Furthermore, the study integrates the identified MLC compound-related targets with publicly available databases to elucidate common drug disease targets.Additionally, the R programming language is utilized to predict the central targets and molecular pathways that MLC may impact in the treatment of UC.Finally,MD are conducted using AutoDock Vina software to assess the affinity of bioactive components to the main targets and confirm their therapeutic potential.RESULTS Firstly, through a comprehensive analysis of UHPLC-QE-MS data and public database resources,we identified 146 drug-disease cross targets related to 11 bioactive components. The Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis highlighted that common disease drug targets are primarily involved in oxidative stress management, lipid metabolism, atherosclerosis, and other processes. They also affect AGE-RAGE and apoptosis signaling pathways.Secondly,by analyzing the differences in diseases,we identified key research targets. These core targets are related to 11 active substances, including active ingredients such as quercetin and luteolin. Finally,MD analysis revealed the stability of compound-protein binding, particularly between JUNLuteolin,JUN-Quercetin,HSP90AA1-Wogonin,and HSP90AA1-Rhein.Therefore,this suggests that MLC may help alleviate intestinal inflammation in UC,restore abnormal lipid accumulation, and regulate the expression levels of core proteins in the intestine.CONCLUSION The utilization of MLC has demonstrated notable therapeutic efficacy in the management of UC by means of the compound target interaction pathway. The amalgamation of botanical resources, metabolomics, natural products,MD,and gene chip technology presents a propitious methodology for investigating therapeutic targets of herbal medicines and discerning novel bioactive constituents. 展开更多

Objective:To uncover the underlying mechanisms of action of the Yinlai decoction on high-calorie dietinduced pneumonia through proteomics analysis. Methods:Based on the Gene Expression Omnibus(GEO)database, lung tissue samples from normal and high-fat diet(HFD)fed mice in the GSE16377 dataset were selected as test cohorts to identify differentially expressed genes and conduct bioinformatics analyses. In the animal experiments, mice were randomly divided into the control(N),high-calorie diet pneumonia(M),and Yinlai decoction treatment(Y)groups.Mice in the M group received high-calorie feed and a 0.5 mg/mL lipopolysaccharide solution spray for 30 min for 3 d.The mice in the Y group were intragastrically administered 2 mL/10 g Yinlai decoction twice daily for 3 d.Pathological evaluation of the lung tissue was performed.Differentially expressed proteins(DEPs)in the lung tissue were identified using quantitative proteomics and bioinformatics analyses. The drug-target relationships between Yinlai decoction and core DEPs in the lung tissue were verified using AutoDock Vina and Molecular Graphics Laboratory(MGL)Tools.DEPs were verified by western blot. Results:GEO data mining showed that an HFD altered oxidative phosphorylation in mouse lung tissue. The Yinlai decoction alleviated pathological damage to lung tissue and pneumonia in mice that were fed a high-calorie diet.A total of 47 DEPs were identified between the Y and M groups.Enrichment analysis revealed their association with energy metabolism pathways such as the tricarboxylic acid cycle(TCA)and oxidative phosphorylation. The protein-protein interaction network revealed that Atp5a1,Pdha1,and Sdha were the target proteins mediating the therapeutic effects of Yinlai decoction.Molecular docking results suggested that the mechanism of the therapeutic effect of Yinlai decoction involves the binding of brassinolide, praeruptorin B,chrysoeriol, and other components in Yinlai decoction to Atp5a1.Conclusion:The Yinlai decoction alleviated lung tissue damage and pneumonia in mice that were fed a high-calorie diet by regulating the TCA and oxidative phosphorylation. Our study highlights the importance of a healthy diet for patients with pneumonia and provides a scientific basis for the prevention and treatment of pneumonia through dietary adjustments. 展开更多

Dragon Boat Festival herbal markets in the Qianxinan Buyi and Miao Autonomous Prefecture of southwestern Guizhou have a long well-conserved history. These markets, which are a feature of Buyi and Miao traditional medicines, contain a rich diversity of medicinal plants and traditional medical knowledge.Today, people in southwestern Guizhou still believe that using herbs during the Dragon Boat Festival prevents and can treat disease. In this study,we identified the fresh herbal plants sold at the herbal markets of Xingren City and Zhenfeng County in Qianxinan Buyi and Miao Autonomous Prefecture and quantified their importance. We identified 141 plant species(belonging to 114 genera and 61 families). The plant family with the most species was Asteraceae(14 species).Informants reported that most medicinal plants are herbaceous, with 95.7%of plants used for decoction and 30.5%used for medicinal baths.Medicinal plants are most commonly used to treat rheumatism, injury, and abdominal diseases. The utilization frequency index and relative importance values indicated that Artemisia argyi and Acorus calamus are the most important plants sold at herbal markets during the Dragon Boat Festival. The price of medicinal materials sold in the market may serve as an indicator of the conservation status of species in the region. These findings indicate that the Dragon Boat Festival herbal markets in the Qianxinan Buyi and Miao Autonomous Prefecture fully embodies the characteristics of indigenous ethnomedicine and culture, and also exhibits the diversity of plant resources. We recommend that rare and endangered plants in this region be domesticated and protected. 展开更多

Aim:As conflict caused by cultural diversity among patients in China continues to rise, hospitals are in urgent need for improvement of transcultural efficacy among nurses. This study aims to evaluate the transcultural self-efficacy of nurses working in the tertiary general hospital in Guizhou Province,an ethnic minority region in western China, and to identify whether nurses'demographic characteristics affect their transcultural self-efficacy.Method:We used the Chinese version of the Transcultural Self-Efficacy Tool(TSET-CV)to survey 1,190 inservice nurses. Results:Results showed that the level of transcultural self-efficacy of the nurses was generally moderate;few of the nurses had high or low transcultural self-efficacy.The nurses'transcultural self-efficacy was affected by demographic variables, including age, marital status, employment type, income, work experience, and whether or not they were head nurses.Having a stable work environment,a stable marriage,a good educational background, and a high-ranked professional title were associated with increased transcultural self-efficacy.Conclusion:Nursing administrators in hospitals should offer continuing education on transcuitural nursing according to nurses'demographic characteristics and the SEST scores. 展开更多

Background:Crossbow medicine needle is a traditional external therapy. However, acupuncture pain, inaccurate administration, secondary infection, and poor patient compliance limit the application of specialty therapies.Since the most effective components of crossbow medicine are lipid-soluble,the crossbow medicine needle was prepared as an innovative O/W-type microemulsion to solve these problems. In this study, the crossbow medicine microemulsion preparation method was established and its quality was evaluated. Methods:A single factor and pseudoternary phase diagram, combined with a D-optimal mixture design, optimized the formulation of crossbow medicine microemulsion through particle size, polydispersity index, zeta potential, microstructure, and contents of benzoyl neoaconitine, benzoyl hypoaconitine, neoaconitine, periplocin, and methyl salicylate to evaluate the final prescription. Results:The best prescription for preparing crossbow medicine microemulsion is 0.1500 g crossbow medicine dry extract,0.1500 g isopropyl myristate,0.8625 g polyoxyethylene castor oil-35,0.2875 g ethanol,3.700 g water, and 5.1500 g microemulsion. The prepared microemulsion is an oil-in-water type with a clear and transparent appearance. Its average particle size is 19.83±0.78 nm,average polydispersity index is 0.15±0.02,and average zeta potential is−0.51±0.1 mV.The pH value is 5.17±0.02.The microstructure of crossbow medicine microemulsion illustrated a uniform distribution of spherical oil globules. The average contents of benzoyl neoaconitine, benzoyl hypoaconitine, neoaconitine, periplocin, and methyl salicylate were 13.34,113.05,43.45,628.27 and 27.93μg/g,respectively.Conclusion:The preparation process of crossbow medicine microemulsion is reasonable and feasible. The method to determine the content is accurate and straightforward, laying the foundation for further research. 展开更多
