Effect of Ganduqing Granule on metabolic disorder and liver dysfunction in patients with chronic hepatitis B

目的:探讨肝毒清胶囊改善慢性乙型肝炎患者肝功能异常的潜在机制.方法:采集30例乙型肝炎患者经肝毒清胶囊治疗前、后的血浆以及30例正常健康人的血浆,分别检测其肝功能,并使用超高效液相色谱-质谱联用法检测胆汁酸、脂质和肝脏氧化应激... 目的:探讨肝毒清胶囊改善慢性乙型肝炎患者肝功能异常的潜在机制.方法:采集30例乙型肝炎患者经肝毒清胶囊治疗前、后的血浆以及30例正常健康人的血浆,分别检测其肝功能,并使用超高效液相色谱-质谱联用法检测胆汁酸、脂质和肝脏氧化应激代谢产物的变化.结果:与正常对照组比较,慢性乙肝患者的肝功能明显异常,血浆中有8种代谢产物(3-硫酸甘氨鹅脱氧胆酸、甘氨鹅脱氧胆酸-3-葡萄糖醛酸,牛磺鹅脱氧胆酸,胆烷酸,二羟基雄甾烯酮硫酸,溶血磷脂酰胆碱(C18:3),溶血磷脂酰乙醇胺(C22:1);2,6-二氨基-4-羟基-5-N-甲基甲酰嘧啶)明显升高(P<0.05).经肝毒清胶囊治疗3周后,这8种升高的代谢产物均显著降低(P<0.05),并趋于正常水平.其中,3-硫酸甘氨鹅脱氧胆酸、甘氨鹅脱氧胆酸-3-葡萄糖醛酸,牛磺鹅脱氧胆酸和胆烷酸与胆汁酸代谢有关;二羟基雄甾烯酮硫酸,溶血磷脂酰胆碱(C18:3)和溶血磷脂酰乙醇胺(C22:1)与脂质代谢有关;2,6-二氨基-4-羟基-5-N-甲基甲酰嘧啶与肝脏的氧化应激相关.同时,血清中丙氨酸转氨酶,天冬氨酸转氨酶,总胆红素;直接胆红素和HBV病毒水平明显降低(均P<0.05).结论:肝毒清胶囊通过改善慢性乙肝患者的脂质代谢、胆汁酸代谢及氧化应激反应等,进而改善其肝功能. 展开更多

A clinical study on the treatment of obese polycystic ovary syndrome by electroacupuncture combined with moving cupping

Objective:Effect analysis of electroacupuncture combined with recircupping in treatment of obese polycystic ovary syndrome.Methods: From October 2016 to September 2018, 68 obese PCOS patients were diagnosed and treated in the department of gynaecology, affiliated hospital of shaanxi university of traditional Chinese medicine.Treatment group given cupping and tired walking tank with metformin therapy, the control group only oral metformin treatment, after continuous treatment of three menstrual cycles, observation comprehensive curative effect of two groups before and after treatment, TCM syndrome score, body weight, body mass index (BMI), waist hip ratio (WHR), sex hormone [luteinizing hormone (LH), testosterone (T)], blood sugar [fasting plasma glucose (FPG), fasting insulin (FINS), insulin resistance index (HOMA IR)] and blood lipid [triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), total cholesterol (TC)] metabolic index, on two groups of curative effect are compared and discussed. Results:The total effective rate (83.33%) of the treatment group was higher than that of the control group (66.67%)(P<0.05).After treatment, TCM syndrome score of the two groups decreased significantly(P<0.01).The treatment group was significantly improved compared with the control group (P<0.01).Body weight, BMI, WHR of the two groups decreased significantly after treatment (P<0.01).The treatment group was better than the control group (P<0.05).After treatment, the LH and T values of the two groups decreased significantly (P<0.01).There was no significant difference in T value between the two groups (P>0.05), but the efficacy of LH in the treatment group was better than that in the control group (P<0.01).The levels of FINS, HOMA-IR and FPG in the two groups were significantly decreased after treatment (P<0.01).There was no significant difference in FPG between the two groups (P > 0.05), but the improvement of FINS and HOMA-IR in the treatment group was better than that in the control group (P< 0.01).After treatment, TG, LDL-C and TC levels in the two groups decreased (P< 0.01), while HDL-C levels in the treatment group increased (P< 0.01).There was no significant difference in HDL-C, LDL-C and TC between the two groups (P>0.05), but the efficacy of TG in the treatment group was better than that in the control group (P< 0.01).Conclusion: The curative effect of electroacupuncture and moving cupping combined with metformin is better than that of metformin alone, should be widely used. 展开更多

Method: Our method was divided into five steps: preparation, heart ext raction, perfusion, mechanical dissociation and pu rifica tion and r ecalcifica ti on. Fir stly, make a preparation, including solutions, equipment and so on. Secondly, after anesthesia satisfactorily, open the chest of rat and take the heart out. Thirdly, transect the aorta and hang the aorta onto the Langendroff perfusion system such that the tip is just distal to the aortic valve. For th, perfuse the hea rt with diges tion solution for 17-20 min afte r Ca2+ free tyrode solution for 5 min. Finally, remove the heart from the system quickly and dissociate mechanically in the KB solution and recalcification after filter. Result: Though the microscope, good-state cardiomyocytes (clear stripes, strong sense of three-dimensional, stick well) can be observed. Conclusion: Those cells can be used to undergo medical electrophysiological experiment, including the study of iron channel. 展开更多

本文旨在研究缺氧对大鼠心肌细胞持续钠电流的影响,以探讨心肌细胞缺氧发生的离子通道机制。急性酶解法分离大鼠心肌细胞,一组未做处理,使用全细胞膜片钳技术记录钠电流;一组进行缺氧处理,使用全细胞膜片钳技术记录钠电流。结果显示:与... 本文旨在研究缺氧对大鼠心肌细胞持续钠电流的影响,以探讨心肌细胞缺氧发生的离子通道机制。急性酶解法分离大鼠心肌细胞,一组未做处理,使用全细胞膜片钳技术记录钠电流;一组进行缺氧处理,使用全细胞膜片钳技术记录钠电流。结果显示:与正常细胞比较,缺氧细胞钠电流形态和I-V曲线的形态轨迹特征不变;相对于正常细胞,缺氧细胞的I-V曲线上移明显,INa峰值电流相对正常细胞上调了15.68%(P<0.0001);正常与缺氧细胞激活电位均在-40mV左右,最大电流峰值对应刺激电压为-25mV。上述结果提示,缺氧情况下,大鼠心肌细胞持续钠电流增大。 展开更多
