The main purpose of this study was to investigate the active components of the Chinese medicine formula Shenqi San(SS) by high performance liquid chromatography with diode array detector and electrospray ionization-hybrid quadrupole time-of-flight mass spectrum(HPLC-DADESI-QTOF-MS), and demonstrate the anticancer mechanism of SS on human lung adenocarcinoma A549 cells by evaluating the cell proliferation and apoptosis induction. The chloroform extraction of SS(CE-SS) was extracted from SS, while HPLC-DAD-ESI-QTOF-MS assay was performed to identify components of CE-SS. MTT assay was used to quantify the proliferation of A549 cells with the treatment of CE-SS. Apoptosis analysis was carried out by detecting phosphatidylserine(PS) externalization using the Annexin V-FITC Apoptosis Detection Kit and the stained cells were analyzed with a flow cytometer. DAPI staining assay was carried out to observe morphological characteristics of apoptotic cells. Western blotting was used to detect the expression of important signaling proteins including caspase-3,-8,-9, p53, Bax and Bcl-2. Eight compounds were identified through HPLC-DAD-ESI-QTOF-MS analysis and 3-pyridine carboxylic acid, barbatin C, scutebarbatine F and barbatine D might be the main compounds responsible for the antitumor effect of CE-SS. CE-SS suppressed the proliferation of lung cancer A549 cells in a time-and dose-dependent manner. By Annexin V-FITC/PI double staining, we found that treatment with CE-SS induced apoptosis in A549 cells. After 24-h exposure to CE-SS, the expression of cleaved-caspase-9, cleaved-caspase-8 and cleaved-caspase-3 protein was activated, the expression of p53 protein increased while the ratio of Bax/Bcl-2 also increased. This study identified the eight compounds of CE-SS, and demonstrated their anticancer effect on human lung adenocarcinoma A549 cells via induction of apoptosis. 展开更多

Infectious diseases are the common enemies of mankind. In the course of historical development, they persistently threaten human health and safety.Even today, despite the developments in medical science,we cannot escape the fear and suffering caused by infectious diseases.Whether in ancient or modern times, the source of infection, route of transmission, and a susceptible population are the three key conditions for the prevalence and spread of infectious diseases. All factors closely related to these three conditions can affect the prevalence of infectious diseases.China is one of the cradles of world civilization. The ancient people accumulated a great deal of experience and lessons in the long struggle against infectious diseases. In the face of the current threat posed by widespread infectious disease,it is imperative to review and summarize ancient Chinese ideas and health policies on epidemic prevention and control to inspire contemporary efforts in the prevention and control of infectious disease. The combination of prevention-oriented epidemic prevention ideology and traditional medicine provides valuable insights, especially for impoverished and medically underserved regions. 展开更多

Polygonatum sibiricum has been widely used due to its excellent biological activities. We prepared a novel polysaccharide from P.sibiricum(PSP)in this study.According a monosaccharide composition analysis,PSP was mainly composed of fructose and glucose with a molar percentage of 93.81:5.12.The main linkage types were identified asα-D-Glcp-1→and→2-β-D-Fruf-1→.The molecular weight of PSP showed no significant change after simulated salivary and gastrointestinal digestion. However,PSP could be broken down by intestinal bacteria. Our findings revealed that PSP administration increased the abundance of probiotics such as Bifidobacterium.Furthermore, the results showed that gut microbes could utilize PSP to produce short-chain fatty acids including acetic acid, propionic acid, and butyric acid. Also, the PSP fermentation broth displayed an excellent scavenging effect on free radicals, including 2,2-diphenyl-1-picrylhydrazyl radical, superoxide radical, and hydroxyl radical. In summary, this study will help to promote the application of PSP as prebiotics in functional food and the medical industry. 展开更多

Emerging research suggests a potential association of progression of Alzheimer's disease(AD)with alterations in synaptic currents and mitochondrial dynamics. However, the specific associations between these pathological changes remain unclear. In this study,we utilized Aβ42-induced AD rats and primary neural cells as in vivo and in vitro models. The investigations included behavioural tests, brain magnetic resonance imaging(MRI),liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)analysis,Nissl staining,thioflavin-S staining,enzyme-linked immunosorbent assay,Golgi-Cox staining, transmission electron microscopy(TEM),immunofluorescence staining, proteomics, adenosine triphosphate(ATP)detection, mitochondrial membrane potential(MMP)and reactive oxygen species(ROS)assessment, mitochondrial morphology analysis, electrophysiological studies,Western blotting, and molecular docking. The results revealed changes in synaptic currents, mitophagy, and mitochondrial dynamics in the AD models.Remarkably, intervention with Dengzhan Shengmai(DZSM)capsules emerged as a pivotal element in this investigation.Aβ42-induced synaptic dysfunction was significantly mitigated by DZSM intervention, which notably amplified the frequency and amplitude of synaptic transmission. The cognitive impairment observed in AD rats was ameliorated and accompanied by robust protection against structural damage in key brain regions, including the hippocampal CA3,primary cingular cortex, prelimbic system, and dysgranular insular cortex.DZSM intervention led to increased IDE levels, augmented long-term potential(LTP)amplitude, and enhanced dendritic spine density and length. Moreover,DZSM intervention led to favourable changes in mitochondrial parameters, including ROS expression,MMP and ATP contents, and mitochondrial morphology. In conclusion, our findings delved into the realm of altered synaptic currents, mitophagy, and mitochondrial dynamics in AD,concurrently highlighting the therapeutic potential of DZSM intervention. 展开更多

Allergic rhinitis(AR)and is a type of allergic diseases caused by an inflammation of the nasal mucosa caused by allergens.Clinically,AR is a common allergic disease and is difficult to treat.Due to differences in individual constitutions also known as body type, the severity of the disease is also different and it may even affect the daily life of the patient if it is severe. The effective treatment of AR has become a hot topic in current allergic disease research.Traditional Chinese Medicine’s understanding of constitution is based on the“Yellow Emperor’s Internal Classic”book, and Professor Wang Qi put forward the“physical constitution theory of traditional Chinese medicine”.The constitution is a key internal factor of the occurrence and development of diseases. It is believed in Traditional Chinese medicine(TCM)that the constitution can be regulated, and rectifying the patient’s imbalanced constitution has a positive effect on the diagnosis, treatment, and prognosis of the disease. Therefore, based on the“physical constitution theory of TCM”,this study considers the idea of treating body differently applying TCM constitution in the treatment of AR as the breakthrough point, and explores the correlation between AR and TCM constitution. The results show that TCM constitution is an intrinsic factor affecting the pathogenesis of AR.The use of TCM constitution theory in the treatment of AR has a positive effect and should be popularized in application. 展开更多

The most common cytotoxic effects of chemotherapy drugs alone include bone marrow suppression, gastrointestinal symptoms, and liver and kidney function problems. With the continuous research on non-small cell lung cancer(NSCLC),various treatment modalities such as targeted therapy and immunotherapy have emerged in recent years, but there is no denying that chemotherapy remains one of the most basic treatment modalities.Shen Jia et al.,in exploring the efficacy of detoxification and yin clearing soup(Astragalus membranaceus(Fisch.)Bunge,Atractylodes macrocephala Koidz,et al.),found that the addition of the soup treatment significantly reduced the number of adverse reactions such as bone marrow suppression, nausea and vomiting, and abnormal liver and kidney function, and the incidence of adverse reactions was also significantly lower than that of the group with chemotherapy drugs alone[1]. 展开更多

Objective:To classify the subtypes of metabolic-associated fatty liver disease(MAFLD)and provide new insights into the heterogeneity of MAFLD.Methods:Electronic medical records(EMR)of MAFLD diagnosed in accordance with the diagnostic criteria of Hubei Provincial Hospital of Traditional Chinese Medicine from 2016-2020 were included in the study.for physical annotation, and the data on each clinical phenotype was normalized according to corresponding aspirational standards. The MAFLD heterogeneous medical record network(HEMnet)was constructed using sex, age, disease diagnosis, symptoms, and Western medicine prescriptions as nodes and the co-occurrence times between phenotypes as edges.K-means clustering was used for disease classification.Relative risk(RR)was used to assess the specificity of each phenotype.Statistical methods were used to compare differences in laboratory indicators among subtypes. Results:A total of patients(12,626)with a mean age of 55.02(±14.21)years were included in the study.MAFLD can be divided into five subtypes:digestive diseases(C0),mental disorders and gynecological diseases(C1),chronic liver diseases and decompensated complications(C2),diabetes mellitus and its complications(C3),and immune joint system diseases(C4).Conclusions:Patients with MAFLD experience various symptoms and complications. The classification of MAFLD based on the HEMnet method is highly reliable. 展开更多
