Over-expression of glutathione S-transferase(GST)can promote Cisplatin resistance in hepatocellular carcinoma(HCC)treatment.Hence,inhibiting GST is an attractive strategy to improve Cisplatin sensitivity in HCC therapy.Although several synthesized GST inhibitors have been developed,the side effects and narrow spectrum for anticancer seriously limit their clinical application.Considering the abundance of natural compounds with anticancer activity,this study developed a rapid fluorescence technique to screen“green”natural GST inhibitors with high specificity.The fluorescence assay demonstrated that schisanlactone B(hereafter abbreviated as C1)isolated from Xue tong significantly down-regulated GST levels in Cisplatin-resistant HCC cells in vitro and in vivo.Importantly,C1 can selectively kill HCC cells from normal liver cells,effectively improving the therapeutic effect of Cisplatin on HCC mice by downregulating GST expression.Considering the high GST levels in HCC patients,this compound demonstrated the high potential for sensitizing HCC therapy in clinical practice by down-regulating GST levels. 展开更多

This paper presents a bi-level hybrid local search(BHLS)algorithm for the three-dimensional loading problem with balancing constraints(3DLP-B),where several rectangular boxes with even densities but different sizes are loaded into a single cubic bin to meet the requirements of the space or capacity utilization and the balance of the center of gravity.The proposed algorithm hybridizes a novel framed-layout procedure in which the concept of the core block and its generation strategy are introduced.Once the block-loading sequence has been determined,we can load one block at a time by the designed construction heuristic.Then,the double-search is introduced;its external search procedure generates a list of compact packing patterns while its internal search procedure is used to search the core-block frames and their best distribution locations.The approach is extensively tested on weakly to strongly heterogeneous benchmark data.The results show that it has better performance in improving space utilization rate and balanced condition of the placement than existed techniques:the overall averages from 79.85%to 86.45%were obtained for the balanced cases and relatively high space-usage rate of 89.44%was achieved for the unbalanced ones. 展开更多

A key requirement of the cloud platform is the reasonable deployment of its large-scale virtual machine infrastructure. The mapping relation between the virtual node and the physical node determines the specific resource distribution strategy and reliability of the virtual machine deployment. Resource distribution strategy has an important effect on performance, energy consumption, and guarantee of the quality of service of the computer, and serves an important role in the deployment of the virtual machine. To solve the problem of meeting the fault-tolerance requirement and guarantee high reliability of the application system based on the full use of the cloud resource under the prerequisite of various demands, the deployment framework of the feedback virtual machine in cloud platform facing the individual user's de- mands of fault-tolerance level and the corresponding deploy- ment algorithm of the virtual machine are proposed in this paper. Resource distribution strategy can deploy the virtual machine in the physical nodes where the resource is mutu- ally complementary according to the users' different require- ments on virtual resources. The deployment framework of the virtual machine in this paper can provide a reliable com- puter configuration according to the specific fault-tolerance requirements of the user while considering the usage rate of the physical resources of the cloud platform. The experimental result shows that the method proposed in this paper can provide flexible and reliable select permission of faulttolerance level to the user in the virtual machine deployment process, provide a pertinent individual fault-tolerant deployment method of the virtual machine to the user, and guarantee to meet the user service in a large probability to some extent. 展开更多
