Some morphological characters P. kamii is closely related to P. sheni, P. megalophthalmus, P. retrofasciatus, P. rubrifasciatus, P. knappi, P. helenae, P. pelicieri, P. jothyi, P.retrofasciatus and P. randalli in sharing of body width, upper jaw length, pelvic spine length and orbit diameter.Meristic count characters of P. kamii differ from P. sheni, P. pilicieri, P. megalophthalmus, P. retrofasciatus and P.rubrifasciatus in having more numerous dorsal spine(18 vs. 15–17) and below lateral line(33–34 vs. 29–33) and differ from P. megalophthalmus and P. rubrifasciatus in having more numerous pored scales in lateral line(13 vs.14–15) and shorter of anal spine. Eddy-centric composites reveal that the horizontal distribution of temperature anomaly associated with eddy in winter is more of a dipole pattern in upper 50 m and tends to be centrosymmetric below 50 m, while in summer the distribution pattern is centrosymmetric in the entire water column. The horizontal distribution of eddy-induced salinity anomaly exhibits similar seasonal characteristics, except that the asymmetry of the salinity anomaly is weaker. The vertical distribution of temperature anomaly associated with eddy shows a monolayer structure, while the salinity anomaly demonstrates a triple-layer structure. Further analysis indicates that the vertical distribution of the anomalies is related to the vertical structure of background temperature and salinity fields, and the asymmetry of the anomalies in upper 50 m is mainly caused by the horizontal advection of background temperature and salinity.展开更多
We studied the depth distribution of periphyton,growing on inundated dead trees in Kenyir Lake,Malaysia in June 1995.The algal floral composition and structure manifested changes down the depth gradient in terms of sp...We studied the depth distribution of periphyton,growing on inundated dead trees in Kenyir Lake,Malaysia in June 1995.The algal floral composition and structure manifested changes down the depth gradient in terms of species richness,abundance,diversity and cell density.In regression analysis,all these algal attributes were negatively correlated with the depth gradients at P<0.05.In terms of species richness,the bacillariophytes showed dominance over the cyanophytes and chlorophytes;whereas with respect to standing crop,the cyanophytes showed dominance over the bacillariophytes and chlorophytes.The chlorophyll a was higher at the mid and bottom-depths than the surface-depth in both the downstream and upstream sites,which showed that vertical productivity or biomass accumulation was greater in low light irradiance.The product-moment correlation analysis showed that conductivity,turbidity,dissolved oxygen,reactive phosphate and ammonium-nitrogen were highly correlated with the algal assemblage data.However,photosynthetic active radiation(PAR) showed poor correlation with the community data.These observations have cast some light on the autoecological characteristics,habitat preferences and environmental responses of tropical periphytic communities.展开更多
Global warming has been the subject of concern in today’s world with elevating temperature causing the melting of polar ice and increasing sea level.The aim of this study was to investigate the physiological and phot...Global warming has been the subject of concern in today’s world with elevating temperature causing the melting of polar ice and increasing sea level.The aim of this study was to investigate the physiological and photosynthetic performance of two polar Chlorella,namely Chlorella UMACC 250 and Chlorella UMACC 234 to elevating temperatures as might be experienced under future warming scenarios.The cultures were exposed to three different temperatures of 4℃,8℃and 12℃.The growth and photosynthetic activity were determined every 2 d for a period of 10 d.At the end of the experiment,the cultures were harvested and analysed for biochemical composition.Both Chlorella strains were able to tolerate higher temperatures than their ambient temperature.The final pigments content showed an increasing trend with increased temperatures for both strains.The photosynthetic activities were measured using pulse-amplitude modulation(PAM)fluorometer.The photosynthetic parameters including maximum quantum efficiency(Fv/Fm),maximum relative electron transport rate(r ETRmax),light harvesting efficiency(a)and photoadaptive index(Ek)were derived from the rapid light curves(RLCs).Both Chlorella strains showed a slight decline in growth and photosynthetic activities at the initial part of the experiment.However,they showed the ability to recuperate with Chlorella UMACC 250 recovers better compared to Chlorella UMACC 234.Both Chlorella strains showed similar trend in their carbohydrate content at 12℃,while the protein content of Chlorella UMACC 234 decreased when exposed to increasing temperatures.The results indicated that polar Chlorella are able to survive at increased temperatures throughout the experiment.展开更多

The impact of temperature on the photosynthesis of microalgae is of great interest as the primary production of algal biomass is strongly dependent on the photosynthetic rates in a dynamic environment. Here, we examine the effects of elevated temperature on Chlorella strains originating from different latitudes, namely Antarctic, Arctic, temperate and tropical regions. Chlorophyll fluorescence was used to assess the photosynthetic responses of the microalgae. Rapid light curves(RLCs) and maximum quantum yield( F_v/F_m) were recorded. The results showed that Chlorella originating from different latitudes portrayed different growth trends and photosynthetic performance. The Chlorella genus is eurythermal, with a broad temperature tolerance range, but with strain-specific characteristics. However, there was a large overlap between the tolerance range of the four strains due to their "eurythermal adaptivity". Changes in the photosynthetic parameters indicated temperature stress. The ability of the four strains to reactivate photosynthesis after inhibition of photosynthesis under high temperatures was also studied. The Chlorella strains were shown to recover in terms of photosynthesis and growth(measured as Chl a) when they were returned to their ambient temperatures. Polar strains showed faster recovery in their optimal temperature compared to that under the ambient temperature from which they were isolated.展开更多
The effect of sonication pretreatment condition on Eucheuma cottonii and Gelidium amansii seaweed towards agar extraction wae studied.Four parameters were changed during sonication to investigate the effects on agar y...The effect of sonication pretreatment condition on Eucheuma cottonii and Gelidium amansii seaweed towards agar extraction wae studied.Four parameters were changed during sonication to investigate the effects on agar yield and quality.These parameters include the time interval,concentration ratio,frequency,and intensity.The highest amount of agar extracted from Eucheuma cottonii species could be obtained from the time interval of 30min,seaweed weight to solvent volume ratio of 1:20,the frequency of 35Hz,and the sonication power intensity of 30%.For Gelidium amansii species,the best agar yield also could be obtained from the time interval of 30 min,1:20 of seaweed weight to water volume ratio,the frequency of 35Hz,and power intensity of 30%.From the experiment,sonication pretreatment signifi cantly influenced the yield and properties of extracted agar.The sonication with autoclaved seaweed produced agar containing less sulfate content,which is an excellent chemical property for gel electrophoresis applications.The gelstrength of sonication with autoclaving for both seaweeds,Eucheuma and Gelidium species was the highest among those by sonication with direct heating,which proved that sonication pretreatment with autoclaving could enhance the physical properties of the agar.展开更多

Mesenchymal stem cells (MSCs...Background: There is considerable interest in using goats as models for genetically engineering dairy animals and also for using stem cells as therapeutics for bone and cartilage repair. Mesenchymal stem cells (MSCs) have been isolated and characterized from various species, but are poorly characterized in goats. Results: Goat MSCs isolated from bone marrow (BM-MSCs) and adipose tissue (ASCs) have the ability to undergo osteogenic, adipogenic and chondrogenic differentiation. Cytochemical staining and gene expression analysis show that ASCs have a greater capacity for adipogenic differentiation compared to BM-MSCs and fibroblasts. Different methods of inducing adipogenesis also affect the extent and profile of adipogenic differentiation in MSCs. Goat fibroblasts were not capable of osteogenesis, hence distinguishing them from the MSCs. Goat MSCs and fibroblasts express CD90, CD105, CD73 but not CD45, and exhibit cytoplasmic localization of OCT4 protein. Goat MSCs can be stably transfected by Nucleofection, but, as evidenced by colony-forming efficiency (CFE), yield significantly different levels of progenitor cells that are robust enough to proliferate into colonies of integrants following G418 selection. BM-MSCs expanded over increasing passages in vitro maintained karyotypic stability up to 20 passages in culture, exhibited an increase in adipogenic differentiation and CFE, but showed altered morphology and amenability to genetic modification by selection. Conclusions: Our findings provide characterization information on goat MSCs, and show that there can be significant differences between MSCs isolated from different tissues and from within the same tissue. Fibroblasts do not exhibit trilineage differentiation potential at the same capacity as MSCs, making it a more reliable method for distinguishing MSCs from fibroblasts, compared to cell surface marker expression.展开更多
The morphology of seven specimens of Chrysaora chinensis(Scyphomedusae,Semaestomae)obtained from four sites off the coast of Peninsular Malaysia was examined.Morphological characteristics of C.chinensis that encompass...The morphology of seven specimens of Chrysaora chinensis(Scyphomedusae,Semaestomae)obtained from four sites off the coast of Peninsular Malaysia was examined.Morphological characteristics of C.chinensis that encompasses structures such as the bell,tentacles,oral arms,stomach,manubrium,radial canals and gonads were described in detail.A total of 107 specimens that represented C.chinensis populations of four coastal areas of Peninsular Malaysia(East-Central,East-North,West-Central,and West-North)were also analysed for shape variation using geometric morphometric analysis.Procrustes superimposition,Principal Component Analysis(PCA)and Canonical Variate Analysis(CVA)were applied to the images of gastrovascular pouches of C.chinensis to extract the shape information.Independent contrasts were used for comparisons between shapes.There were no significant differences in shape variation between all the specimens based on the PCA results.However,CVA results showed shape variations between specimens taken from the four areas of Peninsular Malaysia,especially with higher magnitudes of Mahalanobis distances between the east and west coast areas,including between East-Central and East-North,but lower magnitudes were detected between the West-Central and West-North.展开更多

The diversity of fish community in the se...Fish species composition and spatio-temporal variability of the community were studied in a tropical seagrass meadow located in a lagoon in the eastern part of North Sulawesi. The diversity of fish community in the seagrass meadows was relatively high, with the Shannon-Wiener index ranging from 1.57 to 3.69. The family Apogonidae was the most dominant in abundance(8.27 ind./(100 m2)) and biomass(28.49 g/(100 m2)). At the species level,Apogon lateralis and Sphaeramia orbicularis were the most dominant species in abundance and biomass,respectively. For spatial distribution on species, the end, middle and mouth of the lagoon clustered together as a whole, which may be due to the substrate types found in those zones. The fish species, fish abundance and fish biomass were greater in the dry and wet seasons than in the transition season, which is explained by the strong monsoon, which provides a more suitable environment and food for the fish. The maximum length of 93.10% of the captured species was less than their length at maturity, indicating that seagrass meadows are nursery habitats for many fishes. Therefore, protection of the seagrass meadows is essential for fisheries and sustainable resource utilization.展开更多
In tropical waters where temperatures are relatively stable,we investigated whether the relationship between phytoplankton growth and grazing loss rate across diff erent habitats around Peninsular Malaysia can be attr...In tropical waters where temperatures are relatively stable,we investigated whether the relationship between phytoplankton growth and grazing loss rate across diff erent habitats around Peninsular Malaysia can be attributed to its eutrophication states.We measured phytoplankton growth(μ)and grazing loss(g)rates in waters off Bachok Marine Research Station(BMRS),located northeast of Peninsular Malaysia.Chlorophyll-a(chl-a)concentration ranged from 2.90 to 15.78μg/L and was dominated by nanoand micro-phytoplankton(>2μm in size).Using the Landry and Hassett dilution method,μat BMRS ranged from 1.02 to 1.58/d whereas g varied from 0.07 to 0.88/d.Grazing accounted for 35%of the primary production at BMRS.A systematic review of available data in waters around Peninsular Malaysia,revealed howμfl uctuated over a wide range(0.01–1.80/d)and correlated with chl a distribution(R 2=0.181,P<0.001).However,the relationship was only signifi cant at<9μg/L chl a for mesotrophic waters and<16μg/L chl a for eutrophic waters.In contrast,g ranged from 0.00 to 1.01/d,and correlated withμat all locations.The g/μslope ranged from 19%to 84%,and was generally similar for waters around Peninsular Malaysia.However,all the g/μslopes had a positive y-intercept except for BMRS,and this seemed to suggest the availability of alternative prey supporting grazing at the other stations.展开更多

In this study, photosynthetic effciency, biochemical pro?les and non-targeted metabolic pro?ling were studied to compare between the nitrogen-replete and deplete conditions. Slowed growth, change in photosynthetic pigments and lowered photosynthetic effciency were observed in response to nitrogen deprivation. Biochemical pro?les of the cultures showed an increased level of carbohydrate, lipids and total fatty acids, while the total soluble protein content was lowered. A trend of fatty acid saturation was observed in the nitrogen-deplete culture with an increase in the level of saturated fatty acids especially C16:0 and C18:0, accompanied by a decrease in proportions of monounsaturated and polyunsaturated fatty acids. Fifty-nine metabolites, including amino acids, lipids, phytochemical compounds, vitamins and cofactors were signi?cantly dysregulated and annotated in this study. Pathway mapping analysis revealed a rewiring of metabolic pathways in the cells, particularly purine, carotenoid, nicotinate and nicotinamide, and amino acid metabolisms. Within the treatment period of nitrogen deprivation, the key processes involved were reshu ? ing of nitrogen from proteins and photosynthetic machinery, together with carbon repartitioning in carbohydrates and lipids.展开更多
Water samples were collected in order to study the spatial variation of photosynthetic pigments and phytoplankton community composition in the Lembeh Strait(Indonesia) and the Kelantan River Estuary(Malaysia)during Ju...Water samples were collected in order to study the spatial variation of photosynthetic pigments and phytoplankton community composition in the Lembeh Strait(Indonesia) and the Kelantan River Estuary(Malaysia)during July and August 2016, respectively. Phytoplankton photosynthetic pigments were detected using high performance liquid chromatography combining with the CHEMTAX software to confirm the Chl a biomass and community composition. The Chl a concentration was low at surface in the Lembeh Strait, which it was 0.580–0.682 μg/L, with the average(0.620±0.039) μg/L. Nevertheless, the Chl a concentration fluctuated violently at surface in the Kelantan River Estuary, in which the biomass was 0.299–3.988 μg/L, with the average(0.922±0.992) μg/L. The biomass at bottom water was higher than at surface in the Kelantan River Estuary, in which the Chl a concentration was 0.704–2.352 μg/L, with the average(1.493±0.571) μg/L. Chl b, zeaxanthin and fucoxanthin were three most abundant pigments in the Lembeh Strait. As a consequence, phytoplankton community composition was different in the two study areas. In the Lembeh Strait, prasinophytes(26.48%±0.83%) and Synechococcus(25.73%±4.13%) occupied ~50% of the Chl a biomass, followed by diatoms(20.49%±2.34%) and haptophytes T8(15.13%±2.42%). At surface water in the Kelantan River Estuary, diatoms(58.53%±18.44%)dominated more than half of the phytoplankton biomass, followed by Synechococcus(27.27%±14.84%) and prasinophytes(7.00%±4.39%). It showed the similar status at the bottom water in the Kelantan River Estuary,where diatoms, Synechococcus and prasinophytes contributed 64.89%±15.29%, 16.23%±9.98% and 8.91%±2.62%,respectively. The different phytoplankton community composition between the two regions implied that the bottom up control affected the phytoplankton biomass in the Lembeh Strait where the oligotrophic water derived from the West Pacific Ocean. The terrigenous nutrients supplied the diatoms growing, and pico-phytoplankton was grazed through top down control in the Kelantan River Estuary.展开更多

Length-weight relationship provides basic information of a fi sh’s condition while the morphometric data of otolith has been traditionally used to determine the range of fi sh sizes for various purposes.The length-we...Length-weight relationship provides basic information of a fi sh’s condition while the morphometric data of otolith has been traditionally used to determine the range of fi sh sizes for various purposes.The length-weight relationship of Terapon jarbua(Forskål,1775)was examined from 165 specimens,while 132 specimens were used in sagittal otolith biometry measurements.Fish specimens were obtained from various locations in Malaysia including Pahang,Selangor,Sarawak,and Sabah.The total length of fi sh ranged 12.5–32.5 cm,while the total weight ranged 26.28–444.30 g.The otolith length ranged 0.423–1.070 cm,and the otolith weight was 0.0103–0.1572 g.The specimens displayed a mixture of positive and negative allometry growth as indicated by fl uctuating b values between 2.74 and 3.15.The overall coeffi cient of determination(R 2)between fi sh length and weight was 0.9729,which implies a strong correlation.All regressions between the fi sh length and otolith dimensions were highly signifi cant(P<0.001).The otolith dimensions found to be most closely related to the fi sh total length was the otolith weight.This was supported by the regression model showing a high R 2 value of 86.57%.The aspect ratio of otolith was 1.80±0.14(mean±SD).Results confi rm that the otolith growth refl ects the somatic growth of T.jarbua.The length-weight relationship explained the growth conditions of the fi shes in each location while the overall b value of 2.91 indicates that the condition of T.jarbua grown in Malaysian waters during the study period was generally in good condition.This study contributed to the database on the morphometric data of otolith of T.jarbua as well as the fi rst report on fi sh length-weight relationship for T.jarbua in Malaysia.展开更多

The microalgae are treated as spherical naturally buoyant...In this work, we study the regional dependence of transport behavior of microalgae Chlorella vulgaris inside microflu- idic channel on applied fluid flow rate. The microalgae are treated as spherical naturally buoyant particles. Deviation from the normal diffusion or Brownian transport is characterized based on the scaling behavior of the mean square displacement (MSD) of the particle trajectories by resolving the displacements in the streamwise (flow) and perpendicular directions. The channel is divided into three different flow regions, namely center region of the channel and two near-wall boundaries and the particle motions are analyzed at different flow rates. We use the scaled Brownian motion to model the transitional characteristics in the scaling behavior of the MSDs. We find that there exist anisotropic anomalous transports in all the three flow regions with mixed sub-diffusive, normal and super-diffusive behavior in both longitudinal and transverse directions.展开更多

Ultraviolet(UV)filters are widely used in sunscreen and personal care products due to their ability to give protection to our skin from UV radiation.Oxybenzone,commonly known as benzophenone-3,is one type of UV filter...Ultraviolet(UV)filters are widely used in sunscreen and personal care products due to their ability to give protection to our skin from UV radiation.Oxybenzone,commonly known as benzophenone-3,is one type of UV filter found as the active ingredient in many pharmaceutical products.Although oxybenzone has been extensively studied as an environmental toxicant in the ecosystem,little is known about its toxicity effects on microalgae.The effects of oxybenzone on growth(measured as OD620 nm,chlɑand carotenoids)and macromolecular composition of polar microalgae(Chlorella UMACC 400 and Chlorella UMACC 401)and temperate microalgae(Chlorella sp.,Chlamydomonas reinhardtii,Scenedesmus quadricauda)were investigated.These microalgae were cultured in triplicate and exposed to different oxybenzone concentrations(0,100,200,300 and 400 mg·L-1),at 4℃and 18 for polar and temperate species respectively,for 96 h.The oxybenzone concentrations used℃represent a range from environmental to extreme concentrations to understand the impact of this toxicant on microalgae.The results showed that the highest concentration of oxybenzone(300 and 400 mg·L-1)had adverse effects on growth rate and biomass of these microalgae.However,exposure to oxybenzone concentrations ranging from 200 mg·L-1 to 400 mg·L-1 did not have significant effects on S.quadricauda growth.The exposure to oxybenzone at higher concentrations also led to changes in cell structure after 96 h.Generally,protein and carbohydrate content of all microalgae except S.quadricauda increased with increasing oxybenzone concentrations.Protein content increased significantly when cells were exposed to oxybenzone,though effects were greater in the polar species,suggesting that it could be one of the adaptive strategies that enabled these microalgae to tolerate oxybenzone.Further investigation is required to determine the effects of oxybenzone on other features of microalgal performance at relevant environmental concentrations.展开更多

Thermocline lifting was observed at 104?E from a number of parallel transects. To confirm the presence of upwelling, satellite remote sensing data were used, and numerical model experiments were conducted. A cooler sea-surface temperature along the coast was spotted from both in-situ and satellite data while upward movement from the model agreed with field data. The southwesterly wind that blows along PM from June to September is believed to be the important mechanism that contributed to this upwelling through an Ekman dynamics process.展开更多

文摘Three specimens of the serranid fish(Serranidae), Plectranthias kamii Randall, 1980 were collected from fish market, Bitung, North Sulawesi on May and June 2010. Some morphological characters P. kamii is closely related to P. sheni, P. megalophthalmus, P. retrofasciatus, P. rubrifasciatus, P. knappi, P. helenae, P. pelicieri, P. jothyi, P.retrofasciatus and P. randalli in sharing of body width, upper jaw length, pelvic spine length and orbit diameter.Meristic count characters of P. kamii differ from P. sheni, P. pilicieri, P. megalophthalmus, P. retrofasciatus and P.rubrifasciatus in having more numerous dorsal spine(18 vs. 15–17) and below lateral line(33–34 vs. 29–33) and differ from P. megalophthalmus and P. rubrifasciatus in having more numerous pored scales in lateral line(13 vs.14–15) and shorter of anal spine. The present anthiine fish collected from Bitung, Indonesia was described as new record and bringing the total number of species of this genus known in Indonesia to seven.

文摘The seasonal characteristics and formation mechanism of the thermohaline structure of mesoscale eddy in the South China Sea are investigated using the latest eddy dataset and ARMOR3D data. Eddy-centric composites reveal that the horizontal distribution of temperature anomaly associated with eddy in winter is more of a dipole pattern in upper 50 m and tends to be centrosymmetric below 50 m, while in summer the distribution pattern is centrosymmetric in the entire water column. The horizontal distribution of eddy-induced salinity anomaly exhibits similar seasonal characteristics, except that the asymmetry of the salinity anomaly is weaker. The vertical distribution of temperature anomaly associated with eddy shows a monolayer structure, while the salinity anomaly demonstrates a triple-layer structure. Further analysis indicates that the vertical distribution of the anomalies is related to the vertical structure of background temperature and salinity fields, and the asymmetry of the anomalies in upper 50 m is mainly caused by the horizontal advection of background temperature and salinity.

文摘We studied the depth distribution of periphyton,growing on inundated dead trees in Kenyir Lake,Malaysia in June 1995.The algal floral composition and structure manifested changes down the depth gradient in terms of species richness,abundance,diversity and cell density.In regression analysis,all these algal attributes were negatively correlated with the depth gradients at P<0.05.In terms of species richness,the bacillariophytes showed dominance over the cyanophytes and chlorophytes;whereas with respect to standing crop,the cyanophytes showed dominance over the bacillariophytes and chlorophytes.The chlorophyll a was higher at the mid and bottom-depths than the surface-depth in both the downstream and upstream sites,which showed that vertical productivity or biomass accumulation was greater in low light irradiance.The product-moment correlation analysis showed that conductivity,turbidity,dissolved oxygen,reactive phosphate and ammonium-nitrogen were highly correlated with the algal assemblage data.However,photosynthetic active radiation(PAR) showed poor correlation with the community data.These observations have cast some light on the autoecological characteristics,habitat preferences and environmental responses of tropical periphytic communities.

文摘Global warming has been the subject of concern in today’s world with elevating temperature causing the melting of polar ice and increasing sea level.The aim of this study was to investigate the physiological and photosynthetic performance of two polar Chlorella,namely Chlorella UMACC 250 and Chlorella UMACC 234 to elevating temperatures as might be experienced under future warming scenarios.The cultures were exposed to three different temperatures of 4℃,8℃and 12℃.The growth and photosynthetic activity were determined every 2 d for a period of 10 d.At the end of the experiment,the cultures were harvested and analysed for biochemical composition.Both Chlorella strains were able to tolerate higher temperatures than their ambient temperature.The final pigments content showed an increasing trend with increased temperatures for both strains.The photosynthetic activities were measured using pulse-amplitude modulation(PAM)fluorometer.The photosynthetic parameters including maximum quantum efficiency(Fv/Fm),maximum relative electron transport rate(r ETRmax),light harvesting efficiency(a)and photoadaptive index(Ek)were derived from the rapid light curves(RLCs).Both Chlorella strains showed a slight decline in growth and photosynthetic activities at the initial part of the experiment.However,they showed the ability to recuperate with Chlorella UMACC 250 recovers better compared to Chlorella UMACC 234.Both Chlorella strains showed similar trend in their carbohydrate content at 12℃,while the protein content of Chlorella UMACC 234 decreased when exposed to increasing temperatures.The results indicated that polar Chlorella are able to survive at increased temperatures throughout the experiment.

文摘Elevated temperatures as a consequence of global warming have significant impacts on the adaptation and survival of microalgae which are important primary producers in many ecosystems. The impact of temperature on the photosynthesis of microalgae is of great interest as the primary production of algal biomass is strongly dependent on the photosynthetic rates in a dynamic environment. Here, we examine the effects of elevated temperature on Chlorella strains originating from different latitudes, namely Antarctic, Arctic, temperate and tropical regions. Chlorophyll fluorescence was used to assess the photosynthetic responses of the microalgae. Rapid light curves(RLCs) and maximum quantum yield( F_v/F_m) were recorded. The results showed that Chlorella originating from different latitudes portrayed different growth trends and photosynthetic performance. The Chlorella genus is eurythermal, with a broad temperature tolerance range, but with strain-specific characteristics. However, there was a large overlap between the tolerance range of the four strains due to their "eurythermal adaptivity". Changes in the photosynthetic parameters indicated temperature stress. The ability of the four strains to reactivate photosynthesis after inhibition of photosynthesis under high temperatures was also studied. The Chlorella strains were shown to recover in terms of photosynthesis and growth(measured as Chl a) when they were returned to their ambient temperatures. Polar strains showed faster recovery in their optimal temperature compared to that under the ambient temperature from which they were isolated.

文摘The effect of sonication pretreatment condition on Eucheuma cottonii and Gelidium amansii seaweed towards agar extraction wae studied.Four parameters were changed during sonication to investigate the effects on agar yield and quality.These parameters include the time interval,concentration ratio,frequency,and intensity.The highest amount of agar extracted from Eucheuma cottonii species could be obtained from the time interval of 30min,seaweed weight to solvent volume ratio of 1:20,the frequency of 35Hz,and the sonication power intensity of 30%.For Gelidium amansii species,the best agar yield also could be obtained from the time interval of 30 min,1:20 of seaweed weight to water volume ratio,the frequency of 35Hz,and power intensity of 30%.From the experiment,sonication pretreatment signifi cantly influenced the yield and properties of extracted agar.The sonication with autoclaved seaweed produced agar containing less sulfate content,which is an excellent chemical property for gel electrophoresis applications.The gelstrength of sonication with autoclaving for both seaweeds,Eucheuma and Gelidium species was the highest among those by sonication with direct heating,which proved that sonication pretreatment with autoclaving could enhance the physical properties of the agar.

文摘Background: There is considerable interest in using goats as models for genetically engineering dairy animals and also for using stem cells as therapeutics for bone and cartilage repair. Mesenchymal stem cells (MSCs) have been isolated and characterized from various species, but are poorly characterized in goats. Results: Goat MSCs isolated from bone marrow (BM-MSCs) and adipose tissue (ASCs) have the ability to undergo osteogenic, adipogenic and chondrogenic differentiation. Cytochemical staining and gene expression analysis show that ASCs have a greater capacity for adipogenic differentiation compared to BM-MSCs and fibroblasts. Different methods of inducing adipogenesis also affect the extent and profile of adipogenic differentiation in MSCs. Goat fibroblasts were not capable of osteogenesis, hence distinguishing them from the MSCs. Goat MSCs and fibroblasts express CD90, CD105, CD73 but not CD45, and exhibit cytoplasmic localization of OCT4 protein. Goat MSCs can be stably transfected by Nucleofection, but, as evidenced by colony-forming efficiency (CFE), yield significantly different levels of progenitor cells that are robust enough to proliferate into colonies of integrants following G418 selection. BM-MSCs expanded over increasing passages in vitro maintained karyotypic stability up to 20 passages in culture, exhibited an increase in adipogenic differentiation and CFE, but showed altered morphology and amenability to genetic modification by selection. Conclusions: Our findings provide characterization information on goat MSCs, and show that there can be significant differences between MSCs isolated from different tissues and from within the same tissue. Fibroblasts do not exhibit trilineage differentiation potential at the same capacity as MSCs, making it a more reliable method for distinguishing MSCs from fibroblasts, compared to cell surface marker expression.

文摘The morphology of seven specimens of Chrysaora chinensis(Scyphomedusae,Semaestomae)obtained from four sites off the coast of Peninsular Malaysia was examined.Morphological characteristics of C.chinensis that encompasses structures such as the bell,tentacles,oral arms,stomach,manubrium,radial canals and gonads were described in detail.A total of 107 specimens that represented C.chinensis populations of four coastal areas of Peninsular Malaysia(East-Central,East-North,West-Central,and West-North)were also analysed for shape variation using geometric morphometric analysis.Procrustes superimposition,Principal Component Analysis(PCA)and Canonical Variate Analysis(CVA)were applied to the images of gastrovascular pouches of C.chinensis to extract the shape information.Independent contrasts were used for comparisons between shapes.There were no significant differences in shape variation between all the specimens based on the PCA results.However,CVA results showed shape variations between specimens taken from the four areas of Peninsular Malaysia,especially with higher magnitudes of Mahalanobis distances between the east and west coast areas,including between East-Central and East-North,but lower magnitudes were detected between the West-Central and West-North.

文摘Fish species composition and spatio-temporal variability of the community were studied in a tropical seagrass meadow located in a lagoon in the eastern part of North Sulawesi. The diversity of fish community in the seagrass meadows was relatively high, with the Shannon-Wiener index ranging from 1.57 to 3.69. The family Apogonidae was the most dominant in abundance(8.27 ind./(100 m2)) and biomass(28.49 g/(100 m2)). At the species level,Apogon lateralis and Sphaeramia orbicularis were the most dominant species in abundance and biomass,respectively. For spatial distribution on species, the end, middle and mouth of the lagoon clustered together as a whole, which may be due to the substrate types found in those zones. The fish species, fish abundance and fish biomass were greater in the dry and wet seasons than in the transition season, which is explained by the strong monsoon, which provides a more suitable environment and food for the fish. The maximum length of 93.10% of the captured species was less than their length at maturity, indicating that seagrass meadows are nursery habitats for many fishes. Therefore, protection of the seagrass meadows is essential for fisheries and sustainable resource utilization.
文摘In tropical waters where temperatures are relatively stable,we investigated whether the relationship between phytoplankton growth and grazing loss rate across diff erent habitats around Peninsular Malaysia can be attributed to its eutrophication states.We measured phytoplankton growth(μ)and grazing loss(g)rates in waters off Bachok Marine Research Station(BMRS),located northeast of Peninsular Malaysia.Chlorophyll-a(chl-a)concentration ranged from 2.90 to 15.78μg/L and was dominated by nanoand micro-phytoplankton(>2μm in size).Using the Landry and Hassett dilution method,μat BMRS ranged from 1.02 to 1.58/d whereas g varied from 0.07 to 0.88/d.Grazing accounted for 35%of the primary production at BMRS.A systematic review of available data in waters around Peninsular Malaysia,revealed howμfl uctuated over a wide range(0.01–1.80/d)and correlated with chl a distribution(R 2=0.181,P<0.001).However,the relationship was only signifi cant at<9μg/L chl a for mesotrophic waters and<16μg/L chl a for eutrophic waters.In contrast,g ranged from 0.00 to 1.01/d,and correlated withμat all locations.The g/μslope ranged from 19%to 84%,and was generally similar for waters around Peninsular Malaysia.However,all the g/μslopes had a positive y-intercept except for BMRS,and this seemed to suggest the availability of alternative prey supporting grazing at the other stations.

文摘A freshwater green microalgae Chlorella sp., UMACC344 was shown to produce high lipid content and has the potential to be used as feedstock for biofuel production. In this study, photosynthetic effciency, biochemical pro?les and non-targeted metabolic pro?ling were studied to compare between the nitrogen-replete and deplete conditions. Slowed growth, change in photosynthetic pigments and lowered photosynthetic effciency were observed in response to nitrogen deprivation. Biochemical pro?les of the cultures showed an increased level of carbohydrate, lipids and total fatty acids, while the total soluble protein content was lowered. A trend of fatty acid saturation was observed in the nitrogen-deplete culture with an increase in the level of saturated fatty acids especially C16:0 and C18:0, accompanied by a decrease in proportions of monounsaturated and polyunsaturated fatty acids. Fifty-nine metabolites, including amino acids, lipids, phytochemical compounds, vitamins and cofactors were signi?cantly dysregulated and annotated in this study. Pathway mapping analysis revealed a rewiring of metabolic pathways in the cells, particularly purine, carotenoid, nicotinate and nicotinamide, and amino acid metabolisms. Within the treatment period of nitrogen deprivation, the key processes involved were reshu ? ing of nitrogen from proteins and photosynthetic machinery, together with carbon repartitioning in carbohydrates and lipids.

文摘Water samples were collected in order to study the spatial variation of photosynthetic pigments and phytoplankton community composition in the Lembeh Strait(Indonesia) and the Kelantan River Estuary(Malaysia)during July and August 2016, respectively. Phytoplankton photosynthetic pigments were detected using high performance liquid chromatography combining with the CHEMTAX software to confirm the Chl a biomass and community composition. The Chl a concentration was low at surface in the Lembeh Strait, which it was 0.580–0.682 μg/L, with the average(0.620±0.039) μg/L. Nevertheless, the Chl a concentration fluctuated violently at surface in the Kelantan River Estuary, in which the biomass was 0.299–3.988 μg/L, with the average(0.922±0.992) μg/L. The biomass at bottom water was higher than at surface in the Kelantan River Estuary, in which the Chl a concentration was 0.704–2.352 μg/L, with the average(1.493±0.571) μg/L. Chl b, zeaxanthin and fucoxanthin were three most abundant pigments in the Lembeh Strait. As a consequence, phytoplankton community composition was different in the two study areas. In the Lembeh Strait, prasinophytes(26.48%±0.83%) and Synechococcus(25.73%±4.13%) occupied ~50% of the Chl a biomass, followed by diatoms(20.49%±2.34%) and haptophytes T8(15.13%±2.42%). At surface water in the Kelantan River Estuary, diatoms(58.53%±18.44%)dominated more than half of the phytoplankton biomass, followed by Synechococcus(27.27%±14.84%) and prasinophytes(7.00%±4.39%). It showed the similar status at the bottom water in the Kelantan River Estuary,where diatoms, Synechococcus and prasinophytes contributed 64.89%±15.29%, 16.23%±9.98% and 8.91%±2.62%,respectively. The different phytoplankton community composition between the two regions implied that the bottom up control affected the phytoplankton biomass in the Lembeh Strait where the oligotrophic water derived from the West Pacific Ocean. The terrigenous nutrients supplied the diatoms growing, and pico-phytoplankton was grazed through top down control in the Kelantan River Estuary.

文摘Antibiotics released into the environment through anthropogenic activities exert selective pressure,driving bacteria towards increasing antimicrobial resistance.The prevalence of antibiotics and the ecological risks posed in the riverine estuarine of Larut River and Sangga Besar River,which included wastewater effl uents from hospital,zoo,and poultry slaughterhouse sources were investigated.Solid phase extraction(SPE)followed by high-performance liquid chromatography tandem mass chromatography(HPLC-MS/MS)were used to extract and quantify the antibiotic residues from 22 antibiotics belonging to six major antibiotic classes(sulfonamide,macrolide,fl uoroquinolone,phenicol,trimethoprim,and tetracycline).Sixteen antibiotic residues were detected with concentrations ranging from limit of detection(LOD)to 1262.3 ng/L.Fluoroquinolones and macrolides were the most frequently detected compounds.Erythromycin,clarithromycin,and ofl oxacin detected in hospital and zoo effl uents posed a high risk to algae while tetracycline had low to medium ecological risks toward all the relevant organisms from aquatic environments(algae,invertebrate Daphnia magna,and fi sh).

文摘Length-weight relationship provides basic information of a fi sh’s condition while the morphometric data of otolith has been traditionally used to determine the range of fi sh sizes for various purposes.The length-weight relationship of Terapon jarbua(Forskål,1775)was examined from 165 specimens,while 132 specimens were used in sagittal otolith biometry measurements.Fish specimens were obtained from various locations in Malaysia including Pahang,Selangor,Sarawak,and Sabah.The total length of fi sh ranged 12.5–32.5 cm,while the total weight ranged 26.28–444.30 g.The otolith length ranged 0.423–1.070 cm,and the otolith weight was 0.0103–0.1572 g.The specimens displayed a mixture of positive and negative allometry growth as indicated by fl uctuating b values between 2.74 and 3.15.The overall coeffi cient of determination(R 2)between fi sh length and weight was 0.9729,which implies a strong correlation.All regressions between the fi sh length and otolith dimensions were highly signifi cant(P<0.001).The otolith dimensions found to be most closely related to the fi sh total length was the otolith weight.This was supported by the regression model showing a high R 2 value of 86.57%.The aspect ratio of otolith was 1.80±0.14(mean±SD).Results confi rm that the otolith growth refl ects the somatic growth of T.jarbua.The length-weight relationship explained the growth conditions of the fi shes in each location while the overall b value of 2.91 indicates that the condition of T.jarbua grown in Malaysian waters during the study period was generally in good condition.This study contributed to the database on the morphometric data of otolith of T.jarbua as well as the fi rst report on fi sh length-weight relationship for T.jarbua in Malaysia.

文摘In this work, we study the regional dependence of transport behavior of microalgae Chlorella vulgaris inside microflu- idic channel on applied fluid flow rate. The microalgae are treated as spherical naturally buoyant particles. Deviation from the normal diffusion or Brownian transport is characterized based on the scaling behavior of the mean square displacement (MSD) of the particle trajectories by resolving the displacements in the streamwise (flow) and perpendicular directions. The channel is divided into three different flow regions, namely center region of the channel and two near-wall boundaries and the particle motions are analyzed at different flow rates. We use the scaled Brownian motion to model the transitional characteristics in the scaling behavior of the MSDs. We find that there exist anisotropic anomalous transports in all the three flow regions with mixed sub-diffusive, normal and super-diffusive behavior in both longitudinal and transverse directions.

文摘Excessive rainfall is one of the triggers for the flooding phenomenon,especially in the tropics with flat or concave areas.Some critical points in the South Tangerang region,which are currently one of the most rapidly developing cities,cannot be ignored from the flooding problem.Floods cause disturbing human activities,loss of life and property,and in turn affect the economic stretch in an area.This paper aimed to predict rainfall by exploring the application of artificial intelligence techniques such as ANFIS(Adaptive Neuro Fuzzy Inference System).The proposed technique combines neural network learning abilities with transparent linguistic representations of fuzzy systems.The ANFIS model with various input structures and membership functions was built,trained,and tested to evaluate the capability of a model.Analyses of six-year rainfall data on a monthly basis in South Tangerang City,Banten found that rainfall prediction based on ANFIS time series is promising where 80%of data testing is well predicted.

文摘Ultraviolet(UV)filters are widely used in sunscreen and personal care products due to their ability to give protection to our skin from UV radiation.Oxybenzone,commonly known as benzophenone-3,is one type of UV filter found as the active ingredient in many pharmaceutical products.Although oxybenzone has been extensively studied as an environmental toxicant in the ecosystem,little is known about its toxicity effects on microalgae.The effects of oxybenzone on growth(measured as OD620 nm,chlɑand carotenoids)and macromolecular composition of polar microalgae(Chlorella UMACC 400 and Chlorella UMACC 401)and temperate microalgae(Chlorella sp.,Chlamydomonas reinhardtii,Scenedesmus quadricauda)were investigated.These microalgae were cultured in triplicate and exposed to different oxybenzone concentrations(0,100,200,300 and 400 mg·L-1),at 4℃and 18 for polar and temperate species respectively,for 96 h.The oxybenzone concentrations used℃represent a range from environmental to extreme concentrations to understand the impact of this toxicant on microalgae.The results showed that the highest concentration of oxybenzone(300 and 400 mg·L-1)had adverse effects on growth rate and biomass of these microalgae.However,exposure to oxybenzone concentrations ranging from 200 mg·L-1 to 400 mg·L-1 did not have significant effects on S.quadricauda growth.The exposure to oxybenzone at higher concentrations also led to changes in cell structure after 96 h.Generally,protein and carbohydrate content of all microalgae except S.quadricauda increased with increasing oxybenzone concentrations.Protein content increased significantly when cells were exposed to oxybenzone,though effects were greater in the polar species,suggesting that it could be one of the adaptive strategies that enabled these microalgae to tolerate oxybenzone.Further investigation is required to determine the effects of oxybenzone on other features of microalgal performance at relevant environmental concentrations.
文摘Upwelling off the east coast of Peninsular Malaysia (PM) was detected from recent cruise data collected during the southwest monsoon. Thermocline lifting was observed at 104?E from a number of parallel transects. To confirm the presence of upwelling, satellite remote sensing data were used, and numerical model experiments were conducted. A cooler sea-surface temperature along the coast was spotted from both in-situ and satellite data while upward movement from the model agreed with field data. The southwesterly wind that blows along PM from June to September is believed to be the important mechanism that contributed to this upwelling through an Ekman dynamics process.

文摘Despite more than a decade of study,there are still significant obstacles to overcome before graphene can be successfully produced on a large scale for commercial use.Chemical oxidation of graphite to produce graphene oxide(GO),followed by a subsequent reduction process to synthesize reduced graphene oxide(rGO),is considered the most practical method for mass production.Microorganisms,which are abundant in nature and inexpensive,are one of the potential green reductants for rGO synthesis.However,there is no recent review discussing the reported microbial reduction of GO in detail.To address this,we present a comprehensive review on the reduction of GO by a range of microorganisms and compared their efficacies and reaction conditions.Also,presented were the mechanisms by which microorganisms reduce GO.We also reviewed the recent advancements in using microbially reduced GO as the anode and cathode material in the microbial fuel cell(MFC)and algal biophotovoltaics(BPV),as well as the challenges and future directions in microbial fuel cell research.
