[Objective]The paper was to identify,isolate,and characterize porcine astrovirus in Shandong Province between 2021 and 2023.[Method]A total of 1025 samples of porcine diarrhea samples were collected from various regions of Shandong Province between January 2021 and October 2023.The samples were tested by RT-PCR,followed by sequencing and phylogenetic analyses of the polymerase.[Result]The total positive rate of PAstV was 34.6%(355/1025).The respective proportions of individuals infected with PAstV-1,PAstV-2,PAstV-4 and PAstV-5 were 25.4%(90/355),28.2%(100/355),35.2%(125/355)and 22.5%(80/355),respectively.Additionally,mixed infection was observed.Meanwhile,849 samples of healthy pigs were tested by RT-PCR,and the results demonstrated that the total positive rate of PAstV was 8.13%(69/849).Of these,the proportion of PAstV-1,PAstV-2 and PAstV-4 infection was 27.5%(19/69),37.7%(26/69)and 40.6%(28/69),and a mixed infection also existed.Further sequencing and characterization of some the selected isolates revealed low sequence identities(56.2%)with known PAstV strains,indicating the presence of novel types or genotypes of PAstVs.Furthermore,the isolation conditions of porcine astrovirus were optimized,resulting in the purification of a pure PAstV-4 strain(designated PAstV-4-GRF1).The virus was found to exhibit typical astroviral morphology,with nucleotide identity ranging from 89.9 to 95.4%with previously published PAstV-4 strains.Then,macrovirus transcriptome sequencing showed that 88.30%of the CRF1 samples were mammalian astroviruses.By species classification,PAstV 4 and PAstV 2 accounted for 21.79%and 0.32%,respectively.Phylogenetic tree analysis showed that the c15050 fragment was identical to the GRF-1 sequencing fragment of the isolated strain,and exhibited the highest homology with the Hunan PAstV-4 sequence MK460231 in China.[Conclusion]As the inaugural isolated PAstV-4 strain,it furnishes pivotal materialfor the investigation ofthe biological and pathogenic properties of this virus as well as for the prospectivedevelopment of relevant biological and diagnostic reagents. 展开更多

The trial lasted 50 d.[Result] The final weight, daily gain and feed intake of experimental group were significantly higher than those of control group(P<0.05), but the feed/gain ratio of experimental group was markedly lower than that of control group(P<0.05). Compared with the control group, the gross profit of experimental group significantly increased( P <0.05).[Conclusion] Pelleted total mixed ration led to better production performance of growing Hainan black goat. 展开更多

[Method] A tntal of 36 Hainan black pigs with the body weight of (46.34±3.06) kg were randomly divided into three groups: con- trol group (protein level 15%, lysine level 0.83%), experimental grnup 1 (protein level 13%, 0.83%), and experimental group II (protein level 13%, lysine level 1.03), each group had three replicates of four pigs. The trial lasted 35 d. [Result] The growth performance of Hainan black pig in grnup II had no significan! difference with that in contrail group (P〉0.05). The apparent digestibility of chide protein and phosphorus of Hainan black pigs in Group II increased significantly (P〈0.05), which were increased by 10.08% and 11.85% respectively compared to the control group; the apparent digestibility of calcium was increased, but the difference was not signifieant (P〉0.05). [Conclusion] It will not reduce the growth performance of Hainan blaek pig when the protein level of diet is reduced by 2% and Ihe lysine level is increased to 1.03%. [Objective] The study was carried out to investigate the effects of dietary crude fiber on growth performance and serum biochemical in-dex of Dingan geese aged from 35 to 70 days.[Method] A total of 120 Dingan geese on day 28 with the same genetic background and similar initial weight(1 384.83.73±0.76 g) were randomly assigned into 3 groups, with 4 replicates per group and 10 geese per replicate. Geese were fed the diets containing 4.29%, 5.29% and 6.29 % crude fiber, respectively. The experiment lasted 35 d.[Result] Final weight and daily gain of 5.29 %CF group were significantly higher than those of 6.29 % CF group(P<0.05);Feed intake was markedly higher than those of 4.29% and 6.29 % CF groups(P<0.05);Feed gain ratio was significantly lower than that of 6.29 % CF group(P<0.05). Compared with 4.29% and 6.29 % CF groups, serum albumin concentrations of 5.29 % CF group increased significantly(P<0.05), while serum total protein and globin contents were higher(P>0.05), but serum uric acid level was lower(P>0.05).[Conclusion] 5.29 % crude fiber could lead to higher growth performance of Dingan geese. 展开更多

In order to compare the slaughter performance and meat quality of Hainan black goats and hybrid offsprings of Nubian Black Goat and Hainan Black Goat, six 7-month-old Hainan black goats and six hybrid offsprings were selected to determine slaughter performance and muscle quality. The results showed that hybridization of Nubian black goat and Hainan black goat significantly improved the production performance and slaughter performance of hybrid offsprings (P>0.05). The meat quality of F1 hybrid black goat had no significant difference with that of Hainan black goat except for the hardness and elasticity (P<0.05). Therefore, the crossbred progenies of Nubian black goat and Hainan black goat have advantages in meat yield and meat performance, and the main meat quality indexes are not significantly different, which meets the market requirements for high-yield and high-quality hybrid combinations, and can be used for cross promotion. 展开更多

N-Linked glycosylation of hemagglutinin(HA) has been demonstrated to regulate the virulence and receptor-binding specificity of avian influenza virus(AIV).In this study,we characterized the variation trend of naturally isolated H9 N2 viruses for the potential N-linked glycosylation sites in HA proteins,and explored any important role of some glycosylation sites.HA genes of 19 H9 N2 subtype AIV strains since 2001 were sequenced and analyzed for the potential glycosylation sites.The results showed that the viruses varied by losing one potential glycosylation site at residues 200 to 202,and having an additional one at residues 295 to 297 over the past few years.Further molecular and single mutation analysis revealed that the N200 Q mutation lost an N-linked glycosylation at positions 200 to 202 of the HA protein and affected the human-derived receptor affinity.We further found that this N-linked glycosylation increased viral productivity in the lung of the infected mice.These findings provide a novel insight on understanding the determinants of host adaption and virulence of H9 N2 viruses in mammals. 展开更多

Goats were fed with Juncao+concentrate(control group), pel-leted total mixed ration I(particle size 6 mm, experimental group I) and II(particle size 5 mm, experimental group II), respectively. The trial lasted 45 d.[Result](1) The final weight and daily gain of experimental group II were significantly higher than those of control group( P<0.05), and the daily gain of experimental group II was also higher than that of experimental group I(P<0.05);the feed intake of experimental group II was markedly higher than that of control group(P<0.05), but was significantly lower than that of experimental group I(P<0.05). The feed/gain ratio of experimental group II was significantly lower than those of control group and experimental group I(P<0.05).(2) The feed gain cost of experimental group II was markedly lower than that of control group(P<0.05), but the gross profit was significantly higher than those of control group and experimental group I(P<0.05).(3) Compared with the control group, the serum total protein content of experimental group II increased markedly( P<0.05) while the urea nitrogen level decreased(P>0.05).[Conclusion] The pelleted total mixed ration with the particle size of 5 mm obtained better fattening ef-fect, and obviously improved the serum total protein level. 展开更多

To detect the effects of the polyclonal antibodies raised in sheep against porcine adipocyte plasma membranes on the porcine carcass composition and meat quality, 30 pigs assigned into 6 treatment groups were given intraperitoneal injections of sheep antipig adipocyte plasma membrane immunoglobulin (ASIg) or sheep nonimmune serum immunoglobulin (NSIg). At the end of the experiment, the pigs were slaughtered at 90 kg body weight, and carcasses and meat quality were evaluated. The results showed that when pigs intraperitoneally immunized with 20 or 30 mg ASIg at 15 kg body weight, 20 mg purified ASIg twice at 15 and 60 kg body weight, or 20 mg purified ASIg at 60 kg body weight, respectively, their lean meat percentage, fat meat percentage, backfat thickness, loin eye area leaf fat weight, caul fat weight, heart weight, liver weight, and kidney weight were significantly affected. However, the kidney weight, lurrg weight, dressing percentage, and spleen weight did not remarkably change. Our results indicated that pigs intraperitoneally immunized with 20 or 30 mg ASIg at 15 kg body weight, and 20 mg ASIg twice at 15 and 60 kg body weight, have significantly different drip loss rate, cooked meat ratio, tenderness, storage loss rate, muscle fiber diameter, moisture content, dry matter content, crude protein content, and crude fat content from the control group that received 20 mg NSIg at 15 kg body weight. However, meat pH, meat color value, meat marbling score, inosinate, and myohemoglobin were not significantly affected. Our results indicated ASIg could not significantly affect the content of most muscular amino acids and intramuscular fatty acids. 展开更多

The results showed that the genotype frequencies of ESR and PRLR genes were in Hardy-Weinberg equilibrium other than the genotype frequency of FSHβ gene. Allele A of FSHβ, ESR and PRLR gene had a positive additive effect on reproductive traits. The favorable genotypes of FSHβ and ESR genes was AA- type, while in PRLR gene was AB-type in the 112 pig individuals detected. With regard to FSHβ and ESR genes, AA-type individuals' TNB of were respectively 1.54 (P 〈0.05) and 2.13 (P 〈 0.01 ) higher than the BB-type individuals'; AA-type individuals' AN B of the two genes were respectively 1.55 ( P 〈 0.05) and 1.82 ( P 〈 0.01 ) higher than the BB-type individuals'. Concerning PRLR gene, AB-type individuals' TNB was 0.77 ( P 〉 0.05 ) and 1.59(P〈0.05) higher than that of AA and BB-type individuals respectively, and ANB was 0.65(P〉0.05) and 1.8( P〈0.01 ) higher than AA and BB-type individuals' respectively. Moreover, our results also showed that the AAAABB combination genotype of FSHβ , ESR, PRLR genes bore more than all other combination genotypes, but only 2 individuals were proved to be AAAABB genotype. Consequently, pyramiding FSHβ and ESR genes could to a large extent enhance TNB and NBA of individuals. Therefore, we advise, in next generation that those individuals with AAAA combination genotype of FSHβ and ESR should be selected to increase frequency of allele A, whether PRLR gene is proper for pyramiding with the other genes or not is still to be studied further. 展开更多

Numbers of vertebrae is an important economic trait associated with body size and meat productivity in animals.However,the genetic basis of vertebrae number in donkey remains to be well understood.The aim of this study was to identify candidate genes affecting the number of thoracic(TVn)and the number of lumbar vertebrae(LVn)in Dezhou donkey.A genome-wide association study was conducted using whole genome sequence data imputed from low-coverage genome sequencing.For TVn,we identified 38 genome-wide significant and 64 suggestive SNPs,which relate to 7 genes(NLGN1,DCC,SLC26A7,TOX,WNT7A,LOC123286078,and LOC123280142).For LVn,we identified 9 genome-wide significant and 38 suggestive SNPs,which relate to 8 genes(GABBR2,FBXO4,LOC123277146,LOC123277359,BMP7,B3GAT1,EML2,and LRP5).The genes involve in the Wnt and TGF-βsignaling pathways and may play an important role in embryonic development or bone formation and could be good candidate genes for TVn and LVn. 展开更多

The product of PCK was cloned into pMD18-T vector, and then transfected Escherichia Coli JMI00. The recombinant plasmids were amplified by PCR and were sequenced. From the nucleotide sequence of the amplified products, phylogenetie analyses were performed and genotypes or subgenotypes were identified. The results indicated that the E2 gene fragment of HB-DCZ strain contained 1277bp nucleotides, and had 89.4%, 70.7%, 97.6%, 68.9%, 67.2% sequence similarity with Osloss, OregonC24V, Changchun184, ZM195, NADL, respectively. Since late 2005, the swine industry in Shandong province has experienced a significant increase in postweaning multisystemic wasting syndrome (PMWS). Porcine circovirus type 2 (PCV2) is one of the major etiologic agents of PMWS. In order to understand the genetic diversity of PCV2 in swine herds in Shandong province, we sequenced six complete genomes of PCV2 between 2005 and 2007. Sequence analysis revealed that all the six PCV2 strains belonged to one genotype (named PCV 2b). The deduced amino acid sequences of the replication associated protein (coded by ORFI) showed that all the six PCV2 strains belonged to genotype 2a and could be further divided into two clusters. Based on the analysis of the capsid protein (coded by ORF2), the six PCV2 strains belonged to genotype 2b. All the six strains belonged to recombinant viruses between genotype PCV2a and genotype PCV2b. This study implied that such recombinant PCV2 spread widely in Shandong area and might be the reason that pig death rate associated with PMWS increased in recent years. 展开更多

The enzyme △3,△2-dienoyl-CoA isomerase (ECI1) plays a crucial role in the mitochondria113-oxidation of fatty acids with a double-bond in odd and even positions. The ECll gene might be a qualified candidate for studies pertaining to lipid deposition and meat quality in swine. In the present study, ECI1 cDNA of the Tibetan pig was obtained by in silico cloning and verified by PCR analysis. Single-nucleotide polymorphisms (SNPs) of ECll were screened by PCR-sequencing and genotypes of those SNPs were tested by PCR-restriction fragment length polymorphism (PCR-RFLP) in Diannan small-ear pigs (DSP, n=40), Tibetan pigs (TP, n=60) and Yorkshire pigs (YP, n=30). The expression levels of ECll were analyzed by real-time quantitative PCR and Western blotting in tissues of the liver, backfat, and Iongissimus dorsi (LD) muscle of DSP (n=8), TP (n=8) and YP (n=8). Single factor linear correlation analysis was applied separately for each breed to evaluate correlations between ECll gene expression in the LD muscle and intramuscular fat (IMF) content. We obtained an ECll gene length of 1 401 bp from the cDNA that contained a full coding region of 909 bp. Three novel SNPs (g.42425337G〉A; g.42424666A〉G; and g.42422755A〉G) were detected, and only g.42424666A〉G exhibited three genotypes among the three breeds. The ECll expression levels in the LD muscle of DSP and TP were significantly higher than that of YP (P〈0.05). Moreover, TP had the highest ECI1 expression in backfat (P〈O.01), and a positive correlation was observed between gene expression and IMF content. The results suggest that differences in ECI1 gene expression might be related to lipid depo- sition and meat quality in pig. 展开更多

[Objective] This paper was to study effects of pelleted total mixed ration with different concentrate-roughage ratios on production performance and serum biochemical parameters of fattening Hainan black goats.[Method] Thirty-six fattening Hainan black goats with the same genetic background and similar initial weight of(14.08±0.13) kg were randomly assigned into three treatments. Goats were fed with Juncao + concentrate(control group), pelleted total mixed ration I(concentrate: roughage 50 ∶50, group Ⅰ) and Ⅱ(concentrate: roughage 45∶55, group Ⅱ), respectively. The test lasted 45 d.[Result] The final weight, daily gain and feed intake of rams in experimental group Ⅰ were significantly higher than those of control group and experimental group Ⅱ(P<0.05);the final weight and daily gain of rams in experimental group Ⅰ were also markedly higher than those of experimental group Ⅱ(P<0.05);the feed/gain of rams in experimental group Ⅰ was significantly lower than those in control group and experimental group Ⅱ(P<0.05). The feed gain cost of rams in experimental group Ⅰ was markedly lower than that in experimental group Ⅱ( P<0.05), but was significantly higher than that in experimental group Ⅰ(P<0.05);the gross profit of rams in experimental group Ⅰ was significantly higher than those in control group and experimental group Ⅱ(P<0.05). Compared with control group and experimental group Ⅱ, the serum total protein and glucose lev-els in experimental group Ⅱ increased significantly(P<0.05), but the urea nitrogen content reduced(P>0.05).[Conclusion] When the concentrateroughage ratio of pelleted total mixed ration was 50∶50, the fattening effect of Hainan black goat was better, and the serum total protein and glucose levels were evidently improved. 展开更多

The aim of this study was to determine the genomic evolutionary pattern of virulent equine infectious anemia virus (EIAV) during persistent infection. The evolutionary dynamics of proviral genomes were examined by challenging an EIAV seronegative equine (pony 1) and three EIAV vaccinated equines (ponies 4, 7, and 8) with the Chinese virulent strain EIAV- L. Ponies 1 and 7 succumbed to disease and were called progressors, while ponies 4 and 8 lacked clinical symptoms and were considered nonprogressors. Sequences spanning the V3, V4, and V5 hyper-variable regions of the EIAV-L envelope gp90 gene were sequenced from each pony as evolutionary markers of the provirus. The proviral genome of the EIAV-L inoculum evolved during persistent infection and displayed different patterns between EIA progressors and nonprogressors. Inoculum-like variants were isolated from nonprogressors during persistent infection, but only from progressors during acute infection. Variant mutations from nonprogressors were dispersed throughout the sequenced region, while those from progressors were predominantly localized to V3. Humoral immunity and virus variant population selection analyses indicated that immune selection was positive in chronically infected progressors and weak in nonprogressors. In-frame stop codons were frequently localized to a defect "hot spot". The high number of defective variants in nonprogressors may promote disease survival. 展开更多

The results showed that the homology of the nucleotide sequences of protein N was 99.7% between the isolate and the classical strains (VR2332 and SD1), and 93.4% between the isolate and the highly pathogenic strains (JXA1, HuN4, SX-1 and TJM). The results suggested that the virus was classical PRRSV in the pig farm. The research would provide a reference for the diagnosis and targeted therapy of classical PRRSV infection in swine production. 展开更多

In order to obtain the TLR 4 gene sequence of Hainan Wuzhishan pig and analyze its molecular structure characteristics,its genomic sequence was cloned and sequenced by PCR and subjected to related bioinformatics analysis with the genomic sequence of the TLR 4 gene of pig in GenBank as a reference sequence.The full-length DNA sequence of Wuzhishan pig TLR 4 gene was 10 435 bp in length,of which CDS was 2 526 bp in length and encoded 841 amino acids.Compared with the common pig TLR 4 gene sequence published by GenBank,there were 38 mutations,of which two mutations were in the CDS region.The results of homology analysis showed that the amino acid sequence of the TLR 4 gene of Wuzhishan pig shared 99.9%,80.7%,79.9%,74.8%,74.7%,73.1%,73.0%,72.7%,62.5% and 43.4% homology with those of common pig,cattle,sheep,dog,horse,macaque,human,chimpanzee,mouse and chicken,respectively.The protein encoded by the TLR 4 of Wuzhishan pig belonged to secreted protein and transmembrane protein.It was hydrophilic,and had 8 glycosylation modification sites,17 serines (ser),7 threonines (Thr) and 8 tyrosine (TYR) phosphorylation sites.The amino acid phylogenetic tree analysis showed that the TLR 4 genes of different species are highly conserved during evolution.This result lays a foundation for further study of the function of TLR 4 gene. 展开更多
