Background Recently, arsenic trioxide (As 2O 3) was considered as a novel anti tumor agent However, it showed severe toxicity effect on normal tissue at the same time To improve its therapeutic efficacy and decrease its toxicity,we prepared arsenic trioxide loaded albuminutes immuno nanospheres [As 2O 3 (HAS NS) BDI 1] targeted with nonoclonal antibody (McAb) BDI 1 and tested its specific killing effect against bladder cancer cell Methods As 2O 3 HAS NS was prepared by chemical cross linking method Monoclonal antibody BDI 1 was purified with ammonium sulphate saltingout and chromatography Albuminutes microspheres were conjugated with McAb by SPDP cross linking method Concentration of As in As 2O 3 (HAS NS) BDI 1 and As 2O 3 HAS NS was measured by atomic fluometry method As 2O 3 (HAS NS) BDI 1 and its activity were detected by SDS PAGE reduction electrophoresis, indirect immunofluorescence test, light microscope and scanning electron microscope observation Acridine orange staining and tritiated thymidine ( 3H TdR) incorporation tests were used to indicate specific killing activity of As 2O 3 (HAS NS) BDI 1 in vitro Results In As 2O 3 (HAS NS) BDI 1 groups, we saw two protein bands in SDS PAGE reduction electrophoresis Albuminutes immuno nanospheres were rounded with clear green fluorescence by immunofluorescence test Under microscope, we observed that BIU 87 cells were covered with the As 2O 3 (HAS NS) BDI 1 and that As 2O 3 (HAS NS) BDI 1 moved with the BIU 87 cells The albuminutes immuno nanospheres were tightly junctioned with the BIU 87 cells Specific killing activity of As 2O 3 (HAS NS) BDI 1 on bladder tumor cells was observed by acridine orange staining and 3H TdR incorporation assays Conclusions As 2O 3 (HAS NS) BDI 1 might bind specifically against BIU 87 cells, thus leading to high activity of killing bladder tumor cells 展开更多

We demonstrate that a specific protein-protein... Wnt signaling directs cell-fate choices during embryonic development and tissue tumorigenesis. T cell fac-tor 4 (TCF4) plays a pivotal role in the Wnt signaling path-way. We demonstrate that a specific protein-protein interac-tion occurs between TCF4 and ATF5 (activating transcrip-tion factor 5) —— a new member of cAMP response element binding protein (CREB) with the yeast two-hybrid system. The N-terminal and DNA binding domain of TCF4 (TCF4ND, 1—495 aa) and the C-terminal spanning bZIP domain of ATF5 (162—282 aa) were found to be responsible for the interaction, and the C-terminal of ATF5 (ATF5/C) showed a much stronger interaction with TCF4ND than the full-length of ATF5 by detecting the b-gal activity. Further-more, overexpression of ATF5/C enhanced transcriptional activation by TCF4 proteins in luciferase assay by transient transfection. Taken together, these data suggest that ATF5 may function as a co-activator to potentiate the ability of TCF4 to activate transcription. 展开更多

To assess the intravesical application of immunotoxin as adjuvant therapy to prevent recurrence after tumor resection in bladder cancer patients Methods An anti human immunotoxin against bladder carcinoma, BDI 1 RT, was prepared and its in vitro targeting cytotoxicity estimated The immunoreactivity of BDI 1 RT with human bladder cancer tissue of different grades and stages was detected by immunohistochemical analysis After safety test, intravesical administration of BDI 1 RT was performed in 31 patients while mitomycin C (MMC) was used in 36 patients serving as a control group The recurrence rates and side effects in both groups were recorded In addition, the development of human anti mouse antibodies (HAMA) was determined by ELISA, to assess the potential safety of this immuotoxin Results In our study, BDI 1 RT had immunoreactivity with 81 6% of bladder transitional cell carcinomas The immunoreactivity of BDI 1 RT correlated with tumor grade High grade carcinoma had stronger staining than low grade ( P <0 05) There was no significant difference between the BDI 1 RT group (10%) and MMC group (19 3%) in recurrence rate ( P >0 05) Side effects, including systemic and local, were more frequent in the MMC group (11 of 36 patients versus 2 of 31, P <0 05) HAMA was not detected in any of 7 patients Conclusion Immunotoxin may have considerable potential in the prophylaxis of bladder transition cell carcinoma 展开更多
