This study aims at the effects of an intake of low molecular weight corn peptides(LMCPs) prepared from zein on alcohol metablism in rats. LMCPs(1.0 g/kg body weight) in 15% ethanol(10 mL/kg body weight) were given to Wister rats by intragastric gavage. The assay of blood ethanol was conducted by using the enzyme based assay kit. The amino acid analysis was made with an amino acid analyzer. The data of the animal experiments showed that LMCPs could accelerate the metabolism of alcohol in rats. In the control group, the blood ethanol concentration reached the maximum level of (827.0±77.3) mg/L after ethanol loading for 30 min, then gradually decreased. In contrast, the blood ethanol concentration only reached (527.25±47.0) mg/L after 30 min in the group of LMCPs taken. These results indicate that LMCPs could decrease ethanol concentration in blood rapidly. 展开更多

A Pb 2+ precipitation method was designed to get rid of the impure proteins in a lipase. The results show that it was a simple way in the primary treatment of the crude samples and about 20% impure proteins were removed in the precipitation step. Further, continuous elution electrophoresis was also applied as a preparative technique for attaining the highly pure lipase. During the continuous elution electrophoresis, the enzyme was eluted as a single peak and 5.7-fold purification was achieved in a yield of 54.3%. The two steps finally yielded an electrophoretically homogeneous enzyme. 展开更多

To enhance the relative movement of domains, we inserted a random sequence of fifteen-peptide into the three domains of L-aspartase. By means of directed screening, the three isoforms of monomeric, dimmeric and tetrameric enzymes were obtained. Compared to the wild-type tetrameric L-asparease, these mutants remained 19.7%, 42.3%, and 92% of the enzyme activity, respectively. Moreover, the examination of enzyme properties revealed that their k_ cat and K_M changed in varying degrees, and the optimum pH shifted towards acidic pH, while the dependence of the activity of enzyme on Mg 2+ concentration and thermostability increased. Therefore this strategy provides a novel approach to directed evolution of enzymes. 展开更多
