The success of deep transfer learning in fault diagnosis is attributed to the collection of high-quality labeled data from the source domain.However,in engineering scenarios,achieving such high-quality label annotatio...The success of deep transfer learning in fault diagnosis is attributed to the collection of high-quality labeled data from the source domain.However,in engineering scenarios,achieving such high-quality label annotation is difficult and expensive.The incorrect label annotation produces two negative effects:1)the complex decision boundary of diagnosis models lowers the generalization performance on the target domain,and2)the distribution of target domain samples becomes misaligned with the false-labeled samples.To overcome these negative effects,this article proposes a solution called the label recovery and trajectory designable network(LRTDN).LRTDN consists of three parts.First,a residual network with dual classifiers is to learn features from cross-domain samples.Second,an annotation check module is constructed to generate a label anomaly indicator that could modify the abnormal labels of false-labeled samples in the source domain.With the training of relabeled samples,the complexity of diagnosis model is reduced via semi-supervised learning.Third,the adaptation trajectories are designed for sample distributions across domains.This ensures that the target domain samples are only adapted with the pure-labeled samples.The LRTDN is verified by two case studies,in which the diagnosis knowledge of bearings is transferred across different working conditions as well as different yet related machines.The results show that LRTDN offers a high diagnosis accuracy even in the presence of incorrect annotation.展开更多
Artificial immune detection can be used to detect network intrusions in an adaptive approach and proper matching methods can improve the accuracy of immune detection methods.This paper proposes an artificial immune de...Artificial immune detection can be used to detect network intrusions in an adaptive approach and proper matching methods can improve the accuracy of immune detection methods.This paper proposes an artificial immune detection model for network intrusion data based on a quantitative matching method.The proposed model defines the detection process by using network data and decimal values to express features and artificial immune mechanisms are simulated to define immune elements.Then,to improve the accuracy of similarity calculation,a quantitative matching method is proposed.The model uses mathematical methods to train and evolve immune elements,increasing the diversity of immune recognition and allowing for the successful detection of unknown intrusions.The proposed model’s objective is to accurately identify known intrusions and expand the identification of unknown intrusions through signature detection and immune detection,overcoming the disadvantages of traditional methods.The experiment results show that the proposed model can detect intrusions effectively.It has a detection rate of more than 99.6%on average and a false alarm rate of 0.0264%.It outperforms existing immune intrusion detection methods in terms of comprehensive detection performance.展开更多

We...Objective: Verify if the educational technology based on Just in Time Education is relevant to Permanent Health Education in Workers’ Health. Method: This study is exploratory and descriptive qualitative research. Were performed semi-structured interviews were carried out with the worker’s health team of a large company from the South of Brazil region. The collected data were carried out in 2021, between January and May, through the virtual environment, with eight participants from the Work Health Team of Eletrosul, located in Florianópolis, who agreed to participate in the research by signing the Free and Informed Consent Term, in accordance with the data saturation criterion. The research was approved by the Ethics Committee (n. 4079133) and the data were submitted to Bardin for thematic content analysis. Result: The results are presented from three categories: 1) Expansion and inclusion in Permanent Education needs;2) Tools adopted in educational processes;3) Just in Time Education: educational technology for workers’ health. Conclusion: The active methodology Just in Time Education helps the processes of Permanent Education in Health, which moved from a face-to-face model to e-learning and required the knowledge renewal in the technology area to maintain efficient communication. This methodology allows quick and accurate access to information from a reliable, adequate, and up-to-date source, causing a positive impact on the qualified and problem-solving work process, in line with the needs of the employer and the company. Therefore, Just in Time Education can provide standardization, organization, and informational and operational quality necessary in this context, in addition to serving as an indicator and data generator (management tool).展开更多

When severe, EIB can ...Background: Exercise-Induced Bronchospasm (EIB) is an inflammatory condition characterized by severe airway constriction following the mobilization of inflammatory cells and interleukin-6 (IL-6). When severe, EIB can require the use of pressurized salbutamol to treat athletes. This study investigated the nature of the systemic changes in inflammatory cells and post-exercise IL-6 concentrations after salbutamol treatment in EIB-susceptible distance runners. Materials and Methods: This was an experimental study that enrolled 12 long-distance runners. In Session A, the participants completed a treadmill exercise test, and those who had a maximum expiratory volume per second (FEV1) that was decreased by at least 10% compared to their base value were placed in the EIB-susceptible group (EIB+) (n = 6). Those whose FEV1 did not meet this criterion were placed in the nonresponsive (EIB?) group (n = 6). Before the Session B exercise, athletes in the BIE+ group inhaled two puffs of salbutamol (EIB+ Salb), while their EIB? counterparts received no treatment. Spirometry was performed before and after the exercise using a Spirobank G portable spirometer. Blood samples were taken before, immediately after and 2 hours after the stress test. Results: The mean post-exercise FEV1 values were not significantly different (p > 0.05) between the EIB+ Salb group and the EIB? group. The systemic changes in inflammatory cells and IL-6 concentrations in the EIB+ runners after salbutamol treatment were similar to those observed in their EIB? counterparts. Conclusion: Salbutamol pretreatment improved the systemic immune status of EIB-susceptible athletes.展开更多

Because of the progressive neurodegeneration, individual cognitive and behavioral functions are impaired, affecting the quality of life of millions of people. Although the exact pathogenesis of AD has not been fully elucidated, amyloid plaques, neurofibrillary tangles (NFTs), and sustaining neuroinflammation dominate its characteristics. As one of the major tau kinases leading to hyperphosphorylation and aggregation of tau, glycogen synthase kinase-3β (GSK-3β) has been drawing great attention in various AD studies. Another research focus of AD in recent years is the inflammasome, a multiprotein complex acting as a regulator in immunological reactions to exogenous and endogenous danger signals, of which the Nod-like receptor (NLR) family, pyrin domain-containing 3 (NLRP3) inflammasome has been studied mostly in AD and proven to play a significant role in AD development by its activation and downstream effects such as caspase-1 maturation and interleukin (IL)-1β release. Studies have shown that the NLRP3 inflammasome is activated in a GSK-3β-dependent way and that inhibition of the NLRP3 inflammasome downregulates GSK-3β, suggesting that these two important proteins are closely related. This article reviews the respective roles of GSK-3β and the NLRP3 inflammasome in AD as well as their relationship and interaction.展开更多

This study ai...Context: Fine particles (PM2.5 and PM10) can accumulate in classrooms and in schoolyards located near urban roads. PM2.5 and PM10 can initiate, develop and exacerbate exercise-induced bronchospasm (EIB). This study aimed to assess the concentration levels of PM2.5 and PM10 in schools and to determine the rate of sensitivity to EIB among schoolchildren. Methods: A total of 128 students (67 girls and 61 boys) with an average age of 11 participated in this study. An ISAAC II questionnaire on respiratory symptoms was administered. PM2.5 and PM10 were measured. A 6-minute stress test was performed. Spirometry was performed. The ratio (I/O) of the concentrations of PM2.5 and PM10 recorded inside (I) the classrooms to those obtained outside (O) the classrooms was equal to 1 (I/O = 1). Results: The concentrations of PM2.5 and PM10 recorded inside the classrooms and those obtained outside the classrooms were higher than the values recommended by the WHO. 29 students out of 128 were diagnosed as sensitive to EIB [EIB (+)], i.e., a rate of sensitivity to EIB of 22.66%. A drop in postexercise PEF of 17.396% was observed among EIB (+) students. Conclusion: Schoolchildren in schools located near highways are exposed to high levels of PM2.5 and PM10 concentrations. Exposure to PM2.5 and PM10 played an important role in the initiation and exacerbation of exercise-induced bronchospasm in schoolchildren from schools located near highways. Effective programs for improving air quality in schools must be put in place to reduce the effects of particulate pollution on the respiratory health of school children.展开更多
Objective Post-stroke cognitive impairment(PSCI)develops in approximately one-third of stroke survivors and is associated with ingravescence.Nonetheless,the biochemical mechanisms underlying PSCI remain unclear.The st...Objective Post-stroke cognitive impairment(PSCI)develops in approximately one-third of stroke survivors and is associated with ingravescence.Nonetheless,the biochemical mechanisms underlying PSCI remain unclear.The study aimed to establish an ischemic mouse model by means of transient unilateral middle cerebral artery occlusions(MCAOs)and to explore the biochemical mechanisms of p25/cyclin-dependent kinase 5(CDK5)-mediated tau hyperphosphorylation on the PSCI behavior.Methods Cognitive behavior was investigated,followed by the detection of tau hyperphosphorylation,mobilization,activation of kinases and/or inhibition of phosphatases in the lateral and contralateral cerebrum of mice following ischemia in MACO mice.Finally,we treated HEK293/tau cells with oxygen-glucose deprivation(OGD)and a CDK5 inhibitor(Roscovitine)or a GSK3βinhibitor(LiCl)to the roles of CDK5 and GSK3βin mediating ischemia-reperfusion-induced tau phosphorylation.Results Ischemia induced cognitive impairments within 2 months,as well as causing tau hyperphosphorylation and its localization to neuronal somata in both ipsilateral and contralateral cerebra.Furthermore,p25 that promotes CDK5 hyperactivation had significantly higher expression in the mice with MCAO than in the shamoperation(control)group,while the expression levels of protein phosphatase 2(PP2A)and the phosphorylation level at Tyr307 were comparable between the two groups.In addition,the CDK5 inhibitor rescued tau from hyperphosphorylation induced by OGD.Conclusion These findings demonstrate that upregulation of CDK5 mediates tau hyperphosphorylation and localization in both ipsilateral and contralateral cerebra,contributing to the pathogenesis of PSCI.展开更多
Objective Schizophrenia(SZ)is associated with cognitive impairment,and it is known that the activity of cAMP response element binding protein(CREB)decreases in the brain of SZ patients.The previous study conducted by ...Objective Schizophrenia(SZ)is associated with cognitive impairment,and it is known that the activity of cAMP response element binding protein(CREB)decreases in the brain of SZ patients.The previous study conducted by the investigators revealed that the upregulation of CREB improves the MK801-related SZ cognitive deficit.The present study further investigates the mechanism on how CREB deficiency is associated with SZ-related cognitive impairment.Methods MK-801 was used to induce SZ in rats.Western blotting and immunofluorescence were performed to investigate CREB and the CREB-related pathway implicated in MK801 rats.The long-term potentiation and behavioral tests were performed to assess the synaptic plasticity and cognitive impairment,respectively.Results The phosphorylation of CREB at Ser133 decreased in the hippocampus of SZ rats.Interestingly,among the upstream kinases of CREB,merely ERK1/2 was downregulated,while CaMKII and PKA remained unchanged in the brain of MK801-related SZ rats.The inhibition of ERK1/2 by PD98059 reduced the phosphorylation of CREB-Ser133,and induced synaptic dysfunction in primary hippocampal neurons.Conversely,the activation of CREB attenuated the ERK1/2 inhibitor-induced synaptic and cognitive impairment.Conclusion These present findings partially suggest that the deficiency of the ERK1/2-CREB pathway is involved in MK801-related SZ cognitive impairment.The activation of the ERK1/2-CREB pathway may be therapeutically useful for treating SZ cognitive deficits.展开更多

The present study aims to evaluate the level of concentrations of fine particles (PM2.5 and PM10) near road traffic and the effects of exposure to automobile pollutants on pulmonary function and arteriolar blood oxygen saturation among sellers around road traffic. Materials and Methods: The study recruited 48 healthy people carrying out a sales activity near road traffic. PM2.5 and PM10 measurements were taken from 6 a.m. to 6 p.m. using a Temtop Airing-1000 portable particle detector. Spirometric measurements were taken in the morning and in the evening from a portable Spirobank G spirometer. Oxygen saturation measurements were also taken in the morning and evening using a Pulse oxymeter CMS50D pulse oxymeter. Results: Mean values of forced vital capacity (FVC), forced expiratory volume in one second (FEV1), peak expiratory flow (PEF), and forced expiratory flow between 25% and 75% of forced vital capacity (FEF25 - 75) recorded in the evening were significantly lower than those obtained in the morning in the subjects of the total group and in the men. Only the mean values of PEF and FEF25 - 75 obtained in the evening were significantly lower than those recorded in the morning in women. Minimum PM2.5 values recorded near road traffic were negatively correlated with evening PEF in men, while maximum PM10 values were negatively correlated with evening PEF in women. The blood oxygen saturation recorded in the evening was significantly lower than that obtained in the morning. Conclusion: Short-term exposure to automobile pollutants has adverse effects on lung function and oxygen saturation in people carrying out a sales activity near road traffic.展开更多

Background:Autophagy dysfunction plays a crucial role in tau accumulation and neurodegeneration in Alzheimer’s disease(AD).This study aimed to investigate whether and how the accumulating tau may in turn affect autop...Background:Autophagy dysfunction plays a crucial role in tau accumulation and neurodegeneration in Alzheimer’s disease(AD).This study aimed to investigate whether and how the accumulating tau may in turn affect autophagy.Methods:The primary hippocampal neurons,N2a and HEK293T cells with tau overexpression were respectively starved and treated with vinblastine to study the effects of tau on the initiating steps of autophagy,which was analysed by Student’s two-tailed t-test.The rapamycin and concanamycin A were employed to inhibit the mammalian target of rapamycin kinase complex 1(mTORC1)activity and the vacuolar H+-ATPase(v-ATPase)activity,respectively,which were analysed by One-way ANOVA with post hoc tests.The Western blotting,co-immunoprecipitation and immunofuorescence staining were conducted to gain insight into the mechanisms underlying the tau effects of mTORC1 signaling alterations,as analysed by Student’s two-tailed t-test or One-way ANOVA with post hoc tests.The autophagosome formation was detected by immunofuorescence staining and transmission electron microscopy.The amino acids(AA)levels were detected by high performance liquid chromatography(HPLC).Results:We observed that overexpressing human full-length wild-type tau to mimic AD-like tau accumulation induced autophagy deficits.Further studies revealed that the increased tau could bind to the prion-related domain of T cell intracellular antigen 1(PRD-TIA1)and this association significantly increased the intercellular level of amino acids(Leucine,P=0.0038;Glutamic acid,P=0.0348;Alanine,P=0.0037;Glycine,P=0.0104),with concordant upregulation of mTORC1 activity[phosphorylated eukaryotic translation initiation factor 4E-binding protein 1(p-4EBP1),P<0.0001;phosphorylated 70 kD ribosomal protein S6 kinase 1(p-p70S6K1),P=0.0001,phosphorylated unc-51-like autophagyactivating kinase 1(p-ULK1),P=0.0015]and inhibition of autophagosome formation[microtubuleassociated protein light chain 3 II(LC3 II),P=0.0073;LC3 puncta,P<0.0001].As expected,this tau-induced deficit of autophagosome formation in turn aggravated tau accumulation.Importantly,we also found that blocking TIA1 and tau interaction by overexpressing PRD-TIA1,downregulating the endogenous TIA1 expression by shRNA,or downregulating tau protein level by a small proteolysis targeting chimera(PROTAC)could remarkably attenuate tau-induced autophagy impairment.Conclusions:Our findings reveal that AD-like tau accumulation inhibits autophagosome formation and induces autophagy deficits by activating the TIA1/amino acid/mTORC1 pathway,and thus this work reveals new insight into tau-associated neurodegeneration and provides evidence supporting the use of new therapeutic targets for AD treat-ment and that of related tauopathies.展开更多
Background Methamphetamine(METH)addiction causes a huge burden on society.The prefrontal cortex(PFC),associated with emotion and cognitive behaviours,is also involved in addiction neurocircuitry.Although bulk RNA sequ...Background Methamphetamine(METH)addiction causes a huge burden on society.The prefrontal cortex(PFC),associated with emotion and cognitive behaviours,is also involved in addiction neurocircuitry.Although bulk RNA sequencing has shown METH-induced gene alterations in the mouse PFC,the impact on different cell types remains unknown.Aims To clarify the effects of METH treatment on different cell types of the PFC and the potential pathways involved in METH-related disorders.Methods We performed single-nucleus RNA sequencing(snRNA-seq)to examine the transcriptomes of 20465 nuclei isolated from the PFC of chronic METH-treated and control mice.Main cell types and differentially expressed genes(DEGs)were identified and confirmed by RNA fluorescence in situ hybridization(FISH).Results Six main cell types were identified depending on the single-cell nucleus sequencing;of particular interest were the mature oligodendrocytes in the PFC.The DEGs of mature oligodendrocytes were enriched in the myelin sheath,adenosine triphosphate(ATP)metabolic process,mitochondrial function and components,and so on.The messenger RNA levels of Aldoc and Atp5l(FISH)and the protein level of the mitochondrial membrane pore subunit TOM40(immunofluorescence)decreased in the mature oligodendrocytes.Fast blue staining and transmission electron microscopy image indicated myelin damage,and the myelin thickness decreased in METH brains.Conclusions snRNA-seq reveals altered transcriptomes of different cell types in mouse PFC induced by chronic METH treatment,underscoring potential relationships with psychiatric disorders.展开更多

Tribological properties of impregnated graphite are greatly influenced by preparation technology and working conditions and it’s highly susceptible to corrosion environmental impacts,but the experimental research abo...Tribological properties of impregnated graphite are greatly influenced by preparation technology and working conditions and it’s highly susceptible to corrosion environmental impacts,but the experimental research about it are few.In this paper,three kinds of impregnated graphite samples are prepared with different degree of graphitization,the tribological properties of these samples in the dry friction environment and in a corrosive environment are analyzed and contrasted.The tribo-test results show that the friction coefficient of samples is reduced and the amount of wear of samples increase when the graphitization degree of samples increases in dry friction condition.While in a corrosive environment(samples are soaked N2O4),the friction coefficient and amount of wear are changed little if the graphitization degree of samples are low.If the degree of graphitization increase,the friction coefficient and amount of wear of samples increase too,the amount of wear is 2 to 3 times as the samples tested in the non-corrosive environment under pv value of 30MPa?m/s.The impregnated graphite,which friction coefficient is stable and graphitization degree is in mid level,such#2,is more appropriate to have a work in the corrosion conditions.In this paper,preparation and tribological properties especially in corrosive environment of the impregnated graphite is studied,the research conclusion can provide an experimental and theoretical basis for the selection and process improvement of graphite materials,and also provide some important design parameters for contact seal works in a corrosive environment.展开更多

Two types of mainly used tube bundles, the planar elastic tube bundle and the conical spiral tube bundle were comprehensively compared in the condition of the same shell side diameter. The natural mode characteristics, the effect of fluid-structure interaction, the stress distribution, the comprehensive heat transfer performance and the secondary fluid flow of the two elastic tube bundles were all concluded and compared. The results show that the natural frequency and the critical velocity of vibration buckling of the planar elastic tube bundle are larger than those of the conical spiral tube bundle, while the stress distribution and the comprehensive heat transfer performance of the conical spiral tube bundle are relatively better.展开更多

文摘The success of deep transfer learning in fault diagnosis is attributed to the collection of high-quality labeled data from the source domain.However,in engineering scenarios,achieving such high-quality label annotation is difficult and expensive.The incorrect label annotation produces two negative effects:1)the complex decision boundary of diagnosis models lowers the generalization performance on the target domain,and2)the distribution of target domain samples becomes misaligned with the false-labeled samples.To overcome these negative effects,this article proposes a solution called the label recovery and trajectory designable network(LRTDN).LRTDN consists of three parts.First,a residual network with dual classifiers is to learn features from cross-domain samples.Second,an annotation check module is constructed to generate a label anomaly indicator that could modify the abnormal labels of false-labeled samples in the source domain.With the training of relabeled samples,the complexity of diagnosis model is reduced via semi-supervised learning.Third,the adaptation trajectories are designed for sample distributions across domains.This ensures that the target domain samples are only adapted with the pure-labeled samples.The LRTDN is verified by two case studies,in which the diagnosis knowledge of bearings is transferred across different working conditions as well as different yet related machines.The results show that LRTDN offers a high diagnosis accuracy even in the presence of incorrect annotation.

文摘Artificial immune detection can be used to detect network intrusions in an adaptive approach and proper matching methods can improve the accuracy of immune detection methods.This paper proposes an artificial immune detection model for network intrusion data based on a quantitative matching method.The proposed model defines the detection process by using network data and decimal values to express features and artificial immune mechanisms are simulated to define immune elements.Then,to improve the accuracy of similarity calculation,a quantitative matching method is proposed.The model uses mathematical methods to train and evolve immune elements,increasing the diversity of immune recognition and allowing for the successful detection of unknown intrusions.The proposed model’s objective is to accurately identify known intrusions and expand the identification of unknown intrusions through signature detection and immune detection,overcoming the disadvantages of traditional methods.The experiment results show that the proposed model can detect intrusions effectively.It has a detection rate of more than 99.6%on average and a false alarm rate of 0.0264%.It outperforms existing immune intrusion detection methods in terms of comprehensive detection performance.
文摘Objective: Verify if the educational technology based on Just in Time Education is relevant to Permanent Health Education in Workers’ Health. Method: This study is exploratory and descriptive qualitative research. Were performed semi-structured interviews were carried out with the worker’s health team of a large company from the South of Brazil region. The collected data were carried out in 2021, between January and May, through the virtual environment, with eight participants from the Work Health Team of Eletrosul, located in Florianópolis, who agreed to participate in the research by signing the Free and Informed Consent Term, in accordance with the data saturation criterion. The research was approved by the Ethics Committee (n. 4079133) and the data were submitted to Bardin for thematic content analysis. Result: The results are presented from three categories: 1) Expansion and inclusion in Permanent Education needs;2) Tools adopted in educational processes;3) Just in Time Education: educational technology for workers’ health. Conclusion: The active methodology Just in Time Education helps the processes of Permanent Education in Health, which moved from a face-to-face model to e-learning and required the knowledge renewal in the technology area to maintain efficient communication. This methodology allows quick and accurate access to information from a reliable, adequate, and up-to-date source, causing a positive impact on the qualified and problem-solving work process, in line with the needs of the employer and the company. Therefore, Just in Time Education can provide standardization, organization, and informational and operational quality necessary in this context, in addition to serving as an indicator and data generator (management tool).

文摘In recent years,intelligent data-driven prognostic methods have been successfully developed,and good machinery health assessment performance has been achieved through explorations of data from multiple sensors.However,existing datafusion prognostic approaches generally rely on the data availability of all sensors,and are vulnerable to potential sensor malfunctions,which are likely to occur in real industries especially for machines in harsh operating environments.In this paper,a deep learning-based remaining useful life(RUL)prediction method is proposed to address the sensor malfunction problem.A global feature extraction scheme is adopted to fully exploit information of different sensors.Adversarial learning is further introduced to extract generalized sensor-invariant features.Through explorations of both global and shared features,promising and robust RUL prediction performance can be achieved by the proposed method in the testing scenarios with sensor malfunctions.The experimental results suggest the proposed approach is well suited for real industrial applications.

文摘In this work,we numerically study the structure of the turbulent/nonturbulent(T/NT)interface in a fully developed spatially evolving axisymmetric wake by means of direct numerical simulations.There is a continuous and contorted pure shear layer(PSL)adjacent to the outer edge of the T/NT interface.The local thickness of the PSLδ_(PSL)exhibits a wide range of scales(from the Kolmogorov scale to the Taylor microscale)and the conditional mean thickness<δ_(PSL)>I/η_(c)≈6 withη_(c)being the centerline Kolmogorov scale is the same as the viscous superlayer.In the viscous superlayer,the pure shear motions without rotation are overwhelmingly dominant.It is also demonstrated that the physics of the turbulent sublayer is closely related to the PSL with a large thickness.Another significant finding is that the time averaged area of the rotational regionA R,and the pure shear region<A_(S)>at different streamwise locations scale with the square of the wake-width b_(U)^(2).This study opens an avenue for a better understanding of the structures of the T/NT interface.

文摘Dear Editor,This letter is concerned with visual feedback disturbance rejection control for an amphibious bionic stingray subject to actuator saturation with internal and external disturbances. A visual feedback control method is designed for a dynamic nonlinear system on yaw angle of the amphibious bionic stingray via a finite-time extended-stateobserver,which is proved to achieve finite-time stability via a Lyapunov method. Finally, simulation results verify the effectiveness on the visual feedback control method.

文摘Prediction of students’engagement in aCollaborative Learning setting is essential to improve the quality of learning.Collaborative learning is a strategy of learning through groups or teams.When cooperative learning behavior occurs,each student in the group should participate in teaching activities.Researchers showed that students who are actively involved in a class gain more.Gaze behavior and facial expression are important nonverbal indicators to reveal engagement in collaborative learning environments.Previous studies require the wearing of sensor devices or eye tracker devices,which have cost barriers and technical interference for daily teaching practice.In this paper,student engagement is automatically analyzed based on computer vision.We tackle the problem of engagement in collaborative learning using a multi-modal deep neural network(MDNN).We combined facial expression and gaze direction as two individual components of MDNN to predict engagement levels in collaborative learning environments.Our multi-modal solution was evaluated in a real collaborative environment.The results show that the model can accurately predict students’performance in the collaborative learning environment.
文摘Background: Exercise-Induced Bronchospasm (EIB) is an inflammatory condition characterized by severe airway constriction following the mobilization of inflammatory cells and interleukin-6 (IL-6). When severe, EIB can require the use of pressurized salbutamol to treat athletes. This study investigated the nature of the systemic changes in inflammatory cells and post-exercise IL-6 concentrations after salbutamol treatment in EIB-susceptible distance runners. Materials and Methods: This was an experimental study that enrolled 12 long-distance runners. In Session A, the participants completed a treadmill exercise test, and those who had a maximum expiratory volume per second (FEV1) that was decreased by at least 10% compared to their base value were placed in the EIB-susceptible group (EIB+) (n = 6). Those whose FEV1 did not meet this criterion were placed in the nonresponsive (EIB?) group (n = 6). Before the Session B exercise, athletes in the BIE+ group inhaled two puffs of salbutamol (EIB+ Salb), while their EIB? counterparts received no treatment. Spirometry was performed before and after the exercise using a Spirobank G portable spirometer. Blood samples were taken before, immediately after and 2 hours after the stress test. Results: The mean post-exercise FEV1 values were not significantly different (p > 0.05) between the EIB+ Salb group and the EIB? group. The systemic changes in inflammatory cells and IL-6 concentrations in the EIB+ runners after salbutamol treatment were similar to those observed in their EIB? counterparts. Conclusion: Salbutamol pretreatment improved the systemic immune status of EIB-susceptible athletes.

文摘Alzheimer’s disease (AD) is the most prevalent cause of dementia worldwide. Because of the progressive neurodegeneration, individual cognitive and behavioral functions are impaired, affecting the quality of life of millions of people. Although the exact pathogenesis of AD has not been fully elucidated, amyloid plaques, neurofibrillary tangles (NFTs), and sustaining neuroinflammation dominate its characteristics. As one of the major tau kinases leading to hyperphosphorylation and aggregation of tau, glycogen synthase kinase-3β (GSK-3β) has been drawing great attention in various AD studies. Another research focus of AD in recent years is the inflammasome, a multiprotein complex acting as a regulator in immunological reactions to exogenous and endogenous danger signals, of which the Nod-like receptor (NLR) family, pyrin domain-containing 3 (NLRP3) inflammasome has been studied mostly in AD and proven to play a significant role in AD development by its activation and downstream effects such as caspase-1 maturation and interleukin (IL)-1β release. Studies have shown that the NLRP3 inflammasome is activated in a GSK-3β-dependent way and that inhibition of the NLRP3 inflammasome downregulates GSK-3β, suggesting that these two important proteins are closely related. This article reviews the respective roles of GSK-3β and the NLRP3 inflammasome in AD as well as their relationship and interaction.
文摘Context: Fine particles (PM2.5 and PM10) can accumulate in classrooms and in schoolyards located near urban roads. PM2.5 and PM10 can initiate, develop and exacerbate exercise-induced bronchospasm (EIB). This study aimed to assess the concentration levels of PM2.5 and PM10 in schools and to determine the rate of sensitivity to EIB among schoolchildren. Methods: A total of 128 students (67 girls and 61 boys) with an average age of 11 participated in this study. An ISAAC II questionnaire on respiratory symptoms was administered. PM2.5 and PM10 were measured. A 6-minute stress test was performed. Spirometry was performed. The ratio (I/O) of the concentrations of PM2.5 and PM10 recorded inside (I) the classrooms to those obtained outside (O) the classrooms was equal to 1 (I/O = 1). Results: The concentrations of PM2.5 and PM10 recorded inside the classrooms and those obtained outside the classrooms were higher than the values recommended by the WHO. 29 students out of 128 were diagnosed as sensitive to EIB [EIB (+)], i.e., a rate of sensitivity to EIB of 22.66%. A drop in postexercise PEF of 17.396% was observed among EIB (+) students. Conclusion: Schoolchildren in schools located near highways are exposed to high levels of PM2.5 and PM10 concentrations. Exposure to PM2.5 and PM10 played an important role in the initiation and exacerbation of exercise-induced bronchospasm in schoolchildren from schools located near highways. Effective programs for improving air quality in schools must be put in place to reduce the effects of particulate pollution on the respiratory health of school children.

文摘Objective Post-stroke cognitive impairment(PSCI)develops in approximately one-third of stroke survivors and is associated with ingravescence.Nonetheless,the biochemical mechanisms underlying PSCI remain unclear.The study aimed to establish an ischemic mouse model by means of transient unilateral middle cerebral artery occlusions(MCAOs)and to explore the biochemical mechanisms of p25/cyclin-dependent kinase 5(CDK5)-mediated tau hyperphosphorylation on the PSCI behavior.Methods Cognitive behavior was investigated,followed by the detection of tau hyperphosphorylation,mobilization,activation of kinases and/or inhibition of phosphatases in the lateral and contralateral cerebrum of mice following ischemia in MACO mice.Finally,we treated HEK293/tau cells with oxygen-glucose deprivation(OGD)and a CDK5 inhibitor(Roscovitine)or a GSK3βinhibitor(LiCl)to the roles of CDK5 and GSK3βin mediating ischemia-reperfusion-induced tau phosphorylation.Results Ischemia induced cognitive impairments within 2 months,as well as causing tau hyperphosphorylation and its localization to neuronal somata in both ipsilateral and contralateral cerebra.Furthermore,p25 that promotes CDK5 hyperactivation had significantly higher expression in the mice with MCAO than in the shamoperation(control)group,while the expression levels of protein phosphatase 2(PP2A)and the phosphorylation level at Tyr307 were comparable between the two groups.In addition,the CDK5 inhibitor rescued tau from hyperphosphorylation induced by OGD.Conclusion These findings demonstrate that upregulation of CDK5 mediates tau hyperphosphorylation and localization in both ipsilateral and contralateral cerebra,contributing to the pathogenesis of PSCI.

文摘Objective Schizophrenia(SZ)is associated with cognitive impairment,and it is known that the activity of cAMP response element binding protein(CREB)decreases in the brain of SZ patients.The previous study conducted by the investigators revealed that the upregulation of CREB improves the MK801-related SZ cognitive deficit.The present study further investigates the mechanism on how CREB deficiency is associated with SZ-related cognitive impairment.Methods MK-801 was used to induce SZ in rats.Western blotting and immunofluorescence were performed to investigate CREB and the CREB-related pathway implicated in MK801 rats.The long-term potentiation and behavioral tests were performed to assess the synaptic plasticity and cognitive impairment,respectively.Results The phosphorylation of CREB at Ser133 decreased in the hippocampus of SZ rats.Interestingly,among the upstream kinases of CREB,merely ERK1/2 was downregulated,while CaMKII and PKA remained unchanged in the brain of MK801-related SZ rats.The inhibition of ERK1/2 by PD98059 reduced the phosphorylation of CREB-Ser133,and induced synaptic dysfunction in primary hippocampal neurons.Conversely,the activation of CREB attenuated the ERK1/2 inhibitor-induced synaptic and cognitive impairment.Conclusion These present findings partially suggest that the deficiency of the ERK1/2-CREB pathway is involved in MK801-related SZ cognitive impairment.The activation of the ERK1/2-CREB pathway may be therapeutically useful for treating SZ cognitive deficits.
文摘Context: Automobile pollution is becoming a potential threat to the cardiorespiratory health of the urban population of sub-Saharan Africa. The present study aims to evaluate the level of concentrations of fine particles (PM2.5 and PM10) near road traffic and the effects of exposure to automobile pollutants on pulmonary function and arteriolar blood oxygen saturation among sellers around road traffic. Materials and Methods: The study recruited 48 healthy people carrying out a sales activity near road traffic. PM2.5 and PM10 measurements were taken from 6 a.m. to 6 p.m. using a Temtop Airing-1000 portable particle detector. Spirometric measurements were taken in the morning and in the evening from a portable Spirobank G spirometer. Oxygen saturation measurements were also taken in the morning and evening using a Pulse oxymeter CMS50D pulse oxymeter. Results: Mean values of forced vital capacity (FVC), forced expiratory volume in one second (FEV1), peak expiratory flow (PEF), and forced expiratory flow between 25% and 75% of forced vital capacity (FEF25 - 75) recorded in the evening were significantly lower than those obtained in the morning in the subjects of the total group and in the men. Only the mean values of PEF and FEF25 - 75 obtained in the evening were significantly lower than those recorded in the morning in women. Minimum PM2.5 values recorded near road traffic were negatively correlated with evening PEF in men, while maximum PM10 values were negatively correlated with evening PEF in women. The blood oxygen saturation recorded in the evening was significantly lower than that obtained in the morning. Conclusion: Short-term exposure to automobile pollutants has adverse effects on lung function and oxygen saturation in people carrying out a sales activity near road traffic.

文摘Background:Autophagy dysfunction plays a crucial role in tau accumulation and neurodegeneration in Alzheimer’s disease(AD).This study aimed to investigate whether and how the accumulating tau may in turn affect autophagy.Methods:The primary hippocampal neurons,N2a and HEK293T cells with tau overexpression were respectively starved and treated with vinblastine to study the effects of tau on the initiating steps of autophagy,which was analysed by Student’s two-tailed t-test.The rapamycin and concanamycin A were employed to inhibit the mammalian target of rapamycin kinase complex 1(mTORC1)activity and the vacuolar H+-ATPase(v-ATPase)activity,respectively,which were analysed by One-way ANOVA with post hoc tests.The Western blotting,co-immunoprecipitation and immunofuorescence staining were conducted to gain insight into the mechanisms underlying the tau effects of mTORC1 signaling alterations,as analysed by Student’s two-tailed t-test or One-way ANOVA with post hoc tests.The autophagosome formation was detected by immunofuorescence staining and transmission electron microscopy.The amino acids(AA)levels were detected by high performance liquid chromatography(HPLC).Results:We observed that overexpressing human full-length wild-type tau to mimic AD-like tau accumulation induced autophagy deficits.Further studies revealed that the increased tau could bind to the prion-related domain of T cell intracellular antigen 1(PRD-TIA1)and this association significantly increased the intercellular level of amino acids(Leucine,P=0.0038;Glutamic acid,P=0.0348;Alanine,P=0.0037;Glycine,P=0.0104),with concordant upregulation of mTORC1 activity[phosphorylated eukaryotic translation initiation factor 4E-binding protein 1(p-4EBP1),P<0.0001;phosphorylated 70 kD ribosomal protein S6 kinase 1(p-p70S6K1),P=0.0001,phosphorylated unc-51-like autophagyactivating kinase 1(p-ULK1),P=0.0015]and inhibition of autophagosome formation[microtubuleassociated protein light chain 3 II(LC3 II),P=0.0073;LC3 puncta,P<0.0001].As expected,this tau-induced deficit of autophagosome formation in turn aggravated tau accumulation.Importantly,we also found that blocking TIA1 and tau interaction by overexpressing PRD-TIA1,downregulating the endogenous TIA1 expression by shRNA,or downregulating tau protein level by a small proteolysis targeting chimera(PROTAC)could remarkably attenuate tau-induced autophagy impairment.Conclusions:Our findings reveal that AD-like tau accumulation inhibits autophagosome formation and induces autophagy deficits by activating the TIA1/amino acid/mTORC1 pathway,and thus this work reveals new insight into tau-associated neurodegeneration and provides evidence supporting the use of new therapeutic targets for AD treat-ment and that of related tauopathies.

文摘Background Methamphetamine(METH)addiction causes a huge burden on society.The prefrontal cortex(PFC),associated with emotion and cognitive behaviours,is also involved in addiction neurocircuitry.Although bulk RNA sequencing has shown METH-induced gene alterations in the mouse PFC,the impact on different cell types remains unknown.Aims To clarify the effects of METH treatment on different cell types of the PFC and the potential pathways involved in METH-related disorders.Methods We performed single-nucleus RNA sequencing(snRNA-seq)to examine the transcriptomes of 20465 nuclei isolated from the PFC of chronic METH-treated and control mice.Main cell types and differentially expressed genes(DEGs)were identified and confirmed by RNA fluorescence in situ hybridization(FISH).Results Six main cell types were identified depending on the single-cell nucleus sequencing;of particular interest were the mature oligodendrocytes in the PFC.The DEGs of mature oligodendrocytes were enriched in the myelin sheath,adenosine triphosphate(ATP)metabolic process,mitochondrial function and components,and so on.The messenger RNA levels of Aldoc and Atp5l(FISH)and the protein level of the mitochondrial membrane pore subunit TOM40(immunofluorescence)decreased in the mature oligodendrocytes.Fast blue staining and transmission electron microscopy image indicated myelin damage,and the myelin thickness decreased in METH brains.Conclusions snRNA-seq reveals altered transcriptomes of different cell types in mouse PFC induced by chronic METH treatment,underscoring potential relationships with psychiatric disorders.

文摘Tribological properties of impregnated graphite are greatly influenced by preparation technology and working conditions and it’s highly susceptible to corrosion environmental impacts,but the experimental research about it are few.In this paper,three kinds of impregnated graphite samples are prepared with different degree of graphitization,the tribological properties of these samples in the dry friction environment and in a corrosive environment are analyzed and contrasted.The tribo-test results show that the friction coefficient of samples is reduced and the amount of wear of samples increase when the graphitization degree of samples increases in dry friction condition.While in a corrosive environment(samples are soaked N2O4),the friction coefficient and amount of wear are changed little if the graphitization degree of samples are low.If the degree of graphitization increase,the friction coefficient and amount of wear of samples increase too,the amount of wear is 2 to 3 times as the samples tested in the non-corrosive environment under pv value of 30MPa?m/s.The impregnated graphite,which friction coefficient is stable and graphitization degree is in mid level,such#2,is more appropriate to have a work in the corrosion conditions.In this paper,preparation and tribological properties especially in corrosive environment of the impregnated graphite is studied,the research conclusion can provide an experimental and theoretical basis for the selection and process improvement of graphite materials,and also provide some important design parameters for contact seal works in a corrosive environment.

文摘Elastic heat transfer tube bundles are widely used in the field of flow-induced vibration heat transfer enhancement. Two types of mainly used tube bundles, the planar elastic tube bundle and the conical spiral tube bundle were comprehensively compared in the condition of the same shell side diameter. The natural mode characteristics, the effect of fluid-structure interaction, the stress distribution, the comprehensive heat transfer performance and the secondary fluid flow of the two elastic tube bundles were all concluded and compared. The results show that the natural frequency and the critical velocity of vibration buckling of the planar elastic tube bundle are larger than those of the conical spiral tube bundle, while the stress distribution and the comprehensive heat transfer performance of the conical spiral tube bundle are relatively better.
