Nesting behaviour of sea turtles remains a subject to study, due to their enigmatic pattern of seasonal breeding activities. In the present study, characteristics of the breeding turtle, Olive Ridley (Lepidochelys olivacea) at Ramnagar along N-E coast of North Andaman Islands were investigated, for the nesting periods 2016-2017. Olive Ridley is the dominant sea turtles, with more than 300 individuals nesting every season with at least a 75% hatching success rate. It is one of the conducive, highly protective and undisturbed nesting sites for Olive Ridley on North Andaman coast. In the present study an initiative is made, to identify nature’s cues and the biological characteristics of Olive Ridley, which leads it to nest at Ramnagar beach, with a high hatchling success rate. 展开更多

The Jiangchuan Biota from the Jiucheng Member(Mb.)of the Dengying Formation(Fm.),discovered in Jiangchuan,eastern Yunnan,China,is marked by copious macrofossils at the apex of the Ediacaran strata.This fauna features benthic algae with varied holdfasts and other fossils of indeterminate taxonomic affinity and is compositionally unique compared to the Shibantan and Gaojiashan biotas of the Dengying Fm.and the Miaohe and Wenghui biotas of the Doushantuo Fm.,elsewhere in China.One novel benthic saccular macroalgal fossil,named here Houjiashania yuxiensis gen.and sp.nov.,from the Jiangchuan Biota is based on fossils that are sausage-shaped,elongate,tubular,ranging from 0.3 to 4 cm in length,and up to 0.8 cm in diameter.One terminus is blunt and rounded to an obtuse angle,the other is bent with a spread-out surface resembling a holdfast,suggesting a three-dimensional thallus.Thin,stipe-shaped outgrowths,likely vestiges of sessile saccular life forms,are prevalent in macroalgal fossils of analogous size and shape,as well as present brown algae Scytosiphonaceae,such as Colpomenia and Dactylosiphon.The new findings augment the diversity of benthic algae,such as those known from the Early Neoproterozoic Longfengshan Biota in North China.The benthic algal macrofossils in the Jiucheng Mb.add to knowledge of Late Ediacaran metaphyte diversification and offer more clues about the evolutionary positioning of primitive macroalgae.The co-occurrence of numerous planktonic and benthic multicellular algae and planktonic microbes might have facilitated ecologically the more extensive later Cambrian explosion evidenced by the Chengjiang Biota in Yunnan. 展开更多

Mammalian T-cell responses require synergism between the first signal and co-stimulatory signal.However,whether and how dual signaling regulates the T-cell response in early vertebrates remains unknown.In the present study,we discovered that the Nile tilapia(Oreochromis niloticus)encodes key components of the LAT signalosome,namely,LAT,ITK,GRB2,VAV1,SLP-76,GADS,and PLC-γ1.These components are evolutionarily conserved,and CD3εmAb-induced T-cell activation markedly increased their expression.Additionally,at least ITK,GRB2,and VAV1 were found to interact with LAT for signalosome formation.Downstream of the first signal,the NF-κB,MAPK/ERK,and PI3K-AKT pathways were activated upon CD3εmAb stimulation.Furthermore,treatment of lymphocytes with CD28 mAbs triggered the AKT-mTORC1 pathway downstream of the co-stimulatory signal.Combined CD3εand CD28 mAb stimulation enhanced ERK1/2 and S6 phosphorylation and elevated NFAT1,c-Fos,IL-2,CD122,and CD44 expression,thereby signifying T-cell activation.Moreover,rather than relying on the first or co-stimulatory signal alone,both signals were required for T-cell proliferation.Full T-cell activation was accompanied by marked apoptosis and cytotoxic responses.These findings suggest that tilapia relies on dual signaling to maintain an optimal T-cell response,providing a novel perspective for understanding the evolution of the adaptive immune system. 展开更多

Proteomic response of Phaeocystis globosa to nitrogen limitation

Phaeocystis globosa is an important unicellular eukaryotic alga that can also form colonies.P.globosa can cause massive harmful algal blooms and plays an important role in the global carbon or sulfur cycling.Thus far,the ecophysiology of P.globosa has been investigated by numerous studies.However,the proteomic response of P.globosa to nitrogen depletion remains largely unknown.We compared four protein preparation methods of P.globosa for two-dimensional electrophoresis(2-DE)(Urea/Triton X-100 with trichloroacetic acid(TCA)/acetone precipitation;TCA/acetone precipitation;Radio Immuno Precipitation Assay(RIPA)with TCA/acetone precipitation;and Tris buffer).Results show that the combination of RIPA with TCA/acetone precipitation had a clear gel background and showed the best protein spot separation effect,based on which the proteomic response to nitrogen depletion was studied using 2-DE.In addition,we identified six differentially expressed proteins whose relative abundance increased or decreased more than 1.5-fold(P<0.05).Most proteins could not be identified,which might be attributed to the lack of genomic sequences of P.globosa.Under nitrogen limitation,replication protein-like,RNA ligase,and sn-glycerol-3-phosphate dehydrogenase were reduced,which may decrease the DNA replication level and ATP production in P.globosa cells.The increase of endonucleaseⅢand transcriptional regulator enzyme may affect the metabolic and antioxidant function of P.globosa cells and induce cell apoptosis.These findings provide a basis for further proteomic study of P.globosa and the optimization of protein preparation methods of marine microalgae. 展开更多

The deep-sea clam Calyptogena marissinica is widely distributed in the Haima cold seep ecosystem on the northwes-tern slope of the South China Sea with low pH values,low temperature and high pressure.Limited information is available on the biomineralization of this species.In this research,we generated a comprehensive transcript dataset of C.marissinica’s mantle tissue,and a total of 19821 unigenes were assembled.Fourteen shell matrix proteins(SMP)-related genes were identified.The qPCR results showed that four out of six prismatic matrix genes(MSP2,MSP5,prisilkin-39,and shematrin),four out of the six nacreous matrix genes(perlucin,pif,pif97,and papilin),and two extrapallial fluid proteins(SPARC and calmodulin)were significantly expressed in the mantle.Both the nacreous and the prismatic layers are chrysanthemum-shaped,which are stacked on the top of each other to form a laminated nacreous structure.The alignment and phylogenetic analysis of MSP-5,Prisilkin-39,Perlucin,and Pif homologues showed that some amino acids of C.marissinica that differed from those detected in other molluscs may cause the different shape of the nacreous and prismatic layers,but do not lead to a change in the species’evolutionary status.These results indicated the conservation of the functions of SMP-related genes in C.marissinica,and the specific shape of the prismatic and nacreous layers of this deep-sea mollusc,which will contribute to the research on the molecular regulation mechanisms of biomineralization in C.marissinica and provide a new perspective to investigate biomineralization in deep-sea clams in general. 展开更多

Long non-coding RNAs(lncRNAs)function as key modulators in mammalian immunity,particularly due to their involvement in lncRNA-mediated competitive endogenous RNA(ceRNA)crosstalk.Despite their recognized significance in mammals,research on lncRNAs in lower vertebrates remains limited.In the present study,we characterized the first immune-related lncRNA(pol-lnc78)in the teleost Japanese flounder(Paralichthys olivaceus).Results indicated that pol-lnc78 acted as a ceRNA for pol-miR-n199-3p to target the sterile alpha and armadillo motif-containing protein(SARM),the fifth discovered member of the Toll/interleukin 1(IL-1)receptor(TIR)adaptor family.This ceRNA network regulated the antibacterial responses of flounder via the Toll-like receptor(TLR)signaling pathway.Specifically,SARM acted as a negative regulator and exacerbated bacterial infection by inhibiting the expression of inflammatory cytokines IL-1βand tumor necrosis factor-α(TNF-α).Pol-miR-n199-3p reduced SARM expression by specifically interacting with the 3’untranslated region(UTR),thereby promoting SARM-dependent inflammatory cytokine expression and protecting the host against bacterial dissemination.Furthermore,pol-lnc78 sponged pol-miR-n199-3p to ameliorate the inhibition of SARM expression.During infection,the negative regulators pol-lnc78 and SARM were significantly down-regulated,while pol-miR-n199-3p was significantly up-regulated,thus favoring host antibacterial defense.These findings provide novel insights into the mechanisms underlying fish immunity and open new horizons to better understand ceRNA crosstalk in lower vertebrates. 展开更多

In this study,a calcium-phosphate coating was formed on a Mg-Mn-Ce alloy by the plasma electrolytic oxidation(PEO).The antibiotic vancomycin,widely used in the treatment of infections caused by Staphylococcus aureus(S.aureus),was impregnated into the coating.Samples with vancomycin showed high bactericidal activity against S.aureus.The mechanical and electrochemical properties of the formed coatings were studied,as well as in vitro cytotoxicity tests and in vivo tests on mature male rats were performed.According to SEM,EDS,XRD and XPS data,coatings had a developed morphology and contained hydroxyapatite,which indicates high biocompatibility.The analysis of roughness of coatings without and with vancomycin did not reveal any differences,confirming the high roughness of the samples.During electrochemical tests,an increase in corrosion resistance by more than two times after the application of PEO coatings was revealed.According to the results of an in vivo study,after 28 days of the implantation of samples with calcium phosphate PEO coating and vancomycin,no signs of inflammation were observed,while an inflammatory reaction developed in the area of implantation of bare alloy,followed by encapsulation.Antibiotic release tests from the coatings show a sharp decrease in the concentration of the released antibiotic on day 7 and then a gradual decrease until day 28.Throughout the experiment,no significant deviations in the condition and behavior of the animals were observed;clinical tests did not reveal a systemic toxic reaction. 展开更多

One specimen belonging to the family Comatellinae was collected from the Zhenbei Seamount(332.5–478.2 m)in the South China Sea in July 2022.Based on the morphological characters,the specimen was identified as Palaeocomatella hiwia McKnight,1977.It is first recorded from China Sea and redescribed in detail.This specimen differs from the original description from New Zealand for never showing syzygy at br4+5 or br5+6 on interior and br1+2 on exterior arms.However,it is much conform to the redescription to specimens from Indonesia,with only differences in position of the second syzygy and distalmost pinnule comb.Specimen is deposited in the Institute of Oceanology,Chinese Academy of Sciences.Phylogenetic analyses based on the mitochondrial c oxidase subunit I(COI)and 16S rRNA genes indicated that P.hiwia was nested within the tribe Phanogeniini and clustered with Aphanocomaster pulcher.Furthermore,P.hiwia showed same morphological features in terms of mouth placement,comb location,and number of comb teeth rows as other genera of Phanogeniini.Therefore,we suggest that the genus Palaeocomatella should be put in the tribe Phanogeniini. 展开更多

As ectotherms, fish are highly sensitive to temperature fluctuations, which can profoundly impact their reproductive cycles. In this study, we investigated the fertility and histological characteristics of zebrafish(Danio rerio) ovaries exposed to a temperature gradient ranging from the thermopreferendum temperature of the species,27℃, to lower temperatures of 22℃, 20℃, and 13℃ over a period of two weeks. Comparative metabolomic(six biological replicates for each temperature) and transcriptomic(four biological replicates for each temperature) analyses were conducted under the four temperature conditions. Results indicated that lower temperatures inhibited oocyte development and differential metabolites were involved in steroid hormone production,antioxidant function, and lipid and protein catabolism.Disrupted reproductive hormones, increased proteolysis,and lipid degradation significantly impeded oocyte development and egg maturation. Notably, a significant increase in bile acid content was noted in the ovaries of the cold-treated fish, indicating that bile acids play a critical role in ovarian failure. Overall, these findings provide valuable insights into the mechanisms governing the reproductive response of fish to cold stress. 展开更多

Marine sediments collected from the Zhujiang(Pearl) River Estuary(ZRE) and South China Sea(SCS) were utilized to study the occurrence and spatial distribution of tetrabromobisphenol A(TBBPA) and hexabromocyclododecane(HBCDD).The levels of TBBPA and HBCDD in sediments ranged from not detected(nd) to 6.14 ng/g dry weight(dw) and nd to 0.42 ng/g dw.TBBPA concentrations in marine sediments were substantially higher than HBCDD.The concentrations of TBBPA and HBCDD in the ZRE sediments were significantly greater than those in the SCS.α-HBCDD(48.7%) and γ-HBCDD(46.2%) were the two main diastereoisomers of HBCDD in sediments from the ZRE,with minor contribution of β-HBCDD(5.1%).HBCDD were only found in one sample from the northern SCS.The enantiomeric fraction of α-HBCDD in sediments from the ZRE was obviously greater than 0.5,indicating an accumulation of(+)-α-HBCDD.The enantiomers of HBCDD were not measured in sediments from the SCS.This work highlighted the environmental behaviors of TBBPA and HBCDD in marine sediments. 展开更多

Avirulence effectors(Avrs),encoded by plant pathogens,can be recognized by plants harboring the corresponding resistance proteins,thereby initiating effector-triggered immunity(ETI).In susceptible plants,however,Avrs can function as effectors,facilitating infection via effector-triggered susceptibility(ETS).Mechanisms of Avr-mediated ETS remain largely unexplored.Here we report that the Magnaporthe oryzae effector Avr-PikD enters rice cells via the canonical cytoplasmic secretion pathway and suppresses rice basal defense.Avr-PikD interacts with an LSD1-like transcriptional activator AKIP30 of rice,and AKIP30 is also a positive regulator of rice immunity,whereas Avr-PikD impedes its nuclear localization and suppresses its transcriptional activity.In summary,M.oryzae delivers Avr-PikD into rice cells to facilitate ETS by inhibiting AKIP30-mediated transcriptional regulation of immune response against M.oryzae. 展开更多

Coilia nasus,a migratory fish species found in the middle and lower reaches of the Yangtze River and along offshore areas of China,possesses considerable aquacultural and economic potential.However,the species faces challenges due to significant variation in the gonadal development rate among females,resulting in inconsistent ovarian maturation times at the population level,an extended reproductive period,and limitations on fish growth rate due to ovarian prematurity.In the present study,we combined genome-wide association study(GWAS)and comparative transcriptome analysis to investigate the potential single nucleotide polymorphisms(SNPs)and candidate genes associated with population-asynchronous ovarian development in C.nasus.Genotyping of the female population based on whole-genome resequencing yielded 2120695 high-quality SNPs,39 of which were suggestively associated with ovarian development.Of note,a significant SNP peak on LG21 containing 30 suggestively associated SNPs was identified,with cpne5a determined as the causal gene of the peak.Therefore,single-marker and haplotype association analyses were performed on cpne5a,revealing four genetic markers(P<0.05)and seven haplotypes(r2>0.9)significantly associated with the phenotype.Comparative transcriptome analysis of precociously and normally maturing individuals screened out 29 and 426 overlapping differentially expressed genes in the brain and ovary,respectively,between individuals of different body sizes.Integrating the GWAS and transcriptome analysis results,this study identified genes and pathways related to hypothalamic-pituitary-gonadal axis hormone secretion,extracellular matrix,angiogenesis,and gap junctions involved in population-asynchronous ovarian development.The insights gained from this study provide a basis for a deeper understanding of the molecular mechanisms underlying ovarian development in fish and may facilitate the genetic breeding of C.nasus strains exhibiting population-synchronous ovarian development in the future. 展开更多

During the production of nitrile rubber, significant amounts of nitrogen in the form of ammonium are generated in the wastewater. The discharge of this high-nitrogen wastewater can lead to serious environmental issues, including eutrophication, disruption of aquatic ecosystems, and groundwater contamination. To mitigate these impacts, this research explored the bioremediation capabilities of the macroalgae Ulva lactuca (Chlorophyta) for removing nitrogen from nitrile rubber production wastewater. The study employed single-phase and Michaelis-Menten decay models based on ammonium consumption, using various dilutions of wastewater to identify the optimal concentration for treatment. The physiological state of the macroalgae was monitored by measuring the photosynthetic capacity and specific growth rate during the experiments. In the presence of U. lactuca, ammonium concentrations decreased in all treatment groups, confirming that the ammonium kinetics conformed to both applied models. Our results show that U. lactuca effectively reduces ammonium concentrations, with an approximate removal rate of 0.020 µM·g−1·min−1 across different wastewater concentrations (70%, 80%, 90%, and 100%). Notably, the treatments with 70%, 80%, and 90% wastewater strength achieved about 67% reduction in ammonium, demonstrating the alga’s capacity to treat high-nitrogen wastewater. The photosynthetic performance of U. lactuca initially declined in control conditions but stabilized across all treatments, highlighting its adaptability. The kinetic analysis using the Michaelis-Menten model indicated a Vmax of 1342 μM·g−1·DMh−1, suggesting a robust capacity for ammonium uptake when fully saturated. Our study underscores the potential of Ulva lactuca as a cost-effective and efficient agent for wastewater bioremediation, particularly in settings with high nitrogen loads. 展开更多

Stable isotope analysis is a widely used method for gathering ecological insights into the diet and feeding habitats of various species. While captive studies often limit lethal sampling and differ from wild conditions, they offer valuable insights into inherent isotopic variations among individuals, which are often assumed to reflect differences between natural populations. In the Sea Turtle Conservation Program, loggerhead turtle hatchlings from different nests were fed. Necropsies were conducted on turtles that died during this period, obtaining bone fragments for analysis. We evaluated the isotopic variation of carbon (δ<sup>13</sup>C) and nitrogen (δ<sup>15</sup>N) in bone tissue across six turtle nests (n = 66 samples) and assessed differences in Straight Carapace Length (SCL, n = 71 samples). Using SIBER and nicheROVER in R, we calculated niche width and overlap, while the simmr package determined primary prey assimilation. Despite feeding the hatchlings the same prey, we observed variations in nitrogen isotope assimilation between nests. Nests 4 and 6 had a niche width >1.8‰, indicating consistent consumption frequencies across all prey and >70% niche overlap with other nests. In contrast, nests 1 and 2 showed a narrower niche width (Mugil sp. constituted the primary diet component (>40%) across all groups. This study demonstrates how factors like competition or prey preference can influence the assimilation of diet, even when the source remains constant (inherent variation). 展开更多

A chemical investigation of the ethyl acetate extract of the fermentation broth of Alternaria tenuissima EN- 192, an endophytic fungus obtained from the stems of the marine mangrove plant Rhizophora stylosa, resulted in the isolation of nine known secondary metabolites, including four indole-diterpenoids: penijanthine A (1), paspaline (2), paspalinine (3), and penitrem A (4); three tricycloalternarene derivatives: tricycloalternarene 3a (5), tricycloalternarene lb (6), and tricycloalternarene 2b (7); and two alternariol congeners: djalonensone (8) and alternariol (9). The chemical structures of these metabolites were characterized through a combination of detailed spectroscopic analyses and their comparison with reports from the literature. The inhibitory activities of each isolated compound against four bacteria were evaluated and compounds 5 and 8 displayed moderate activity against the aquaculture pathogenic bacterium Vibrio anguillarum, with inhibition zone diameters of 8 and 9 mm, respectively, at 100 gg/disk. To the best of our knowledge, this is the first report on the secondary metabolites of mangrove-derived Alternaria tenuissima and also the first report of the isolation ofindole-diterpenoids from fungal genus Alternaria. 展开更多

The toxic mechanism of herbicide butachlor to induce extremely high lethality in marine flatfish flounder, Paralichthys Olivaceus, was analyzed by histopathological examination, antioxidant enzymes activities and ATP content assay. Histopathological examination of gill, liver and kidney of exposed fishes showed that gill was a target organ of butachlor. The butachlor seriously impaired the respiration of gills by a series of lesions such as edema, lifting and detachment of lamellar epithelium, breakdown of pillar cells, and blood congestion. The dysfunction of gill respiration caused suffocation to the exposed flounder with extremely high acute lethality. Antioxidant enzyme activity assay of the in vitro cultured flounder gill (FG) ceils exposed to butachlor indicated that butachlor markedly inhibited the antioxidant enzyme activities of Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX). Furthermore, along with the decline of antioxidant enzyme activities, ATP content in the exposed FG cells decreased, too. This infers that the oxidative stress induced by butachlor can inhibit the production of cellular ATP. Similar decrease of ATP content was also observed in the exposed flounder gill tissues. Taken together, as in FG cells, butachlor possibly induced a short supply of ATP in pillar cells by inhibiting the antioxidant enzyme activities and then affecting the contractibility of the pillar cells, which in turn resulted in the blood congestion and suffocation of exposed flounder. 展开更多

Twenty-four compounds including eight steroids (1-8), nine triterpenoids (9-16, 24), three flavonoids (20-22), and four benzenecarboxylic derivatives (17-19, 23) were isolated and identified from stems and twigs of medicinal mangrove plant Sonneratia caseolaris. The structures of the isolated compounds were determined by extensive analysis of their spectroscopic data. Among these rnetabolites, compounds 1, 4-20 and 22-24 were isolated and identified for the first time from S. caseolaris. In the in vitro cytotoxic assay against SMMC-7721 human hepatoma cells, compound 21 (3',4',5,7-tetrahydroxyflavone) exhibited significant activity with IC50 2.8 μg/mL, while oleanolic acid (14), 3,3'-di-O-methyl ether ellagic acid (18), and 3,3',4-O-tri-O-methyl ether ellagic acid (19) showed weak activity. The soft coral S. po... Soft corals are an important group of organisms that live in reef ecosystems. In this study, the chemical defense of soft coral Sinularia polydactyla against biofilm-forming bacteria was assessed. The soft coral S. polydactyla was collected from the Obhur creek of the Red Sea and the bioactive compounds were extracted under different conditions using methanol and hexane as solvents. Results revealed that the bioactive compounds produced by the soft coral S. polydactyla were active against the growth, hydrophobicity and extracellular polymeric substances production of the biofilm-forming bacteria. However, the damage or injury in soft coral tissue caused a decrease in the activity against biofilm-forming bacteria. GC-MS analysis revealed that sesquiterpenes were abundant in normal coral sample extract while cembranoids were abundant in damaged coral sample extracts. In general, the results indicated that the soft corals either under stress or with damage may not have a strong chemical defense against the colonizing marine organisms and competitors. 展开更多
