Oxidative damage of chronic PM2.5 exposure to rat lung tissue

Objective:To investigate the oxidative damage caused by chronic exposure to PM2.5 in rat lung tissues according to the PM2.5 concentration in the atmospheric environment.Methods:Forty healthy SD rats were randomly divided into 4 groups,the control group and the 5 mg/kg,10 mg/kg,20 mg/kg PM2.5 exposure groups.The rats in each group were administered by intratracheal instillation with 1ml/kg,2 times per week.After 8 weeks of exposure,the changes of lung histopathology,superoxide dismutase(SOD),malondialdehyde(MDA),catalase(CAT)and reduced glutathione/oxidized glutathione(GSH/GSSG)were detected.Results:Compared with the control group,the histopathological changes in the lung tissue of the three dose groups exposed to PM2.5 were significant.Masson staining showed obvious collagen fibrosis.The activities of SOD and CAT in the lung tissue of the exposed group were decreased,and the contents of MDA were increased with statistically significant(P<0.05),showing that the difference was the most obvious in the middle-dose group.The GSH/GSSG ratio in the lung tissue decreased,indicating that the degree of oxidative damage was aggravated.Conclusion:Chronic exposure to PM2.5 could induce oxidative stress in the lung tissue of rats and caused different degrees of oxidative damage. 展开更多

Objective:To study the effect of Baihe Dihuang Decoction in improving anxiety and its mechanism of action.Methods: Male mice were randomly divided into 5 groups: blank control group, Baihe Dihuang Decoction high, medium, low-dose group and diazepam group. After continuous intragastric administration for 14 d, the behavioral test of the mouse elevated plus maze experiment and opening test was performed. The brain tissue GABA and Glu content was measured by immunosorbent assay (ELISA).Results: Compared with the blank control group, the middle and high doses of Baihe Dihuang Decoction could increase the time and frequency of movement of the mice in the open cross labyrinth in the open arm and increase the number of times that the mouse entered the central area in the opening test. The anti-anxiety effect also showed an increase in the brain tissue GABA content in mice, ignificantly decreased Glu contents in mice.Conclusion: Baihe Dihuang Decoction has some anxiolytic effect, and anti-anxiety effect may be related to increasing brain tissue GABA content, decreased Glu contents in mice. 展开更多

Objective:To study the correlation of anxiety state with blood glucose control, microinflammation and oxidative stress in patients with type 2 diabetes mellitus.Methods:A total of 138 patients with type 2 diabetes mellitus were divided into the non-anxiety group (n=43) (SAS score < 50 points), mild-to-moderate anxiety group (n=71) (SAS score 50-69 points) and severe anxiety group (n=24) (SAS score 70 points) according to the self-rating anxiety scale (SAS) score. The differences in levels of glucose metabolism indexes, microinflammation indexes and oxidative stress indexes were compared among the three groups.Results: Peripheral blood FPG, P2hPG, HOMA-IR, CRP, ASAA, IL-6, MDA and ROS levels of mild-to-moderate anxiety group and severe anxiety group were higher than those of non-anxiety group while CAT and GSH-Px contents were lower than those of non-anxiety group;peripheral blood FPG, P2hPG, HOMA-IR, CRP, ASAA, IL-6, MDA and ROS levels of severe anxiety group were higher than those of mid-to-moderate anxiety group while CAT and GSH-Px contents were lower than those of mid-to-moderate anxiety group.Conclusion:The increase of anxiety in patients with type 2 diabetes mellitus is a direct factor leading to the abnormal glucose metabolism as well as the aggravation of microinflammatory state and oxidative stress state. 展开更多
