METHODS Fifteen human cancer cell lines were subjected to various concentrations of oridonin in culture medium. The inhibitory rate of cell growth was measured by the MTT assay, and compared with a negative control and 5-Fu-positive control. RESULTS The 50% inhibiting concentration (IC50) and maximal inhibition (Imax) of oridonin shown by studying the growth of the cancer cell lines were as follows: leukemias (HL60 cells: 3.9 μg/ml and 73.8%, K562 cells: 4.3 μg/ml and 76.2%); esophageal cancers(SHEEC cells: 15.4 μg/ml and 99.2%, Eca109 cells: 15.1 μg/ml and 84.6%, TEl cells: 4.0 μg/ml and 70.2%); gastric cancers (BGC823 cells: 7.6 μg/ml and 98.7%, SGC7901 cells: 12.3 μg/ml and 85.7%); colon cancers (HT29 cells: 13.6 μg/ml and 97.2%, HCT cells: 14.5 μg/ml and 96.5%); liver cancers (Bel7402 cells: 15.2 μg/ml and 89.2%, HepG2 cells: 7.1 μg/ml and 88.3%); pancreatic cancer (PC3 cells: 11.3 μg/ml and 68.4%); lung cancer (A549 cells: 18.6 μg/ml and 98.0% ); breast cancer (MCF7 cells: 18.4 μg/ml and 84.7%); uterine cervix cancer (Hela cells: 13.7 μg/ml and 98.5%). CONCLUSION Oridonin had a relatively wide anti-tumor spectrum, and a relatively strong inhibitory effect on the growth of the 15 human cancer cells. To solve the contravention between the theory of collaboration decision design and development requirement in distributed C2 system,three-stage design approach is proposed to research coherence and optimization by which decision-maker carries out decision regulations. First,getting information and decision process are described; decision indexes and regulation models of collaboration are established. And then,a test circumstance is designed and established for measuring various decision-maker’s capabilities of carrying out decision regulation by simulation and getting their load capability parameters. Finally,the obtained parameters from the experiment are disposed and substituted into the original models for proving the coherence of decision regulations. As a result,it is feasible for three-stage approach to design collaboration decision,and decision regulations can satisfy various decision-maker requirements.展开更多
OBJECTIVE This study investigated transcriptional regulation of the main chemical con.stituents of Polygonum multiflorum Thunb.including Stilbene Glucoside(THSG) and anthraquinone constituents(Emodin,Rhein,Aloeemodin,...OBJECTIVE This study investigated transcriptional regulation of the main chemical con.stituents of Polygonum multiflorum Thunb.including Stilbene Glucoside(THSG) and anthraquinone constituents(Emodin,Rhein,Aloeemodin,Chrysophanol and Physcion) and six potential liver injury constituents(gallic acid,quercetin,luteolin,kaempferol,resveratrol) on mediated by PXR CYP3A4.Early establishment of pregnane X receptor mediated CYP3A4 drug induced rapid screening technique was used to determine the effects of these constituents.METHODS First,effect of constituents on the cell activity was detected by MTS cell viability assay.IC50 was calculated.Second,the expression vector and reporter vector were co-transfected into Hep G2 cells,10 μmol·L^(-1) Rifampicin as a positive control,10 μmol·L^(-1) Ketoconazole as a negative control.After treated with different concentrations of(the an.thraquinone constituents concentrations were 2.5,5 and 10 μmol·L^(-1);the concentrations of Gallic Acid,Quercetin,Luteolin,Kaempferol,Apigenin,Resveratrol concentrations were 5,10 and 20 μmol·L^(-1)) for24 h,the cells were tested for dual luciferase activity.RESULTS The results show that the inhibitory ef.fect of THSG,Chrysophanol,Emodin,Rhein and Aloeemodin on CYP3A4 was inhibited by co-transfec.tion of pcDNA3.1 and pGL4.17-CYP3A4.The expression of pcDNA3.14-PXR and pGL4.17-CYP3A4 was induced by the four constituents.Besides,Emodin has a directly inducing effect.Four anthraqui.none constituentscan induce the effect of CYP3A4 by PXR,but Emodin can directly induce CYP3A4.THSG can inhibit CYP3A4,but in the presence of PXR plasmid can induce CYP3A4.For the six poten.tial liver injury constituents,results show that the plasmid pcDNA3.1 was cotransfected with pGL4.17-CYP3A4 regulation of Gallic Acid and Resveratrol on CYP3A4 inhibitory effects of Quercetin,Luteolin,Kaempferol have an induce effect;after pcDNA3.14-PXR and pGL4.17-CYP3A4 cotransfected,Quercetin,Luteolin,Kaempferol,Apigenin,Resveratrol have induced effect,three constituents' induc.tion effect had significant difference.CONCLUSION 12 kinds of Polygonum multiflorum Thunb.constit.uents have inhibitory or activating effects on CYP3A4,after the participation of PXR,9 components have induced effects on CYP3A4,and the induction effect of 6 components has significant difference.The results suggested that we should pay attention to potential drug interactions when combined with Polygonum multiflorum Thunb.,and improve safety and efficacy.展开更多

However, many studies have shown that radiation can also enhance the ability of tumor cells metastasis, although the lung adenocarcinoma could be killed. The increased metastasis induced by radiation is associated with the activation of STAT3?in lung adenocarcinoma cells. Based on the importance role of?STAT3?in cell proliferation and survival, we can construct an adenovirus vector of?PIAS3 overexpress driven by radiation-induced promoter to inhibit the activation of STAT3 specifically. In this way, when?STAT3 was activated by radiation, the expression of?PIAS3 will be increased at the same time;this lead to the inhibition of invasion and metastasis caused by?STAT3 in lung adenocarcinoma cells. These researches are expected to develop a novel target and method for radiotherapy and molecular therapy of lung adenocarcinoma.展开更多
Objective:To elucidate the relationship between surgical time and postoperative complications in senile patients with hip fractures, and try to find out other factors which are related to these complications.Methods...Objective:To elucidate the relationship between surgical time and postoperative complications in senile patients with hip fractures, and try to find out other factors which are related to these complications.Methods: Sixty-two patients, 28 males aged from 65 to 72 years with a mean age of 76.3 years and 34 females aged from 65 to 95 years with a mean age of 78.1 years, who had undergone orthopedic surgery because of hip fractures,were enrolled in a retrospective cohort study. The surgical time and pattern, the type of fracture, preoperative comorbidities, American Society of Anesthesiologists (ASA) score and the volume of blood transfusion during operation were obtained from these patients who were followed up by telephone calls for postoperative complications.All the patients were followed up at least for 1 year and were divided into subgroups according to their clinical characteristics and the results were analyzed by the Statistical Analysis System software.Results:There was no significant difference in the morbidity of postoperative eomplications with the gender,age,surgical time and pattern,or ASA score. There was significant difference in the morbidity of postoperative complications related to preoperative comorbidities and the volume of blood transfusion. There was a significant causality between preoperative comorbidities and postoperative complications. The morbidity of postoperative complications was 1.651 times higher in patients with preoperative comorbidities than those without.Conclusions:There is no relationship between the surgical time and postoperative complications in senile patients who received surgery for hip fracture within 1 year.No correlation is found between the postoperative complications and gender,age,type of fracture, surgical pattern,ASA score and the volume of blood transfusion. Preoperative comorbidities are an independent predictor for postoperative complications.展开更多
As mechanical devices for moving or controlling mechanisms or systems,actuators have attracted increasing attention in various fields.Compared to traditional actuators with rigid structures,soft actuators made up of s...As mechanical devices for moving or controlling mechanisms or systems,actuators have attracted increasing attention in various fields.Compared to traditional actuators with rigid structures,soft actuators made up of stimulus-responsive soft materials are more adaptable to complex working conditions due to soft bodies and diverse control styles.Different from plate-shaped soft actuators,which have the limited deformations between two dimensional(2D)and 3D-configurations such as bending and twisting,fiber-shaped soft actuators(FSAs)own intriguing deformation modes to satisfy diverse practical applications.In this mini review,the recent progress on the controlled fabrication of the FSAs is presented.The advantages and disadvantages of each fabrication method are also demonstrated.Subsequently,the as-developed actuation mechanisms of the FSAs are displayed.Additionally,typical examples of the related applications of the FSAs in different fields have been discussed.Finally,an outlook on the development tendency of the FSAs is put forward as well.展开更多
