Characterization and Comparison of Microbial Soil Diversity in Two Andean Peatlands in Different States of Conservation-Vega Tocorpuri

The presence of a more or less constant supply of water conditions the existence of characteristic vegetation systems known as bogs (bofedales, vegas and marshes). These wetlands have a cultural, environmental and economic social importance. As a result of the exploitation of aquatic rights, peatlands began to dry up with the consequent loss of natural resources and damage to ancestral rights, and natural resources. The activities of microorganisms in wetlands play an important role in biogeochemical processes. The interaction between microbial diversity and soil, influences to the ability of the ecosystem to recover from stress (resilience). In the present work, the soil characteristics and the associated microbial biodiversity were studied, comparing samples of active and deteriorated peatland. It was seen that the loss of water causes great changes in the physical-chemical characteristics of the soil, which leads to a modification of the microbiota Proteobacteria decreased by 18% in deteriorated peatlands, which are evident more sensible to extreme conditions while Acidobacteria, Actinobacteria increased in these sample showing a better adaptation to the change of conditions. In view of the fact that high Andean Peatlands are exposed to increasing environmental impact, this preliminary comparative study of pristine and altered soil could guide the research directed to recovery of dead peatlands strategies. 展开更多

In this work a hybri... Bone tissue engineering aims to use biodegrade able scaffolds to replace damaged tissue. This scaffold must be gradually degraded and replaced by tissue as similar as possible to the original one. In this work a hybrid porous scaffold containing chitosan, polyvinyl alcohol and bioactive glass was successfully obtained and subsequently characterized by scanning electron microscopy. The scaffold presented satisfactory pore size range and open interconnected pores, which are essential for tissue ingrowth. A cytotoxicity assay showed that this biomaterial allows adequate cell viability, so that it was considered suitable for an in vivo experiment. Promising results were obtained with the implant of the scaffold in an experimental model of a New Zealand rabbit femur bone lesion. Clinical and biochemical parameters measured such as complete blood count, total serum proteins, albumin, alanine aminotransferase and aspartate aminotransferase were similar between animals in the control group at all time periods studied. Histological and histometric studies showed that the scaffold was coated with a cement-like substance, exhibiting many areas of mineralized structures. Very few osteocyte-like cells or lining-like cells were found inside the amorphous mineralized deposit. In vivo results allow us to consider this scaffold as a promising biomaterial to be applied in bone tissue engineering. 展开更多

Effluents of dying industries are specially colored and could cause severe damage to the environment. The anaerobic treatment of textile dying effluents is nowadays the preferred option, but it could generate carcinogenic aromatic amines. Recently, yeasts have become a promising altemative, combining unicellular growth with oxidative mechanisms. This work reports the characterization of the first methylotrophic yeast with dye decolorizing ability, Candida boidinii MM 4035 and some insights into its decoloration mechanism. The analysis of two selected media revealed a possible two stages mechanism of Reactive Black 5 decoloration. In glucose poor media, decoloration is incomplete and only the first stage proceeds, leading to the accumulation of a purple compound. In media with higher glucose concentrations, the yeast is able to decolorize totally an initial concentration of 200 mg/L. The entire process is co-metabolic, being largely dependent on glucose concentration but being able to proceed with several nitrogen sources. Manganese dependent peroxidase but not laccase activity could be detected during decoloration. Aromatic amines do not accumulate in culture media, supporting an oxidative decoloration mechanism of unknown ecophysiological relevance. 展开更多
