Objective:To evaluate antidiabetic and antioxidant potential of Hybanthus enneaspermus in different models.Methods:The oral glucose tolerance test(OGTT) and normoglycemic effect of alcoholic extract of Hybanthus enneaspermus(AHE) were evaluated at a dose of 125,250 and 500mg/kg p.o.while hypoglycemic activity and effect on body weight were tested at 250 and 500 mg/kg p.o.per day for 21 days in Streptozotocin(STZ) induced diabetic rats.Further,glucose uptake by hemidiaphram was also evaluated.The total polyphenolic and flavonoid were determined and their correlation with various antioxidant assays was also determined.Results:The results showed high level of phenolic content in AHE.AHE also exhibited higher total antioxidant capacity,good reducing power and a significant scavenger of reactive oxygen species like DPPH radical,nitric oxide,hydrogen peroxide and deoxyribose.Furthermore there was a significant increase in the body weight and decrease in the blood glucose level on treatment with the AHE.AHE increased glucose uptake on isolated rat hemi-diaphragm compared to control group.Conclusions:AHE reduce blood glucose level in STZ-induced diabetic model.It does not show significant effect in normoglycemic study but showes significant effect in OGT.AHE has significant antioxidant activity,which may be attributed to high phenolic content. 展开更多

Diabetes mellitus is one of the common metabolic disorders acquiring around 2.8%of the world's population and is anticipated to cross 5.4%by the year 2025.Since long back herbal medicines have been the highly esteemed source of medicine therefore,they have become a growing part of modern,high-tech medicine.In view of the above aspects the present review provides profiles of plants(65 species) with hypoglycaemic properties,available through literature source from various database with proper categorization according to the parts used,mode of reduction in blood glucose(insulinomimetic or insulin secretagugues activity) and active phyloconsliluents having insulin mimetics activity.From the review it was suggested that,plant showing hypoglycemic potential mainly belongs to the family Leguminoseae,Lamiaceae,Liliaceae,Cucurbitaceae, Asteraceae,Moraceae,Rosaceae and Araliaceae.The most active plants are Allium sativum. Gymnema sylvestre,Citrullus colocynthis,Trigonella foenum greacum,Momordica charantia and Ficuts bengalensis.The review describes some new bioactive drugs and isolated compounds from plants such as roseoside,epigallocatechin gallate,beta-pyrazol-1-ylalanine,cinchonain Ib,leucocyandin 3-O-beta-d-galactosyl cellobioside,leucopelargonidin-3- O-alpha-L rhamuoside,glycyrrhetinic acid,dehydrotrametenolic acid,strictinin,isostrictinin,pedunculagin, epicatechin and christinin-A showing significant insulinomimetic and antidiabetic activity with more efficacy than conventional hypoglycaemic agents.Thus,from the review majorly,the antidiabetic activity of medicinal plants is attributed to the presence of polyphenols,flavonoida, terpenoids,coumarins and other constituents which show reduction in blood glucose levels.The review also discusses the management aspect of diabetes mellitus using these plants and their active principles. 展开更多

Diabetes mellitus is not a single disease but is a group of metabolic disorders affecting a huge numlier of population in the world.It is mainly characterized by chronic hyperglycemia,resulting from defects in insulin secretion or insulin action.It is predicated that the number of diabetes person in the world could reach upto 366 million by the year 2030.Even though the cases of diabetes are increasing day by day,except insulin and oral hypoglycemic drugs no other way of treatment has been successfully developed so far.Thus,the objective of the present review is to provide an insight over the pathophysiological and etiological aspects of diabetes mellitus along with the remedies available for this metabolic disorder.The review also contains brief idea about diabetes mellitus and the experimental screening model with their relevant mechanism and significance mainly used nowadays.Alloxan and streptozotocin are mainly used for evaluating the antidiabetic activity of a particular drug.This review contain list of medicinal plants which have been tested for their antidiabetic activity in the alloxan induced diabetic rat model.From the available data in the literature,it was found that plant having antidiabetic activity is mainly due to the presence of the secondary metabolite.Thus,the information provided in this review will help the researchers for the development of an alternative methods rather than insidin and oral hypoglycemic agents for the treatment of diabetes niellitus,which will minimize the complication associated with the diabetes and related disorder. 展开更多

Objective:To examine the antioxidant activity and total phenolic content of different solvent fractions of Pedulium murex(P.murex)Linn fruits(Family:Pedaliaceae)as well as the correlation between the total antioxidant capacity and total phenolic content.Methods:In the present study,the antioxidant activities of P.murex were evaluated using six in-vitro assays,namely total antioxidant assay,DPPH assay,reducing power,nitric oxide scavenging,hydrogen peroxide scavenging and deoxyribose scavenging assays,and total phenol contents were also investigated.Results:The ethyl acetate(EA)fraction was found to have high levels of phenolic content(298.72±2.09 mg GAE/g).The EA fraction exhibit higher total antioxidant capacity,higher percentage of DPPH radical scavenging activity(135.11±2.95μg/mL),nitric oxide(200.57±4.5lμg/mL),hydrogen peroxide(2I7.91±6.12μg/mL),deoxyribose(250.01±4.68μg/mL)and higher reducing power.Correlation coefficient(r^2=0.914)was found to be significant between total phenolic content and total antioxidant activity.Conclusions:In general,the results indicate that the EA fractions are rich in phenolic antioxidants with potent free radical scavenging activity implying their importance to human health. 展开更多

Methods:Different fractions of H.enneaspermus were tested for total phenolic content,and in vitro antioxidant activity was measured by total antioxidant assay,DPPH assay,reducing power, nitric oxide(NO),hydrogen peroxide(H<sub>2</sub>O<sub>2</sub>) scavenging assays.Results:The ethyl acetate(EA) fraction was found to have high levels of phenolic content[(212.15±0.79) mg GAE/g].The EA fraction exhibited higher total antioxidant capacity,higher percentage of DPPH radical scavenging activity[(127.07±2.29) Fg/mL],nitric oxide[(245.16±1.44)μg/mLJ,hydrogen peroxide[(227.38±7.18)μg/mL],deoxyribose[(270.61±8.72)μg/mL]and higher reducing power.There was a significant correlation between total phenolic content and total antioxidant activity(r =0.972). Conclusions:These results reveal that EA fraction of H.enneaspermus has strong antioxidant potential compared with other fractions.Our further study has been extended to the isolation of the possible compound that is responsible for having antioxidant property. 展开更多

Objective:To evaluate the aldose reductase inhibitory(ARI)activity of different fractions of Hybanthus enneaspermus for potential use in diabetic cataract.Methods:Total phenol and flavonoid content of different fractions was determined.ARI activity of different fractions in rat lens was investigated in vitro.Results:The results showed significant level of phenolic and flavonoid content in ethyl acetate fraction[total phenol(212.15±0.79 mg/g),total flavonoid(39.11±2.27mg/g)]and aqueous fraction[total phenol(140.62±0.57mg/g),total flavonoid(26.07±1.49 mg/g)]as compared with the chloroform fraction[total phenol(68.56±0.51mg/g),total flavonoid(13.41±0.82mg/g)]and petrolium ether fraction[total phenol(36.68±0.43mg/g),total flavonoid(11.55±1.06mg/g)].There was a significant difference in the ARI activity of each fraction,and it was found to be the highest in ethyl acetate fraction[IC_(50)(49.26±1.76μg/mL)]followed by aqueous extract[IC_(50)(70.83±2.82μg/mL)]and it was least in the petroleum ether fraction[IC_(50)(118.89±0.71μg/mL)].Chloroform fraction showed moderate activity[IC_(50)(98.52±1.80μg/mL)].Conclusions:Different fractions showed significanct amount of ARI activity,where in ethyl acetate fraction it was found to be maximum which may be due to its high phenolic and flavonoid content.The extract after further evaluation may be used in the treatment of diabetic cataract. 展开更多

Bioautography is a means of target-directed isolation of active molecules on chromatogram. Organic solvents employed in chromatographic separation process can be completely removed before biological detection because these solvents cause inactivation of enzymes and/or death of living organisms. They offer a rapid and easy identification of bioactive lead/scaffolds in complex matrices of plant extracts. Bioautography is a technique to isolate hit(s)/lead(s) by employing a suitable chromatographic process followed by a biological detection system. This review critically describes the methodologies to identify antimicrobial, antioxidant, enzyme inhibitor lead/scaffolds by employing bioautography. Objetive: To explore the potential insecticidal, ovipositor deterrent and antifeedant effects of ethyl acetate extract of the seeds of Senna tora(Syn. Cassia tora) against cowpea weevil(Callosobruchus maculatus).Method: The activities were evaluated using standard protocols.In these bioassays, the cowpea seeds were used directly as an insect feed. The activity of the extract and isolated compounds were tested at concentrations of 100, 200 and 300 μg/mL and compared to neem oil and cinnamaldehyde(as standard positive controls). Phytochemical analysis of the ethyl acetate extract was done through a number of chromatographic techniques and the structures of the isolated compounds were established through comprehensive spectroscopic analysis including 2 D-NMR and ESI-MS studies.Results: Fractionation of the active ethyl acetate extract resulted in the isolation of one known anthraquinone,aurantio-obtusin(1) and a novel compound that was named as cassiatorin(2). Compounds1 and 2 showed comparable insect antifeedant properties with the positive controls while their insecticidal and ovipositor deterrent effects were far superior to the standard controls.Conclusions: It is thus concluded that Senna tora extracts and the isolated compounds(1 and2) may be employed in the postharvest management of stored cowpea seeds and as other crop protectants. 展开更多

Methods:Phytochemical screening and in vitro antioxidant activities of A.campanulatus tuber n-hexane extract(ACHE) and methanolic extract(ACME) were evaluated using DPPH,hydroxyl radical,reducing power and total antioxidant capacity assays.The total phenolic and flavonoid contents were also investigated.The protective potential of two different doses of ACME(125 and 250 mg/kg) was also evaluated against thioaeetamide(TAA) induced oxidative stress in rats. Silymarin used as a standard drug control.Hepatotoxicitv was assessed by quantifying the serum levels of aspartate aminotransferase(AST),alanine aminotransferase(AIT),alkaline phosphatase (ALP) and lactate dehydrogenase(LDH).The antioxidant potential of ACME were also evaluated by the estimation of catalase(CAT),glutathione peroxidase(GPx),glutathione reductase (OR),glutathione-S-transferase(GST),reduced glutathione(GSH) and lipid peroxidation (Thiobarbituric acid reactive substances) in hepatic and renal tissues.Histopathologic changes of liver were also evaluated.Results:In vitro studies revealed that ACME has higher antioxidant and radical scavenging activity than ACHE,which may be attributed to its higher phenolic and flavonoid content.ACME significandy prevented the elevation of serum AST,ALT.ALP,LDH,and tissue malondialdehyde levels(P 【 0.05).Hepatic and renal GSH.GST.GR,GPx,and catalase levels were remarkably increased by the treatment with the extract.Quantification of histopathological changes also supported the dose dependent protective effects of ACME.Conclusions:The results do suggest that A.campanulatus tuber could be considered as a potential source of natural antioxidant. 展开更多

Objective: To discover the mechanism behind ameliorative effects of Michelia champaca(M. champaca) in gastrointestinal, respiratory and cardiovascular disorders. Methods: Antispasmodic potential was evaluated by trying the M. champaca extract(aqueous:ethanolic) on rabbit aorta, trachea and jejunum in vitro. Isotonic and isometric transducers coupled with Power Lab data acquisition system was used to record the responses of isolated tissues. Results: M. champaca extract relaxed the spontaneous and high K^+(80 mmol/L)-induced contractions of isolated jejunum preparation of rabbit showing a Ca^(2+) channel blocking mechanism. Moreover, extract shifted calcium concentration response curves towards right like standard calcium channel blocker verapamil. In rabbit tracheal preparation, M. champaca relaxed both carbachol(1 μmol/L) and high K^+-induced contractions, likewise verapamil. In rabbit aorta preparation, M. champaca relaxed phenylephrine(1 μmol/L) and high K^+-induced contractions similar to verapamil. Conclusion: M. champaca possesses spasmolytic, airways relaxant and vasodilator actions mediated perhaps due to blocking of Ca^(2+) channels, hence validating its therapeutic usage in diarrhea, asthma and hypertension. 展开更多

Cancer is an abnormal state of cells where they undergo uncontrolled proliferation and produce aggressive malignancies that causes millions of deaths every year.With the new understanding of the molecular mechanism(s)of disease progression,our knowledge about the disease is snowballing,leading to the evolution of many new therapeutic regimes and their successive trials.In the past few decades,various combinations of therapies have been pro-posed and are presently employed in the treatment of diverse cancers.Targeted drug therapy,immunotherapy,and personalized medicines are now largely being employed,which were not common a few years back.The field of cancer discoveries and therapeutics are evolving fast as cancer type-specific biomarkers are progressively being identified and several types of cancers are nowadays undergoing systematic therapies,extending patients’disease-free survival thereafter.Although growing evidence shows that a systematic and targeted approach could be the future of cancer medicine,chemotherapy remains a largely opted therapeutic option despite its known side effects on the patient’s physical and psychological health.Chemother-apeutic agents/pharmaceuticals served a great purpose over the past few decades and have remained the frontline choice for advanced-stage malignancies where surgery and/or radiation therapy cannot be prescribed due to specific reasons.The present report succinctly reviews the existing and contemporary advancements in chemotherapy and assesses the status of the enrolled drugs/pharmaceuticals;it also comprehensively discusses the emerging role of specific/targeted therapeutic strategies that are presently being employed to achieve better clinical success/survival rate in cancer patients. 展开更多

AIM:A decoction of Elephantopus scaber(Asteraceae) root is used to treat liver disorders in Indian and Chinese traditional medicine.The study was designed to examine the dose response effects of E.scaber methanolic extract on rats exposed to N-nitrosodiethylamine(NDEA) induced hepatotoxicity(0.02% NDEA in water five days per week,per oral) in preventive and curative models.METHODS:In preventive groups,NDEA was administered for six weeks.Daily doses of E.scaber methanolic extract(200 and 100 mg kg 1) started one week before the onset of NDEA intoxication and continued for six weeks.In curative animals,NDEA was administered for six weeks followed by treatment with the methanolic n-hexane extract of E.scaber(200 and 100 mg kg 1) for ten days.RESULTS:E.scaber extract treatment significantly(P ≤ 0.05) reduced the levels of AST,ALT,and MDA in both experimental groups.The extract also enhanced the antioxidant enzyme and protein levels in rats intoxicated with NDEA.Treatment with the extract dose dependently protected the liver from NDEA-induced hepatotoxicity with normal hepatocytes and uniform sinusoids,but in some areas showed degenerating hepatic cells in both treatment groups.CONCLUSION:E.scaber methanolic extract dose dependently prevented and reversed the hepatotoxicity induced by NDEA in both experimental models. 展开更多

Aldose reductase (AR), a cytosolic, monomeric oxidoreductase, is a key enzyme in the polyol pathway which controls the conversion of glucose to sorbitol. The accumulation of sorbitol by the activation of AR enzymes in lens, retina, and sciatic nerves leads to the cause of diabetic defects resulting in various secondary complications, viz. retinopathy, neuropathy, nephropathy and Alzheimer's disease. Thus, reduction of the polyol pathway flux by AR inhibitors could be a potential therapeutic opening in the treatment and prevention of diabetic complications. At present, the AR inhibitors belong to two different chemical classes. One is the hydantoin derivatives, such as Sorbinil, Dilantin, and Minalrestat, and the other is the carboxylic acid derivatives, such as Epalrestat, Alrestatin, and Tolrestat. However, it is known that most of these synthethic compounds have unacceptable side-effects. Well known medicinal plants like Chrysanthemum indicum, Chrysanthemum morifolium, Prunus mume, Myrcia multiflora, Centella asiatica, and Salacia reticulata, Salacia oblonga, and Salacia chinensis exhibited potent AR inhibitory activity. The present review summarizes the list of plant material, and their isolated phytoconstituents which have been tested for their AR inhibitory activity. This litreature review covers the period to 2011, and a total of 72 plants are listed. 展开更多

Muell belonging to the family Violaceae, popularly known as Ratanpurus (Hindi) is a herb or a shrub distributed in the tropical and subtropical regions of the world. In the Ayurvedic literature, the plant is reported to cure conditions of "Kapha" and "Pitta", urinary calculi, strangury, painful dysentery, vomiting, burning sensation, wandering of the mind, urethral discharge, blood trouble, asthma, epilepsy, cough, and to give tone to the breasts. Phytochemically, the plant contains a considerable amount of dipeptide alkaloids, aurantiamide acetate, isoarborinol, and β-sitosterol, sugars, flavonoids, steroids, triterpenes, phenols, flavones, catachins, tannins, anthraquinones and amino acids. Pharmacologically, the plant is reported to possess antidiabetic, antiplasmodial, antimicrobial, anticonvulsant, nephroprotective, aphrodisiac, hepatoprotective, antiinflammatory, aldose reductase inhibitory and free radical scavenging activities. The information provided in this review will be worthwhile to know the applicability of H. enneaspermus for the treatment of various acute or chronic diseases with a diverse nature of phytoconstituents. The overall data in this review article were collected from various scientific sources on the research of H. enneaspermus. 展开更多
