METHODS: In vitro antioxidant activity was studied in 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assay. Phenolic content was determined using Folin-Ciocalteu's reagent. Reducing ability was evaluated by ferric reducing power assay. Peripherally and centrally acting analgesic activity was studied in three different in vivo models, namely, acetic acid-induced writhing, hot-plate test, and tail-flick test in Swiss albino mice. In vivo antidiarrheal activity was evaluated in castor oil- and magnesium sulfate-induced diarrhea in mice. Gastrointestinal motility test was also carried out in mice. All studies in mice were undertaken at the doses of 250 and 500 mg/kg body weight. Antibacterial activity was assessed by disk diffusion assay against some Gram-positive and Gram-negative bacterial strains. Acute toxicity test was conducted to assess the safe doses of the extract. RESULTS: The extract showed 50% inhibitory concentration value of 12.65 μg/mL in DPPH radical- scavenging assay. Phenolic content was found to be 589.83 mg gallic acid equivalent per 100 g of dried fruits extract. Reducing power was in a concentration-dependent manner, and strongly comparable with the standard ascorbic acid. The extract demonstrated significant inhibition of writhing in acetic acid-induced writhing test at both dose levels (P〈0.01). The extract also raised pain threshold in both hot-plate and tail-flick test in a dose-dependent manner, and the results were statistically significant (P〈0.01). The extract significantly (P〈0.01) increased latent period, and decreased defecation in both castor oil- and magnesium sulfate-induced diarrhea. The extract also decreased gastrointestinal motility in mice. In disk diffusion assay, the extract showed potential antibacterial activity against all the tested bacterial strains. CONCLUSION: The results suggest that the ethanol extract of M. seminifera fruit has potential antioxidant, analgesic, antidiarrheal, and antibacterial activities.展开更多

文摘Background:Blumea lacera(Family Asteraceae)is a herb found in the uncultivated lands of many Asian countries. This plant is used by quack doctors for some medicinal purposes. Objective:Works presented in this manuscript were conducted on the ethanol extract of aerial parts of B.lacera.Various pharmacological tests like antioxidant,analgesic,anti-inflammatory,antidiarrheal,diuretic,antibacte-rial,anthelmintic and toxicological tests like brine shrimp lethality and acute toxicity were evaluated. Methods:Antioxidant test was conducted by determining secondary metabolites content and DPPH free radical scavenging assay.Acetic acid-induced writhing method,xylene-induced ear edema method, castor oil-induced diarrheal method, and urinary volume test were performed to evaluate the analgesic,anti-inflammatory,an-tidiarrheal and diuretic effects, respectively. The antibacterial test was performed by disk diffusion method, anthelmintic activity by recording the paralysis and death time of P.cervi and finally, cytotoxic activity was performed by brine shrimp lethality bioassay. Results:TPC,TFC and TTC contents of B.lacera extract were found to be 9 mg GAE/g,31 mg QE/g and 18 mg GAE/g.The SC 50 value of the DPPH radical scavenging assay was found to be higher than 3000μg·mL^(-1).The LD 50 value of B.lacera extract in mice was higher than 5000 mg·kg^(-1).This extract significantly inhibited writhing reflexes by 24.5%and 43.6%and reduced ear edema up to 24.6%and 41%at 250 and 500 mg·kg^(-1) bw doses, respectively. It also increased the latency of the first defecation period up to 52.8 min and 106.6 min as well as decreased the stool count by 34.1%and 48.2%at 250 and 500 mg·kg-1 bw doses, respectively. It also showed an increase in urinary output in the diuretic test.Good antibacterial activity of B.lacera extract was confirmed by retardation in bacterial growth and significantly killed P.cervi in a dose-dependent manner. In brine shrimp lethality bioassay, the LC 50 value was found to be 5.4μg·mL-1.Conclusion:B.lacera showed mild analgesic,anti-inflammatory and antidiarrheal properties. It could be used as a diuretic, antibacterial, anthelmintic and cytotoxic drug which justify the plant’s traditional uses in folk medicine.

文摘Objective:To assess ethanolic extract of the leaves of Melia azedarach L.(Family-Meliaceae)for its possible antioxidant, analgesic and antibacterial activities in association with performing phytochemical evaluation. Methods:A number of phytochemical tests of this extract were utilized to distinguish the existence of different sorts of compounds.Qualitative and quantitative antioxidant activities were assessed by TLC and DPPH scavenging assay respectively.Acetic acid induced writhing test in mice and disk diffusion assay of the leaves extract were carried out to demonstrate the analgesic and antibacterial activities respectively. Results:The phytochemical assessment revealed the existence of alkaloids, flavonoids, glycosides, saponins and tannins like compounds.Ethanolic extract of the leaves demonstrated antioxidant, analgesic and antibacterial activities in vitro. Conclusion:Considering the study, this could justify the leaves extract’s bioactivities but,to substantiate the activity of individual compound further investigation is necessary.
