BACKGROUND The circular RNA circ-PRKCI is an endogenous non-coding RNA that forms a covalently closed ring after reverse splicing, which plays a key role in the occurrence and development of multiple digestive system ...BACKGROUND The circular RNA circ-PRKCI is an endogenous non-coding RNA that forms a covalently closed ring after reverse splicing, which plays a key role in the occurrence and development of multiple digestive system tumors.AIM To investigate the role and mechanism of circ-PRKCI in the occurrence and development of hepatocellular carcinoma(HCC).METHODS This study used real-time polymerase chain reaction to detect the expression of circ-PRKCI in tumor tissues, tumor adjacent tissues, and blood in patients with HCC and other digestive system tumor cells. A series of functional tests were performed to explore whether circ-PRKCI affects the growth of HCC cells and what is its mechanism in HCC. Meanwhile, fluorescence in situ hybridization was used to detect the subcellular localization of circ-PRKCI. Survival analysis was performed to predict the correlation between circ-PRKCI and the prognosis of HCC. Chi-square test and t-test were performed for statistical analyses.RESULTS The level of circ-PRKCI was significantly higher in HCC tissues than in tumor adjacent tissues, and in HCC cell lines than in cells lines of esophageal, liver,stomach, and colon cancers. A series of functional tests showed that circ-PRKCI substantially inhibited cell apoptosis and promoted cell invasion. It was foundthat circ-PRKCI can act as the sponge of miRNA-545 to reduce the expression of AKT3 protein. Moreover, the result of survival analysis showed that circ-PRKCI target gene E2 F7 can reduce liver cancer patients' survival rate. And clinical data suggested that the distribution of circ-PRKCI rose with the depth of invasion,lymph node metastasis, distant metastasis, and TNM stage, indicating that circPRKCI may affect the survival and prognosis of patients with HCC by regulating E2 E7.CONCLUSION This study explores the role and mechanism of circ-PRKCI in HCC, which provides a new research direction and theoretical basis for the treatment of HCC.展开更多

To verify the e ffectiveness of tiaojing decoction on diminished ovarian reserve and uterine artery blood flow parameters, explore the correlation between uterine artery blood flow parameters and sex hormones. Methods: Total 60 diminished ovarian reserve patients with deficiency of Kidney-Yin were randomly divided into the experimental group (n=30, taking tiaojing decoction and climen) and the control group (n=30, taking climen) and they were treated in 3 months. Observe bFSH, bE2, uterine artery PSV, EDV, S/D, RI before and after the treatment in the 2-4 day of menstruation. Results: After treatment, serum sex hormone levels and uterine artery blood flow parameters of the two groups were significantly different (P < 0.05), and the experimental group was significantly better than the control group. There was a linear correlation between PSV and FSH, PSV and E2, FSH and E2. Conclusion: Tiaojing decoction combined with climen has significant effect on diminished ovarian reserve patients with deficiency of Kidney-Yin. Methods: Sixty cases with night crying were treated with Three-character- scripture School pediatric tuina, followed by observing the efficacy. Results: Forty-nine cases were recovered, 7 cases improved, and 4 cases failed. The total effective rate was 93.3%. Conclusion: Three-character-scripture School pediatric tuina is effective for night crying of infants. Methods AMI model was establi...The study aimed to investigate the effects of ivabradine on the levels of hypoxia-inducible factor-lalpha (HIF-1α) and VEGF in serum of rabbit with acute myocardial infarction (AMI). Methods AMI model was established by ligating the left anterior descending branch of the coronary artery in New Zealand white rabbits. Twenty five rabbits were randomly divided into 4 groups: sham-operated (S), myocardial-infarction (M) with bisoprolol treatment (M + B) and ivabradine-treated (I + M). The medical treatment began immediately after infarction and continued for 3 weeks. Serum of each rabbit was obtained at the following time points (24 h before the operation, 24 h, 3 d, 1 week, 2 weeks and 3 weeks after the operation). ELISA was used to measure the levels of HIF-1α and VEGF of each sample. ECG and heart rates (before and after treatment) were analyzed. Results Baseline heart rate showed no significant differences between the 3 infarcted groups (M, M + B, M + I). Three weeks later the heart rates were significantly lower in group M + B and group M + I than in group M. However, there was no statistic difference between the two drug-treated groups (P = 0.848). The levels of HIF-1α and VEGF in groups M, M + B and M + I) increased significantly compared with group S (P 〈 0.01). The productions of HIF-1α and VEGF were lower in group M + B and group M + I compared with group M (P 〈 0.01). There was no statistical difference between the group M + B and group M + I (P 〉 0.05), and the correlative analysis revealed that the production of HIF-1α was positively correlated with that of VEGF (r = 0.732, P 〈 0.01). Conclusion Ivabradine can reduce heart rate and meanwhile decrease the serum levels of HIF-1α and VEGF after AMI.展开更多
