Here,we aimed to study the changes in proteome of golden pompano fillets during post-mortem storage.Tandem mass tags(TMT)-labeled quantitative proteomic strategy was applied to investigate the relationships between protein changes and quality characteristics of modified atmosphere packaging(MAP)fillets during superchilling(-3°C)storage.Scanning electron microscopy was used to show that the muscle histology microstructure of fillets was damaged to varying degrees,and low-field nuclear magnetic resonance was used to find that the immobilized water and free water in the muscle of fillets changed significantly.Total sulfhydryl content,TCA-soluble peptides and Ca2+-ATPase activity also showed that the fillet protein had a deterioration by oxidation and denaturation.The Fresh(FS),MAP,and air packaging(AP)groups were set.Total of 150 proteins were identified as differential abundant proteins(DAPs)in MAP/FS,while 209 DAPs were in AP/FS group.The KEGG pathway analysis indicated that most DAPs were involved in binding proteins and protein turnover.Correlation analysis found that 52 DAPs were correlated with quality traits.Among them,8 highly correlated DAPs are expected to be used as potential quality markers for protein oxidation and water-holding capacity.These results provide a further understanding of the muscle deterioration mechanism of packaging golden pompano fillets during superchilling. 展开更多

Lotus(Nelumbo nucifera),an ancient aquatic plant,possesses a unique pharmacological activity that is primarily contributed by benzylisoquinoline alkaloids(BIAs).However,only few genes and enzymes involved in BIA biosynthesis in N.nucifera have been isolated and characterized.In the present study we identified the regiopromiscuity of an O-methyltransferase,designated NnOMT6,isolated from N.nucifera;NnOMT6 was found to catalyze the methylation of monobenzylisoquinoline 6-O/7-O,aporphine skeleton 6-O,phenylpropanoid 3-O,and protoberberine 2-O.We further probed the key residues affecting NnOMT6 activity via molecular docking and molecular dynamics simulation.Verification using site-directed mutagenesis revealed that residues D316,N130,L135,N176A,D269,and E328 were critical for BIA O-methyltransferase activities;furthermore,N323A,a mutant of NnOMT6,demonstrated a substantial increase in catalytic efficiency for BIAs and a broader acceptor scope compared with wild-type NnOMT6.To the best of our knowledge,this is the first study to report the O-methyltransferase activity of an aporphine skeleton without benzyl moiety substitutions in N.nucifera.The study findings provide biocatalysts for the semisynthesis of related medical compounds and give insights into protein engineering to strengthen O-methyltransferase activity in plants. 展开更多

Chemotherapy can significantly reduce follicle counts in ovarian tissues and damage ovarian stroma,causing endocrine disorder,reproductive dysfunction,and primary ovarian insufficiency(POI).Recent studies have suggested that extracellular vesicles(EVs)secreted from mesenchymal stem cells(MSCs)exert therapeutic effects in various degenerative diseases.In this study,transplantation of EVs from human induced pluripotent stem cell-derived MSCs(iPSC-MSC-EVs)resulted in significant restoration of ovarian follicle numbers,improved granulosa cell proliferation,and inhibition of apoptosis in chemotherapy-damaged granulosa cells,cultured ovaries,and in vivo ovaries in mice.Mechanistically,treatment with i PSC-MSC-EVs resulted in up-regulation of the integrinlinked kinase(ILK)-PI3K/AKT pathway,which is suppressed during chemotherapy,most likely through the transfer of regulatory microRNAs(miRNAs)targeting ILK pathway genes.This work provides a framework for the development of advanced therapeutics to ameliorate ovarian damage and POI in female chemotherapy patients. 展开更多

With gradually diminishing Fe grade in tandem with the ever-increasing demand for high-grade iron ores,iron ore industries are now focusing on the beneficiation of low-grade iron ore fines,mainly considered waste.Besides,the scarcity of water at many of the mines’sites and the new water conservation policies of the governments have necessitated research on suitable dry beneficiation routes.In this context,an effort has been made to evaluate the efficacy of a dry classification unit,such as the VSK separator,in upgrading the iron values of two low-grade Indian iron ore fines,named Sample 1 and Sample 2.The mineralogical studies,involving scanning electron microscopy and X-ray diffraction,suggest that Sample 1 is a low-grade blue dust sample(51.2wt%Fe)containing hematite and quartz as the major minerals,while Sample 2(53.3wt%Fe)shows the presence of goethite in addition to hematite and quartz.The experiments,carried out using Box-Benkhen statistical design,indicate that blower speed,followed by feed rate,is the most influencing operating parameter in obtaining a good product in the VSK separator.At optimum levels of the operating factors,a fines product with~55wt%Fe at a yield of~40%can be obtained from Sample 1,while Sample 2 can be upgraded to~56wt%Fe at a yield of~85%.The results suggest that the VSK separator can be employed as an efficient intermediate unit operation in a processing circuit to upgrade the iron contents of iron ore fines. 展开更多

In this paper, a free-surface synthetic schlieren(FS-SS) method was performed to detect the free surface disturbances. It is a purely optical method that uses refraction of light to reconstruct the height changes of water surface. The theory was developed based on Moisy's research, but has mainly been used in small-scale applications like painting and coating industry. Based on the methods and theories of the literature review, an in-depth investigation was conducted to optimize the FS-SS method and verify its feasibility on a relatively large-scale(e.g., wake region of shallow flow). The experimental setup was simplified which is approachable in most laboratories. Through proper experimental setting and an optimized post-processing routine, the quality of image was highly improved and ensured the accuracy of results. A drop test was performed proving the continuity of FS-SS method in the time domain. Also, a comparison test with flow around a cylinder at two speeds showed the ability of FS-SS method to reconstruct the irregular water surface in relative large-scale flow structures. 展开更多

The theoretical capacity of Li Fe PO4 based on one electron reaction is 170 m Ah g-1at the stable voltage plateau of 3.5 V vs. Li/Li+. However, the instinct drawbacks of olivine structure induce a poor rate performance, resulting from the low lithium ion diffusion rate and low electronic conductivity.In this review, we summarize the methods for enhancing the rate performance of Li Fe PO4 cathode materials,including carbon coating, elements doping, preparation of nanosized materials, porous materials and composites,etc. Meanwhile, the advantages and disadvantages of above methods are also discussed. 展开更多

The monographs of European Pharmacopoeia (Ph. Eur.) and United States Pharmacopoeia (USP) specify six impurities, viz.;impurities A, B, C, D, E and F, respectively for its active pharmaceutical ingredient (API). The identification and synthesis of all impurities except impurity E are well described in the literature;however, there is no report related to impurity E. The prospects to the formation and controlling of impurity E up to -0.03% in the synthesis of pantoprazole sodium sesquihydrate (PAN) were discussed in detail for the first time. The present work described the journey towards the successful development of an optimal preparation procedure of dimer impurity E. Mg-9Al-xPr(x=0.4,0.8 and 1.2,mass fraction,%)magnesium alloys were prepared by high-pressure die-casting technique.The effects of Pr on the microstructures of die-cast Mg-9Al based alloy were investigated by XRD and SEM.Needle-like Al11Pr3 phase and polygon Al6Mn6Pr phase are found in the microstructure.With 0.4%Pr addition,fine needle-like Al11Pr3 phase and a small amount of polygon Al6Mn6Pr phase near the grain boundary are found in the microstructure.Increasing Pr addition to 0.8%, lots of coarse needle-like Al11Pr3 phase within grain and polygon Al6Mn6Pr phase on grain boundary are observed.Further increasing Pr addition,the size of needle-like Al11Pr3 phase decreases,while the size of polygon Al6Mn6Pr relatively increases.The mass fraction of Pr at around 0.8%is considered to be suitable to obtain the optimal mechanical properties.The optimal mechanical properties are mainly resulted from grain boundary strengthening obtained by precipitates and solid solution. 展开更多

The objective of the study is to investigate the suitability of using Pulse-coherent Acoustic Doppler Profiler (PCADP) to estimate suspended sediment concentration (SSC). The acoustic backscatter intensity was corrected for spreading and absorption loss, then calibrated with OBS and finally converted to SSC. The results show that there is a good correlation between SSC and backscatter intensity with R value of 0.74. The mean relative error is 22.4%. Then the time span of little particle size variation was also analyzed to exclude the influence of size variation. The correlation coefficient increased to 0.81 and the error decreased to 18.9%. Our results suggest that the PCADP can meet the requirement of other professional instruments to estimate SSC with the errors between 20% and 50%, and can satisfy the need of dynamics study of suspended particles. 展开更多

Coffee is commonly consumed beverage in the world and it has been suggested to have beneficial effect.Chlorogenic acids(CGAs)are main ingredient of coffee beans which has been extensively used in nutraceuticals and medicine.Recently,various therapeutic effects of chlorogenic acids have been investigated.However,there are limited studies to investigate its anticancer properties.In the present study,we have used chlorogenic acid complex(CGA7)a decaffeinated water soluble green coffee bean extract to evaluate its cytotoxic effect on human and mouse cancer cell lines by using different approaches.From our results we found CGA7 treatment induces cell death in a dose and time dependent manner in different cancer cell lines.Further,CGA7 induced apoptosis was characterized by DNA fragmentation,PARP-1 cleavage,caspase-9 activation,and down regulation of Bcl-2,an anti-apoptotic protein and up regulation of pro-apoptotic protein BAX.Overall findings indicated that CGA7 complex a potent anticancer molecule found in green coffee beans could be a safe bioactive ingredient for prevention of cancer. 展开更多

Proposed structure of impurity was unambiguously confirmed by synthesis followed by characterization using 1H, 13C nuclear magnetic resonance spectrometry (NMR), 1H-1H correlation spectro-scopy (COSY) and infrared spectroscopy (IR). Subarachnoid hemorrhage is a devastating disease with significant mortality and morbidity,despite advances in treating cerebral aneurysms.There has been recent progress in the intensive care management and monitoring of patients with subarachnoid hemorrhage,but the results remain unsatisfactory.Microglia,the resident immune cells of the brain,are increasingly recognized as playing a significant role in neurological diseases,including subarachnoid hemorrhage.In early brain injury following subarachnoid hemorrhage,microglial activation and neuroinflammation have been implicated in the development of disease complications and recovery.To understand the disease processes following subarachnoid hemorrhage,it is important to focus on the modulators of microglial activation and the pro-inflammatory/anti-inflammatory cytokines and chemokines.In this review,we summarize research on the modulators of microglia-mediated inflammation in subarachnoid hemorrhage,including transcriptome changes and the neuroinflammatory signaling pathways.We also describe the latest developments in single-cell transcriptomics for microglia and summarize advances that have been made in the transcriptome-based classification of microglia and the implications for microglial activation and neuroinflammation. 展开更多

The Upper Ordovician Wufeng-Lower Silurian Longmaxi and the Lower Cambrian Qiongzhusi shales are the major targets for shale gas exploration and development in China.Although the two organic-rich shales share similar distribution ranges and thicknesses,they exhibit substantially different exploration and development results.This work analyzed the nanopore structures of the shale reservoirs in this region.Pore development of 51 shale samples collected from various formations and locations was compared using the petromineralogical,geochemical,structural geological and reservoir geological methods.The results indicate that the reservoir space in these shales is dominated by organic pores and the total pore volume of micropores,mesopores,macropores in different tectonic areas and formations show different trends with the increase of TOC.It is suggested that organic pores of shale can be well preserved in areas with simple structure and suitable preservation conditions,and the shale with smaller maximum ancient burial depth and later hydrocarbongeneration-end-time is also more conducive to pore preservation.Organic pore evolution models are established,and they are as follows:①Organic matter pore development stage,②Early stage of organic matter pore destruction,and③late stage of organic matter pore destruction.The areas conducive to pore development are favorable for shale gas development.Research results can effectively guide the optimization and evaluation of favorable areas of shale gas. 展开更多

The experimental parameters were set as the flow rate of decane from 0 mL/min to 10 mL/min and the DC charging voltage from DC 0 V to 12 V with a pulse repetition rates of 200 Hz. The flow regime of decane was observed and the volume-to-electrical charge ratio was measured. Unlike a conventional EHDA system, the results show that a corona discharge was initiated at the edge of the hollow electrode at a specific corona on-set voltage of -17 kV or -20 kV in the case with or without decane flow, respectively. The spread of SARS-CoV-2 as an emerging novel coronavirus disease (COVID-19) had progressed as a worldwide pandemic since the end of 2019. COVID-19 affects firstly lungs tissues which are known for their very slow regeneration. Afterwards, enormous cytokine stimulation occurs in the infected cells immediately after a lung infection which necessitates good management to save patients. Exosomes are extracellular vesicles of nanometric size released by reticulocytes on maturation and are known to mediate intercellular communications. The exosomal cargo serves as biomarkers in diagnosing various diseases;moreover, exosomes could be employed as nanocarriers in drug delivery systems. Exosomes look promising to combat the current pandemic since they contribute to the immune response against several viral pathogens. Many studies have proved the potential of using exosomes either as viral elements or host systems that acquire immune-stimulatory effects and could be used as a vaccine or drug delivery tool. It is essential to stop viral replication, prevent and reverse the massive storm of cytokine that worsens the infected patients’ situations for the management of COVID-19. The main benefits of exosomes could be;no cells will be introduced, no chance of mutation, lack of immunogenicity and the damaged genetic material that could negatively affect the recipient is avoided. Recovery of alginate extracted from aerobic granular sludge(AGS)has given rise to a novel research direction.However,these extracted alginate solutions have a water content of nearly 100%.Alternately,ultrafiltration(UF)is generally used for concentration of polymers.Furthermore,the introduction of multivalent metal ions into alginate may provide a promising method for the development of novel nanomaterials.In this study,membrane fouling mitigation by multivalent metal ions,both individually and in combination,and properties of recycled materials were investigated for UF recovery of sodium alginate(SA).The filtration resistance showed a significantly negative correlation with the concentration of metal ions,arranged in the order of Mg^2+<Ca^2+<Fe^3+<Al^3+(filtration resistance mitigation),and the moisture content of recycled filter cake showed a marked decrease.For Ca^2+,Mg^2+,Fe^3+,and Ca^2++Fe^3+,the filtration resistances were almost the same when the total charge concentration was less than 5 mmol·L^–1.However,when the total charge concentration was greater than 5 mmol·L^–1,membrane fouling mitigation increased significantly in the presence of Ca^2+or Fe^3+and remained constant for Mg^2+with the increase of total charge concentration.The filtration resistance mitigation was arranged in the order of Fe^3+>Fe^3++Ca^2+>Ca^2+>Mg^2+.Three mechanisms were proposed in the presence of Fe^3+,such as the decrease of SA concentration,change in p H,and production of hydroxide iron colloids from hydrolysis.The properties of recycled materials(filter cake)were investigated via optical microscope observation,dynamic light scattering,Fourier transform infrared,X-ray photoelectron spectroscopy(XPS),and scanning electron microscopy.The results provide further insight into UF recoveries of alginate extracted from AGS. 展开更多

This paper considers nonlinear dynamics of teth- ered three-body formation system with their centre of mass staying on a circular orbit around the Earth, and applies the theory of space manifold dynamics to deal with the nonlinear dynamical behaviors of the equilibrium configurations of the system. Compared with the classical circular restricted three body system, sixteen equilibrium configurations are obtained globally from the geometry of pseudo-potential energy sur- face, four of which were omitted in the previous research. The periodic Lyapunov orbits and their invariant manifolds near the hyperbolic equilibria are presented, and an iteration procedure for identifying Lyapunov orbit is proposed based on the differential correction algorithm. The non-transversal intersections between invariant manifolds are addressed to generate homoclinic and heteroclinic trajectories between the Lyapunov orbits. (3,3)- and (2,1)-heteroclinic trajecto- ries from the neighborhood of one collinear equilibrium to that of another one, and (3,6)- and (2,1)-homoclinic trajecto- ries from and to the neighborhood of the same equilibrium, are obtained based on the Poincar6 mapping technique. 展开更多

RAPD (randomly amplified polymorphic DNA) markers were employed to characterize polymorphisms among 5 provenances of Acacia leucophloea and to detect genetic relatedness of the species with 6 other acacias (A. holosericea, A. auriculiformis, A. mangium, A. dealbata, A. ferruginea, and A. nilotica) widely grown in India. Of 194 markers scored for the provenances, 29.38% exhibited polymorphism. Also, 326 markers were generated among 7 species of Acacia, accounting for 55.82% of the polymorphisms. The fifteen 10-mer primers employed were capable of producing 1-8 polymorphic bands for the provenances, and 6-17 for all seven species of Acacia. The genetic similarity coefficient based on Jaccard' s coefficient revealed that provenances Thirumangalam and Dharmapuri were closely related. The dendrogram based on a sequential agglomerative hierarchical non-overlapping (SAHN) clustering analysis grouped 4 provenances of A. leucophloea (Dharapuram, Thirumangalam, Pudukottai and Dharmapuri) into one cluster and the other provenance, Sendurai, into a separate cluster. The genetic similarity matrix for 7 Acacia species showed that A. nilotica and A. dealbata were distantly related, while A. holosericea and A. ferruginea were very closely related. Cluster analysis grouped the species of Acacias into 3 major groups of which A. dealbata alone formed a separate group. The RAPD markers generated 36 provenance-specific markers and 162 species-specific markers that could have strong applications for species identification and tree breeding programs for A. leucophloea and for other Acacia species included in this study. 展开更多

Purpose–This research aims to improve the performance of rail fastener defect inspection method for multi railways,to effectively ensure the safety of railway operation.Design/methodology/approach–Firstly,a fastener region location method based on online learning strategy was proposed,which can locate fastener regions according to the prior knowledge of track image and template matching method.Online learning strategy is used to update the template library dynamically,so that the method not only can locate fastener regions in the track images of multi railways,but also can automatically collect and annotate fastener samples.Secondly,a fastener defect recognition method based on deep convolutional neural network was proposed.The structure of recognition network was designed according to the smaller size and the relatively single content of the fastener region.The data augmentation method based on the sample random sorting strategy is adopted to reduce the impact of the imbalance of sample size on recognition performance.Findings–Test verification of the proposed method is conducted based on the rail fastener datasets of multi railways.Specifically,fastener location module has achieved an average detection rate of 99.36%,and fastener defect recognition module has achieved an average precision of 96.82%.Originality/value–The proposed method can accurately locate fastener regions and identify fastener defect in the track images of different railways,which has high reliability and strong adaptability to multi railways. 展开更多

The method CPCM1 combineschemical stabilization and vacuum preloading (VP), while CPCM2 is similar to CPCM1 but includes boththe application of surcharge and use of geo-bags to provide confinement during surcharge preloading.The key advantage of CPCM2 using geo-bags is that the surcharge can be immediately applied on thechemically stabilized slurries. Two types of geo-bags were investigated under simulated land filling anddyke conditions, respectively. The test results show that the shear strength (cu) of treated slurry byCPCM2 is generally much higher than that by CPCM1. Besides, the use of CPCM2 can significantly reducethe treatment time due to the short drainage paths created by geo-bags. Overall, CPCM2 allows fasterconsolidation and higher preloading that help to achieve higher mechanical properties of the stabilizedslurry.
