Here,we aimed to study the changes in proteome of golden pompano fillets during post-mortem storage.Tandem mass tags(TMT)-labeled quantitative proteomic strategy was applied to investigate the relationships between protein changes and quality characteristics of modified atmosphere packaging(MAP)fillets during superchilling(-3°C)storage.Scanning electron microscopy was used to show that the muscle histology microstructure of fillets was damaged to varying degrees,and low-field nuclear magnetic resonance was used to find that the immobilized water and free water in the muscle of fillets changed significantly.Total sulfhydryl content,TCA-soluble peptides and Ca2+-ATPase activity also showed that the fillet protein had a deterioration by oxidation and denaturation.The Fresh(FS),MAP,and air packaging(AP)groups were set.Total of 150 proteins were identified as differential abundant proteins(DAPs)in MAP/FS,while 209 DAPs were in AP/FS group.The KEGG pathway analysis indicated that most DAPs were involved in binding proteins and protein turnover.Correlation analysis found that 52 DAPs were correlated with quality traits.Among them,8 highly correlated DAPs are expected to be used as potential quality markers for protein oxidation and water-holding capacity.These results provide a further understanding of the muscle deterioration mechanism of packaging golden pompano fillets during superchilling. 展开更多

This study provides a comprehensive analysis of the concepts and assessment processes of water resources in China,focusing on the characteristics of water resources and variations in water cycle fluxes.It reveals that the distribution of water resources in China is uneven,with more south and less north,and human activities have led to a decline in water resources,particularly in northern arid and semi-arid regions.Further analysis shows that duplicated measurements of surface water and groundwater significantly affect water balance calculation and water resource assessments,serving as a crucial factor guiding water resource development and utilization.The study also finds that consistency correction of runoff series is insufficient to meet the requirements of accurate water resource assessment.It is urgent to strengthen fundamental research in hydrology and hydrogeology,and to establish a dynamic assessment system for the efficient management and rational use of surface water and groundwater in the current changing environment. 展开更多

The potential of cooling slope casting process to produce EN AW 6082 forging stock for the manufacture of EN AW 6082 suspension components was investigated.EN AW 6082 billets cast over a cooling plate offer a fine uniform structure that can be forged even without a separate homogenization treatment.This is made it possible by the limited superheat of the melt at the start of casting and the fractional solidification that occurs already on the cooling plate.Suspension parts forged from cast and homogenized billets with or without Cr all showed a umform structure,and the hardness reached HV 110 after the standard artificial ageing treatment. 展开更多

Olivine lithium iron phosphate(Li Fe PO4) is considered as a promising cathode material for high power density lithium ion battery due to its high capacity, long cycle life, environmental friendly, low cost, and safety consideration. The theoretical capacity of Li Fe PO4 based on one electron reaction is 170 m Ah g-1at the stable voltage plateau of 3.5 V vs. Li/Li+. However, the instinct drawbacks of olivine structure induce a poor rate performance, resulting from the low lithium ion diffusion rate and low electronic conductivity.In this review, we summarize the methods for enhancing the rate performance of Li Fe PO4 cathode materials,including carbon coating, elements doping, preparation of nanosized materials, porous materials and composites,etc. Meanwhile, the advantages and disadvantages of above methods are also discussed. 展开更多

Pantoprazole sodium, a substituted benzimidazole derivative, is an irreversible proton pump inhibitor which is primarily used for the treatment of duodenal ulcers, gastric ulcers, and gastroesophageal reflux disease (GERD). The monographs of European Pharmacopoeia (Ph. Eur.) and United States Pharmacopoeia (USP) specify six impurities, viz.;impurities A, B, C, D, E and F, respectively for its active pharmaceutical ingredient (API). The identification and synthesis of all impurities except impurity E are well described in the literature;however, there is no report related to impurity E. The prospects to the formation and controlling of impurity E up to -0.03% in the synthesis of pantoprazole sodium sesquihydrate (PAN) were discussed in detail for the first time. The present work described the journey towards the successful development of an optimal preparation procedure of dimer impurity E. Coffee is commonly consumed beverage in the world and it has been suggested to have beneficial effect.Chlorogenic acids(CGAs)are main ingredient of coffee beans which has been extensively used in nutraceuticals and medicine.Recently,various therapeutic effects of chlorogenic acids have been investigated.However,there are limited studies to investigate its anticancer properties.In the present study,we have used chlorogenic acid complex(CGA7)a decaffeinated water soluble green coffee bean extract to evaluate its cytotoxic effect on human and mouse cancer cell lines by using different approaches.From our results we found CGA7 treatment induces cell death in a dose and time dependent manner in different cancer cell lines.Further,CGA7 induced apoptosis was characterized by DNA fragmentation,PARP-1 cleavage,caspase-9 activation,and down regulation of Bcl-2,an anti-apoptotic protein and up regulation of pro-apoptotic protein BAX.Overall findings indicated that CGA7 complex a potent anticancer molecule found in green coffee beans could be a safe bioactive ingredient for prevention of cancer. 展开更多

Mg-9Al-xPr(x=0.4,0.8 and 1.2,mass fraction,%)magnesium alloys were prepared by high-pressure die-casting technique.The effects of Pr on the microstructures of die-cast Mg-9Al based alloy were investigated by XRD and SEM.Needle-like Al11Pr3 phase and polygon Al6Mn6Pr phase are found in the microstructure.With 0.4%Pr addition,fine needle-like Al11Pr3 phase and a small amount of polygon Al6Mn6Pr phase near the grain boundary are found in the microstructure.Increasing Pr addition to 0.8%, lots of coarse needle-like Al11Pr3 phase within grain and polygon Al6Mn6Pr phase on grain boundary are observed.Further increasing Pr addition,the size of needle-like Al11Pr3 phase decreases,while the size of polygon Al6Mn6Pr relatively increases.The mass fraction of Pr at around 0.8%is considered to be suitable to obtain the optimal mechanical properties.The optimal mechanical properties are mainly resulted from grain boundary strengthening obtained by precipitates and solid solution. 展开更多

A new coordination polymer,Zn(bpy)L(BUC‐21),(H2L=cis‐1,3‐dibenzyl‐2‐imidazolidone‐4,5‐dicarboxylic acid,bpy=4,4′‐bipyridine),has been synthesized under hydrothermal conditions,and characterized by single‐crystal X‐ray analysis,Fourier transform infrared spectroscopy,thermogravimetric analyses,CNH elemental analysis and UV‐Vis diffuse reflectance spectroscopy.BUC‐21exhibited an excellent performance for photocatalytic Cr(VI)reduction with a conversion efficiency of96%,better than that of commercial P25(39%),under UV light irradiation for30min.BUC‐21could also be used to conduct photocatalytic degradation of organic dyes including methylene blue,rhodamine B,methyl orange and reactive red X‐3B.Also,the photocatalytic activity of BUC‐21remained high across a wide pH range from2.0to12.0.It is interesting to note,however,that BUC‐21was unable to achieve simultaneous reduction of Cr(VI)and degradation of an organic pollutant in a mixed matrix,which can be attributed to the competition between Cr(VI)and the organic dyes for access to the photo‐excited electrons. 展开更多

The structure of unknown impurity was postulated based on liquid chromatography mass spectrometry using electrospray ionization and ion trap analyzer (LC/ESI-IT/MS) analysis. Proposed structure of impurity was unambiguously confirmed by synthesis followed by characterization using 1H, 13C nuclear magnetic resonance spectrometry (NMR), 1H-1H correlation spectro-scopy (COSY) and infrared spectroscopy (IR). Based on the spectroscopic and spectrometric data, unknown impurity was characterized as 5-carbamoyl-10,11-dihydro-5H-dibenzo[b,f]azepin-10-yl propionate. 展开更多

Subarachnoid hemorrhage is a devastating disease with significant mortality and morbidity,despite advances in treating cerebral aneurysms.There has been recent progress in the intensive care management and monitoring of patients with subarachnoid hemorrhage,but the results remain unsatisfactory.Microglia,the resident immune cells of the brain,are increasingly recognized as playing a significant role in neurological diseases,including subarachnoid hemorrhage.In early brain injury following subarachnoid hemorrhage,microglial activation and neuroinflammation have been implicated in the development of disease complications and recovery.To understand the disease processes following subarachnoid hemorrhage,it is important to focus on the modulators of microglial activation and the pro-inflammatory/anti-inflammatory cytokines and chemokines.In this review,we summarize research on the modulators of microglia-mediated inflammation in subarachnoid hemorrhage,including transcriptome changes and the neuroinflammatory signaling pathways.We also describe the latest developments in single-cell transcriptomics for microglia and summarize advances that have been made in the transcriptome-based classification of microglia and the implications for microglial activation and neuroinflammation. 展开更多

The objective of the study is to investigate the suitability of using Pulse-coherent Acoustic Doppler Profiler (PCADP) to estimate suspended sediment concentration (SSC). The acoustic backscatter intensity was corrected for spreading and absorption loss, then calibrated with OBS and finally converted to SSC. The results show that there is a good correlation between SSC and backscatter intensity with R value of 0.74. The mean relative error is 22.4%. Then the time span of little particle size variation was also analyzed to exclude the influence of size variation. The correlation coefficient increased to 0.81 and the error decreased to 18.9%. Our results suggest that the PCADP can meet the requirement of other professional instruments to estimate SSC with the errors between 20% and 50%, and can satisfy the need of dynamics study of suspended particles. 展开更多

In this paper, the effectiveness, applicability and validity of chemicalephysical combined methods(CPCMs) for treatment of marine clay (MC) slurries were evaluated. The method CPCM1 combineschemical stabilization and vacuum preloading (VP), while CPCM2 is similar to CPCM1 but includes boththe application of surcharge and use of geo-bags to provide confinement during surcharge preloading.The key advantage of CPCM2 using geo-bags is that the surcharge can be immediately applied on thechemically stabilized slurries. Two types of geo-bags were investigated under simulated land filling anddyke conditions, respectively. The test results show that the shear strength (cu) of treated slurry byCPCM2 is generally much higher than that by CPCM1. Besides, the use of CPCM2 can significantly reducethe treatment time due to the short drainage paths created by geo-bags. Overall, CPCM2 allows fasterconsolidation and higher preloading that help to achieve higher mechanical properties of the stabilizedslurry. Lotus(Nelumbo nucifera),an ancient aquatic plant,possesses a unique pharmacological activity that is primarily contributed by benzylisoquinoline alkaloids(BIAs).However,only few genes and enzymes involved in BIA biosynthesis in N.nucifera have been isolated and characterized.In the present study we identified the regiopromiscuity of an O-methyltransferase,designated NnOMT6,isolated from N.nucifera;NnOMT6 was found to catalyze the methylation of monobenzylisoquinoline 6-O/7-O,aporphine skeleton 6-O,phenylpropanoid 3-O,and protoberberine 2-O.We further probed the key residues affecting NnOMT6 activity via molecular docking and molecular dynamics simulation.Verification using site-directed mutagenesis revealed that residues D316,N130,L135,N176A,D269,and E328 were critical for BIA O-methyltransferase activities;furthermore,N323A,a mutant of NnOMT6,demonstrated a substantial increase in catalytic efficiency for BIAs and a broader acceptor scope compared with wild-type NnOMT6.To the best of our knowledge,this is the first study to report the O-methyltransferase activity of an aporphine skeleton without benzyl moiety substitutions in N.nucifera.The study findings provide biocatalysts for the semisynthesis of related medical compounds and give insights into protein engineering to strengthen O-methyltransferase activity in plants. 展开更多

The spread of SARS-CoV-2 as an emerging novel coronavirus disease (COVID-19) had progressed as a worldwide pandemic since the end of 2019. COVID-19 affects firstly lungs tissues which are known for their very slow regeneration. Afterwards, enormous cytokine stimulation occurs in the infected cells immediately after a lung infection which necessitates good management to save patients. Exosomes are extracellular vesicles of nanometric size released by reticulocytes on maturation and are known to mediate intercellular communications. The exosomal cargo serves as biomarkers in diagnosing various diseases;moreover, exosomes could be employed as nanocarriers in drug delivery systems. Exosomes look promising to combat the current pandemic since they contribute to the immune response against several viral pathogens. Many studies have proved the potential of using exosomes either as viral elements or host systems that acquire immune-stimulatory effects and could be used as a vaccine or drug delivery tool. It is essential to stop viral replication, prevent and reverse the massive storm of cytokine that worsens the infected patients’ situations for the management of COVID-19. The main benefits of exosomes could be;no cells will be introduced, no chance of mutation, lack of immunogenicity and the damaged genetic material that could negatively affect the recipient is avoided. Additionally, it was found that exosomes are static with no ability for in vivo reproduction. The current review article discusses the possibilities of using exosomes for detecting novel coronavirus and summarizes state of the art concerning the clinical trials initiated for examining the use of COVID-19 specific T cells derived exosomes and mesenchymal stem cells derived exosomes in managing COVID-19. 展开更多

Mitochondria are crucial for the production of primary and secondary metabolites,which largely determine the quality of fruit.However,a method for isolating high-quality mitochondria is currently not available in citrus fruit,preventing high-throughput characterization of mitochondrial functions.Here,based on differential and discontinuous Percoll density gradient centrifugation,we devised a universal protocol for isolating mitochondria from the pulp of four major citrus species,including satsuma mandarin,ponkan mandarin,sweet orange,and pummelo.Western blot analysis and microscopy confirmed the high purity and intactness of the isolated mitochondria.By using this protocol coupled with a label-free proteomic approach,a total of 3353 nonredundant proteins were identified.Comparison of the four mitochondrial proteomes revealed that the proteins commonly detected in all proteomes participate in several typical metabolic pathways(such as tricarboxylic acid cycle,pyruvate metabolism,and oxidative phosphorylation)and pathways closely related to fruit quality(such asγ-aminobutyric acid(GABA)shunt,ascorbate metabolism,and biosynthesis of secondary metabolites).In addition,differentially abundant proteins(DAPs)between different types of species were also identified;these were found to be mainly involved in fatty acid and amino acid metabolism and were further confirmed to be localized to the mitochondria by subcellular localization analysis.In summary,the proposed protocol for the isolation of highly pure mitochondria from different citrus fruits may be used to obtain high-coverage mitochondrial proteomes,which can help to establish the association between mitochondrial metabolism and fruit storability or quality characteristics of different species and lay the foundation for discovering novel functions of mitochondria in plants. 展开更多

This paper considers nonlinear dynamics of teth- ered three-body formation system with their centre of mass staying on a circular orbit around the Earth, and applies the theory of space manifold dynamics to deal with the nonlinear dynamical behaviors of the equilibrium configurations of the system. Compared with the classical circular restricted three body system, sixteen equilibrium configurations are obtained globally from the geometry of pseudo-potential energy sur- face, four of which were omitted in the previous research. The periodic Lyapunov orbits and their invariant manifolds near the hyperbolic equilibria are presented, and an iteration procedure for identifying Lyapunov orbit is proposed based on the differential correction algorithm. The non-transversal intersections between invariant manifolds are addressed to generate homoclinic and heteroclinic trajectories between the Lyapunov orbits. (3,3)- and (2,1)-heteroclinic trajecto- ries from the neighborhood of one collinear equilibrium to that of another one, and (3,6)- and (2,1)-homoclinic trajecto- ries from and to the neighborhood of the same equilibrium, are obtained based on the Poincar6 mapping technique. 展开更多

The Upper Ordovician Wufeng-Lower Silurian Longmaxi and the Lower Cambrian Qiongzhusi shales are the major targets for shale gas exploration and development in China.Although the two organic-rich shales share similar distribution ranges and thicknesses,they exhibit substantially different exploration and development results.This work analyzed the nanopore structures of the shale reservoirs in this region.Pore development of 51 shale samples collected from various formations and locations was compared using the petromineralogical,geochemical,structural geological and reservoir geological methods.The results indicate that the reservoir space in these shales is dominated by organic pores and the total pore volume of micropores,mesopores,macropores in different tectonic areas and formations show different trends with the increase of TOC.It is suggested that organic pores of shale can be well preserved in areas with simple structure and suitable preservation conditions,and the shale with smaller maximum ancient burial depth and later hydrocarbongeneration-end-time is also more conducive to pore preservation.Organic pore evolution models are established,and they are as follows:①Organic matter pore development stage,②Early stage of organic matter pore destruction,and③late stage of organic matter pore destruction.The areas conducive to pore development are favorable for shale gas development.Research results can effectively guide the optimization and evaluation of favorable areas of shale gas. 展开更多

Experimental investigation was conducted to study the flow regimes and discharge characteristics of plasma electrohydrodynamic atomization (EHDA) for decane (CloH22) under pulsed applied negative voltage. The experimental parameters were set as the flow rate of decane from 0 mL/min to 10 mL/min and the DC charging voltage from DC 0 V to 12 V with a pulse repetition rates of 200 Hz. The flow regime of decane was observed and the volume-to-electrical charge ratio was measured. Unlike a conventional EHDA system, the results show that a corona discharge was initiated at the edge of the hollow electrode at a specific corona on-set voltage of -17 kV or -20 kV in the case with or without decane flow, respectively. Although multiple microscopic techniques have been applied to horticultural research,few studies of individual organelles in living fruit cells have been reported to date.In this paper,we established an efficient system for the transient transformation of citrus fruits using an Agrobacterium-mediated method.Kumquat(Fortunella crassifolia Swingle)was used;it exhibits higher transformation efficiency than all citrus fruits that have been tested and a prolonged-expression window.Fruits were transformed with fluorescent reporters,and confocal microscopy and live-cell imaging were used to study their localization and dynamics.Moreover,various pH sensors targeting different subcellular compartments were expressed,and the local pH environments in cells from different plant tissues were compared.The results indicated that vacuoles are most likely the main organelles that contribute to the low pH of citrus fruits.In summary,our method is effective for studying various membrane trafficking events,protein localization,and cell physiology in fruit and can provide new insight into fruit biology research. 展开更多

Recovery of alginate extracted from aerobic granular sludge(AGS)has given rise to a novel research direction.However,these extracted alginate solutions have a water content of nearly 100%.Alternately,ultrafiltration(UF)is generally used for concentration of polymers.Furthermore,the introduction of multivalent metal ions into alginate may provide a promising method for the development of novel nanomaterials.In this study,membrane fouling mitigation by multivalent metal ions,both individually and in combination,and properties of recycled materials were investigated for UF recovery of sodium alginate(SA).The filtration resistance showed a significantly negative correlation with the concentration of metal ions,arranged in the order of Mg^2+<Ca^2+<Fe^3+<Al^3+(filtration resistance mitigation),and the moisture content of recycled filter cake showed a marked decrease.For Ca^2+,Mg^2+,Fe^3+,and Ca^2++Fe^3+,the filtration resistances were almost the same when the total charge concentration was less than 5 mmol·L^–1.However,when the total charge concentration was greater than 5 mmol·L^–1,membrane fouling mitigation increased significantly in the presence of Ca^2+or Fe^3+and remained constant for Mg^2+with the increase of total charge concentration.The filtration resistance mitigation was arranged in the order of Fe^3+>Fe^3++Ca^2+>Ca^2+>Mg^2+.Three mechanisms were proposed in the presence of Fe^3+,such as the decrease of SA concentration,change in p H,and production of hydroxide iron colloids from hydrolysis.The properties of recycled materials(filter cake)were investigated via optical microscope observation,dynamic light scattering,Fourier transform infrared,X-ray photoelectron spectroscopy(XPS),and scanning electron microscopy.The results provide further insight into UF recoveries of alginate extracted from AGS. 展开更多
