Due to the expected rise in the world population,an increase in the requirements for quality and safety of food and feed is expected,which leads to the growing demand for new sources of sustainable and renewable protein.Insect protein is gaining importance as a renewable material for several reasons,reflecting its potential contribu-tions to sustainability,resource efficiency,and environmental conservation.Some insect species are known to be able to efficiently convert organic waste into high-value products such as protein,requiring less land and water compared to traditional livestock.In addition,insect farming produces fewer greenhouse gas emissions,contri-buting to mitigating climate change.Insects are considered as a major potential alternative to animal or plant protein due to their many nutritional benefits,including high protein,mineral,and vitamin contents.On average,the protein content of insects ranges between 35%and 60%dry weight,which exceeds plant protein sources,such as cereal,soybeans,and lentils.As the acceptance of insect protein grows and technologies advance,the food and feed industries continue to explore and expand their applications,offering consumers diverse and sustainable pro-tein choices.In this review,we discuss the recentfindings relating to insect protein focusing on its characteristics,extraction methods,applications,and opportunities along with some trade-offs and uncertainties. 展开更多

Down Regulation of Survivin Gene and Up Regulation of p53 Gene expression by siRNA Induces Apoptosis in human Hepatocellular Carcinoma cell Line HepG2

Survivin gene may be a good target for cancer gene therapy because it is over expressed in a variety of human tumors including human hepatocellular carcinoma but not in differen- tiated adult tissues. To explore the effects of the siRNA of survivin gene inducing apoptosis in human hepatocellular cancer cells, three siRNAs cpusiRNA1, cpusiRNA2 and cpusiRNA3 were designed and transferred into human hepatocellular carcinoma cell line HepG2 (HepG2) by lipofection. MTT test showed that the growth of HepG2 decreased when it was transfected with 25nM, 50nM, 100nM, 150nM, 200nM, 400nM siRNA respectively after 48 hours. And the change of mRNA and protein of survivin gene and p53 gene had been detected by RT-PCR and Western blot. Cells presented an increase in apoptosis index was assayed by flow cytometry. Small interfering RNA can exert a knockdown of survivin gene expression and up regulation of p53 gene to induce apoptosis and to inhibit the growth of HepG2. 展开更多

The lateral flow immuno assay (LFIA) has emerged as a powerful tool for rapid scree ning owing to its simplicity and flexibility for detection of various biomarkers. However, conventional LFIA strips have several disadvantages, including limits in quantitative analysis and low sensitivity. Here we developed a novel surface-e nhanced Rama n scatteri ng LFIA based on non spherical gap-e nhanced Raman tags (GERTs), with Raman molecules (RMs) embedded in a 1-nm gap between Au nanorod core and Au shell. Such tags have a strong and uniform surface-enhanced Rama n scattering (SERS) resp on se, an order of mag nitude higher than that of other comm on SERS tags such as Au nano rods, nano stars, Au nano shells with surface-adsorbed RMs, or spherical GERTs with embedded RMs. The feasibility of the tags was dem on strated by the semiqua ntitative and sen sitive detecti on of the heart disease biomarker cardiac tropo nin I (cTnI). GERTs were conjugated with mono clonal antibodies and used for LFIA in the same way as ordinary functionalized colloidal gold. The presenee of the target antigen, cTnI, was identified by Raman microscopy mapping of the test zone. With the SERS-based LFIA, the limit of cTnI detection was about 0.1 ng/mL. This value is within the diagnostic range of cTnI in the blood serum of patients with heart infarction and is 30 times lower than that of the colorimetric LFIA test using the same antibodies and either GERTs or colloidal gold as labels. 展开更多
