文摘Hepatocyte nuclear factor 1 alpha(HNF1A),hepatocyte nuclear factor 4 alpha(HNF4A),and forkhead box protein A2(FOXA2)are key transcription factors that regulate a complex gene network in the liver,cre-ating a regulatory transcriptional loop. The Encode and ChIP-Atlas databases identify the recognition sites of these transcription factors in many glycosyltransferase genes. Our in silico analysis of HNF1A,HNF4A.and FOXA2 binding to the ten candidate glyco-genes studied in this work confirms a significant enrich-ment of these transcription factors specifically in the liver. Our previous studies identified HNF1A as a master regulator of fucosylation, glycan branching, and galactosylation of plasma glycoproteins. Here,we aimed to functionally validate the role of the three transcription factors on downstream glyco-gene transcriptional expression and the possible effect on glycan phenotype. We used the state-of-the-art clus-tered regularly interspaced short palindromic repeats/dead Cas9(CRISPR/dCas9)molecular tool for the downregulation of the HNF1A,HNF4A,and FOXA2 genes in HepG2 cells-a human liver cancer cell line. The results show that the downregulation of all three genes individually and in pairs affects the transcrip-tional activity of many glyco-genes,although downregulation of glyco-genes was not always followed by an unambiguous change in the corresponding glycan structures. The effect is better seen as an overall change in the total HepG2 N-glycome,primarily due to the extension of biantennary glycans. We propose an alternative way to evaluate the N-glycome composition via estimating the overall complexity of the glycome by quantifying the number of monomers in each glycan structure. We also propose a model showing feedback loops with the mutual activation of HNF1A-FOXA2 and HNF4A-FOXA2 affecting glyco-genes and protein glycosylation in HepG2 cells.

文摘This article presents an 8-element dual-polarized phased-array transceiver(TRX)front-end IC for millimeter-wave(mm-Wave)5G new radio(NR).Power enhancement technologies for power amplifiers(PA)in mm-Wave 5G phased-array TRX are discussed.A four-stage wideband high-power class-AB PA with distributed-active-transformer(DAT)power combining and multi-stage second-harmonic traps is proposed,ensuring the mitigated amplitude-to-phase(AM-PM)distortions across wide carrier frequencies without degrading transmitting(TX)power,gain and efficiency.TX and receiving(RX)switching is achieved by a matching network co-designed on-chip T/R switch.In each TRX element,6-bit 360°phase shifting and 6-bit 31.5-dB gain tuning are respectively achieved by the digital-controlled vector-modulated phase shifter(VMPS)and differential attenuator(ATT).Fabricated in 65-nm bulk complementary metal oxide semiconductor(CMOS),the proposed TRX demonstrates the measured peak TX/RX gains of 25.5/21.3 dB,covering the 24−29.5 GHz band.The measured peak TX OP1dB and power-added efficiency(PAE)are 20.8 dBm and 21.1%,respectively.The measured minimum RX NF is 4.1 dB.The TRX achieves an output power of 11.0−12.4 dBm and error vector magnitude(EVM)of 5%with 400-MHz 5G NR FR2 OFDM 64-QAM signals across 24−29.5 GHz,covering 3GPP 5G NR FR2 operating bands of n257,n258,and n261.

文摘This paper investigates the low earth orbit(LEO)satellite-enabled coded compressed sensing(CCS)unsourced random access(URA)in orthogonal frequency division multiple access(OFDMA)framework, where a massive uniform planar array(UPA)is equipped on the satellite. In LEO satellite communications, unavoidable timing and frequency offsets cause phase shifts in the transmitted signals, substantially diminishing the decoding performance of current terrestrial CCS URA receiver. To cope with this issue,we expand the inner codebook with predefined timing and frequency offsets and formulate the inner decoding as a tractable compressed sensing(CS)problem.Additionally,we leverage the inherent sparsity of the UPA-equipped LEO satellite angular domain channels, thereby enabling the outer decoder to support more active devices.Furthermore, the outputs of the outer decoder are used to reduce the search space of the inner decoder, which cuts down the computational complexity and accelerates the convergence of the inner decoding.Simulation results verify the effectiveness of the proposed scheme.

文摘Roles of oligodendrocyte precursor cells in the central nervous system:Oligodendrocyte precursor cells(OPCs)have long been recognized for their critical role as precursors to oligodendrocytes, the primary myelin-producing cells. As precursors,OPCs mature and differentiate into oligodendrocytes, which contribute significantly to the formation of myelin sheaths around axons. This myelination, which is critical for the conduction of salutatory nerve impulses in the cerebral white matter, underscores the classical role of oligodendrocytes in central nervous system(CNS)functionality.Importantly, because oligodendrocytes are differentiated cells that cannot proliferate.

文摘Aqueous-phase reforming(APR)is an attractive process to produce bio-based hydrogen from waste biomass streams, during which the catalyst stability is often challenged due to the harsh reaction conditions. In this work, three Pt-based catalysts supported on C,AlO(OH),and ZrO_(2)were investigated for the APR of hydroxyacetone solution in afixed bed reactor at 225℃and 35 bar.Among them, the Pt/C catalyst showed the highest turnover frequency for H_(2)production(TOF of 8.9 molH_(2)molPt^(-1)min^(-1))and the longest catalyst stability.Over the AlO(OH)and ZrO_(2)supported Pt catalysts, the side reactions consuming H_(2),formation of coke, and Pt sintering result in a low H_(2)production and the fast catalyst deactivation. The proposed reaction pathways suggest that a promising APR catalyst should reform all oxygenates in the aqueous phase, minimize the hydrogenation of the oxygenates, maximize the WGS reaction, and inhibit the condensation and coking reactions for maximizing the hydrogen yield and a stable catalytic performance.

文摘In this paper,a statistical cluster-based simulation channel model with a finite number of sinusoids is proposed for depicting the multiple-input multiple-output(MIMO)communications in vehicleto-everything(V2X)environments.In the proposed sum-of-sinusoids(SoS)channel model, the waves that emerge from the transmitter undergo line-of-sight(LoS)and non-line-of-sight(NLoS)propagation to the receiver, which makes the model suitable for describing numerous V2X wireless communication scenarios for sixth-generation(6G).We derive expressions for the real and imaginary parts of the complex channel impulse response(CIR),which characterize the physical propagation characteristics of V2X wireless channels. The statistical properties of the real and imaginary parts of the complex CIRs,i.e.,autocorrelation functions(ACFs),Doppler power spectral densities(PSDs),cross-correlation functions(CCFs),and variances of ACFs and CCFs,are derived and discussed.Simulation results are generated and match those predicted by the underlying theory, demonstrating the accuracy of our derivation and analysis. The proposed framework and underlying theory arise as an efficient tool to investigate the statistical properties of 6G MIMO V2X communication systems.

文摘BACKGROUND Early diagnosis of pancreatic ductal adenocarcinoma(PDAC)has been a longstanding challenge. The prognosis of patients with PDAC depends on the stage at diagnosis. It is necessary to identify biomarkers for the detection and differentiation of pancreatic tumors and optimize PDAC sample preparation procedures for DNA and RNA analysis. Most molecular studies are done using paraffin-embedded blocks;however, the integrity of DNA and RNA is often compromised in this format. Moreover,RNA isolated from human pancreatic tissue samples is generally of low quality,in part, because of the high concentration of endogenous pancreatic RNAse activity present.AIM To assess the potential of endoscopic ultrasound-guided fine-needle aspiration(EUS-FNA)to obtain specimens from pancreatic neoplasms for subsequent RNA molecular profiling, including next-generation sequencing(NGS).METHODS Thirty-four EUS-FNA samples were included in this study:PDAC(n=15),chronic pancreatitis(n=5),pancreatic cysts(n=14),mucinous cysts(mucinous cystic neoplasia/intraductal papillary mucinous neoplasia)n=7,serous cystic neoplasms n=5,and pseudocysts n=2.Cyst material consisted of cyst fluid and cyst wall samples obtained by through-the-needle biopsy(TTNB).Samples were stored at -80℃ until analysis.RNA purity(A260/230,A260/280 ratios),concentration, and integrity(RIN)were assessed.Real-time polymerase chain reaction was conducted on all samples, and small RNA libraries were prepared from solid mass samples.RESULTS RNA was successfully extracted from 29/34(85%)EUS-FNA samples:100% pancreatic adenocarcinoma samples,100% chronic pancreatitis samples,70% pancreatic fluid cyst samples, and 50%TTNB samples. The relative expression of GAPDH and HPRT were obtained for all successfully extracted RNA samples(n=29)including lowquality RNA specimens.Low concentration and nonoptimal RIN values(no less than 3)of RNA extracted from EUS-FNA samples did not prevent NGS library preparation. The suitability of cyst fluid samples for RNA profiling varied. The quality of RNA extracted from mucinous cyst fluid had a median RIN of 7.7(5.0-8.2),which was compatible with that from solid neoplasms[6.2(0-7.8)],whereas the quality of the RNA extracted from all fluids of serous cystic neoplasms and TTNB samples had a RIN of 0.CONCLUSION The results demonstrate the high potential of EUS-FNA material for RNA profiling of various pancreatic lesions, including low-quality RNA specimens.

文摘An externally generated resonant magnetic perturbation can induce complex non-ideal MHD responses in their resonant surfaces. We have studied the plasma responses using Fitzpatrick's improved two-fluid model and program LAYER.We calculated the error field penetration threshold for J-TEXT.In addition,we find that the island width increases slightly as the error field amplitude increases when the error field amplitude is below the critical penetration value. However, the island width suddenly jumps to a large value because the shielding effect of the plasma against the error field disappears after the penetration.By scanning the natural mode frequency,we find that the shielding effect of the plasma decreases as the natural mode frequency decreases. Finally,we obtain the m/n=2/1 penetration threshold scaling on density and temperature.
文摘The Advanced Glycation End Products(AGE)binding with its receptor can increase reactive oxygen species(ROS)generation through specific signaling mediators. The effect of superoxide(O2-)and O2-mediated ROS and reactive nitrogen species depends on their concentration and location of formation.Nitric oxide(NO)has anti-inflammatory and anticoagulant properties and a vasodilation effect, but NO can be deactivated by reacting with O_(2)^(-).This reaction between NO and O2-produces the potent oxidant ONOO−.Therefore,ONOO-'s regulatory role in AGEs in diabetic cardiovascular complications must considered as a regulator of cardiovascular complications in diabetes.

文摘Manipulating strain mode and degree that can be applied to epitaxial complex oxide thin films have been a cornerstone of strain engineering. In recent years,lift-off and transfer technology of the epitaxial oxide thin films have been developed that enabled the integration of heterostructures without the limitation of material types and crystal orientations. Moreover, twisted integration would provide a more interesting strategy in artificial magnetoelectric heterostructures.A specific twist angle between the ferroelectric and ferromagnetic oxide layers corresponds to the distinct strain regulation modes in the magnetoelectric coupling process, which could provide some insight in to the physical phenomena. In this work, the La_(0.67)Sr_(0.33)MnO_(3)(001)/0.7Pb(Mg_(1/3)Nb_(2/3))O_(3)-0.3PbTiO_(3)(011)(LSMO/PMN-PT)heterostructures with 45.and 0.twist angles were assembled via water-etching and transfer process. The transferred LSMO films exhibit a fourfold magnetic anisotropy with easy axis along LSMO<110>.A coexistence of uniaxial and fourfold magnetic anisotropy with LSMO[110]easy axis is observed for the 45°Sample by applying a 7.2 kV cm^(−1)electrical field, significantly different from a uniaxial anisotropy with LSMO[100]easy axis for the 0°Sample.The fitting of the ferromagnetic resonance field reveals that the strain coupling generated by the 45°twist angle causes different lattice distortion of LSMO,thereby enhancing both the fourfold and uniaxial anisotropy. This work confirms the twisting degrees of freedom for magnetoelectric coupling and opens opportunities for fabricating artificial magnetoelectric heterostructures.

文摘Dear Editor,In this letter,a novel hierarchical fusion framework is proposed to address the imperfect data property in complex medical image analysis(MIA)scenes. In particular,by combining the strengths of convolutional neural networks(CNNs)and transformers, the enhanced feature extraction, spatial modeling, and sequential context learning are realized to provide comprehensive insights on the complex data patterns.Integration of information in different level is enabled via a multi-attention fusion mechanism, and the tensor decomposition methods are adopted so that compact and distinctive representation of the underlying and high-dimensional medical image features can be accomplished[1].It is shown from the evaluation results that the proposed framework is competitive and superior as compared with some other advanced algorithms, which effectively handles the imperfect property of inter-class similarity and intra-class differences in diseases, and meanwhile, the model complexity is reduced within an acceptable level, which benefits the deployment in clinic practice.MIA has assumed a pivotal role in numerous critical clinical scenarios, where sophisticated image analysis techniques have proven instrumental in augmenting medical decision-making,facilitating individualized therapeutic interventions, and enhancing patient prognostication[2]−[4].

文摘In this paper, the channel impulse response matrix(CIRM)can be expressed as a sum of couplings between the steering vectors at the base station(BS)and the eigenbases at the mobile station(MS).Nakagami distribution was used to describe the fading of the coupling between the steering vectors and the eigenbases.Extensive measurements were carried out to evaluate the performance of this proposed model.Furthermore, the physical implications of this model were illustrated and the capacities are analyzed. In addition, the azimuthal power spectrum(APS)of several models was analyzed. Finally, the channel hardening effect was simulated and discussed. Results showed that the proposed model provides a better fit to the measured results than the other CBSM,i.e.,Weichselberger model. Moreover, the proposed model can provide better tradeoff between accuracy and complexity in channel synthesis. This CIRM model can be used for massive MIMO design in the future communication system design.

文摘After reconstructing the anterior cruciate ligament(ACL),unsatisfactory bone tendon interface healing may often induce tunnel enlargement at the early healing stage. With good biological features and high formability,Magnesium-Zinc-Gadolinium(ZG21)wires are developed to bunch the tendon graft for matching the bone tunnel during transplantation.Microstructure, tensile strength, degradation, and cytotoxicity of ZG21 wire are evaluated. The rabbit model is used for assessing the biological effects of ZG21 wire by Micro-CT,histology,and mechanical test. The SEM/EDS,immunochemistry,and in vitro assessments are performed to investigate the underlying mechanism.Material tests demonstrate the high formability of ZG21 wire as surgical suture.Micro-CT shows ZG21 wire degradation accelerates tunnel bone formation, and histologically with earlier and more fibrocartilage regeneration at the healing interface. The mechanical test shows higher ultimate load in the ZG21 group. The SEM/EDS presents ZG21 wire degradation triggered calcium phosphate(Ca-P)deposition.IHC results demonstrate upregulation of Wnt3a,BMP2,and VEGF at the early phase and TGFβ3 and Type II collagen at the late phase of healing. In vitro tests also confirmed the Ca-P in the metal extract could elevate the expression of Wnt3a,βcatenin,ocn and opn to stimulate osteogenesis.Ex vivo tests of clinical samples indicated suturing with ZG21 wire did not weaken the ultimate loading of human tendon tissue. In conclusion, the ZG21 wire is feasible for tendon graft bunching. Its degradation products accelerated intra-tunnel endochondral ossification at the early healing stage and therefore enhanced bone-tendon interface healing in ACL reconstruction.
