Nevertheless, the precise molecular mechanisms by which basolateral amygdala astrocytes regulate chronic stress–induced anxiety-like behaviors remain to be fully elucidated. In this study, we found that in a mouse model of anxiety triggered by unpredictable chronic mild stress, the expression of excitatory amino acid transporter 2 was upregulated in the basolateral amygdala. Interestingly, our findings indicate that the targeted knockdown of excitatory amino acid transporter 2 specifically within the basolateral amygdala astrocytes was able to rescue the anxiety-like behavior in mice subjected to stress. Furthermore, we found that the overexpression of excitatory amino acid transporter 2 in the basolateral amygdala, whether achieved through intracranial administration of excitatory amino acid transporter 2agonists or through injection of excitatory amino acid transporter 2-overexpressing viruses with GfaABC1D promoters, evoked anxiety-like behavior in mice. Our single-nucleus RNA sequencing analysis further confirmed that chronic stress induced an upregulation of excitatory amino acid transporter 2 specifically in astrocytes in the basolateral amygdala. Moreover, through in vivo calcium signal recordings, we found that the frequency of calcium activity in the basolateral amygdala of mice subjected to chronic stress was higher compared with normal mice. After knocking down the expression of excitatory amino acid transporter 2 in the basolateral amygdala, the frequency of calcium activity was not significantly increased, and anxiety-like behavior was obviously mitigated. Additionally, administration of an excitatory amino acid transporter 2 inhibitor in the basolateral amygdala yielded a notable reduction in anxiety level among mice subjected to stress. These results suggest that basolateral amygdala astrocytic excitatory amino acid transporter 2 plays a role in in the regulation of unpredictable chronic mild stress-induced anxiety-like behavior by impacting the activity of local glutamatergic neurons, and targeting excitatory amino acid transporter 2 in the basolateral amygdala holds therapeutic promise for addressing anxiety disorders.展开更多

Although the vast majority of meta-analyses have confirmed that exercise interventions can increase brain-derived neurotrophic factor levels in children and adolescents, the effects of specific types of exercise on brain-derived neurotrophic factor levels are still controversial. To address this issue, we used meta-analytic methods to quantitatively evaluate, analyze, and integrate relevant studies. Our goals were to formulate general conclusions regarding the use of exercise interventions, explore the physiological mechanisms by which exercise improves brain health and cognitive ability in children and adolescents, and provide a reliable foundation for follow-up research. We used the Pub Med, Web of Science, Science Direct, Springer, Wiley Online Library, Weipu, Wanfang, and China National Knowledge Infrastructure databases to search for randomized controlled trials examining the influences of exercise interventions on brain-derived neurotrophic factor levels in children and adolescents. The extracted data were analyzed using Review Manager 5.3. According to the inclusion criteria, we assessed randomized controlled trials in which the samples were mainly children and adolescents, and the outcome indicators were measured before and after the intervention. We excluded animal experiments, studies that lacked a control group, and those that did not report quantitative results. The mean difference(MD;before versus after intervention) was used to evaluate the effect of exercise on brain-derived neurotrophic factor levels in children and adolescents. Overall, 531 participants(60 children and 471 adolescents, 10.9–16.1 years) were included from 13 randomized controlled trials. Heterogeneity was evaluated using the Q statistic and I^(2) test provided by Review Manager software. The meta-analysis showed that there was no heterogeneity among the studies(P = 0.67, I^(2) = 0.00%). The combined effect of the interventions was significant(MD = 2.88, 95% CI: 1.53–4.22, P < 0.0001), indicating that the brain-derived neurotrophic factor levels of the children and adolescents in the exercise group were significantly higher than those in the control group. In conclusion, different types of exercise interventions significantly increased brain-derived neurotrophic factor levels in children and adolescents. However, because of the small sample size of this meta-analysis, more high-quality research is needed to verify our conclusions. This metaanalysis was registered at PROSPERO(registration ID: CRD42023439408).展开更多

Due to the diffraction, however, the acquired nano-resolution phase signal may suffer splitting problem, which impedes the direct reconstruction of phase contrast CT(nPCT) images. To overcome, a new model-driven nPCT image reconstruction algorithm is developed in this study. In it, the diffraction procedure is mathematically modeled into a matrix B, from which the projections without signal splitting can be generated invertedly. Furthermore, a penalized weighted least-square model with total variation(PWLSTV) is employed to denoise these projections, from which nPCT images with high accuracy are directly reconstructed.Numerical experiments demonstrate that this new algorithm is able to work with phase projections having any splitting distances. Moreover, results also reveal that nPCT images of higher signal-to-noise-ratio(SNR) could be reconstructed from projections having larger splitting distances. In summary, a novel model-driven nPCT image reconstruction algorithm with high accuracy and robustness is verified for the Lau interferometer-based hard x-ray nano-resolution phase contrast imaging.展开更多

The hysteresis was reversed by reversing the current direction, which illustrates the spin–momentum locking feature of Cd_(3)As_(2). Furthermore, we realized the on–off states of the spin signals through electric modulation of the Fermi arc via the three-terminal configuration, which enables the great potential of Cd_(3)As_(2) in spin field-effect transistors.展开更多

文摘Painful stimuli elicit first-line reflexive defensive reactions and,in many cases, also evoke second-line recuperative behaviors, the latter of which reflects the sensing of tissue damage and the alleviation of suffering. The lateral parabrachial nucleus(lPBN),composed of external-(elPBN),dorsal-(dlPBN),and central/superior-subnuclei(jointly referred to as slPBN),receives sensory inputs from spinal projection neurons and plays important roles in processing affective information from external threats and body integrity disruption. However, the organizational rules of lPBN neurons that provoke diverse behaviors in response to different painful stimuli from cutaneous and deep tissues remain unclear. In this study,we used region-specific neuronal depletion or silencing approaches combined with a battery of behavioral assays to show that slPBN neurons expressing substance P receptor(NK1R)(lPBNNK1R)are crucial for driving pain-associated self-care behaviors evoked by sustained noxious thermal and mechanical stimuli applied to skin or bone/muscle,while elPBN neurons are dispensable for driving such reactions.Notably,lPBNNK1R neurons are specifically required for forming sustained somatic pain-induced negative teaching signals and aversive memory but are not necessary for fear-learning or escape behaviors elicited by external threats.Lastly, both lPBNNK1R and elPBN neurons contribute to chemical irritant-induced nocifensive reactions. Our results reveal the functional organization of parabrachial substrates that drive distinct behavioral outcomes in response to sustained pain versus external danger under physiological conditions.

文摘Hepatocellular carcinoma(HCC)is a prevalent and aggressive liver malignancy. The interplay between bile acids(BAs)and the gut microbiota has emerged as a critical factor in HCC development and progression.Under normal conditions,BA metabolism is tightly regulated through a bidirectional interplay between gut microorganisms and BAs.The gut microbiota plays a critical role in BA metabolism, and BAs are endogenous signaling molecules that help maintain liver and intestinal homeostasis.Of note, dysbiotic changes in the gut microbiota during pathogenesis and cancer development can disrupt BA homeostasis, thereby leading to liver inflammation and fibrosis, and ultimately contributing to HCC development. Therefore, understanding the intricate interplay between BAs and the gut microbiota is crucial for elucidating the mechanisms underlying hepatocarcinogenesis. In this review,we comprehensively explore the roles and functions of BA metabolism, with a focus on the interactions between BAs and gut microorganisms in HCC.Additionally, therapeutic strategies targeting BA metabolism and the gut microbiota are discussed, including the use of BA agonists/antagonists,probiotic/prebiotic and dietary interventions, fecal microbiota transplantation, and engineered bacteria. In summary, understanding the complex BA-microbiota crosstalk can provide valuable insights into HCC development and facilitate the development of innovative therapeutic approaches for liver malignancy.

文摘Modern medicine is reliant on various medical imaging technologies for non-invasively observing patients’anatomy. However, the interpretation of medical images can be highly subjective and dependent on the expertise of clinicians. Moreover, some potentially useful quantitative information in medical images, especially that which is not visible to the naked eye,is often ignored during clinical practice. In contrast, radiomics performs high-throughput feature extraction from medical images, which enables quantitative analysis of medical images and prediction of various clinical endpoints.Studies have reported that radiomics exhibits promising performance in diagnosis and predicting treatment responses and prognosis, demonstrating its potential to be a non-invasive auxiliary tool for personalized medicine. However, radiomics remains in a developmental phase as numerous technical challenges have yet to be solved, especially in feature engineering and statistical modeling. In this review,we introduce the current utility of radiomics by summarizing research on its application in the diagnosis, prognosis, and prediction of treatment responses in patients with cancer. We focus on machine learning approaches, for feature extraction and selection during feature engineering and for imbalanced datasets and multi-modality fusion during statistical modeling.Furthermore,we introduce the stability, reproducibility, and interpretability of features, and the generalizability and interpretability of models. Finally,we offer possible solutions to current challenges in radiomics research.

文摘Brain injuries due to trauma or stroke are major causes of adult death and disability.Unfortunately, few interventions are effective for post-injury repair of brain tissue. After a long debate on whether endogenous neurogenesis actually happens in the adult human brain, there is now substantial evidence to support its occurrence.Although neurogenesis is usually significantly stimulated by injury, the reparative potential of endogenous differentiation from neural stem/progenitor cells is usually insufficient.Alternatively, exogenous stem cell transplantation has shown promising results in animal models, but limitations such as poor long-term survival and inefficient neuronal differentiation make it still challenging for clinical use.Recently,a high focus was placed on glia-to-neuron conversion under single-factor regulation.Despite some inspiring results, the validity of this strategy is still controversial. In this review,we summarize historical findings and recent advances on neurogenesis strategies for neurorepair after brain injury. We also discuss their advantages and drawbacks,as to provide a comprehensive account of their potentials for further studies.

文摘The conventional perception of astrocytes as mere supportive cells within the brain has recently been called into question by empirical evidence, which has revealed their active involvement in regulating brain function and encoding behaviors associated with emotions.Specifically, astrocytes in the basolateral amygdala have been found to play a role in the modulation of anxiety-like behaviors triggered by chronic stress. These results suggest that basolateral amygdala astrocytic excitatory amino acid transporter 2 plays a role in in the regulation of unpredictable chronic mild stress-induced anxiety-like behavior by impacting the activity of local glutamatergic neurons, and targeting excitatory amino acid transporter 2 in the basolateral amygdala holds therapeutic promise for addressing anxiety disorders.

文摘Brain-derived neurotrophic factor is a crucial neurotrophic factor that plays a significant role in brain health. This metaanalysis was registered at PROSPERO(registration ID: CRD42023439408).

文摘The organ-specific toxicity resulting from microplastic(MP)exposure has been extensively explored, particularly concerning the gut, liver, testis, and lung. However, under natural conditions, these effects are not restricted to specific organs or tissues.Investigating whether MP exposure presents a systemic threat to an entire organism, impacting factors such as lifespan, sleep, and fecundity,is essential. In this study,we investigated the effects of dietary exposure to two different doses of MPs(1–5μm)using the terrestrial model organism Drosophila melanogaster. Results indicated that the particles caused gut damage and remained within the digestive system.Continuous MP exposure significantly shortened the lifespan of adult flies.Even short-term exposure disrupted sleep patterns, increasing the length of daytime sleep episodes.Additionally, one week of MP exposure reduced ovary size, with a trend towards decreased egg-laying in mated females.Although MPs did not penetrate the brain or ovaries, transcriptome analysis revealed altered gene expression in these tissues. In the ovary,Gene Ontology(GO)analysis indicated genotoxic effects impacting inflammation, circadian regulation, and metabolic processes, with significant impacts on extracellular structure-related pathways. In the brain,GO analysis identified changes in pathways associated with proteolysis and carbohydrate metabolism.Overall, this study provides compelling evidence of the systemic negative effects of MP exposure, highlighting the urgent need to address and mitigate environmental MP pollution.

文摘The remarkable properties of carbon nanotubes(CNTs)have led to promising applications in the field of electromagnetic inter-ference(EMI)shielding.However,for macroscopic CNT assemblies, such as CNT film, achieving high electrical and mechanical properties remains challenging, which heavily depends on the tube-tube interac-tions of CNTs.Herein,we develop a novel strategy based on metal-organic decomposition(MOD)to fabricate a flexible silver-carbon nanotube(Ag-CNT)film.The Ag particles are introduced in situ into the CNT film through annealing of MOD,leading to enhanced tube-tube interactions. As a result, the electrical conductivity of Ag-CNT film is up to 6.82×10^(5) S m^(-1),and the EMI shielding effectiveness of Ag-CNT film with a thickness of~7.8μm exceeds 66 dB in the ultra-broad frequency range(3-40 GHz).The tensile strength and Young’s modulus of Ag-CNT film increase from 30.09±3.14 to 76.06±6.20 MPa(~253%)and from 1.12±0.33 to 8.90±0.97 GPa(~795%),respectively.Moreover,the Ag-CNT film exhibits excellent near-field shield-ing performance, which can effectively block wireless transmission. This innovative approach provides an effective route to further apply macroscopic CNT assemblies to future portable and wearable electronic devices.

文摘DEAR EDITOR,The macaques belongs to the genus Macaca, consisting of at least 23 species(Roos et al.,2019).Among all congeners, rhesus macaque(M.mulatta)is regarded as the widest distributed non-human primate species in the world. Its native range spans in East Asia, northern part of Southeast Asia and Indian subcontinent(Liu et al.,2018).Listed as“Least Concern”on the IUCN Red List, this species is locally threatened due to habitat loss and degradation in China and Thailand(Lu et al.,2018).Nevertheless,pet release resulting in hybridization with other congeners(e.g.,rhesus macaque×crab-eating macaque(M.fascicularis))was documented in Hong Kong SAR,China(Wong&Ni,2000),threatening genetic integrity of wild populations.

文摘CO_(2) emulsions used for EOR have received a lot of interest because of its good performance on CO_(2)mobility reduction. However, most of them have been focusing on the high quality CO_(2) emulsion(high CO_(2) fraction),while CO_(2) emulsion with high water cut has been rarely researched. In this paper,we carried out a comprehensive experimental study of using high water cut CO_(2)/H_(2)O emulsion for enhancing oil recovery.Firstly,a nonionic surfactant, alkyl glycosides(APG),was selected to stabilize CO_(2)/H_(2)O emulsion, and the corresponding morphology and stability were evaluated with a transparent PVT cell.Subsequently, plugging capacity and apparent viscosity of CO_(2)/H_(2)O emulsion were measured systematically by a sand pack displacement apparatus connected with a 1.95-m long capillary tube.Furthermore,a high water cut(40 vol%) CO_(2)/H_(2)O emulsion was selected for flooding experiments in a long sand pack and a core sample, and the oil recovery, the rate of oil recovery, and the pressure gradients were analyzed. The results indicated that APG had a good performance on emulsifying and stabilizing CO_(2) emulsion. An inversion from H_(2)O/CO_(2) emulsion to CO_(2)/H_(2)O emulsion with the increase in water cut was confirmed.CO_(2)/H_(2)O emulsions with lower water cuts presented higher apparent viscosity, while the optimal plugging capacity of CO_(2)/H_(2)O emulsion occurred at a certain water cut.Eventually, the displacement using CO_(2)/H_(2)O emulsion provided 18.98% and 13.36% additional oil recovery than that using pure CO_(2) in long sand pack and core tests, respectively. This work may provide guidelines for EOR using CO_(2) emulsions with high water cut.

文摘The attention mechanism can extract salient features in images, which has been proved to be effective in improving the performance of person re-identification(Re-ID).However,most of the existing attention modules have the following two shortcomings:On the one hand, they mostly use global average pooling to generate context descriptors, without highlighting the guiding role of salient information on descriptor generation, resulting in insufficient ability of the final generated attention mask representation; On the other hand, the design of most attention modules is complicated, which greatly increases the computational cost of the model. To solve these problems, this paper proposes an attention module called self-supervised recalibration(SR)block, which introduces both global and local information through adaptive weighted fusion to generate a more refined attention mask.In particular,a special"Squeeze-Excitation"(SE)unit is designed in the SR block to further process the generated intermediate masks, both for nonlinearizations of the features and for constraint of the resulting computation by controlling the number of channels.Furthermore,we combine the most commonly used Res Net-50 to construct the instantiation model of the SR block, and verify its effectiveness on multiple Re-ID datasets, especially the mean Average Precision(m AP)on the Occluded-Duke dataset exceeds the state-of-the-art(SOTA)algorithm by 4.49%.

文摘Background:Cochlear hair cell injury is a common pathological feature of hearing loss. The basic helix-loop-helix family, member e40(Bhlhe40),a gene belonging to the basic helix-loop-helix(bHLH)family,exhibits strong transcriptional repression activity. Methods:Oxidative damage,in House Ear Institute-Organ of Corti 1(HEI-OC1)cells,was caused using hydrogen peroxide(H2O2).The Ad-Bhlhe40 particles were constructed to overexpress Bhlhe40 in HEI-OC1 cells.Various assays including cell counting kit-8(CCK-8),terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling assay(TUNEL),flow cytometry, immunofluorescence, and corresponding commercial kits were employed to investigate the impacts of Bhlhe40 on cell viability, apoptosis, oxidative stress levels, mitochondrial membrane potential and cellular senescence.Additionally,a dual-luciferase reporter assay was performed to confirm the targeting of the histone deacetylases 2(Hdac2)by Bhlhe40.Results:The results revealed that Bhlhe40 was downregulated in H_(2)O_(2)-treated HEI-OC1 cells, but its overexpression improved cell viability and mitigated H_(2)O_(2)-induced oxidative injury in HEI-OC1 cells with increase of superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GPx)activities and decrease of reactive oxygen species(ROS)levels.Besides, overexpression of Bhlhe40 suppressed H_(2)O_(2)-triggered cell senescence,as evidenced by the fact that the upregulation of P53,P21,and P16 in HEI-OC1 cells treated with H2O2 were all alleviated by Bhlhe40 overexpression. And we further verified that overexpression of Bhlhe40 could inhibit the expression of Hdac2,which may be related to the repression of Hdac2 transcription. Conclusion:This study suggests that Bhlhe40 plays a protective role against senescence and oxidative damage in cochlear hair cells exposed to H2O2.

文摘The low-density imaging performance of a zone plate-based nano-resolution hard x-ray computed tomography(CT)system can be significantly improved by incorporating a grating-based Lau interferometer. In summary, a novel model-driven nPCT image reconstruction algorithm with high accuracy and robustness is verified for the Lau interferometer-based hard x-ray nano-resolution phase contrast imaging.

文摘The strength of microwave(MW)electric field can be observed with high precision by using the standard electromagnetically induced transparency and Aulter–Towns(EIT-AT)technique,when its frequency is resonant or nearly-resonant with the Rydberg transition frequency. As the detuning of MW field increases, one of the transmission peaks(single peak)is easier to measure due to its increased amplitude. It can be found that the central symmetry point of the two transmission peaks f_(1/2)is only related to the detuning of MW field△_(MW)and central symmetry point f_(0)of resonant MW field, satisfying the relation f_(1/2)=△_(MW)/2+f_(0).Thus,we demonstrate a single transmission peak method that the MW E-field can be determined by interval between the position of single peak and f_(1/2).We use this method to measure continuous frequencies in a band from-200 MHz to 200 MHz of the MW field. The experimental results and theoretical analysis are presented to describe the effectiveness of this method. For 50 MHz<△_(MW)<200 MHz,this method solves the problem that the AT splitting cannot be measured by using the standard EIT-AT techniques or multiple atomic-level Rydberg atom schemes.

文摘Spin–momentum locking is a key feature of the topological surface state, which plays an important role in spintronics. The electrical detection of current-induced spin polarization protected by the spin–momentum locking in nonmagnetic systems provides a new platform for developing spintronics, while previous studies were mostly based on magnetic materials. In this study, the spin transport measurement of Dirac semimetal Cd_(3)As_(2) was studied by three-terminal geometry, and a hysteresis loop signal with high resistance and low resistance state was observed. The hysteresis was reversed by reversing the current direction, which illustrates the spin–momentum locking feature of Cd_(3)As_(2). Furthermore, we realized the on–off states of the spin signals through electric modulation of the Fermi arc via the three-terminal configuration, which enables the great potential of Cd_(3)As_(2) in spin field-effect transistors.
