This paper investigates traffic flow of connected and automated vehicles at lane drop on two-lane highway. We evaluate and compare performance of an optimization-based control algorithm(OCA) with that of a heuristic rules-based algorithm(HRA). In the OCA, the average speed of each vehicle is maximized. In the HRA, virtual vehicle and restriction of the command acceleration caused by the virtual vehicle are introduced. It is found that(i) capacity under the HRA(denoted as C_(H)) is smaller than capacity under the OCA;(ii) the travel delay is always smaller under the OCA, but driving is always much more comfortable under the HRA;(iii) when the inflow rate is smaller than C_(H), the HRA outperforms the OCA with respect to the fuel consumption and the monetary cost;(iv) when the inflow rate is larger than C_(H), the HRA initially performs better with respect to the fuel consumption and the monetary cost, but the OCA would become better after certain time. The spatiotemporal pattern and speed profile of traffic flow are presented, which explains the reason underlying the different performance. The study is expected to help for better understanding of the two different types of algorithm. 展开更多

There are nearly 1 000 rice landrace varieties in the Taihu basin, China. To assess the genetic diversity of the rice, 24 intragenic molecular markers(representing 17 starch synthesis-related genes) were investigated in 115 Taihu basin rice landraces and 87 improved cultivars simultaneously. The results show that the average genetic diversity and polymorphism information content values of the landraces were higher than those of improved cultivars. In total, 41 and 39 allele combinations(of the 17 genes) were derived from the landraces and improved cultivars, respectively; only two identical allele combinations were found bet ween the two rice variety sources. Cluster analysis, based on the molecular markers, revealed that the rice varieties could be subdivided into five groups and, within these, the japonica improved rice and japonica landrace rice varieties were in two separate groups. According to the quality reference criteria to classify the rice into grades, some of the landraces were found to perform we ll, in terms of starch quality. For example, according to NY /T595-2002 criteria from the Ministry of Agriculture of China, 25 and 33 landraces reached grade 1, in terms of their apparent amylose content and gel consistency. Th e varieties that had outstanding quality could be used as breeding materials for rice quality breeding programs in the future. Our study is useful for future applications, such as genetic diversity studies, the protection of rice variety and improvment of rice quality in breeding programs. 展开更多

Construction of a Multipurpose M13KE Phage Display System

In this study, a multipurpose M13KE phage display vector was constructed from wild-type M13KE phage for long peptide or protein display libraries without helper phage to expand the scope of targeted high-throughput screening. Based on the relationship between the structure and function of minor coat protein of wild-type MI3KE (wt-plII), a truncated gene III (tglll) encoding minor coat protein from M13KE phage was cloned. A fusion gene fragment harboring a hw/tac promoter, signal peptide and C-terminal region sequence of gill was assembled with SOEing-PCR (splice-overlapping-extension polymerase chain reaction) method and inserted into M13KE vector. SDS-PAGE and Western blot analysis with anti-M13 pIII moneclonal antibody were employed to detect the expression of re- combinant protein, c-Myc and HA tag sequences were fused into the recombinant protein. The results showed that tglll was inserted into an unessential region of M13KE. According to the results of SDS-PAGE and Western blot with anti-M13 pIII antibody, pIII was expressed by wt-gIII and tgIII, glII harboring two tags ex- pressed both c-Myc and HA peptides using SDS-PAGE and Western blot with the corresponding monoclonal antibodies. In this study, a multipurpose M13KE phage display system was successfully constructed, which could express both short and long peptide libraries without helper phage. In future, the obtained M13KE phage display system may be used for targeted high-throughput screening of long peptide libraries without helper phage. 展开更多

In this paper,with 511 rice landraces in the Taihu Lake area as test materials,we select 19 starch synthesis-related intragenic molecular markers to detect the genetic quality of starch,and compare them with 86 bred varieties. The results show that the average polymorphic information content( PIC) of japonica landraces is 0. 1726,slightly higher than the average PIC( 0. 1101) of the bred japonica rice varieties.Based on Nei's genetic distance between materials,UPGMA method is used for clustering,and all study materials are divided into 6 groups.Group I mainly includes indica rice,the bred japonica rice varieties are mainly concentrated in the first half of Group II and Group III,and the japonica landraces are mainly concentrated in the second half of Group III,and Group Ⅳ,Ⅴ,Ⅵ. Both of them are in different regions,and there has been genetic differentiation. According to the national standard of high quality rice,it is found that many rice landraces in the Taihu Lake area have good quality and traits,and these varieties can be used for future high quality breeding. 展开更多

Plant height is an important morphological trait that affects crop yield.Several genes related to plant height and yield have been reported in rice(Oryza sativa L.),however,the molecular mechanism underlying the regulation of these traits is still not completely understood.VP64 is widely used as a transcriptional activator to investigate the biological function of genes encoding transcription factors.Here,we identified a novel bZIP transcription factor OsbZIP62 that is involved in modulating agronomic traits in rice.Overexpression of OsbZIP62-VP64(OsbZIP62V)significantly increases the plant height and yield per plant in rice.RNA-seq analysis showed that some plant height and panicle development related genes(i.e.,OsEATB,OsDSS1 and OsGA3ox2)were up-regulated in OsbZIP62V overexpressing rice plants.Besides,OsbZIP62 could also bind to the promoters of several putative target genes.These results suggested that OsbZIP62 plays a role as transcriptional regulator in regulating the expression of genes associated with agronomic traits,and OsbZIP62 fused with VP64 would be useful in crop genetic modification with improved plant architecture and yield. 展开更多

We present a novel method to achieve the decoupling between the transmission and reflection waves of non-Hermitian doped epsilon-near-zero(ENZ)media by inserting a dielectric slit into the structure.Our method also allows for independent control over the amplitude and the phase of both the transmission and reflection waves through few dopants,enabling us to achieve various optical effects,such as perfect absorption,high-gain reflection without transmission,reflectionless high-gain transmission and reflectionless total transmission with different phases.By manipulating the permittivity of dopants with extremely low loss or gain,we can realize these effects in the same configuration.We also extend this principle to multi-port doped ENZ structures and design a highly reconfigurable and reflectionless signal distributor and generator that can split,amplify,decay and phase-shift the input signal in any desired way.Our method overcomes limitations of optical manipulation in doped ENZ caused by the interdependent nature of the transmission and reflection,and has potential applications in novel photonic devices. 展开更多

Drought stress is one of the major environmental factors affecting crop growth and productivity.Cuticular wax plays essential roles in protecting plants from environmental stress via forming a hydrophobic barrier on leaf epidermis.In this study,we analyzed nine members(OsCUT1‒OsCUT9)ofβ-ketoacyl-CoA synthase,the rate-limiting key enzyme for cuticular wax synthesis in rice by homology search and domain prediction.The expression levels of OsCUT genes under different abiotic stresses were investigated and OsCUT1 down-regulated by abiotic stress was selected for further function validation.Compared to the wild type,overexpression of OsCUT1(OX-OsCUT1)exhibited significantly increased drought resistance.Epicuticular wax was increased on the leaf surface of OX-OsCUT1 and the chlorophyll leaching experiment showed that the cuticular permeability was decreased in the OX-OsCUT1 plants.Moreover,overexpression of OsCUT1 didn’t result in the significant changes of major agronomic traits.In total,these results suggested that OsCUT1 is a promising gene for engineering rice plants with enhanced drought tolerance. 展开更多

In this paper,we propose a intrusion detection algorithm based on auto-encoder and three-way decisions(AE-3WD)for industrial control networks,aiming at the security problem of industrial control network.The ideology of deep learning is similar to the idea of intrusion detection.Deep learning is a kind of intelligent algorithm and has the ability of automatically learning.It uses self-learning to enhance the experience and dynamic classification capabilities.We use deep learning to improve the intrusion detection rate and reduce the false alarm rate through learning,a denoising AutoEncoder and three-way decisions intrusion detection method AE-3WD is proposed to improve intrusion detection accuracy.In the processing,deep learning AutoEncoder is used to extract the features of high-dimensional data by combining the coefficient penalty and reconstruction loss function of the encode layer during the training mode.A multi-feature space can be constructed by multiple feature extractions from AutoEncoder,and then a decision for intrusion behavior or normal behavior is made by three-way decisions.NSL-KDD data sets are used to the experiments.The experiment results prove that our proposed method can extract meaningful features and effectively improve the performance of intrusion detection. 展开更多

Tlie rigid-flexible coupling dynamic modeling theory and the discretization methods of a rotating flexible rectangular thin plate are investigated in this paper.Based on the continuum mechanics,the rigid-flexible coupling dynamic model is established for the flexible rectangular thin plate undergoing large overall rotation,and the coupling term of the deformation which is caused by transverse deformation is considered.Assumed mode method(AMM),spline finite point method(SFPM)and Beizer finite point method(BFPM)are used to describe the deformation of the flexible rectangular plate,and then the dynamic equations of a rotating flexible rectangular thin plate undergoing overall motion are derived by Lagrange^equation of the second kind.The dynamics of a cantilever plate undergoing large overall rotation is simulated via using different dynamic models,and the simulation results of the first order approximation model are compared with those of the traditional zero-order approximation model.It is shown that the first order approximation model with the dynamic stiffening terms can correctly describe the dynamic behavior of the system undergoing large overall rotation,while the zero-order approximation model cannot get the correct results.And AMM.SFPM.BFPM can well describe the deformation of a rotating flexible rectangular plate. 展开更多
