Summary: This paper aimed to study the ability of baicalein to block human cytomegalovirus (HCMV) infection in extravillous cytotrophoblasts (EVT) and its effect on the vasoactive intestinal peptide (VIP) expression in HCMV-infected EVT in vitro. A human trophoblast cell line (HPT-8) was chosen in this study. HCMV with 100 TCIDs0was added into culture medium to infect HPT-8 cells, and then HCMV pp65 antigen was assayed by immunofluorescence staining. The infection status was determined by vi- rus titration. Real-time quantitative polymerase chain reaction (qRT-PCR) was used to detect virus DNA load in the infected cells. The expression of VIP mRNA and protein in the infected cells was measured by qRT-PCR, immunocytochemistry and Western blotting. Concentration of VIP secreted in supernatants was determined by ELISA. Red-stained HCMV pp65 antigens were found in infected HPT-8 cells 48 h after infection. HCMV replicated in large quantity in infected HPT-8 cells 4 days after infection, reaching a peak at day 6 post-infection. After treatment with baicalein, virus DNA load in in- fected HPT-8 cells was decreased (P〈0.05), and the levels of VIP mRNA and protein, and the concen- tration were raised to the normal (P〉0.05). Our study suggested that baicalein exerts a positive effect on the VIP expression in HCMV-infected EVT at maternal-fetal interface. 展开更多

Objective: To analyze the literature status and research hotspots of Science Citation Index (SCI)-related ostomy in the world and to provide references for scientific research and clinical work in the stoma care field. Methods: Based on the Web of Science core database and its own analysis function, HistCite analysis software and Excel were used to study the published research about ostomy patients. Results: A total of 1,262 articles were published between 1910 and 2016 with the authors from 48 countries and regions, 1,347 research institutions, published in 321 journals, with 4,048 first authors and coauthors; globally, there was a trend of slow growth in the number of authors every year. The study in the USA was absolutely in the lead position, and Canada and Turkey were more active. China's circulation volume was the 15th in the world. The periodical that published most often was the Journal of Wound Ostomy and Continence Nursing. The most interdisciplinary surgical studies were surgery and nursing, where these should be considered important. The most prolific author in the field was "Grant", and the highest cited article was entitled as "Living with a stoma: a review of the literature". Conclusions: The related research of global stoma is constantly developing. The research hotspot is nursing before and after stoma surgery. China and the USA are leading countries in research. They should follow the recent trend to improve the depth and breadth of the research in the field. 展开更多

Determining clinical questions is fundamental to the development of clinical practice guidelines(CPGs),which bridges the initial phases and the final recommendations.It is essential for evidence retrieval and the formulation of recommendations.The scientific rigor and precision in determination of clinical questions directly influence the future implementation and applicability of guidelines.In 2020,the World Federation of Acupuncture-Moxibustion Societies initiated the project of clinical practice guideline on acupuncture and moxibustion for adult major depressive disorder(mild-moderate degree)to address clinical and medical decision-making issues in acupuncture treatment for adult mild to moderate major depressive disorder.This CPG provides systematic recommendations based on clinical evidence,patient values,and other factors,aiding decision-makers,clinicians,and patients in selecting appropriate interventions.This paper discusses and analyzes the determination process of clinical questions,and the related issues during the development of this guideline,aiming to provide a reference for determining clinical questions and developing CPGs in the field of acupuncture and exploring more scientific tools and methods for determining clinical questions in future CPGs. 展开更多
