Psoriasis is a kind of immune-mediated, chronic, inflammatory skin disease, which is often associated with different degrees of psychological disorders. Specifically, there is a significant correlation between psoriasis and depression, and they show the relationships of reciprocal causation and mutual promotion. Psoriasis with depression is more harmful than simple psoriasis, and its prognosis is worse, which brings a huge burden to the family and society and is worthy of clinical attention. Based on the pathogenic factors of western medicine and pathogenesis of traditional Chinese medicine in psoriasis with depression, the paper summarized and elaborated the research progress on the mechanism of traditional Chinese medicine in the treatment of psoriasis with depression, in order to provide new ideas for clinical treatment. 展开更多

Recurrent spontaneous abortion is a common disease in gynecology,and it seriously affects women's reproductive health and brings heavy burden and pain to society and families.The cause of recurrent spontaneous abortion is complicated,in addition to the well-defined genetic,anatomical,infection and endocrine factors,and there are still some unknown causes,which is called as unexplained recurrent spontaneous abortion,accounting for 40%of recurrent abortion.At present,there are a lot of researches on the treatment methods of the patients with the unexplained recurrent spontaneous abortion,which also shows that the treatment of traditional Chinese and Western medicine all have certain clinical application effect.Western medicine clinical methods mainly includes immunotherapy,immunosuppressive therapy,anticoagulation therapy,progesterone therapy,etc.Based on the experience of the professor and combined with many years of clinical practice,the author believes that the pathogenesis of this disease in traditional Chinese medicine is mainly due to impaired impulse and deficiency of Spleen,lack of qi and blood,can not nourishing the fetus;deficiency of Kidney Qi,blood flow was delayed,and blood stasis and could not raise the fetus.Clinical treatment is based on invigorating the kidney,tonifying spleen and nourishing blood,promoting blood circulation to remove blood stasis and dredging collaterals.Oral Chinese medicine combined with external acupuncture and moxibustion has achieved excellent effects in improving pregnancy rate.This article reviews the domestic and foreign methods of treating unexplained recurrent miscarriage in order to provide clinical reference.In the future,the combination of Chinese and Western medicine should become the main therapy to increase pregnancy rate. 展开更多

The diversified treatment methods of traditional Chinese medicine have visible clinical effects, especially in terms of reducing toxicity and increasing efficiency. Objective:To explore Professor Yang Suqing’s experience in treating blood stasis syndrome of psoriasis vulgaris and analyze the characteristics of its prescription;Methods:Outpatient prescriptions for the treatment of psoriasis vulgaris with blood stasis syndrome were selected and extracted traditional Chinese medicine for standardized processing.Then analyze the data by using set visualization analysis and Apriori algorithm.Results:A total of 149 traditional Chinese herbs were extracted.Among them,and the top 12 commonly used herbs with frequency>30 were Licorice,Saposhnikovia,Schizonepeta tenuifolia,Angelica sinensis,Paeonia lactiflora,Ligusticum chuanxiong,Rehmannia glutinosa,Cicada slough,Oyster and Keel through the set visualization analysis,herbs association rules analysis,and herbs combination analysis.Saposhnikovia,Liquorice,Schizonepeta tenuifolia,Angelica sinensis and Paeonia Alba are the most commonly used combination of herbs by Professor Yang Suqing in the treatment of blood stasis syndrome of psoriasis vulgaris.Conclusion:In addition to invigorating blood circulation and removing blood stasis herbs,which are preferred for the treatment of blood stasis syndromes,Saposhnikovia and Schizonepeta tenuifolia,etc.,are used to synergistically exert the functions of invigorating blood circulation and removing blood stasis and eliminating wind expression,with remarkable efficacy. 展开更多

Target gene prediction and functional analysis of miRNAs differently expressed in colon cancer primary tumors to metastases formed in the liver

Background:Colon cancer is one of the main tumor-related causes of death worldwide and now surgical resection is still the most effective method for the treatment of colon cancer.However,many colon cancers currently lose the opportunity for surgical treatment because of liver metastases.The possible molecular mechanism of liver metastasis of colon cancer can provide ideas for the prevention and treatment of liver metastasis colon cancer.Several studies have recently indicated the regulatory effects of microRNAs(miRNAs)in cancer metastasis.Bioinformatics methods were used to analyze the differentially expressed miRNAs in primary colon cancer tissues and liver metastases.Then the target genes of differentially expressed miRNAs were predicted.By analyzing the biological processes and signaling pathways that the target gene may participate in,we infer the possible molecular mechanisms of liver metastasis of colon cancer and the effects of target genes on prognosis of patients were explored.Methods:The chip data GSE98406 was selected and differentially expressed miRNAs between primary colon cancer and liver metastatic colon cancer were explored by Morpheus.TargetScan and miRanda were used to predict target genes of differentially expressed miRNAs.The gene oncology and Kyoto Encyclopedia of Genes and Genomes pathway analyses were performed to analyze the biological processes and possible signaling pathways the target genes involved in.Protein-ptotein interaction analysis was performed by String and Cyotscape,the interactions among target genes and hub genes were analyzed.Finally,we explored the effects of differentially expressed miRNAs and their target genes on the prognosis of colon cancer patients.Results:Two differentially expressed miRNAs were screened out,of which miR-122 was upregulated more than 2 folds and miR-143 was downregulated more than twofold in liver metastatic colon cancer.Target genes of miR-122 and miR-143 were mainly involved in energy metabolism.The major signaling pathways involved are epithelial-mesenchymal transition pathways.Ten hub genes were selected by protein interaction analysis.Among them,KRAS,CDK1,CREB1,CS,PC,RAB7A,and CANX were highly expressed in tumor tissues,and CALM1 and MAPK7 were lowly expressed in tumor tissues.The results showed that reduced expression of CS and PC reduced survival of patients with colon cancer.However,the impact of miR-122 and mi-143 on the prognosis of patients with colon cancer is not clear.Conclusion:Differentially expressed miRNAs mainly affect the expression of target genes involved in energy metabolism and cellular transformation in colon cancer.Intracellular metabolic activity is the center of cellular activity,the treatment of metabolic processes in tumor cells may be a new idea for the treatment of tumors. 展开更多

Objective: To investigate the therapeutic effect of Yuchang suppository on experimental ulcerative colitis rats and the effect of PPARγ/NF-KB signaling pathway. Methods: The UC rat model was replicated by 2.4.6-trinitrobenzenesulfonic acid (TNBS)/ethanol method and randomly divided into normal control group, model group, high-dose group of Yuchang suppository, middle dose group , Low-dose group and sulfasalazine group, 8 rats in each group. After 14 days of treatment, the general condition of the rats was observed and the disease activity index was scored. The changes of serum TNF-α, IL-1β and IL-6 levels were detected by ELISA. The colonic mucosal injury index (CMDI) score and colon were observed by colon specimens. Histopathological changes;RT-PCR and Western blot were used to detect the expression of PPARγ and NF-KB genes and proteins in colon tissue. Results: Compared with the normal control group, the DAI and CMDI scores in the model group and the levels of serum TNF-α, IL-1β, IL-6, NF-κB p65 mRNA and protein in colon tissue were significantly increased, PPAR. - γ mRNA and protein expression levels were significantly reduced. The levels of IL-1β, IL-6, TNF-α and the expression of NF-κB p65 mRNA and protein in colon tissue were lower than those in the model group, and the expression levels of PPARγ mRNA and protein were high. In the model group. In the improvement of NF-κBp65 and PPAR-γ gene and protein expression in rat colonic mucosa, the high-dose group and the middle-dose group were significantly better than the low-dose group. Conclusion:Yuchang suppository can significantly improve the symptoms and histopathology of UC model rats. The mechanism may be through activation of PPARγ, blocking the activation of NF-κB signaling pathway, and down-regulating inflammatory factors such as TNF-α and IL-1β. The expression is such that the inflammatory response is alleviated or eliminated, thereby achieving the therapeutic effect of UC. 展开更多

Objective:To explore the effect of acupuncture on the expression of heme oxygenase-1 in rats with acute cerebral hemorrhage.Methods:108 Wistar male rats were randomly divided into sham operation group,model group,and acupuncture combined with model group(referred to as acupuncture group).Each group was divided into 3 subgroups according to 1d,3d and 7d,with 6 rats in each subgroup.The rat model of cerebral hemorrhage was established by autologous blood injection.Acupuncture was given at Baihui(GU20)and Qubin(GB7).Separately,at the 1st,3rd and 7th day,modified neurological severity score was used to evaluate the neurological function of rats,HE staining was used to measure the volume of cerebral hematoma and western blot was used to detect the expression of heme oxygenase-1 protein in cerebral hematoma tissue.Results:Compared with the model group,at each time point,the modified neurological severity score of the acupuncture group was significantly reduced(P<0.01);at the two time points of 3rd and 7th day,the cerebral hematoma volume of the acupuncture group was significantly reduced(P<0.05)and the expression of heme oxygenase-1 protein in brain tissue was significantly increased(P<0.05).Conclusion:Acupuncture may promote the expression of heme oxygenase-1 protein,reduce the volume of hematoma and the score of neurological deficit in rats with intracerebral hemorrhage,improve the performance of neurological deficit,and play a role in brain protection. 展开更多

Objective:To prepare cationic liposome nanoparticles loaded with survivin-siRNA and Cnidium monnieri based on the ability of liposomes to contain both water-soluble and lipid soluble components.Methods:The preparation technology of Osthol cationic liposomes was optimized by orthogonal test with membrane material ratio,drug lipid ratio,ultrasonic time and steaming temperature as factors.The volume ratio of HA-siRNA to protamine and the ratio of HA-siRNA protamine complex to liposome were investigated by control variable method with potential and particle size as indexes.The particle size and zeta potential were measured by potentiometric particle size analyzer,and the shape was observed by transmission electron microscope;The absorbance of different concentrations of FAM-Survivin-siRNA standard solution was measured by microplate analyzer,and the entrapment efficiency of cationic liposomes loaded with FAM-Survivin-siRNA and osthole was calculated.Results:The optimum preparation conditions of osthole loaded cationic liposomes were as follows:the ratio of membrane to material was 3:1,the ratio of drug to lipid was 1:5,the steaming temperature was 30℃,the ultrasonic time was 70 min,and the encapsulation efficiency was 78.34%.The optimum preparation conditions of osthole loaded cationic liposomes loaded FAM-Survivin-siRNA were as follows:the volume ratio of Survivin-siRNA to protamine was 1:1,Protamine complex 25μg.Add 50μL cationic liposomes.The particle size is 132.3±0.2nm,zeta potential is 43.15±0.05mv,and its shape is irregular round;According to the standard curve,the entrapment efficiency of cationic liposome nanoparticles co loaded with Survivin-siRNA and osthole was 81.34±0.041%.Conclusion:The prepared cationic liposome nanoparticles loaded with Survivin-siRNA and osthole have good encapsulation efficiency,particle size and zeta potential. 展开更多

Objective:To investigate the influence of kidney-tonifying and blood-generating method on platelet(PLT)growth in the treatment of aplastic anemia(AA)and analyze the characteristics of the clinical factors affecting PLT growth.We enrolled patients treated from September 2018to March 2021 in the China Academy of Traditional Chinese Medicine(TCM)Xiyuan Hospital,China Academy of TCM hospital gate 19sub-center.We enrolled 128 AA patients,for whom the inclusion criteria were:treatment is given priority to with kidney and heart's-blood method,on the basis of western medicine treatment for kidney fill party avoid Fried instant particles,stay for 3 months for a period of treatment,taking two consecutive period of treatment,changes in the PLT were observed,and the clinical characteristics of AA patients,such as PLT growth and recovery from baseline value,were analyzed.Materials and Methods:A total of 128 AA patients from 19 branches of Xiyuan Hospital,China Academy of Chinese Medical Sciences and Guang'anmen Hospital,China Academy of Chinese Medical Sciences treated from September 2018 to March 2021 were enrolled.The treatment was based on the kidney-tonifying and blood-generating method and lasted for 3 months as a course of treatment.The changes in the number of PLT in patients were observed,and the clinical characteristics of AA affecting PLT growth and recovery from baseline value were analyzed.Results:After 6 months of treatment following the kidney-tonifying and blood-generating method,PLT increased from the baseline values in 75 cases,including 21 cases of AA with normal PLT recovery and 54 cases of AA without normal PLT recovery.There were 53 cases of AA in whom PLT did not increase from the baseline.Analysis of the general data showed that patients with lower Karnofsky Performance Status(KPS)score were more likely to experience PLT growth or even return to normal after therapy(P<0.05).The results of routine blood analysis showed that when hemoglobin(HGB)was≥60 g/L and the reticulocyte(RET)proportion was≥0.3%before treatment,PLTs were more likely to increase,even returning to normal in some patients(P<0.05).Analysis of bone puncture results before treatment showed that when the proliferation of nucleated cells was low or extremely low in bone puncture examination,with the proportion of granulocytes<40%,and the proportion of erythroid cells<20%,the treatment method was more effective at increasing PLT count(P<0.05).Analysis of the chromosome karyotype results before treatment showed that when the chromosome karyotype was normal,PLTs were more likely to increase or even return to normal after treatment(P<0.05).Analysis of T-lymphocyte subsets before treatment showed that when CD3+human leukocyte antigen(HLA)-DR+proportion was≥30%,PLTs were more likely to increase or even return to normal(P<0.05).Conclusions:AA patients with a low KPS score,higher HGB≥60 g/L,RET proportion≥0.3%,low or extremely low proliferation of nucleated cells,granulocytes proportion<40%,erythroid proportion<20%,normal chromosome karyotype,CD3+HLA-DR+≥30%,should be treated with a kidney-tonifying and blood-generating method.Such patients are more likely to show PLT growth and the values may even return to normal. 展开更多

We investigated the pharmacological mechanism of WendanDecoction(WDD)in treating ischemicstroke(IS)using network pharmacology and molecular docking methods.The Gene Expression Omnibusdatabase was searched with“stroke”as akeyword,species as“Homosapiens,”andGeneChip data was usedto identify,screen,and analyze thedifferentially expressed genesof IS.The potential active components and related WDDtarget proteins were screened from the Traditional Chinese medicine systems pharmacology database.Cytoscape(3.8.2)was used to construct the drug-disease-target regulatory network,and Bisogenet and the CytoNCA plug-in were used to construct the protein-protein interaction(PPI).Gene Ontology and the Kyoto Encyclopedia of Genes and Genomesenrichment analyses were performed and AutoDock was used for molecular docking verification.A total of 2497 targets were obtained from 181 active compounds in WDD with8relatedIS targets.PPI network showed thatJUN,NFKBIA,FOS,and CDKN1A have high gene node degree values,withJUN exhibitingthe largest value.This suggestedthat JUN maybe amajor potential target for WDD treatment of IS.Enrichment analysis showed that WDD core targets were involved in the IL-17 and TNF signaling pathway,lipid and atherosclerosis,NF kappa B,toll-like receiver,and nod-like receiver signaling pathways.Network topology analysis showed that the key components of WDDwere luteolin,naringin,ligustrazine,beta sitosterol,quercetin,and stigmasterol;eachplayed a key role and may serve ascore compoundsfor treatingIS.Molecular docking results showed that the active components in WDDbind well with key targets.Our results suggest that various active components of WDD play a therapeutic role in IS by regulating multiple targets such as JUN,NFKBIA,FOS,and CDKN1A in multiple channels such as inflammation,immunity,and lipid metabolism. 展开更多

Inflammation plays an important role in nerve defects caused by intracerebral hemorrhage. Repairing brain damage by inhibiting the macrophage-inducible C-type lectin/spleen tyrosine kinase (Mincle/Syk) signaling pathway is a potential new target for treating cerebral hemorrhage. In this study, we aimed to determine whether acupuncture through Baihui (DU20) to Qubin (GBT) is an effective treatment for intracerebral hemorrhage through the Mincle/Syk signaling pathway. An intracerebral hemorrhage rat model was established by autol- ogous blood infusion into the caudate nucleus. Acupuncture through Baihui to Qubin was performed for 30 minutes, once every 12 hours, for a total of three times. Piceatannol (34.62 mg/kg), a Syk inhibitor, was intraperitoneally injected as a control. Modified neurological severity score was used to assess neurological function. Brain water content was measured. Immunohistochemistry and western blot assay were used to detect immunoreactivity and protein expression levels of Minde, Syk, and CARD9. Real-time polymerase chain reaction was used to determine interleukin-1[~ mRNA levels. Hematoxylin-eosin staining was performed to observe histopathological changes. Our re- suits showed that acupuncture through Baihui to Qubin remarkably improved neurological function and brain water content, and inhibited immunoreactivity and expression of Mincle, Syk, CARDg, and interkeukin-1β Moreover, this effect was similar to piceatannol. These find- ings suggest that acupuncture through Baihui to Qubin can improve neurological impairment after cerebral hemorrhage by inhibiting the Mincle/Syk signaling pathway. 展开更多

Objective To identify significant factors related to the apolipoprotein B/apolipoprotein A1 (apoB/A1) ratio and investigate the association between the apoB/A1 ratio and metabolic syndrome (MS) in polycystic ovary syndrome (PCOS) women. Methods Totally 307 subjects with PCOS were collected and recruited fulfilling the revised 2003 Rotterdam diagnostic criteria. MS was diagnosed by the National Cholesterol Education Program-Adult treatment Panel (NCEP-ATP) III criteria. Results The prevalence of MS in PCOS women was 31.6%, whose average age was 26.2 ___+ 5.2 years. The apoB/A1 ratio was significantly correlated with age, body mass index (BM1), waist circumference (WC), hip circumference, waist-to-hip ratio, blood pressure, metabolic abnormalities, sex hormone binding globulin (SHBG), and free androgen index. In addition, SHBG had a significant association with all five component risk of MS. The increasing apoB/A1 ratio was associated with the prevalence of MS and was one of the risk factors of MS. Conclusion SHBG was considered as an additional potential factor in predicting the metabolic abnormity in PCOS women. The apoB/A1 ratio is associated with SHBG and might be an indicator of MS in PCOS women. 展开更多

Anti-β2 glycoprotein I(anti-β2GPI)antibodies are important contributors to the development of thrombosis.Anti-β2GPI antibody complexes withβ2GPI are well known to activate monocytes and endothelial cells via the intracellular NF-kB pathway with prothrombotic implications.By contrast,the interaction of anti-β2GPI/β2GPI complexes with platelets has not been extensively studied.The p38 mitogen-activated protein kinase(MAPK)pathway has been recognized to be an important intracellular signaling pathway in the coagulation cascade and an integral component of arterial and venous thrombosis.The present study reveals that levels of anti-β2GPI/β2GPI complexes in sera are positively associated with p38MAPK phosphorylation of platelets in thrombotic patients.Furthermore,SB203580 inhibits anti-β2GPI/β2GPI complex-induced platelet activation.Thrombus formation decreased in p38MAPK−/−mice after treatment with anti-β2GPI/β2GPI complexes.In conclusion,p38MAPK may be a treatment target for anti-β2GPI antibody-associated thrombotic events. 展开更多

OBJECTIVE:To explore the effect of electroacupuncture(EA)on rats with chronic fallopian tube inflammation and its potential mechanisms.METHODS:Thirty-six female Sprague-Dawley rats were divided into Control,Model and EA groups.The pathological morphology of the fallopian tubes was observed by hematoxylin-eosin(HE)and Masson staining.The results of transforming growth factor-β1(TGF-β1),P38 mitogen-activated protein kinase(MAPK),phosphorylation(p)-p38MAPK in rat oviduct tissues were detected by immunohistochemistry.Results of P38MAPK,p-P38MAPK and TGF-β1 in rat oviduct tissues were detected by immunofluorescence.The expression level of p38MAPK,p-P38MAPK,TGF-β1 protein in rats was detected by Western blot.Quantitative real-time polymerase chain reaction(RT-q PCR)was used to detect m RNA expression levels of TGF-β1.RESULTS:It found that collagen fibers counts decreased significantly in EA group compared to Model group.The phosphorylation of P38MAPK in EA group was significantly reduced compared to Model group.The serum TGF-β1 expressions in EA group increased decreased significantly.CONCLUSION:Electroacupuncture was able to attenuate chronic salpingitis through down-regulating TGF-β1/MAPK signaling pathway. 展开更多

OBJECTIVE:To investigate pharmacodynamic effects of modified Gexiazhuyu decoction(MGXZYD)and explore the underlying mechanism in the treatment of chronic salpingitis METHODS:Chronic salpingitis model rats were firstly constructed and the blood was collected to detect the whole blood viscosity and plasma viscosity.Rat oviduct were collected to evaluate the macroscopic damage and the pathological injury and fibrosis of oviduct by hematoxylin-eosin(HE)and Masson staining.Elisa assay was to detect the production interleukin-1β(IL-1β)in serum and collagen I(COL-1),matrix metalloprotein 9(MMP-9),tissue inhibitor of metalloproteinases 1(TIMP-1)in oviduct tissue.And immunohistochemical staining with MMP-9 and TIMP-1 in oviduct tissue were examined.Western blot was used to detect the expressions of p38 mitogen-activated protein kinases(p38MAPK),phosphop38MPAK(p-p38MPAK),transforming growth factor-β1(TGF-β1)in oviduct.The expression ofα-smooth muscle actin(α-SMA),p-p38MPAK,in oviduct tissue were detected by immunofluorescence method.The m RNA of p-p38MAPK,α-SMA,COL-1,MMP-9,TIMP-1 was measured by reverse transcription-polymerase chain reaction.RESULTS:Rats administrated with MGXZYD demonstrated decreased the whole blood viscosity and plasma viscosity.MGXZYD obviously improved the tubal wall thickening,swelling and pelvic adhesion.And HE and Masson staining showed MGXZYD improved the pathological injury and fibrosis of oviduct.The results of MTT assay and flow cytometry indicated that MGXZYD could decreased the NIN-3T3 cells viability and improved the apoptosis.Besides,MGXZYD inhibited the protein and/or m RNA of TGF-β1,IL-1β,COL-1,α-SMA,pp38MAPK expressions and increased the production of MMP-9/TIMP-1.CONCLUSION:MGXZYD could prevent the progression of chronic salpingitis by inhibited the fibrocyte and inflammation which via inhibited the p38 MAPK signaling pathway. 展开更多
