Effect of intensity modulated radiation therapy combined with paclitaxel+ endostar chemotherapy on serum malignant molecule levels in patients with locally advanced non-small cell lung cancer 被引量:1

Objective: To discuss the effect of intensity modulated radiation therapy combined with paclitaxel + endostar chemotherapy on serum malignant molecule levels in patients with locally advanced non-small cell lung cancer. Methods: Patients with locally advanced NSCLC who were treated in the hospital between February 2015 and January 2017 were collected and divided into control group (n=59) and research group (n=59) by random number table. Control group received the routine paclitaxel + endostar chemotherapy after the operation, and research group underwent intensity modulated radiation therapy combined with paclitaxel +endostar chemotherapy after the operation. The differences in serum levels of NSCLC-related tumor markers and angiogenesis indexes were compared between the two groups before and after treatment. Results: Before treatment, the differences in serum levels of NSCLC-related tumor markers and angiogenesis indexes were not statistically significant between the two groups. After treatment, serum TK1, CYFRA21-1, Pro-GRP, CEA, CA125 and SCC-Ag levels of research group were lower than those of control group;serum EGFR, COX-2, VEGF, HIF-1 and MMP-2 levels of research group were lower than those of control group. Conclusion:Postoperative intensity modulated radiation therapy combined with paclitaxel + endostar chemotherapy can effectively reduce the serum malignant molecule levels and optimize the illness in patients with local advanced NSCLC. 展开更多

Astragaloside regulates the growth and invasion activity of colon cancer cells in vitro: the experimental study

Objective: To study the regulating effects of astragaloside on the growth and invasion activity of colon cancer cells in vitro. Methods: Lovo colon cancer cell lines were cultured and randomly divided into the control group treated with DMEM without drug or serum and the astragaloside groups treated with serum-free DMEM containing 10 μmol/L, 20 μmol/L and 40μmol/L astragaloside. The cell proliferation activity, the number of invasive cells as well as the expression of proliferating genes and invasion genes were measured after 24 h of treatment. Results: 24 h after treatment, the cell proliferation activity, the number of invasive cells as well as CCND1, β-catenin, ADAM17, MMP11, VEGF and ZEB2 mRNA expression of different concentrations of astragaloside groups were obviously lower than those of blank control group while WTX, PERK, ATF6, TIMP1 and E-cadherin mRNA expression were significantly higher than those of blank control group, and the higher the concentration of astragaloside, the more significant the decreasing trend of the cell proliferation activity, the number of invasive cells as well as CCND1, β-catenin, ADAM17, MMP11, VEGF and ZEB2 mRNA expression and the increasing trend of the WTX, PERK, ATF6, TIMP1 and E-cadherin mRNA expression. Conclusion: Astragaloside can significantly inhibit the growth and invasion of colon cancer cells in vitro. 展开更多

Effect of anxiety after chemotherapy on antitumor immune response and tumor load in patients with advanced gastric cancer

Objective:To study the effect of anxiety after chemotherapy on antitumor immune response and tumor load in patients with advanced gastric cancer.Methods:Patients with gastric cancer who received SOX chemotherapy in the First People's Hospital of Ziyang Sichuan Province between May 2012 and October 2015 were selected and divided into no anxiety group with HAMA score≤7 points, mild anxiety group with HAMA score 7-14 points, moderate anxiety group with HAMA score 14-21 points and severe anxiety group with HAMA score>21 points according to the HAMA score after four courses of chemotherapy. Cytokine and tumor marker levels in serum, immune cell levels in peripheral blood as well as proliferation gene expression in lesions were determined.Results: Peripheral blood Th1 levels as well as serum IL-2 and IFN-γ levels of mild anxiety group, moderate anxiety group and severe anxiety group were significantly lower than those of no anxiety group while peripheral blood Th2, Th17 and Treg levels as well as serum IL-4, IL-5, IL-17, IL-10, CEA, CA19-9, TK-1 and VEGF levels were significantly higher than those of no anxiety group, and EPHA2, c-myc and PCNA mRNA expression in tumor lesions were significantly higher than those of no anxiety group;the severer the anxiety, the lower the peripheral blood Th1 levels as well as serum IL-2 and IFN-γ levels, the higher the peripheral blood Th2, Th17 and Treg levels as well as serum IL-4, IL-5, IL-17, IL-10, CEA, CA19-9, TK-1 and VEGF levels, and the higher the EPHA2, c-myc and PCNA mRNA expression in tumor lesions.Conclusion:The aggravated anxiety in patients with advanced gastric cancer after chemotherapy will inhibit the antitumor immune response and increase the tumor load. 展开更多

Objective:To study the correlation between depression and immune function as well as tumor load in patients with advanced gastric cancer.Methods: 98 patients diagnosed with advanced gastric cancer in our hospital between May 2013 and September 2016 were selected and divided into depression group (n=39) with HAMD scores >50 and non-depression group with HAMD scores≤50 (n=39), serum was collected to detect the levels of cytokines interleukin-2 (IL-2), IL-4, IL-10, IL-17, interferon-γ (IFN-γ) and transforming growth factorβ1 (TGF-β1) as well as tumor markers carcinoembryonic antigen (CEA), carbohydrate antigen 199 (CA199), CA724, TK-1 and sLAG-3, and gastric cancer tissues were collected to determine the expression of cell cycle-related proteins CyclinD1, CDK4 and E2F.Results:Serum IL-2, IFN-γand IL-17 levels of depression group were significantly lower than those of non-depression group (P<0.05) while IL-4, IL-10, TGF-β1, CEA, CA199, CA724, TK-1 and sLAG-3 levels were significantly higher than those of non-depression group (P<0.05);CyclinD1, CDK4 and E2F protein expression in tumor tissues of depression group were significantly higher than those of non-depression group (P<0.05).Conclusions:Depression can inhibit the anti-tumor immune response in patients with advanced gastric cancer, and then promote cancer cell proliferation and increase tumor load. 展开更多

Methods: Patients with advanced bladder cancer who received bladder infusion chemotherapy in the First People's Hospital of Ziyang between May 2014 and December 2016 were selected and divided into the control group without negative emotions, the anxiety group with anxiety, the depression group with depression and the anxiety depression group with both anxiety and depression according to the assessment results of HAMA scale and HAMD scale. The contents of tumor markers in serum, the contents of immune cells in peripheral blood and the expression of apoptosis genes in urine were detected during chemotherapy. Results: DKK-1, DKK-3, OPN and CYFRA21-1 contents in serum as well as PD-1+CD4+T cell, PD-1+CD8+T cell and B7-H1+CD11c+DC cell contents in peripheral blood of anxiety group, depression group and anxiety depression group were significantly higher than those of control group while DAP2IP, Fas, FasL, Caspase-8 and PTEN protein expression in urine were significantly lower than those of control group, and DKK-1, DKK-3, OPN and CYFRA21-1 contents in serum as well as PD-1+CD4+T cell, PD-1+CD8+T cell and B7-H1+CD11c+DC cell contents in peripheral blood of anxiety depression group were significantly higher than those of anxiety group and depression group while DAP2IP, Fas, FasL, Caspase-8 and PTEN protein expression in urine were significantly lower than those of anxiety group and depression group. Conclusion: The anxiety and depression in bladder cancer infusion chemotherapy can suppress the expression of apoptosis genes and the anti-tumor immune response to lead to more vigorous proliferation of cancer cells. 展开更多

Objective:To study the correlation of the anxiety caused by postoperative chemotherapy with endocrine disorder and immune response in patients with colorectal cancer.Methods:A total of 110 patients who received radical operation for colorectal cancer and postoperative chemotherapy in the hospital between August 2014 and December 2016 were divided into the non-anxiety group (SAS<50 points) (n=29), mild-to-moderate anxiety group (50≤SAS≤69) (n=63) and severe anxiety group (SAS 70 points) (n=18) according to self-rating anxiety scale (SAS) score after one course of chemotherapy. The differences in the levels of RAAS indexes, monoamine neurotransmitters, Th1/Th2 cytokines and Th17/Treg cytokines were compared among three groups of patients before and after chemotherapy.Results: Before chemotherapy, the differences in serum levels of RAAS indexes, monoamine neurotransmitters, Th1/Th2 cytokines and Th17/Treg cytokines were not statistically significant among the three groups of patients. After one course of chemotherapy, serum RAAS indexes REN, AngⅡ and ALD levels of mild-to-moderate anxiety group and severe anxiety group were higher than those of non-anxiety group;monoamine neurotransmitters NE and DA levels were higher than those of no anxiety group;serum IL-2 level was higher than that of non-anxiety group while IL-4 level was lower than that of non-anxiety group;serum IL-17 level was higher than of non-anxiety group while IL-10 was lower than that of non-anxiety group;the changes in serum levels of the above indexes of severe anxiety group were greater than those of mild-to-moderate anxiety group.Conclusion: The anxiety caused by postoperative chemotherapy in patients with colorectal cancer can directly lead to the endocrine disorder and the decrease of cellular immune function, and the negative changes increase with the aggravation of anxiety. 展开更多

Objective:To study the correlation of serum Exosome and Dickkopf-1 content with pathological process and gene expression in patients with gastric cancer.Methods:A total of 480 patients diagnosed with gastric cancer in our hospital between May 2015 and July 2016 were selected as the gastric cancer group and 220 healthy volunteers receiving physical examination during the same period were selected as control group. Serum was collected to detect Exosome and Dickkopf-1 content, and gastric cancer tissue and para-carcinoma tissue were collected to detect the expression of pro-proliferation and pro-invasion molecules as well as tumor suppressor genes.Results:Serum Exosome and Dickkopf-1 content of gastric cancer group were significantly higher than those of control group and the deeper the infiltration and the worse the differentiation, the higher the serum Exosome and Dickkopf-1 content, and serum Exosome and Dickkopf-1 content in patients with lymph node metastasis were significantly higher than those in patients without lymph node metastasis;UHRF1, SIRT1 and LOXL2 expression in gastric cancer tissue were significantly higher than those in para-carcinoma tissue and positively correlated with serum Exosome and Dickkopf-1 content while RASAL2, ING5, Period1 and Period2 expression were significantly lower than those in para-carcinoma tissue and negatively correlated with serum Exosome and Dickkopf-1 content. Conclusion:The elevated serum Exosome and Dickkopf-1 in patients with gastric cancer are closely related to the high expression of pro-proliferation and pro-invasion molecules as well as the low expression of proto-oncogene. 展开更多

Relationship between serum Ca2+ level and nerve injury as well as myocardial injury in children with febrile convulsion

Objective:To study the relationship between serum Ca2+ level and nerve injury as well as myocardial injury in children with febrile convulsion.Methods: A total of 32 children with febrile convulsion treated in our hospital between May 2011 and September 2015 were selected as observation group and 30 healthy children receiving vaccination in our hospital during the same period were selected as normal control group. Blood calcium levels of two groups of children were detected, and the median blood calcium level was referred to further divide the observation group into high blood Ca2+ group (n=16) and low blood Ca2+ group (n=16). The differences in nerve injury and myocardial injury were compared between children with blood calcium levels within observation group.Results: Serum Ca2+ level of observation group was lower than that of normal control group;excitatory amino acids Glu, Asp, Lys and Ser contents in peripheral blood of low blood Ca2+ group were higher than those of high blood Ca2+ group while inhibitory amino acids Gly and GABA contents were lower than those of high blood Ca2+ group;cardiac function parameters SV, CI and Vpk levels of low blood Ca2+ group were lower than those of high blood Ca2+ group while SVR level was higher than that of high blood Ca2+ group;myocardial enzyme spectrum indexes CK-MB, LDH andα-HBDH contents in peripheral blood of low blood Ca2+ group were higher than those of high blood Ca2+ group.Conclusion: Serum Ca2+ level is low in children with febrile convulsion and Ca2+ level is negatively correlated with nerve injury and myocardial injury. 展开更多
