Experimental Research of Tankyrase1 Antisense Oligodeoxynucleotides on the Proliferation of Lung Cancer Cell Nodules

OBJECTIVE To observe the effects of sense and antisense oligodeoxynucleotides of tankyrase 1 (TANK1-SODN and TANK1-ASODN) on murine tumor growth following intratumoral injection, investigate the actual suppressing result and mechanism of TANK1-ASODN on cancer cell proliferation, and discuss the possibility of using it in gene therapy on human lung cancer cells. METHODS After BALB/c nude mice had been subcutaneously inoculated with human lung cancer cell line CALU and it had grown into tumor nodules, we distributed these mice randomly into 3 groups: 4 in saline treatment group, and 5 each in TANK1-SODN group, and TANK1-ASODN groups. Then multiple direct intratumoral injections of synthesized TANK1-ASODN given continuously into tumor nodules for 16 days, this was compared with TANK1-SODN and saline control groups. During the experiment we measured the tumor volume every 5 days with vernier calipers; observed the histopathological characteristics of tumor tissues under microscope; went further to detect the minute changing of ultrastructure of cancer cells by electron microscope; tested the expression levels of ki67 and hTERT protein by means of SABC immunohistochemical method; and detected the lung cancer cells' hTERT mRNA expression level by hybridization in situ (ISH) in each group. RESULTS After 16 days of continuous injection, the tumor volume in TANK1-ASODN group was significantly smaller than the other 2 groups (both P 〈 0.01); quite a lot of tumor cell degeneration and necrosis were observed in mice given TANK1-ASODN. The results of electron microscope also showed that TANK1-ASODN has the power to kill cancer cells in various ways. Moreover, statistically signi.cant decreases in the positive expression ratio of Ki67 Labeling Index (P 〈 0.01), hTERT protein (P 〈 0.01), and hTERT mRNA (P 〈 0.01) were consistently observed in the TANK1-ASODN group. CONCLUSION Human lung cancer cell line CALU expressed high telomerase activity. TANK1-ASODN had the ability to decline the high expression level of hTERT; inhibit the activity of telomerase, accelerate tumor cell degeneration and necrosis; and then suppress the proliferation of cancer cells. 展开更多

Lipoxidation is formed during the oxidation of lipids and specific proteins,causing an alteration in proteins function.Although this occurs under physiological conditions,in many cases,it has been associated with pathological processes,including cancer.Oxidative DNA damage is a significant contributor to cancer development,and lipoxidation products such as malondialdehyde(MDA)and 4-hydroxy-2-nonenal(4-HNE)play a role in the induction of mutations responsible for DNA modifications.Elevations of reactive aldehydes can be seen in patients with metastatic disease in comparison to those without metastatic disease.Lipoxidation adducts can modify the immune response,consequently causing either positive or negative alterations in cancer progression.When reactive aldehydes are added to tumor cells,they exert similar effects to chemotherapeutic drugs by forming DNA adducts and consequently steer the tumor cells toward apoptosis.This article highlights the role of botanical compounds in lipid metabolism,lipid peroxidation,and cell cycle arrest and discusses the potential role they may have in oncology treatments. 展开更多

Methods: The 51 outpatients were treated by basic recipe for massage treatment and modified recipe according to syndromes. Results: The total Athens Insomnia Scale (AIS) score 3.2800±1.5784 after treatment is lower than 9.2400±1.4286 before treatment (P<0.01), there is statistical difference in AIS scores before and after treatment. Conclusion: TCM massage can remarkably improve sleep condition of infants with dyssomnia. 展开更多

Methods: Sixty patients with apoplectic hemiplegia were randomly divided into a treatment group of 30 cases and a control group of 30 cases. The treatment group was treated by "remissive stage-reinforcing and acute stage- reducing" acupuncture and the control group by modem rehabilitation training. Before and after treatment, the degree of spasm, the level of motor function and ability for daily life were evaluated by modified Ashwordth spasm grading, Brunnstrom grading and Barthel index now often used in the rehabilitation field. Results: "Remissive stage-reinforcing and acute stage reducing"acupuncture was superior to modem rehabilitation training in relieving apoplectic myospasm.Conchtsion: "Remissive stage-reinforcing and acute stage-reducing"acupuncture has a unique advantage in relieving spasm. 展开更多

Methods: altogether 54 cases were randomly allocated into acupuncture group (30 cases) and control group of western medications (24 cases), the latter was treated with routine western medication, while the former was combined acupuncture based on routine western medication, and the treatment course of the two groups were both 30 days. Results: the effect of treatment group was superior to the control group (P〈0.05). Conclusion: acupuncture can improve symptoms of diabetic nephropathy, lower blood glucose, urine albumin, blood urine nitrogen, and creatinine and improve the function of kidney. 展开更多
